1.The effect of vascular pediceled peripheral nerve tissue grafting in repair spinal cord injury

Objective:To observe the repairment effects of vascular pediceled nervous tissue on adult rat's spinal cord injury. Method:A 5mm cord defect of the left lateral column was made at Ti-3 vertebral level,and then the defect was grafted randomly using vascdarized peripheral nerve (Group VPN),embryonic day 14 fetal spinal cord(Group FSC) or vascular pediceled peripheral nerve and fetal spinal cord(Group V + F).8 weeks after surgery, the survival,differentiation of the grafts, and the ability of repaired host spinal cord were assessed. Result: In VPN-FSC co-graft group, peripheral nerve segment was extensively vascularized and well innervated,plenty of Schwann cells survived and proliferated. In this group, anti-NF, S-100 positives fibers were significantly greater than group VPN (P<0.01). The fetal implant was intimately fused with the host CNS parenchyma, the volume, nerve fibers and neurons density of the fetal implant were significantly higher than Group FSC(P<0.01). The majority of neurons in this group were well differentiated, well-developed and synapses were scattered throughout the neuropil. SEP assessments showed that the latgncy of P1 ,N1 wave was significantly decreased in this group (P<0.01).Conclusion:The vascular pediceled peripheral nerve could prompt the development of fetal implant and support the regeneration of central injured axons to certain degrees.
2.The survival and migration of Schwann cells after peripheral nerve grafted into spinal cord

3.Preliminary application polyaxial locking plate in the treatment of tibial plateau fractures

[Objective]To report the polyaxial locking plate and its primary clinical results in the treatment of the tibial plateau fractures.[Method]From July 2006 to August 2007,38 patients with tibial plateau fractures,including 26 males and 12 females,were treated with surgical reduction and internal fixation with polyaxial locking plate.The mean age of the patients was 30.6 years(18-65 years).According to Schatzker classification,there were 3 of type Ⅰ,5 of type Ⅱ,5 of type Ⅲ,6 of type Ⅳ,16 of type Ⅴ,3 of type Ⅵ,of which 34 were closed fractures and 4 open fractures.The follow-up period ranged from 7 to 18 months with a mean of 13.7 months.Function of the knees were assessed according to HSS system and radiological examination.[Result]Solid arthrodesis were obtained in all patients at 3-6 months after operation.Skin flap necrosis and incision infection happened respectively in 2 cases of type Ⅴ and type Ⅵ fractures.The score of HSS system was 92.8(range 78-98) averagely half year after operation and no reduction loss was observed.[Conclusion]The polyaxial locking plate is an effective fixation technique for the surgical treatment of the tibial plateau fractures because of its convenience and stability.The locking screws can be implanted in various directions and reduction loss is avoided post-operation.
4.Regulation of ? opioid receptor activities

As for the ? opioid receptor, there are some disputes about the domains involved in the selective recognition of ligands, the relative spatial orientation of the amino acids within the TM affect the affinities of selective ligands. Regulation of opioid receptor activities does not appear to involve in their ability to promote the association of GTP onto the G proteins and the subsequent dissociation of heterotrimers. It is not very clear if phosphorylation correlates with agonist induced receptor desensitization. The celluar processing of ? opioid receptors requires the formation of multiple protein complexes, it is clear that interactions of ubiquitous transcriptional factors determine gene transcription.

6.Influencing factors for nicotine dependence among smoking patients with chronic obstructive pulmonary disease

Objective:
To investigate the nicotine dependence of patients with chronic obstructive pulmonary disease (COPD) who smoke and analyze its influencing factors, so as to provide insights into targeted smoking cessation interventions for smoking COPD patients.
Methods:
The smoking COPD patients were selected from three hospitals in Lanzhou City using the convenience sampling method from October 2022 to April 2023. Demographic data, smoking status, and willingness to quit smoking were collected using questionnaire surveys. Nicotine dependence was assessed according to Fagerstrom Test for Nicotine Dependence. The influencing factors of nicotine dependence in smoking COPD patients were analyzed using a multiple linear regression model.
Results:
A total of 200 questionnaires were distributed, and 171 valid questionnaires were collected, with a response rate of 85.50%. The median age was 48.00 (interquartile range, 10.00) years. There were 158 males, accounting for 92.40%; 53 cases with a college degree or above, accounting for 30.99%. The median smoking duration was 15.00 (interquartile range, 11.00) years. There were 73 patients (42.69%) smoking 21 to 30 cigarettes per day, and 159 patients (92.98%) with the willingness to quit smoking. The median nicotine dependence score was 3.00 (interquartile range, 1.00) points. There were statistically significant differences in nicotine dependence scores among smoking COPD patients with different ages, educational levels, occupations, interpersonal relationships, smoking duration, daily cigarette consumption, awareness of smoking hazards and willingness to quit smoking (all P<0.05). Multiple linear regression analysis revealed that nicotine dependence was related with educational level (β=-0.251), interpersonal relationship (β=-0.246), smoking duration (β=0.038), daily cigarette consumption (β=0.518) and willingness to quit smoking (β=-0.173) among smoking COPD patients (all P<0.05).
Conclusion
The nicotine dependence among smoking COPD patients was influenced by educational level, interpersonal relationships, smoking duration, daily cigarette consumption and willingness to quit smoking.
7.Clinical study on diagnosis of laryngeal carcinoma by high frequency ultrasound and CT

Objective To investigate the value of high frequency ultrasound and CT in the diagnosis of laryngeal carcinoma. Methods Sixty-nine cases of laryngeal carcinoma were typed and staged by high frequency ultrasound and CT, and compared with the results of laryngoscopy and postsurgical pathology. Results The typing accuracy was ~94.2 % for ultrasonography and ~91.3 % for CT, higher than that of laryngoscopy(~73.9 %).T staging accuracy of ultrasonography was T_1 ~85.9 %,T_2 ~88.5 %,T_3 ~94.1 % and T_4 ~88.9 %, higher than that of CT (~68.8 %) and laryngoscopy (~75.0 %) in T_3 stage and superior to laryngoscopy (~66.7 %) in T_4 stage. Lymph node metastases were found in 37 cases (~53.6 %) by ultrasonography, higher than those by CT scanning (~27.5 %). Color Doppler blood flow laryngeal carcinoma was shown in 65 cases(~94.2 %),most were low resistance spectrum pattern. Conclusions High frequency ultrasonography is superior to CT and laryngoscopy in the staging of laryngeal carcinoma and is of great value in choosing the therapeutical protocol.

Aim To generate m3AChR-G11 fusion protein in baculovirus-Sf9 cells and test the couping function,the interation and the influence factors be-tween m3AChR and G11 protein,as well as screen the specific ligands for m3AChR. Methods m3AChR-G11 fused DNA was generated through a two-step PCR and then expressed in Sf9 cells to produce fusion protein. The total concentration for membrane protein was de-tected by BCA method,[3H]QNB and [35 S]GTP?S binding experiment as perfomed to study the function of m3AChR-G11 fusion protein. Results The expression level of m3AChR-G11 was 7. 76 ? 10 -9 mol?g -1. The affinity of GDP to G11 partner changed in the presence of different muscarinic ligands. IC50 values of GDP in the presence of ACh,Pilo,CCh,MCN-A-343,Atro,4-Damp and Dafi were 82. 2,93. 70,12. 10,14. 30, 1. 93,1. 37,0. 72 ? 10 -6 mol ? L -1 respectively,and that in the absence of muscarinic was 1. 99 ? 10 -6 mol ?L-1.Concluslons The m3AChR-G11 fusion protein has the pharmacological specificity of m3 receptor and the efficient coupling interaction of the two partners. Affinity of GDP to ligand-bound fusion protein represents the species of muscarinic ligands. This is helpful in screening and detecting the new specific ligands to muscarinic receptors.
10.The status and related factors of daily activities in patients with chronic obstructive pulmonary disease

Objective To determine relationships between daily activities measured as modified version of Pulmonary Functional status and Dyspnea Questionaire (PFSDQ-M) and functional capacity and symptoms experienced in patients with chronic obstructive pulmonary disease (COPD). Methods Convenience sample of 94 COPD patients with stable condition were assessed by interview with PFSDQ-M Chinese version and modified Medical Research council dyspnea scale (MMRC) respectively. Pulmonary function test (PFT) and 6-minute walking test (6MWT) were undergone on the same day or within one week as interview. PFSDQ-M has three subscales,i.e.,change experienced by patient with activities (CA),dyspnea with activities (DA) and fatigue with activities (FA). Results Scores of CA,DA and FA correlated to 6-minute walking distancer= (-0.37)- (-0.42),FEV_1 r=(-0.27)-(-0.32),FEV_1/FVC r= (-0.27)- (-0.32),dyspnea rated by MMRC (r=0.55-0.60) and BODE index (r=0.35-0.40),respectively (all P<0.01). dyspnea explained 26% of the variance in changes of activities. Conclusions Physical activities moderately changed in patients with stable COPD;Dyspnea is the best predictor of limitation of daily activities.
