1.Study on expression of P16, CD44v6 and TGF-β1 in gastric carcinoma and their relationship

Objective To explore the expression of P16, TGF-β1 and CD44v6 in gastric carcinoma and the relationship between their expression and clinicopathological features. Methods The expressions of P16, TGF-β1 and CD44v6 in 78 cases of gastric carcinoma, 20 cases gastric epithelial dysplasia and 25 cases normal gastric tissues were detected by immunohistochemistiy (SP method). Results The expressions of P16 in gastric carcinoma (46. 2% ) and gastric epithelial dysplasia (60. 0% ) were markedly lower than that in normal gastric tissues (92. 0% ). The expressions of CD44v6 in gastric carcinoma (51.3%) and gastric epithelial dysplasia (45.0% ) were markedly higher than that in normal gastric tissues (4.0% ). The expression of TGF-β1 in gastric carcinoma (65.4% ) was much higher than the counterparts in normal gastric group (12. 0% ) and gastric epithelial dysplasia group (35.0% ). P16 was positively correlated with the differentiation degree of gastric carcinoma, the depth of invasion, lymphatic metastasis and TNM stage ( P < 0. 05 ), and it had no significant correlation with tumor size,vessel tumor thrombus and the progression of the disease. CD44v6 was positively related with the invasive depth of gastric carcinoma, lymphatic metastasis, progression and TNM stage ( P <0.05) and had no significant correlation with differentiation degree of tissue, tumor size and vessel tumor thrombus ( P >0.05). TGF-β1l was positively correlated with the differentiation degree of gastric carcinoma, the depth of invasion, lymphatic metastasis and TNM stage ( P < 0.05 ) and had no significant correlation with tumor size, vessel tumor thrombusand the progression of the disease. There was a significant positive correlation between the expressions of CD44v6 and TGF-β1 of on gastric carcinoma ( r = 0. 532, P < 0.05 ). And a negative correlation was found between CD44v6 and P16 ( r = - 0. 615, P < 0. 05 ). Conclusions P16 ,TGF-β1 and CD44v6 may play a role in the tumorigenesis and progression of gastric carcinoma in some degree. Combined detection of P16, TGF-β1 and CD44v6 may be helpful to judge the degree of malignancy and potential lymph node metastasis and evaluate the prognosis.
4.AN EXPERIMENTAL STUDY OF TIMING OF WOUND

Experimental incised wounds 8 mm in diameter were made on the right side of the back of 48 rats under anesthesia using aseptic technique. All animals were randomly divided into 12 groups according to the time of wound biopsies which were taken immediately after skin cutting, lh,12h,24h,2,3,4,5,6,7,8,9 and 10 days after wounding.One rat of each group was given colchicine before animal sacrifice in order to observe cell mitosis. Skin specimens were fixed in 10% formalin,embeded in paraffin and sectioned for HE stain,Gorden and Sweet stain for reticular fiber,Weigert stain for elastic fiber,PTAH stain for fibrin and Van Gieson stain.Macroscopically, wounds were getting smaller daily,and reduced in size markedly on the 6~7th day. The histologic changes were as follows:Acute inflammation on the wall of wounds occurred 1h after skin cutting;profuse fibrin exudation at the top of the base and on the wall of wounds at 24h;new capillary formation and regeneration of the epidermis on the 2nd day;granulation tissue formation and collagen fiber deposition on the 3rd day;small artery formation in the granulation tissue on the 4~5th day;granulation tissue maturation and the regenerating tissue covering the whole wound area on thd 6~7th day;the scar formation and contraction on the 8~9th day;further sear contration on the 10th day.The epidermis regenerated from the proliferation of the epidermis at the edge of the wound with the speed of 1.23mm/day. The regeneration of the epithelium proceeded under the necrotic tissue on both the wall and at the base of wounds. The mechanism of the wound contraction was discussed.
5.Influence of head irradiation on hypothalamo-pituitarygonad axis in adrenalectomized male rats

Head irradiation with 10 Gy X-rays was given to adrenalectomized male rats in the present study to observe the changes of serum LH, FSH and testosterone (TS) levels and testcular cAMP content. 7.Implantation of intrathecal infusion system for intractable pain therapy

Objective To discuss the mechanism,procedures and effect of implantable intrathecal infusion system for intractable pain therapy. Methods Among the 4 patients with intractable pain,2 had cancer pain,2 had failed back syndrome.Before operation,Visual Analogy Scale(VAS) were 8~10.Three had bolus intrathecal injection screening trial,1 had bolus epidural injection screening trial.In the clinical trail,VAS all decreased to 75%~90%.(Results) They all had intrathecal infusion system implantation.After operation,Morphine was prescribed(intrathecally.) The dose was 1~(8 mg).They all had satisfied pain relief without serious drug adverse effects.Conclusion The effect of implantable intrathecal system for intractable pain was satisfied.The drug adverse effects were obviously alleviated.
8.Quality evaluation on different specifications of cervi cornu pantotrichum with its effect on ovariectomized osteoporosis model rats.

OBJECTIVETo study the effect of eight specifications of Cervi Cornu Pantotrichum on the osteoporosis of ovariectomized rats and grade the eight different specifications of Cervi Cornu Pantotrichum.
METHODTotally 100 SD female rats were divided randomly into 10 groups, namely the normal group, the model group and eight Cervi Cornu Pantotrichum groups of different specifications. Their bilateral ovaries were excised to reproduce the osteoporosis model. Meanwhile, the rats were given the eight different specifications of Cervi Cornu Pantotrichum for consecutively 12 weeks. Subsequently, the effects of the different specifications of Cervi Cornu Pantotrichum on bone mineral density and serum biochemical indicators of rats were observed. A clustering analysis was made for the eight specifications of Cervi Cornu Pantotrichum, with the serum content of ALP, BMP-2 and BGP as influencing factors.
RESULTAfter 12 weeks, the eight different specifications of Cervi Cornu Pantotrichum could significantly improve ALP, BMP-2, BGP in serum and bone mineral density of ovariectomized rats. And the cluster analysis showed similar results to the quality classification of traditional commercial herbs Cervi Cornu Pantotrichum.
CONCLUSIONDifferent specifications of Cervi Cornu Pantotrichum can antagonize the osteoporosis of ovariectomized rats, and their effects are related to the quality of commercial herbs.

10.The comparison of feasibility of different plasma substitutes during preoperative acute hypervolemic hemodilution.

Objective To compare the effects of three different plasma substitutes, 6% hydroxyethyl starch (HES 200/0.5), 4% Gelofusine and 5% Polygeline, on blood coagulation and arterial blood gases and electrolytes during acute hypervolemic hemodilution (AHHD) . Methods Seventy ASA Ⅰ - Ⅱ adult patients undergoing elective orthopedic surgery with the intraoperative blood loss predicted to exceed 500 ml were entered in this study. Radial artery was cannulated for BP monitoring and blood sampling before induction of anesthesia. Anesthesia was induced with fentanyl 3 ?g?kg-1, propofol 2 mg?kg-1 and atracurium 0.5 mg? kg-1 and maintained with enflurane (1-2 MAC) . The patients were mechanically ventilated ( VT= 10 ml? kg-1 , RR = 12 bpm) after tracheal intubation. Internal jugular vein was cannulated after induction of anesthesia for CVP monitoring. After induction of anesthesia the patients were randomized to receive 20 ml?kg-1 of either 6% HES (group H, n = 20), 4% Gelofusine (group G, n = 20), 5% Polygeline (group P, n = 20) or lactated Ringer's solution (group R, n = 10) within 20-40 min. Arterial blood samples were taken before AHHD, at the end of and 30 min after the infusion of substitute for determination of activated coagulation time (ACT), thromboelastography (TEG) , blood gas analysis and plasma electrolytes. Blood volume expansion rate was calculated [ blood volume expansion rate = (Hct before AHHD - Hct after AHHD) / Hct before AHHD] .Results The four groups were comparable with respect to sex, age and body weight. Lactated Ringer' s solution was significantly less efficient in expanding intravascular blood volume than the 3 plasma substitutes, but there was no significant difference in blood volume expansion rate among group H, G and P.
