1.Research on wavelength matching of therapeutic laser fiber.

Medical laser adopts optical fibers as spread media, using laser energy for the purpose of diagnostics and therapeutics. The corresponding industry standard is YY/T 0758-2009 General requirements for therapeutic laser fiber, which requires manufacturers to offer applicable wave length (or spectrum range) of the fiber, and the corresponding minimum transmission efficiency of the wave length. This research focuses on the matching of therapeutic laser fiber and laser source, to support and emphasize the importance and reasonability of relative requirements in YY/T 0758-2009.

2.Dynamic expression of intercellular adhesion molecule-1 in laser-induced choroidal neovascularization in Brown Norway rats

Background Extracellular matrix (ECM) has various kinds of types and important biological function.The important role of ECM during choroidal neovascularization (CNV) arouse attention.Intercellular adhesion molecule-1 (ICAM-1), which is one of ECMs, involves in the formation of blood vessels, but the relationship between CNV and ICAM-1 is still unknown.Objective This study was to observe the dynamic expression of ICAM-1 in krypton laser-induced CNV and explore the effect of ICAM-1 on CNV.Methods Forty-eight healthy male clean BN rats were randomly divided into post-photocoagulation 1-week group,2-week group,3-week group,4-week group, 5-week group, 6-week group, 7-week group and 8-week group.Laser-induced CNV models were monocularly established and the fellow eyes served as the normal controls.Fundus fluorescein angiography (FFA) was carried out to quantitate the leakage degree (absorbency).The eyeballs were enucleated on various time points,and CNV areas were assessed by hematoxylin and eosin staining;immunohistochemistry and in situ hybridization assay were employed, respectively, for the detection of relative expression levels of ICAM-1 protein and mRNA (absorbency).The use and care of the animals complied with the Regulations for the Administration of Affair Concerning Experimental Animals by State Science and Technology Committee.Results After laser photocoagulation,the retinal inner and outer nuclear layers were ruptured and invaginated.Disruption of Bruch membrane and macrophages migrating to the photocoagulation zone were seen under the optical microscope.The proliferation of fibroblasts and vascular endothelial cells was exhibited.ICAM-1 protein was mainly expressed in vascular endothelial cells,retinal pigment epithelium (RPE) cells and macrophages,and ICAM-1 mRNA was mainly expressed in the outer nuclear layer.Statistically significant differences were found in the fluorescence leakage degree, the relative expression levels of ICAM-1 protein and mRNA as well as CNV areas among the groups (F=178.839, 739.077,2 463.508,194.434, all at P<0.05).The gradually enlarged CNV area, increased fluorescence leakage, up-regulation of ICAM-1 protein and mRNA expressions were matched with the extension of time after photocoagulation and peaked in the post-photocoagulation 8-week group.Conclusions The expressions of ICAM-1 protein and mRNA up-regulate upon the increase of CNV area and fluorescein leakage, suggesting that ICAM-1 might play an important role in CNV development.
3.Effects of Triamcinolone Acetonide on Choroidal Neovascularization in Brown Norway Rats

Objective To observe the effect of intravitreal triamcinolone acetonide injection on choroidal neovascularization in brown Norway rats. Methods Thirty-six healthy brown Norway rats were divided randomly into control group (n=18) and experimental group (n=18), while one eye was chosen randomly as experimental eye. The choroidal neovascularization model was establishied, while 8μl triamcin-olone acetonide was injected in vitreous body immediately after photocoagulation in the experimental group, and the same volume isotonic balanced salt solution was injected in the control group. The eyes of six rats were enucleated for histological slices in the second, forth, and sixth week, respectively. The expression of nuclear factor-kappa B was detected with immunohistochemical method, and the max area of choroidal neovascularization was also measured. Results The area of choroidal neovascularization was smaller, and the expression of nucle-ar factor-kappa B was lower in the experimental group than in the control group (t>7.450, P<0.001). Conclusion Triamcinolone acetonide could effectively inhibit the development of choroidal neovascularization, and decrease the expression of nuclear factor-kappa B in brown Norway rats.
4.Establishment and preliminary application of a real-time fluorescent quantitative PCR assay for detection of murine norovirus

Objective To establish a real-time fluorescent quantitative PCR ( FQ-PCR ) method for detection of murine norovirus ( MNV) in laboratory mouse and provide the basis for establishment of a standard detection method for MNV.Methods Specific primers were designed and MNV DNA standards were prepared according to the MNV genome sequences published on NCBI .The specificity, sensitivity, repeatability and stability of the established Q-PCR method were tested.The established Q-PCR method was applied to detect 766 mouse caecum content samples to explore preliminarily the infection status of laboratory mice in Beijing .Results No cross reaction showed in human norovirus and feline calicivirus with the established Q-PCR method.The sensitivity was up to 10 copies/μL.The coefficient of variation ( CV) of intra-assay and inter-assay was less than 2%.There were 301 positive cases detected in the 766 samples of laboratory mice.Conclusions The established FQ-PCR method is accurate and effective with high specificity , sensitivity and repeatabiliy in the quantitative detetion of nucleic acid , and can be applied to rapidly and quantitatively screen MNV in laboratory mice .
6.Effect of air pressure wave on the prevention of lower limb deep venous thrombosis in patients with cerebral stroke and the application value of color Doppler ultrasound

Objective To explore the effect of air pressure wave on the prevention of lower limb deep venous thrombosis in patients with cerebral stroke and the application value of color Doppler ultrasound (CDUS).Methods 72 stroke patients were randomly divided into experimental group (n =37) and control group (n =35) by random digits table.The control group received routine nursing,conventional and rehabilitation therapy.The experimental group was treated by the air pressure wave for 15 days on the basis of the control group.DVT formation of lower extremities and hemodynamic changes before and after treatment was observed by CDUS,and the results were compared with the X-ray phlebography examination results.Results CDUS displayed the incidence rate of DVT in the experimental group was significantly lower than that in the control group (2.7% vs 20.0%,x2 =5.449,P <0.05).The changes of blood flow velocity and the average reflux time of femoral,popliteal vein in the experimental group had significant differences between before and after treatment (t =8.503,4.254,8.587,2.062,all P < 0.05) ;and in the control group,only popliteal vein blood flow velocity had statistically significant difference (t =6.895,2.116,4.169,all P < 0.05).Compared with the X-ray angiography,8 cases of DVT diagnosed by CDUS were consistent,but 1 case of DVT in the control group was missed diagnosis.Conclusion Air pressure wave can effectively reduce the occurrence of DVT in patients with cerebral stroke.CDUS can accurately diagnose the DVT formation,and can help to understand the hemodynamic change,it has important clinical value for monitoring the preventive effect of DVT.

Doctor He combined traditional Chinese and western medicine, considering the comprehensive, combined with the individual constitution, emphasized the selection of herbal medicine with multiple functions. The clinical effect in the proven cases was amazing. [Conclusion] Professor He had rich experience treating abdominal mass with infertility, which might be used as reference data in clinical medicine.

9.Application of HPV genotype detection in prevention and treatment of cervical cancer and cervical precancerous lesions

Epidemiological studies have shown that human papillomavirus (HPV) infection is the main cause of cervical cancers and cervical intraepithelial neoplasia(CIN).And different types of HPV have different carcinogenicity,different cervical cancer pathological types and prognoses of cervical cancer.The effective way to decrease the cervical cancer morbidity is to detect and classify the high-risk HPV infection in early time,and carry out intervention therapy in early time.Therefore,detecting HPV subtypes in clinic will have great significance in prevention and treatment of cervical cancer and cervical precancerous.
10.Genetic susceptibility to coronary artery disease

OBJECTIVE: To summarize the advances in the genetic susceptibility to coronary artery disease (CAD) and explore the heritable basis of the disease.DATA SOURCES:Related articles in English from January 1990 to June 2006 were searched in Pubmed and EMCC databases with the terms "gene, coronary artery disease and myocardial infarction". Meanwhile, the CMCC database was searched for the relevant articles published between January 2000 and June 2006 in Chinese.STUDY SELECTrON: After the preliminary selection,relevant literatures on genetics of CAD and myocardial infarction(MI)were selected, and those with obvious indifferent contents or with less correlation were excluded.DATA EXTRACTION: Data of 49 articles were mainly extracted from the selected literatures providing solid evidence to elucidate the genetic susceptibility to CAD or MI.DATA SYNTHESIS: The involved 49 articles showed that CAD is a complex multifactorial disorder which is believed to result from the interplay between a person's genetic makeup and various environmental factors. The heritable basis is increasingly recognized as a crucial component in the development of CAD. Recent work in the field of genetics has lead to determining key genes associated with the susceptibility for CAD and MI through genome-wide linkage scans and large-scale gene-association studies as core human genetics approaches These susceptibility genes involve diverse functions, including dyslipidemia, vascular homeostasis, endothelial dysfunction, inflammation and immunity.CONCLUSION: The identification of genes that predispose to or directly cause CAD provides new insights into the pathogenesis of this disorder. However, the precise role of genetic factor in the CAD or MI events and the personalized gene-specific therapy await further investigation.
