1.Effects of L-carnitine on Inflammatory Factor,Oxidative Stress Index and Cardiac Function of Mainte-nance Hemodialysis Patients

OBJECTIVE:To investigate the effects of L-carnitine on inflammatory factor,oxidant stress index and cardiac func-tion in patients with maintenance hemodialysis. METHODS:One hundred and twenty maintenance hemodialysis patients selected from our hospital during Dec. 2014-Feb. 2016 were randomly divided into observation group and control group,with 60 cases in each group. Control group received maintenance hemodialysis for 1 month,and then was given rhEPO injection 3000 IU subcuta-neously,3 times a week. Observation group was additionally given Levocarnitine for injection 2 g,iv,qod,1 day after hemodialy-sis,on the basis of control group. Two groups were treated for consecutive 3 months. The levels of inflammatory factors (IL-6, TNF-α,CRP),oxidant stress indexes(MDA,GSH-Px,SOD)and cardiac function indexes(CO,LVEF,LVST)were observed in 2 groups before and after treatment;the occurrence of ADR was recorded. RESULTS:There was no statistical significance in in-flammatory factor,oxidant stress index and cardiac fuaction before treatment between 2 groups(P>0.05). After treatment,the lev-els of IL-6,TNF-α,CRP and MDA in observation group were decreased significantly,while the levels of GSH-Px,SOD,CO and LVEF were increased significantly,compared to before treatment;the improvement of above indexes in observation group were sig-nificantly better than control group,with statistical significance (P<0.05). No obvious ADR was found in 2 groups during treat-ment. CONCLUSIONS:L-carnitine can effectively alleviate the micro-inflammatory state in maintenance hemodialysis patients and protect myocardial function with good safety.
2.Quantitative analysis of circulating tumor DNA as a diagnostic marker for breast cancer and its role

Objective To establish the methods for quantitating the circulating tumor DNA with PicoGreen fluorescent nucleic acid stain and investigate the role of the quantitative analysis in diagnosis of breast cancer. Methods Circulating tumor DNA was isolated from serum with QIAmp blood kit and was quantitated by spectrofluorometry with PicoGreen fluorescent stain. Receiver operator characteristic ( ROC) curve and area under the curve were used to estimate the role of DNA quantification in diagnosis of breast cancer. Results The efficiency of QIAamp blood kit isolating DNA from serum was 37. 8%-46. 2%, average 43. 4%. Circulating tumor DNA concentration as low as 1 ng could be detected by PicoGreen spectrofluorometry. and the detected range was 1-500 ng/0. 2 ml. The median concentration of serum DNA in breast cancer group was (169. 70+ 124. 10) μg/L, and that of healthy control and breast benign group were (54. 30±36. 84) μg/L and (51. 70±29. 04) μg/L, respectively (P<0. 01). The area under the ROC curve was 0. 899 (95% CI: 0.848-0.951), and the sensitivity was 78. 2%, the specificity was 90% by using the cutoff value of 96. 0 ng/ml. Conclusions The concentration of circulating tumor DNA can be efficiently quantitated by PicoGreen spectrofluorometry, which indicates the potential of clinical applicability in breast cancer diagnosis.
3.Effects of Acupuncture on Blood Lipids in Hyperlipidemia Mice

Objective To observe the effects of acupuncture on blood lipids of hyperlipidemia mice, and explore the mechanism of acupuncture against endothelial dysfunction. Methods 40 ApoE (-/- ) mice were randomly divided into control group (C), acupuncture at non-acupoint group (D), acupuncture at acupoint group (E), and simvastatin group (F) with 10 cases in each group. After 8 weeks, total cholesterol (TC), the plasma angiotensin II (Ang II), endothelin-1 (ET-1) and nitric oxide (NO) were measured with enzyme-linked immuno sorbent assay (ELISA). The mice myocardial angiotensin II type 1 receptor (AT1R), and endothelin-1 type A receptor (ETAR) protein were detected with Western blotting. Results The blood lipid, the content of plasma Ang II and ET-1, and the level of AT1R and ETAR in the heart tissue were significantly lower, and the content of NO was significantly higher in groups E and F than in group C (P<0.05). Conclusion Acupuncture can inhibit the level of blood lipids in ApoE(-/-) mice, reduce the levels of Ang II and ET-1, increase the level of NO in peripheral blood, and inhibit the expression of AT1R, ETAR in heart tissue.
4.One-stage posterior approach surgical treatment for congenital scoliosis caused by lumbosacral spine deformity in children

Objective To explore the treatment method and clinical effect of congenital scoliosis caused by lumbosacral spine deformity in children.Methods From October 2000 to October 2015,a consecutive series of 21 congenital scoliosis due to lumbosacral spine deformity were treated in Department of Pediatric Orthopedics,the Third Affiliated Hospital of Zhengzhou University,including 12 male and 9 female,and the age was (63.1±47.3) months(3-144 months).The hemivertebrae,bone bridge,and the mixed type were found in 18 cases,2 cases,and 1 case,respectively.Hemivertebraes were on the left in 10 cases,on the right in 9 cases;involved L5 in 8 cases,S1 in 6 cases,multiple vertebrae malformation in 4 cases,wedged-shaped vertebrae involved L5 associated with contralateral bar in 1 case,unilateral unsegmented bar from L4 to S1 in 1 case,from L2 to L5 in 1 case.There were 4 cases combining with thoracic hemivertebrae,3 cases with ribs fusion and contralateral bar,16 cases(76%) with spinal cord malformations,3 cases(17%) with urinary system malformations,no congenital heart malformation.All the 21 patients were operated with one-stage posterior approach.Compared the scoliosis Cobb angle,trunk shift,operative time,blood loss volume,and complications among preoperation,post-operation and final follow-up.Results In this study,operative time was (278.9±83.1) min,blood loss volume was (355.3±249.0) mL,follow-up was (5.1±2.7) years.There were 18 cases operated with pedicle screw fixation,aged (72.9±44.1) months(16-144 months),operative time was (296.2±74.2) min,blood loss volume was(422.1±238.2) mL;the mean coronal Cobb angle of malformation area at preoperation,post operation and the final follow-up were(27.3±10.2)°,(11.0±5.9)°,(9.8±4.2)°,while the correction rate of post-operation and the final follow-up were 59.7%,64.1%.There were 3 cases operated by hemivenebra resection without pedicle screw fixation,aged 7.7 months(3-15 months),operative time was 168.3 min,blood loss volume was 103.3 mL.The mean coronal Cobb angle of malformation area at preoperation,post-operation and the final follow-up were 26.0°,13.6°,12.5°,while the correction rate of post-operation and the final follow-up were 47.7%,51.9%.The follow-up period was (5.1±2.7) years.The coronal Cobb angle of lumbosacral curve at preo-peration,post-operation and the final follow-up were (27.7±10.0)°,(11.3±5.4)°,(10.0±5.0)°,compared with preoperation,the differences of post-operation and the final follow-up were statistically significant (t=6.600,7.230,all P<0.05),with the correction rate of 59.2%,63.9%;the compensatory head side Cobb angle were(25.0±12.8)°,(11.9±8.2)°,(10.3±6.9)°,compared with preoperation,the differences of post-operation and the final follow-up were statistically significant (t=3.934,4.626,all P<0.05),with the correction rate of 52.4%,58.8%.Trunk shift was significantly improved.One case appeared fracture fixation rods breakage after 9 years,1 case appeared iliac screw breakage after 2 years but no complication such as infection,nerve injury.Conclusions The formation of obstacles and vertebral segmentation defects will lead to lumbosacral spine deformity,and scoliosis.Early one-stage posterior hemivertebra resection,breakage the bone bridge combined with pedicle screw fixation can obtain satisfactory clinical outcome.
5.Effects of Electroacupuncture on Morphology Repairment and Basic Fibroblast Growth Factor/Extracellular Signal-regulated Kinase Signal Pathway of Rabbits with Acute Lumbar Muscle Contusion

Objective To observe the effect of electroacupuncture on basic fibroblast growth factor (bFGF), Desmin and extracellular signal-regulated kinase (ERK1/2) expression of rabbits with acute lumbar muscle contusion, and discuss the possible mechanism of muscle tissue regeneration and repair. Methods 40 male New Zealand rabbits were randomly divided into blank group (BG, n=10), model group (MG, n=10), Weizhong (BL40) electroacupuncture group (WG, n=10) and local electroacupuncture group (LG, n=10). Lumbar muscle injury was established with blunt trauma, and the rabbits were assessed with Appearance Score. The WG and LG accepted electroacupuncture for 2 weeks after modeling. The lumbar muscle of each animal was stained with HE to observe pathological changes, and immunohistochemical staining was used to observe bFGF expression, and Western blotting was used to detect the expression of Desmin and ERK1/2 protein.Results Appearance Score was more in the MG, WG and LG than in the BG (P<0.01). Histological score ranked from more to less as MG,LG, WG and BG (P<0.05). The expression of bFGF ranked as LG, WG, MG and BG (P<0.01), Desmin ranked as WG=LG and BG=MG (P<0.05), ERK1/2 as LG=WG, MG and BG (P<0.05). Conclusion Electroacupuncture could promote the regeneration and repair of tissue after acute lumbar muscle contusion, which may be related to the up-regulation of bFGF/ERK signaling pathway.
6.Characteristic of growth decreased CHO(dhfr-) cell line mutated by outer space

ObjectiveTo investigate the changes of the morphology, growth and cycle of CHO(dhfr-) cells after space flight.MethodsCHO(dhfr-) cells were carried in the No.18 recoverable satellite and monocloned harvesting cells before multiplying. 4 cell lines were selected randomly,and the growth characteristics of the most slowly growing one at the fifth passage was observed by methods of MTT and FCM as well as the cells' shape.Results159 cell strains were obtained after monocloning and multiplying. The cells' morphology changes, growth speed decrease and the number of G1 phase increased markedly.ConclusionSpace flight induced morphological changes of cells and it is impossible to screen out finer bioengineering cells.
7.Effect of rehabilitation bridging mode on postoperative functional exercise in patients with breast cancer

Objective:To explore the effect of functional exercise rehabilitation bridging mode on the functional exercise and self-management ability of patients after breast cancer surgery, so as to provide reference for guiding patients to achieve full process management and self-management and rehabilitation.Methods:Using a non randomized grouping quasi experimental design, 202 patients with breast cancer undergoing surgical treatment were selected from the breast surgery department of Yuhuangding Hospital in Yantai from March to July 2021 through convenient sampling. The control group received routine nursing and rehabilitation guidance. The intervention group adopted a rehabilitation bridging mode for postoperative functional exercise on the basis of routine nursing care. The functional exercise compliance rate and self management ability of the two groups of patients after the intervention were evaluated.Results:In the intervention group, the rate of functional exercise reaching the standard, the rate of functional exercise knowledge awareness, correctness and compliance in self-management ability of breast cancer patients were as follows: 93.1%(95/102), 71.6%(73/102), 80.4%(82/102), 73.5%(75/102), which were higher than the control group 84.0%(84/100), 46.0%(46/100), 52.0%(52/100), 17.0%(17/100). The differences were statistically significant ( χ2 values were 4.18-65.06, all P<0.05). Conclusions:The rehabilitation bridging mode could connect the functional exercise needs of patients at different stages of rehabilitation, provide personalized, continuous, and standardized functional exercise guidance for patients, help them improve the effect of functional exercise, and improve their self management ability.
8.Application of outer space cell living system in space flight of rabbit's mesenchymal stem cell

ObjectiveTo investigate the condition and method of a long time mesenchymal stem cell (MSCs) culture with outer space cell living system carried by a recoverable satellite.MethodsMSCs were obtained from 1 week New Zealand rabbits, and seeded to culture flask and outer space cell living system. Under the condition of absence O2, CO2 and medium unable changed, influence of monolayer culture and outer space cell living system upon survival of long time MSCs culture in simulated space various temperature was observed.ResultsUnder the condition of absence O2, CO2 and medium unable changed, monolayer cultured MSCs were maintained to survive for 15 days, and group of space MSCs were successfully maintained to survive for 30 days in outer space cell living system. There was a significant difference between two methods. ConclusionOuter space cell living system can maintain MSCs to survive for 30 days under space flight temperature.
9.A preliminary study on the ultrasonic evaluation of deformity changes during the treatment of congenital clubfoot in infants

Objective:To evaluate the deformity changes in the treatment of congenital clubfoot in infants by ultrasound, and to explore its clinical application value.Methods:Seventeen infants with congenital clubfoot treated in the Department of Pediatric Orthopaedics of the Third Affiliated Hospital of Zhengzhou University from March 2020 to June 2021 and 27 normal infants in the same period were selected. The distance between medial malleolus and scaphoids (MM-N distance) of all feet was measured by ultrasound. The distance from the tangent line of the lateral edge of calcaneus to the midpoint of the lateral edge of the chondroid bone (C-C distance), medial soft tissue thickness and tibial calcaneal angle were measured by ultrasound. The data before, after treatment and during follow-up were statistically analyzed.Results:A total of 88 feet of 44 infants were evaluated. The mean number of cast was 4.7±1.1, the follow-up time was (96.9±59.3)days. The MM-N distance, C-C distance and posterior tibial calcaneal angle in the clubfoot group were improved after treatment and at the last follow-up, and the differences were statistically significant (all P<0.01). During the treatment, 1 case had false correction, and 2 cases recurred in the last follow-up. Conclusions:Ultrasound can clearly display the medial, dorsal, lateral and posterior articular structures of normal and clubfoot, and can observe the deformity changes of clubfoot during the correction process, which can provide guidance for the clinical treatment of clubfoot.
