1.Expression of RhoA in placenta of preeclampsia.

In order to detect the expression of RhoA in placenta from normal pregnancy and preeclampsia and evaluate the role of RhoA in preeclampsia, the expression of RhoA in placenta collected from 40 preeclampsia patients and 20 normotensive controls was determined by immunohistochemistry and RT-PCR. RhoA was found in syncytiotrophoblasts and cytotrophoblasts. The mean density of RhoA protein in placental tissues of mild and severe preeclampsia groups was significantly higher than that in normal pregnancy. The expression level of RhoA mRNA in mild and severe preeclampsia groups was significantly higher than that of normal pregnancy. Increased expression of RhoA in placental tissues might play an important role in the pathogenesis of preeclampsia.
2.Detection of double-position mutation in the basic core promoter of hepatitis B virus with microplate sandwich hybridization

Objective To investigate 1762T、1764A double position mutation in the Basic Core Promoter(BCP) of hepatitis B virus and reveal its relation to clinical symptoms and HBeAg phenotype. Methods microplate sandwich hybridization technique was used to detect BCP double position mutation. One common capture probe and one mutant specific detector probe as well as one wild type detector probe were synthesized and hybridized with amplified HBV DNA from the sera of hepatitis B patients. The results of hybridization were exhibited with ELISA. Results 147 hepatitis B patients who had been confirmed HBV DNA positive were screened. 51 patients were BCP double position mutation, 42 of which were BCP single position mutation and, 9 were mix mutation(both mutation and wild type were positive). BCP mutant was detected in 36 of 117 with chronic hepatitis and, 8 of which were mix mutants. Moreover, BCP mutant was detected in 7 of 30 with acute hepatitis in 25 of 78 with HBeAg positive were mutant and in 26 of 65 with HBeAg negative were mutant.Conclusions (1) The rate of BCP double position mutation in chronic hepatitis B patients is higher than that in acute hepatitis B patients. (2) BCP mutation may impair HBeAg expression. (3) PCR microplate sandwich hybridization ELISA is a sensitive and efficient method for detecting gene mutations.
3.The effect of telmisartan on APN,hs-CRP and TNF-α in hypertensive patients with unstable angina pectoris

Objective To investigate the effect of telmisartan on the expression of APN,hs-CRP,TNF-α and the prognosis of hypertensive patients with unstable angina pectoris. Methods Totally 100 hypertensive patients with unstable angina pectoris were randomly divided into the telmisartan group(n =50) and the losartan group(n =50),each group with oral administration. The blood pressure controlled condition, APN, hs-CRP and TNF-α content changes and the occurrence of cardiovascular accident after treatment were observed in both two groups. Results In both groups, blood pressure returned to normal in 8 weeks,with no significant difference(P > 0.05). After treatment,APN level in the telmisartan group increased faster than that in the losartan group(P <0.05) ,while the hs-CRP,TNF-αdecreased significantly(P < 0.05). After 8 weeks, the occurrence of cardiovascular accident in the telmisartan and losartan groups was 10.0% (5/50) and 24. 5 % (12/49) respectively in the following 4 months (P < 0.05). Conclusion Telmisartan has significant effect in regulating APN, hs-CRP, TNF-α expression in hypertensive patients with unstable angina pectoris, which is able to decrease the short-term occurrence of cardiovascular accident.
4.Detection of human parvovirus B19 nonstrutural protein DNA by nested-polymerase chain reaction in gravida serum and pregnant tissues.

A new nested-polymerase chain reaction (nested-PCR) assay was developed to detect human parvovirus B19 DNA corresponding to the nonstructural protein in clinical specimens in a routine diagnostic laboratory. The sensitivity of this highly specific assay was up to 0.005 fg of B19 DNA. Parvovirus B19 was identified in sera of 20 pregnant women with abnormal pregnant outcome. Among these 20 cases, intrauterine parvovirus infection did exist in 7 pregnant women because parvovirus B19 DNA was detected in the pregnant tissues of them such as placenta tissues, chorionic villi, amniotic fluid, fetal spleen, liver and abdominal fluids.

5.Value of transvaginal ultrasonic in the diagnosis of early cervical carcinoma and cervical neoplasis

Objective To study the clinical diagnostic value of transvaginal ultrasonic in early cervical carcinema and cervical neoplasis.Methotis The suspected patients with cervical carcinoma and cervical neoplasis were detected with transvaginal ultrasonography,liquid based cytology and cervical biopsy.The sonograms of transvaginal ultrasonic were retrospectively analyzed.Results In early cervical carcinoma and cervical neoplasis,the diagnostic sensitivity of transvaglnal ultrasonic was 90.9%and 83.3%;and the diagnostic specifity of transvaginal ultrasonic was 70.6% and 60.0%;the rate of missed diagnosis of transvaginal ultrasonic was 9.1% and 16.7%.Conclusion Transvaginal ultrasonic plays an important part in the clinical diagnosis of early cervical carcinoma.
6.Change of the guanine nucleotide-binding protein in lamina Ⅰ-Ⅲ of spinal cord posterior horn and lateral spinal nucleus of rats after transection of dorsal roots

BACKGROUND: In China, this laboratory is the first one to report such researches, confirming that strong αo-immunoreactive (IR) appears in the substantia gelatinosa (SG) of spinal cord and lateral spinal nucleus which is similar to the distribution of certain neuropeptides that participate in sensory regulation, which suggests that guanine nucleotide binding protein (G protein) may be related to primary afferent informational transfer. OBJECTIVE: To observe the change of αo-IR in gelatinous substance by the method of transection of unilateral spinal dorsal roots.DESIGN: A randomized controlled experiment on animals.SETTING: Staff Room of Neurobiology, Tongji Medical College, Huazhong University of Science and Technology.MATERIALS: The experiment was conducted at the Staff Room of Neurobiology, Tongji Medical College, Huazhong University of Science and Technology from December 1995 to December 1996. Fifteen healthy adult SD rats were selected and divided into 3 groups: ①normal group with five rats (not dealt with any disposal), ②transected dorsal root group with 10 rats (right side) and ③control group (non-transected left sidedness as control).METHODS: Right lumbar 1-3 spinal neural dorsal roots were cut off under the anesthesia of 100 g/L chloral hydrate (300 mg/kg)through intraperitoneal injection in rats, living for 48-60 hours after operation. The subunit αo of guanine nucleotide-binding protein (rabbit polyclonal antiserum) was demonstrated in the αo-IR of rat spinal cord by immunohisto chemical methods. G protein was oriented, and its change was observed after transection ofneural dorsal roots MAIN OUTCOME MEASURES: ①The αo-IR of Ⅰ to Ⅲ of the dorsal horn and lateral spinal nucleus of the normal rats and control rats. ②The αo-IR of Ⅰ to Ⅲ of the dorsal horn and lateral spinal nucleus of rats in the transected dorsal root group. RESULTS: Data of a total of 15 rats were involved in the result analysis. ①In the normal group and control group, intense αo-IR was presented in rexed lamina ( Ⅰ to Ⅲ ) of the dorsal horn of rats, and the highest αo-IR in second lamina (SG). Lateral spinal nucleus of rat revealed higher density of αo-IR containing fiber networks. Following unilateral transection of dorsal roots in SG, αo-IR was markedly decreased. ②Quantitative analysis of absorbance (A) of αo-IR, it was (0.847±0.081) in the inside of the control group, (0.633±0.073)(t=5.71 ,P < 0.001 ) in the inside of transected dorsal root group. It was (0.823±0.089) in the middle area of the control group,(0.660 4±0.074)(t=6.90,P < 0.001 ) in the middle area of the transected dorsal root group. It was (0.915±0.090) in the lumbar region of the control group, and (0.656±0.077)(t=10.31 ,P < 0.001 ) in the lumbar region of the transected dorsal root group. Average value of the control group was (0.852±0.084), and average value of the transected dorsal root group was (0.639±0.078)(t=10.23 ,P < 0.001 ).CONCLUSION: Part of G protein of end-brush neurons related with the primary afferent noxious stimulation in SG derives from primary sensory neurons, which maybe join the adjustment of primary sensory transfer.
7.A DOUBLE LABELLING IMMUNOCYTOCHEMICAL STUDY OF NEUROTENSIN AND SUBSTANCE P IN DORSAL HORN OF SPINAL CORD IN RAT——A LIGHT AND ELECTRON MICROSCOPIC CORRELATIVE STUDY

Immunoreactivity of neurotensin(NT) and substance P (SP) were studied by means of PAP method with simultaneous immunocytochemical double staining on the same sections using diaminobenzidine (DAB) for SP and benzidinc dihydrochloride(BDHC) for NT as the chromogens for light and electron microscopy in dorsal horn of spinal cord of rat treated with colchicinc. In our hands the reaction products of DAB and BDHC is quite discernible, under LM the former is brown and later is blue; under EM DAB reaction product is homogeneous and diffuse electron opaque while BDHC reaction product is patch electron opaque, it is a new but simpler approach to demonstrate two antigens simultaneously in the same ultrathin section. It would be conformable further to the studies of morphological and functional relationships between different kinds of neurons. The results showed that NT-like immunoreactive (NT-LI) perikarya were mainly located in lamina Ⅱi and out layer of lamina Ⅲ and NT-LI terminals were mainly located in laminae Ⅰ-Ⅲ. Under EM the NT-LI axon terminals may synapse with the unlabeled axon or unlabeled dendrites. SP-like immunoreactive (SP-LI) perikarya were mainly located in lamina Ⅱ. The density of SP-LI terminals were higher in laminae Ⅰ-Ⅱ, while axo (SP)-axonic (SP), axo (SP)-somatic (SP) synapses were identified with EM. In double labelling sections, furthermore under EM, it was found that NT-LI axon terminal (BDHC patch like electron opaque reaction products)can synapse or contact with SP-LI axon (DAB diffuse electron opaque reaction products). Our results suggested that NT-and SP-containing neurons and terminals in dorsal horn might participate in regulating process of primary sensory transmission.

Objective To explore the anatomy of optic microvascular and the transcranial microsurgicaloperative strategies for protection of optic nerve function during the operation of the resection of giant invasivepituitary adenoma.Methods Thirty-five patients with giant pituitary adenoma were treated by different surgicalapproaches according to preoperative imaging data.The tumors were removed by employing microsurgicaltechnique via the parachiasmal opticocarotid and carotidotentorial spaces in sellar region and by opening thelamina terminals.Much attention was given to the preservation of perforating arteries from the interal carotidartery,anterior and posterior communicating arteries and arterior choroidal artery to optic nerve and chiasmResults Total removal of tumors Was conducted in 25 cases.Subtotal removal was performed in 10cases.Anatomical preservation of the optic nerve was achieved in all the cases during the operation,with opticnerve function getting better in 20 cases,remaining unchanged in 2 cases,no improvement in 7 cases and gettingworse in 6 cases.Conclusion A reasonable surgical approach is a prerequisite for the giant invasive pituita~adenoma surgery.Understanding the anatomical characteristics of microvascular in the sellar region,carefulprotection of the optic nerve,optic chiasm and microvascular,skilled microsurgical technique during the operationis key to prevent post-operative complications of optic nerve function.
