1.30-YEAR RETROSPECTIVE STUDY OF SCHISTOSOMAL CIRRHOSIS OF THE LIVER IN AN AREA AFTER SCHISTOSOMIASIS CONTROL

The natural history of 383 patients of schistosomal cirrhosis, discovered from 1960-1990 in Donjing township Shanghai suburb, where schistosomiasis had been controlled for more than 10 years, was studied. The cumulative survival rate in 243 patients who had splenecto-my 11-30 years previously (group I ) was much higher than that of 140 patients without splenectomy (group II ), and the time for 75% survival rate of these 2 groups were 18 and 4 years respectively, being much longer in the splenectomized patients without ascites and those operated before age of 30 than those complicated with ascites and those operated between the age of 31 - 50 or after 51 years of age.The complications and mortality rate of group I were significantly lower than that of group I , indicating that splenectomy not only prevented the complication of portal hypertension but improved the general health and resistance of the patients against other pathogens as well.There was no significant difference between group I and group II in the ratio of the avarage death age of the deceased patients to the life expectancy of the general population, suggesting that the life span of a number of patients without splenectomy are not substantially shortened in an area where schistosomiasis had been controlled.
2.ACRIDINE MUTAGEN ICR-170 INDUCED DEVELOPMENTAL SUPPRESSION OF OVARY AND VITELLINE GLAND IN FEMALE SCHISTOSOMA JAPONICUM

Mice of Kunming strain were infected with Schistosoma japonicum cercariae previously incubated with various concentrations of acridine mutagen ICR-170 for different time durations. At 6 weeks after infection, the mice were autopsied. The results showed that 24 out of 28(85. 7%) adult female worms had deformed or lacked ovaries and vitelline glands when the cercariae were treated with the agent at a concentration of 10?g/ml and incubated at 30. 5℃ for 30min. No apparnet changes were observed in the male worms inhabiting the mesenteric and portal veins with the females worms in their gynecophoral canals. The mutagenized female schistosomes obtained from the present experiment might be served as another form of attenuated worms for the induction of protective immunity.
3.Anti-rejection Effect of Tripterygium Glycosides on Allogeneic Islet Transplantation in Mice

Objective To investigate the effect of tripterygium glycosides on the resistance to immune rejection after allogeneic islet transplantation in mice.Methods Twenty C57BL/6 mice were treated with STZ diabetes mellitus and transplanted the islets from Balb/c mice donor,then recipient mice were randomly divided into two groups:triptolide group and model control group(n=10),and were intraperitoneal injected with tripterygium glycoside solutin and equivalent solvent of 5 mg·kg-1·d-1 for 14 days.Blood glucose and body weight were measured within 4 weeks after transplantation.Two weeks later,two groups of grafted islets were stained by HE staining and immunohistochemical staining,the expression of IL-2 protein were detected by Western blotting.Results The level of blood glucose were decreased to normal in the triptolide group and model control group after islet transplantation,but blood glucose gradually increased in the model control group after two weeks.Compared with the model control group,the inflammatory cells were less infiltrated and the immunohistochemical staining of insulin was deeper in the triptolide group.The expression of IL-2 in the triptolide group was significantly decreased(P<0.05).Conclusion Tripterygium glucoside could significantly decrease the inflammatory cell infiltration and inflammation factor expression in the allogeneic islet recipients to reduce the immune rejection and improve graft survival.
5.Theoretical and experimental studies on apparent solubility parameters of multiple components for Chinese materia medica with HPLC fingerprint.

OBJECTIVETo establish the theories and methods to determine apparent solubility paraneters of multiple components for the Chinese materia medica (CMM) with HPLC fingerprint.
METHODThe mathematical functional expresses to determine the apparent solubility parameters for multiple constituents were established according to total quantum geometrical average retention time (TQGART) for HPLC fingerprint that characterized the entirety tendencies for all-over chromatographic peaks, validated by the aloe-emodin's solubility parameters which had been determined. The HPLC for the aloe-emodin's solubility parameters were carried out with an Alltech Apollo C18 as column, Acetone: Water as mobile phase, gradient elution,flow rate as 1.0 mL x min(-1), the detection wave-length as 430 nm and the temperature as 30 degrees C.
RESULTThe mathematical functional model between the TQGART of HPLC chromatographic fingerprint and the total quantum apparent solubility parameters was established and used to determine the aloe-emodin's mixture solubility parameter as 36.12 J(1/2) x cm(-3/2), nearly equal to 35.57, 36.07 J(1/2) x cm(-3/2) calculated by ration of peak area ratios and molecular fraction, respectively.
CONCLUSIONThe TQGART of HPLC fingerprint can be used to determine simultaneously the apparent or single intrinsic solubility parameters for total quantum or intrinsic solubility parameters for single in multiple constitute systems, by which theoretical and technologic platform to study the compatibility rule and dosage form reform of the single CMM will be established.
