1.Sensitivity of Pseudomonas aeruginosato 18 Kinds of Antibacterials:Analysis of 485 Strains

OBJECTIVE To explore the antibiotic activity and changing trend of clinically isolated Pseudomonas aerginosa strains.METHODS The Drug-resistance of 485 clinically isolated to 18 kinds of common antibiotics P.aeruginosa strains was performed by Kirby-Bauer method.The drug sensitiveity test was indged based on the standard of the National Committee for Clinical Laboratory Standards(NCCLS) of the USA from 2004 to 2007.RESULTS Imipenem was sensitire against P.aeruginosa with the total drug-sensitivity rate of 80.8%.The resistance to cefazolin was high(0%).The sensitivity rate of P.aeruginosa was declining year by year.CONCLUSIONS It is very important to select antibiotics correctly according to the results of sensitivity tests.
2.Changes in the constitution of female college students before and after the aerobic exercise

BACKGROUND: Nowadays, aerobic exercise has been a theme of mod ern body-building. Therefore, there are practical significance of under standing and mastering the effect of aerobic exercise on the constitution of body builder in instructing body-building practice. OBJECTIVE: To observe the effect of aerobic exercise on body composi tion, cardiore spiratory function and body figure of female college students. DESIGN: Non-randomized concurrent control experiment. SETTING: China Female College. PARTICIPANTS: The experiment was conducted from May 2003 to Oc tober 2004. 100 female college students were randomly selected from Chi na Female College and other three colleges and taken as the aerobic exer cise group. Another 100 female college students of the same age without systematic aerobic exercise were taken as the control group. METHODS: ①The course for aerobic exercise group were mainly aero bics, ballroom dancing and hexagram boxing, including 5-15 minutes of warming up, 30-45 minutes of aerobic exercise [the target heart rate in (75% maximal heart rate) exercise lasted for at least 15 minutes] and 15-20 minutes of cooling down, which were conducted for 60 minutes each time and twice a week. The exercise should be performed at 2.5 hours before supper for 1.5 years.②Subjects in the control group were not inter fered but kept in same diet and resting time as the aerobic exercise group. MAIN OUTCOME MEASURES: ①Body figure indexes: body height, body mass, chest circumference, waistline, hip circumferences.②Skinfold thickness, fat mass index and lean body mass. ③ Lung capacity, maximal breathing capacity in each minute, stroke volume, cardiac output, ejection fraction, heart rate, ejection time, mean normalized systolic ejection rate (MNSER) and left velocity of circumferential fiber shortening (LVCf). RESULTS: A total of 200 subjects, who had accomplished all tests were involved in the analysis of results. ①The waistline was smaller after exer cise in the aerobic exercise group than that in the control group, while the chest circumference and hip circumferences were bigger than those in the control group (P < 0.05), and the waist-to-hip ratio was smaller than that in the control group (6.50±10.48, 70.39±11.24, P < 0.05). There was no sig nificant difference in body mass index between the two groups (P < 0.05). ②The skinfold thickness in rump, scapular region, thoracic region, abdom inal region as well as femoral region and region of shank were smaller after exercise in the aerobic exercise group than those in the control group (P<0.05-0.005), and the body fat percentage was significantly smaller than that in the control group [(20.5±3.45)%, (26.5±4.58)%,P < 0.005]. However, the lean body mass in the aerobic exercise group was greater than that in the control group [(41.8±9.86), (36.9±8.69) kg,P < 0.01].③ After exercise, the lung capacity, maximal breathing capacity in each min utes, stroke volume, MNSER and LVCf in the aerobic exercise group were remarkably greater than those in the control group (P < 0.05). CONCLUSION: ①Aerobic exercise can effectively ameliorate the body figure of female college students and coordinate three measurements with each other.②Aerobic exercise can greatly reduce the in vivo fat contents of female students and increase their lean body mass. ③Aerobic exercise can remarkably enhance the lung capacity, pulmonary ventilation volume as well as cardiac functions of blood-pumping and contraction etc. In female college students.
3.Health Human Resources of Tertiary General Hospitals in Beijing

Objective To explore a clear picture of the health human resources allocation in tertiary general hospitals in Beijing ,and propose strategies to optimize the configuration of human resources at the same level hospitals ,so as to improve the medical service quality for society. Methods Basic operational data and human resources configuration of 16 tertiary general hospitals with more than 800 beds in Beijing were col?lected by using clustering sampling and literature review ,and then a descriptive analysis of the human resource configuration data at the same level hospitals was carried out by SPSS18.0 and Excel2010. Results The number of patients increased year by year,but the average days of hospitaliza?tion showed a downward trends during 2009?2011. According to the previous way of beds to determine personnel ,there was significantly different from the actual situation. A significant quantitative correlation was foung between the physicians and the nurses,pharmacists,medical laboratory technician,and the ratio between physicians and these staffs is stable. Conclusion With the development of hospital service and increase of health human resources,health manpower ratio has changed significantly in tertiary general hospitals. A stable proportion of physicians can be used as the reference standard for hospital human resources configuration at the same level hospitals.
4.Experimental study of immune responses in mice induced by nucleic acid vaccine of gag gene of Chinese HIV-1 strains

Objective To construct the nucleic acid vaccine(DNA vaccine) plasmid of the gag gene of the prevalent HIV-1 strains in China so as to develop a therapeutic AIDS vaccine against the HIV-1 strains, and to investigate the immune responses induced by HIV-1 DNA vaccine in mice. Methods The recombinant expression vector pCI-neo GAG was constructed by inserting HIV gag gene into the eukaryotic expression vector pCI-neo, which was identified by restrictive enzymes (Xba Ⅰ/Sal Ⅰ) digestion analysis and DNA sequencing. Balb/c mice were immunized with pCI-neo GAG or pCI-neo. Their sera were collected for the assay of the titer of anti-HIV antibody and IFN-? level by ELISA, and their splenic cells were isolated for detecting antigen-specific lymphoproliferative responses and specific cytotoxic T lymphocyte(CTL) response by MTT assay and LDH assay, respectively. Results Restrictive enzymes digestion analysis and DNA sequencing results revealed that the recombinant expression vector pCI-neo GAG had been constructed successfully. 5.Construction and sequencing of human interleukin-18 eukaryotic expression vector

Objective:To construct the eukaryotic expression vector of interleukin-18 gene and carry out analysis of pVAX1-IL-18 sequence . Methods:The recombinant eukaryotic expression vector pVAX1-IL-18 was constructed by inserting interleukin-18 gene into the eukaryotic expression vector pVAX1 with molecular cloning technique . It was confirmed by restrictive enzymes(BamHⅠ/ EcoRⅠ) digestion and analysis of DNA sequencing . Similar nucleotide sequences encoding IL-18 of pVAX1-IL-18 were looked up by Blast means in NCBI GenBank. Results:Restrictive enzymes digestion analysis and DNA sequencing results revealed that the interleukin-18 gene was cloned into the eukaryotic expression vector pVAX1 successfully .The sequence of encoding IL-18 of pVAX1-IL-18 was exactly the same as the reported sequence of encoding human IL-18 in GenBank . Conclusion: The recombinant eukaryotic expression vector pVAX1-IL-18 has been constructed successfully, which lay the foundation for pVAX1-IL-18 as a genetic adjuvant of DNA vaccine.
6.STUDY ON THE CONTENT AND STABILITY OF VITAMIN C OF DATEPLUM PERSIMMON'S LEAVES PROCESSED IN THE FORM OF TEA

The vitamin C content of dateplum persimmon's leaves (Diospyros lotus) collected and processed in some months (May, Jun., Aug., Sept. and Oct.) was determined. It was found that the amount of vitamin C in these products was 2045, 2540, 2650, 2680 and 310 mg per 100 gm respectively. They were all extremely rich in vitamen C and had an average about 2500 mg per 100 gm except that processed in Oct., of which the vitamin C content declined sharply.Observation on the stability of vitamin C in the products packed with several methods and stored in different conditions and durations showed that various environmental factors exerted a considerable influence on vitamin C retention to a certain extent, especially the sunlight and relative humidity which caused seri-ous damage. In general, the vitamin C retention of products packed with various methods were above 75% after two months of storage. Even when the Product was packed by plastic film bag and stored in ordinary condition, the remaining vitamin C was still above 800 mg per 100 gm after six months.After brewing of the product for 5-10 minutes just like making tea, the percentage of vitamin C extracted was about 75% in the first brewing and 11.5% in the second. The total amount of vitamin C extracted in two brews was about 85%.
7.Determination of Total Nitrogen in Seawater by Micro Sequential Injection-Cadmium Column Reduction Spectrophotometry

A cadmium column reduction-azo dyes spectrophotometric method based on micro sequential injection lab-on-valve was established for the determination of total nitrogen in seawater. The experimental parameters were optimized, and the interference experiment was carried out. The results showed that the interference of the main components and salinity in sea water could be eliminated by using a series of standard solution prepared by national standard seawater with certain salinity. The concentration of total nitrogen in seawater was linear with the absorbance in the range of 0 . 03-1 . 00 mg/L with a correlation coefficient of 0. 9993. When determining the national standard seawater at nitrogen concentration of 0. 20 mg/L, the relative standard deviation (RSD) was 4. 9%, the detection limit was 0. 010 mg/L, and the recoveries were 99. 5%-101 . 1%. There were not significance differences between the results of this method and national standard method in the t-test analysis. The method is suitable for the determination of total nitrogen in seawater.
8.Study of genotypic resistance mutations of protease and reverse transcriptase in non-therapeutic acquired immune deficiency syndrome patients in Heiiongjiang Province

Objective To study the presence of drug resistant mutations of protease and reverse transcriptase among human immunodeficiency virus (HIV)-1 strains isolated from treatment naive HIV/ acquired immune deficiency syndrome (AIDS) patients in Heilongjiang Province of China and to provide the baseline data for starting antiretroviral therapy in this area. Methods The protease and reverse transcriptase gene sequences were amplified by nested-polymerase chain reaction (PCR) and then sequenced. The results were compared to the subtype B consensus amino acid sequence and analyzed with Stanford HIV-db drug resistance sequence interpretation. Results The results showed that HIV strains from 49 patients were classified as subtype B'. No primary mutations associated with protease inhibitor were detected. Some secondary mutations associated with protease inhibitor were detected, which included V77I(91.5%), L63P(76.6%), I93L(74.5%), E35D(61.7%), R57K (19.1%), R41K(10.6%), A71V(8.5%), M36I(8.5%), L10I(6.4%), D60E(6.4%), L89M (4.2%) and G16E(2. 1%). Only one case had a primary mutation M184I that was associated with resistance to reverse transcriptase inhibitors. However, many secondary mutations associated with resistance to reverse transcriptase inhibitors were found, including I135L/T/R/V(81.8%), V106I(22.7%), V179D/E(11.4%), R211K(9.1%), L214F(4.5%), V189I(4.5%) and V108I(2. 3%).Conclusions The prevalence of genotypic anti-HIV drug resistance is very low in treatment naive HIV/AIDS patients in Heilongjiang Province. However, closely monitoring on drug resistance mutation is very important for preventing the development and prevalence of multi-drug resistant or cross drug resistant HIV.
10.Correlation between perceived control and subjective well-being in patients with coronary heart disease

Objective To investigate the spiritual well-being (SWB) and perceived control (PC) in adult patients with heart failure (HF),and analyze the relationship between spiritual well-being and perceived control. Methods 125 patients with heart failure were surveyed with questionnaires by convenience sampling, including Control Attitudes Scale-Revised (CAS-R) and Memorial University of Newforndland Scale of Happiness (MUNSH). Results The mean score of the spiritual well-being was(6.34± 2.92), and the mean score of perceived control was (18.40±4.39). The patients’spiritual well-being was closely related to perceived control (P < 0.05). There was a positive correlation between the self-efficacy and the positive indicator of subjective well-being, r=0.46, negative correlation was between the self-efficacy and the negative indicator of subjective well-being, r=-0.17. There was a positive correlation between the locus of control and the positive indicator, r=0.38,while the negative correlation was between the locus of control and the negative indicator, r=-0.05. There was a negative correlation between the learned helpless and the positive indicator, r=-0.43, while the positive correlation was between the learned helpless and the negative indicator, r=0.06. Conclusions The spiritual well-being of patients is at a low level, as well as perceived control. The spiritual well-being of patients can be enhanced by means of improving the ability of perceived control.
