1.Research progress of drugs in treatment of stress gastric ulcer

The stress gastric ulcer is due to a stress-induced gastric erosions, superficial ulcers and bleeding gastric mucosal le-sions characterized by stress. Its etiology is closely associated with lives, the incidence is increasing. In recent years, the the basic re-search of drugs focuses on inhibition of oxidative damage caused by stress, anti-inflammatory cytokine expression, and regulation of gastric acid, etc., and made a lot of success. Treatment choice of stress ulcer drug gradually increased. The effects of plant extracts on stress ul-cer get more and more attention. This article reviews research progress of the treatment drugs of stress ulcer in the current stage.
2.Membrane potential properties in the smooth muscle cells of middle cerebral artery in stroke-prone spontaneously hypertensive rats

BACKGROUND: The small change of the membrane potential (Em) of vascular smooth muscle cells (VSMC) could result in significant alteration in tension, and the membrane depolarization was closely associated with the development of hypertension.OBJECTIVE: To study the resting Em of VSMC from the middle cerebral artery (MCA) of stroke-prone spontaneously hypertensive rats (SHRSP) and the reactivity of Em to KCl and norepinephrine (NE).DESIGN: An animal experiment of comparative analysis.SETTING: Institute of Materia Medica, Shandong Academy of Medical Sciences.MATERIALS: Eighteen male SHRSP and 26 male Wistar rats were used.METHODS: ① Rats in both groups were anesthetized with intraperitoneal injection of ethyl carbamate (1 g/kg), the brains were removed quickly, and the Em of VSMC was recorded by intracellular microelectrodes. ② The effects of different concentrations of KC1 (10, 20, 50 mmol/L) and NE (10-7,10-6, 10-5 mol/L) on the Em of VSMC were observed.MAIN OUTCOME MEASURES: ① Em of VSMC in both groups; ②Effects of different concentrations of KC1 and NE on the Em of VSMC.RESULTS: ① The Em was obviously lower in SHRSP than in the Wistar rats [(-48.2±3.1), (-64.4±4.3) mV]. ② Both KC1 (10, 20, 50 mmol/L)and NE (10-7, 10-6, 10-5 mol/L) could cause the depolarization of Em of MCA in a concentration-dependent manner. The reactivity to MCA of SHRSP was obviously increased as compared with the Wistar rats.CONCLUSION: The resting Em of VSMC from the MCA of SHRSP was lower, and the reactivity of VSMC to KCl and NE was significantly stronger than that of Wistar rats.
3.THE DISTRIBUTION OF CFU-C IN HUMAN FETAL BONE MARROW, LIVER, SPLEEN AND PERIPHERAL BLOOD

40 human fetuses were used in this study. The range of fetal age varied from 3 months to full terms. All fetuses used were normal and their parents were healthy. The CFU-C from bone marrow, liver, spleen and peripheral blood were studied in vitro with agar culture technique modified by Metcalf.Experiments proved that the CFU-C of bone marrow may be observed in the fetuses in 3rd month, however, it increased rapidly in 4th month and maintained higher level until the end of fetal life. The progenitor cell population was detected at very high level in liver from 4th month to 6th month fetuses. The number of CFU-C of fetal liver were low in 7—10th month. The circulating progenitor cells in peripheral blood of 4, 5 and 6-month fetuses Were on high level but it was lower than that of liver and bone marrow.
4.Optimization of Fermentation Process of Compound Chinese Medicine by Response Surface Methodology

Objective To investigate the factors that influence the fermentation process of compound Chinese medicine and determining the optimum fermentation with single factor experiment and response surface methodology. Methods Through controlling the factors in the fermentation process of compound Chinese medicine (such as fermentation bacteria, fermentation time, fermentation temperature, inoculum amount, etc.), with its increase rate of total peak area as evaluation indicator, the alcohol extracts before and after fermentation were monitored and comparative evaluated by HPLC, and the optimum fermentation process was determined by response surface methodology. Results The fermentation process optimized by single factor experiment and response surface methodology was as follows:SZ-2 strain served as the fermentation bacteria, temperature was 33 ℃, inoculum amount was 4%, and time was 3.5 d, the average increase rate of total peak area was 31.24%. Conclusion HPLC can be used to identify and evaluate the fermentation of compound Chinese medicine under the different factors, and to clarify the optimal fermentation process by response surface methodology, which provide reference for the development of fermentation process.
5.Alteration of Wnt/β-catenin signaling pathway in early diabetic rat myocardium

Aim To investigate the alteration of Wnt/β-catenin signaling pathway in early diabetic rat myo-cardium and clarify its role in development of diabetic cardiomyopathy. Methods The diabetes mellitus ( DM) model was prepared by intraperitoneal injection of streptozotocin ( STZ, 60 μg · g-1 ) . The alteration of Wnt/β-catenin signaling pathway was determined by Western blot and immunohistochemistry. HE staining was used to analyze the change of myocardial pathologi-cal structure. Results Cardiac histological analyses revealed that DM induced cardiomyocyte degeneration and necrosis. Myocardial Wnt2, β-catenin and c-Myc were enhanced in 2 wk DM compared with control group while DKK1 showed no significant alteration. Conclusion Wnt/β-catenin signaling pathway is acti-vated in early diabetic myocardial injury. Further re-searches on its role in DM myocardium may find a new therapeutic target for diabetic cardiomyopathy.
6.THE EFFECT OF TADPOLE EXTRACT ON HELA CELLS AND ITS POSSIBLE MECHANISM

The tadpole extract without large molecular protein(T-871) was prepared from dry tadpoles. This extract and RPMI1640 medium were mixed in the ratio of 1:50(V/V) to form T-871 medium. HeLa cells were cultured in the T-871 medium in order to study the possible mechanism of HeLa cell differentiation induced by tadpole extract. We found that there was a decreased acitivity of LDH which may be released through HeLa cell membrane in the 2 day's cultured T-871 medium. After 3 day's culture in the T-871 medium we found that on the HeLa cell membranes, Na-K-ATPase activity reduced and the content of Con A receptors increased. When these cells were analyzed by flow cytometry(FACS-420), the results indicated that HeLa cells accumulated in G_2 and M phases of the cell cycle and the influence on DNA and RNA content of the HeLa cells was insignificant. HeLa cells cultured by RPMI 1640 medium or T-871 medium for 3 days were inoculated into the dorsal hypodermis of 12 nude mice, respectively. Then after 24 hours this extract was injected to each nude mouse at dose of 0.3 ml each day. Our study showed that T-871 could result in decrease of tumor formation such as delay of tumor nodule appearance and reduce of tumor weight. This study suggested that the changes of the cell membrane may be caused by the tadpole extract. It may also reduce malignancy of the HeLa cells.
7.Impact of polysomy 17 of breast cancer on the testing results of human epidermal growth factor receptor 2 (HER-2) and its clinicopathological significance

Objective:To investigate the impact of polysomy 17 of breast cancer on testing results of human epidermal growth factor receptor 2 (HER2) and its clinicopathologic significance. Methods:Seventy-one patients with primary invasive breast carcinoma were studied. The HER2 gene and chromosome 17 copy numbers were determined by dual-color fluorescence in situ hybridization (FISH). The testing results were expressed by absolute HER2 gene copy number or the ratio of HER2 to chromosome 17. Based on the FISH testing results and HER2 protein expression determined by immunohistochemistry the results were compared between different groups divided by related clinicopathologic parameters.Results:All patients who had doubtable FISH results, either by absolute HER2 copy number (14 of 71 patients; 19.7%) or by the ratio HER2/chromosome 17 (2 of 71 patients, 2.8%), displayed polysomy 17. Polysomy 17-positive patients had no significant difference with HER2-negative patients in tumor grade, lymph node metastasis, and estrogen receptor (ER) expression (all P>0.05); but compared with HER2-positive patients, they showed lower tumor grade (50.0% vs 81.5%, P=0.025), higher rate of negative lymph node (55.6% vs 25.9%, P=0.045), and higher rate of ER positive expression (83.3% vs 41.7%, P=0.005) and progesterone receptor(PR)positive expression (87.5% vs 44.4%, P=0.003).Conclusion:Compared with HER2 gene amplification group, polysomy 17-positive group tends to have negative HER2 gene expression. Polysomy 17 influences the testing results of HER2 and may be the main factor that caused doubtable results in FISH examination.
8.Effect of Sangge Mixture on Cerebral Circulation and Metabolism in Rabbits

The effect of Sangge Mixture (SM) on cerebral circulation and metabolism in rabbits was observed.The results showed that SM increased the blood flow of brain and cerebral cortex in a dose-dependent manner and had no effect on the blood flow of femoral arteries. Meanwhile, SM markedly increase the oxygen consumption and glucose intake in rabbit brain. It is suggested that SM can improve cerebral circulation, with a certain degree of selectivity.
9.Expression changes of aquaporin during ischemic reperfusion injury to rat kidney

BACKGROUND: It is a hot investigation to many scholars that how to cure and prevent renal ischemic reperfusion injury in a utility way, but the mechanism is unclear at present. The investigation indicates that aquaporiin-1 plays an important role during this process. OBJECTIVE: To research the correlation between aquaporin-1 expression and renal function change following renal ischemic reperfusion injury. DESIGN, TIME AND SETTING: A randomized controlled animal experiment was performed at Histology and Embryology Laboratory of Dalian Medical University from June 2007 to December 2008. MATERIALS: A total of 80 healthy female adult Wister rats were randomly divided into control group and ischemia-reperfusion group. Rats in each group were observed at days 1, 2, 3, 5, and 7 after operation, with 8 rats for each group. The ischemia-reperfusion injury was established on the left kidney. METHODS: Right kidney was removed. The left renal pedicle was freed and occlused to establish ischemia-reperfusion injury model. After 40 minutes, the blood was re-flowed. If the kidney colored from dark red to bright red within 2-5 minutes, the ischemia-reperfusion injury models were successfully established, and the thrombus was not formed in the kidney vessels. If the kidney was still dark red after 5 minutes, the thrombus was formed, and the rats were excluded from the ischemia-reperfusion group. The abdomen was sutured after 40 minutes.MAIN OUTCOME MEASURES: Rats were sacrificed at days 1, 2, 3, 5, and 7 after ischemia-reperfusion injury. Samples of urine, serum, and kidney were performed with the examinations of urine, renal function, renal pathology and morphology, immunohistological assay of aquaporiin-1, and RT-PCR assay. RESULTS: After ischemia-reperfusion injury, the rats had hydrouria, urine osmotic pressure depress, symptoms of carnine and urea nitrogen increasing. HE staining demonstrated that renal tubular epithelial cells were swelling, necrosis, and desquamate. Aquaporin-1 expression and its mRNA level was decreased; in particular, the expression and level were the lowest at day 1 after ischemia-reperfusion injury and recovered to normal value at day 5 after ischemia-reperfusion injury. CONCLUNSION: The down expression of aquaporin-1 maybe one of the important indicators to reflect renal functional changes of acute renal failure following renal ischemia-reperfusion injury.
10.Clinical efficacy of Deanxit in non-erosive gastroesophageal reflux disease with anxiety and depres-sion

Objective To investigate clinical effects of Deanxit in non-erosive gastroesophageal re-flux disease ( NERD) with anxiety and depression.Methods A total of 76 patients diagnosed NERD with anxiety and depression by endoscopy and hospital anxiety and depression scale ( HAD) were randomly divid-ed into regular treatment group and Deanxit treatment group.Clinical efficacy, symptom score, and anxiety and depression score were observed after one week, four weeks, and eight weeks.Results Deanxit signifi-cantly increased the clinical efficacy after one week ( P <0.01) and remission rate after one week ( P <0.05), four and eight weeks ( P <0.01).It improved symptoms, depression, and anxiety, which showed significant differences ( P <0.01) compared to control group after one week, four and eight weeks adminis-tration.Conclusions Deanxit can be applied in NERD with anxiety and depression treatment, which has quick effect and high remission rate, and improve patient's reflux symptoms and anxiety depression.
