Objective To examine the cellular distribution and the role of a-catenin in epidermal tumors. Methods Expression of a-catenin was investigate d by immunohistochemical staining in 20 patients with Bowen′s disease (BD), 20 squamous cell carcinoma (SCC), 20 basal cell carcinoma (BCC), and 40 normal cont rols. Results Expression of a-catenin was strongest on the cell membranes of b asal cells, but nearly negative in the cytoplasm and the nuclei of epidermal cel ls in normal controls. Objective To investigate the expression and role of CD147in the differentiation of nor-mal keratinocytes and squamous cell carcinoma(SCC)cells.Methods CD147expression was analyzed immunohistochemically in20cases of verruca vulgaris(VV),various benign,premalignant and malignant epidermal tumors including21cases of seborrheic keratosis(SK),20actinic keratosis(AK),20Bowen' s disease(BD)and57squamous cell carcinoma.SCCs were classified using Broder's system of grading.Im-munoblot analysis was used to observe CD147expression in normal keratinocyte(HaCaT)and SCC cell(HSC-5)in vitro during their differentiation process induced by calcium.The effects of high-calcium medi-um culture and CD147antibody on the differentiation-related morphology of HaCaT and HSC-5cells were also observed.Results The same staining pattern of CD147was shown in20VV and21SK specimens as in normal epidermis.Positive CD147staining throughout the lesion was shown in4of20AK and7of20BD specimens.Positive CD147staining at the periphery of tumor nests was shown in8of20gradeⅠSCC specimens.CD147expressed throughout tumor nests in16of20gradeⅡSCC and all of the17gradeⅢSCC specimens.Immunoblot analysis revealed decreased CD147expression in both HaCaT and HSC-5cells during differentiation process induced by calcium.Not only high-calcium medium but also CD147antibody could induce differentiation-related morphology of both HaCaT and HSC-5cells in vitro.Conclusion These results suggest that CD147is a novel low-differentiation marker of keratinocyte,which might inhibit the dif-ferentiation of both normal keratinocyte and SCC cell.
3.Optimization of Fermentation Process of Compound Chinese Medicine by Response Surface Methodology

Objective To investigate the factors that influence the fermentation process of compound Chinese medicine and determining the optimum fermentation with single factor experiment and response surface methodology. Methods Through controlling the factors in the fermentation process of compound Chinese medicine (such as fermentation bacteria, fermentation time, fermentation temperature, inoculum amount, etc.), with its increase rate of total peak area as evaluation indicator, the alcohol extracts before and after fermentation were monitored and comparative evaluated by HPLC, and the optimum fermentation process was determined by response surface methodology. Results The fermentation process optimized by single factor experiment and response surface methodology was as follows:SZ-2 strain served as the fermentation bacteria, temperature was 33 ℃, inoculum amount was 4%, and time was 3.5 d, the average increase rate of total peak area was 31.24%. Conclusion HPLC can be used to identify and evaluate the fermentation of compound Chinese medicine under the different factors, and to clarify the optimal fermentation process by response surface methodology, which provide reference for the development of fermentation process.
4.A clinical study on knee osteonecrosis in patients with systemic lupus erythematosus

Objective To investigate the clinical features of osteonecrosis of the knee (ONK) in patients with systemic lupus erythematosus (SLE).Methods A retrospective chart review was conducted to identify patients of SLE with ONK.The clinical characteristics of these patients were analyzed.Results Between 2008 and 2013,eighty-one patients with SLE were analyzed in the 210th Hospital of People's Liberation Army.Eleven of these patients were diagnosed as ONK.All of these ONK patients were female.The age of onset ranged from 19-57 (36±13) years.The disease duration ranged from 4.5-31.0 (13±6) years.The local clinical symptom included knee swelling with pain in 5 cases,only pain with no swelling in 1 patient,no symptoms in 5 patients.All patients were with aseptic necrosis of the femoral head.For laboratory examination,7 patients had elevated triglyceride lipids in blood.Three of 7 patients accompanied with cholesterol level increased.Total cumulative dosage of oral corticosteroids was (66 824±27 328) mg.Conclusion ONK in patients of SLE is common and the majority of them have impaired renal function and elevated blood lipids,as well as long-term glucocortico-steroid therapy.

Objective To investigate the effects of siRNA targeting CD 147 gene on the expression of CD147 in melanoma cell line A375, and on proliferation of these cells. Methods Previously prepared recombinant plasmid pSUPER/CD147 siRNA was used. In this study, the recombinant was transfected into the A375 cells. The mRNA expression of CD147 was measured by semi-quantitive RT-PCR, the proliferation of the cells by MTT assay. BACKGROUND: It is a hot investigation to many scholars that how to cure and prevent renal ischemic reperfusion injury in a utility way, but the mechanism is unclear at present. The investigation indicates that aquaporiin-1 plays an important role during this process. OBJECTIVE: To research the correlation between aquaporin-1 expression and renal function change following renal ischemic reperfusion injury. DESIGN, TIME AND SETTING: A randomized controlled animal experiment was performed at Histology and Embryology Laboratory of Dalian Medical University from June 2007 to December 2008. MATERIALS: A total of 80 healthy female adult Wister rats were randomly divided into control group and ischemia-reperfusion group. Rats in each group were observed at days 1, 2, 3, 5, and 7 after operation, with 8 rats for each group. The ischemia-reperfusion injury was established on the left kidney. METHODS: Right kidney was removed. The left renal pedicle was freed and occlused to establish ischemia-reperfusion injury model. After 40 minutes, the blood was re-flowed. If the kidney colored from dark red to bright red within 2-5 minutes, the ischemia-reperfusion injury models were successfully established, and the thrombus was not formed in the kidney vessels. If the kidney was still dark red after 5 minutes, the thrombus was formed, and the rats were excluded from the ischemia-reperfusion group. The abdomen was sutured after 40 minutes.MAIN OUTCOME MEASURES: Rats were sacrificed at days 1, 2, 3, 5, and 7 after ischemia-reperfusion injury. Samples of urine, serum, and kidney were performed with the examinations of urine, renal function, renal pathology and morphology, immunohistological assay of aquaporiin-1, and RT-PCR assay. RESULTS: After ischemia-reperfusion injury, the rats had hydrouria, urine osmotic pressure depress, symptoms of carnine and urea nitrogen increasing. HE staining demonstrated that renal tubular epithelial cells were swelling, necrosis, and desquamate. Aquaporin-1 expression and its mRNA level was decreased; in particular, the expression and level were the lowest at day 1 after ischemia-reperfusion injury and recovered to normal value at day 5 after ischemia-reperfusion injury. CONCLUNSION: The down expression of aquaporin-1 maybe one of the important indicators to reflect renal functional changes of acute renal failure following renal ischemia-reperfusion injury.
8.Reduction in thyroid dose from neck CT angiography with 64-slice spiral CT with automatic tube current modulation technique

Objective To assess the effectiveness of Z-axis automatic tube current modulation ( ATCM) technique with respect to dose reduction and image noise in the thyroid regional tissues during neck computed tomography angiography (CTA). Methods A total of 140 patients underwent neck CTA with 64-slice spiral CT, and were randomized into fixed-tube current group and ATCM group. Objective noise, CTDIw, DLP and mA of each group were recorded. Results The image qualities of all the patients were diagnostically acceptable, though the objective noise of neck in the ATCM group was higher than that in the group of fixed-tube current (P< 0.05 ). The radiation doses in the ATCM group [ CTDIw (35.99±1.31 ) mGy, DLP (11 121.39 ±5.51) mGy·cm] were significantly inferior to those in the group of fixed tube current [ CTDIw (43.22±1.42) mGy, DLP (1514.45±5.56) mGy·cm]. Conclusions Compared with fixedtube current technique, ATCM technique could significantly reduce the radiation dose in neck CT with diagnostic acceptability of the image quality.
9.Reproducibility of intravoxel incoherent motion in body DWI technique for normal lumbar disc scanning

Objective To investigate the reproducibility of magnetic resonance intravoxel incoherent motion in body diffusionG weighted imaging (IVIMGDWI)for normal lumbar disc scanning.Methods 50 healthy volunteers were enrolled with informed consent,30 males and 20 females,2 5.20±2.04 years old.Using 3.0T MR on the lumbar spine,the sagittal T1 WI,the sagittal,axial T2 WI and sagittal IVIMGDWI sequences were scanned once,then the second sagittal IVIMGDWI sequence was scanned after 4 hours.The IVIMGDWI sequence used 10 b values (0,10,20,40,60,80,100,200,400,600 s/mm2 ).The discs were graded according to the Pfirrmann grading standard.The ADCfast ,ADCslow ,and f values of each intervertebral disc were measured by two doctors at the postGprocessing workstation.Paired tGtest was used to analyze whether there was a difference between the two scans.The intraGgroup correlation coefficient (ICC)was used to analyze the consistency of the ADCfast ,ADCslow ,and f values of the two acquisitions (P<0.05)and the consistency of the ADCfast ,ADCslow,and f values between the different doctors (P<0.05).The IVIMGDWI imaging was evaluated to measure the repeatability of normal lumbar discs. Results Of the 50 healthy volunteers,230 intervertebral discs matched the criteria(Pfirrmann gradeⅠandⅡ).The ADCslowvalue between the two scans was significantly different (t=2.460,P<0.05),and the differences in ADCfast and f values were not significant (t=-0.418,1.273,P>0.05). The consistency of ADCfast ,ADCslow ,and f values for the two scans were generally (ICC=0.478,0.306,0.316,P<0.05 ).Different observers had good consistency in the measurement of interverG tebral disc ADCfast ,ADCslow,and f values (ICC=0.929,0.909, 0.9 1 1 ,P<0.05).Conclusion The IVIMGDWI imaging has good consistency in the measurement of normal lumbar disc between different observers.The consistency of IVIMGDWI in two scans of normal lumbar intervertebral discs is general,which may be due to the time interval between the two scans.Because ADCslow represents the diffusion of water molecules in tissues,the microenvironment in the lumbar intervertebral disc has changed,resulting in the difference of ADCslow value.As for the problem that different scanning time may lead to the change of IVIMGDWI data,we will study it further.
10.The clinical analysis of ultrasound-guided Mammotome resection of breast lumps through the retromammary space

Objective To evaluate the effect of ultrasound-guided vacuum-assisted minimal invasive resection(Mammotome procedure)of breast lumps through the retromammary space. Methods Seven hundred and eighty-seven patients in Obstetrics and Gynecology Hospital Affiliated to Fudan University from Jan. 2011 to May 2012 were underwent ultrasound-guided Mammotome operation through the retromammary space (retromammary space group,385 cases),or adjacent the lumps,and followed by post-operation visits regularly (Mammotome operation adjacent the lumps group,402 cases). The operation effects were compared between the two groups. Results All cases were followed up for 12 months. The period of Mmmotome operation through the retromam mary space and the rate of resection were(48 ± 6)min and(52 ± 4)min,99. 48%(383 / 385), 99. 25%(399 / 402),perspectively,in group of Mammotome operation through the retromammary space and Mammotome operation adjacent lumpsand. There was no significant difference between the two groups( P> 0. 05). The amount of procedural bleeding,the incidence of ecchymosis,local hematoma and the number of incision in group of Mammotome operation through the retromammary space were(8 ± 3)ml,2. 34%(9 / 385), 0. 52%(2 / 385),(1. 3 ± 0. 6)respectively,which were significantly lower than those in Mammotome operation adjacent the lumps group((14 ± 6)ml,8. 71%(35 / 402),2. 74%(11 / 402),(2. 4 ± 0. 3)respectively). There were statistical difference between two groups( P = 0. 003,P < 0. 001,P = 0. 001,P = 0. 006). The rate of satisfaction in group of Mammotome operation through the retro-mammary space was 98. 70%(380 / 385),which is significantly higher than in group of Mammotome operation adjacent the lumps(89. 30%(359 / 402),P< 0. 01). Conclusion The therapy approach of ultrasound-guided Mammotome operation through the retromammary space has lower hemorrhagic complication,as well as the better effect with special advantages. Therefore it has prospective clinical application.
