Objective: To observe the protective effect of ligustrazine on myocardial ischemia rat induced by isoproterenol. Methods : The model of myocardial ischemia was induced by subtcuaneous injection of isoproterenol (ISP, 5mg?kg -1 ) into rats and repeated the same dose 24 hours later. The activities of Ca 2+ -ATPase、Ca 2+ -Mg 2+ -ATPase and the contents of Ca 2+ in the myocardial mitochondria were observed, respectively. The changes of protein expression of bcl-2 and bax genes were detected by immunohistochemical staining. Results : In the ligustrazine treated group, as compared with those of the model, the activities of Ca 2+ -ATPase、Ca 2+ -Mg 2+ -ATPase in the myocardial mitochondria increased significantly( P 0.05). Conclusion : Ligustrazine possesses protective effects against myocardial ischemia injury via increasing the activities of Ca 2+ -ATPase、Ca 2+ -Mg 2+ -ATPase in the myocardial mitochondria and influencing the expression of bcl-2.

Objective To investigate the morbidity rate and reason of ventricular fibrillation (Vf) undergoing select coronary angiography (SCA).Methods To analysis some correlated factors of Vf during SCA. Results 11 cases with Vf undergoing SCA, including 5 cases with double vessel disease, 6 patients with triple vessel lesion. Of 11 cases, 4 patients with left main coronary artery lesion were accompanied by more other branch lesions of coronary artery. Conclusion Vf is not infrequent in patient with multivessel lesion during SCA. It is important to observe coronary artery pressure and electrocardiogram change undergoing SCA.

Objective To investigate the association of plasma metallothionein 3 (MT3) and its polymorphisms with childhood autism,in order to provide the objective evidence for autistic etiology and molecular diagnosis. Methods A total of 132 autistic children were recruited from several special autism training schools in Wuhan and the Hubei Maternal and Child Health Hospital between January 2011 and November 2014.Three hundred and sixteen healthy children from the out-patients of Zhongnan Hospital of Wuhan University during the same period were enrolled as healthy controls.Enzyme linked immunosorbent assay was utilized to measure plasma MT3 protein levels in a dataset of 81 cases and 80 controls, while eight single nucleotide polymorphisms (SNP) located in MT3 gene were genotyped in another greater dataset that included 132 cases and 236 controls by the matrix-assisted laser desorption/ionization time of flight mass spectrometry within the Sequenom platform. Results Plasma MT3 protein level was significantly lower in autistic group compared to healthy controls [(740.0 ± 327.4) ng/L vs (1 007.1 ± 554.3) ng/L,P < 0.001],particularly for boys when stratified by gender (P =0.005).No difference existed in any allele or genotype frequencies between the 2 groups (all P > 0.05).Conclusions The selected autistic children harbored abnormal expression profiles of plasma MT3 protein, which may have no connection with its gene polymorphisms.

AIM: To select the prescription of harmine hydrochloride cream and to study the anti-itching effect of harmine hydrochloride cream. METHODS: A comprehensive score system, including appearance, homogeneity, coating performance, viscosity, particle diameter,temperature-resistant test, and centrifugal effect were established as evaluative criteria. Then uniform design was used for selecting the prescription of harmine hydrochloride cream. The histamine phosphate and 5-hydroxytryptamine (5-HT) were used to induce mouse cutaneous pruritus. Then the anti-pruritic effect of harmine hydrochloride cream in contrast to Piyanping Ointment as control group. RESULTS: The optimized prescription was as follows: 8% of monostearin,5% of Vaseline,6. 1% of beeswax, 1. 1% of Span-60,1. 9% of Tween-80. The local application of harmine hydrochloride cream could alleviate the itching induced by histamine phosphate and 5-HT,respectively. And the anti-pruritic effect of harmine hydrochloride cream was stronger than the effect of the positive control group. CONCLUSION: The prescription of the harmine hydrochloride cream is available, reasonable, and reproducible. The harmine hydrochloride cream has the anti-pruritic effect.

Chinese Journal of Microbiology and Immunology 2008;28(3):198-202
Objective To knock out the entire LuxS gene of Streptococcus mutans UA159 strain via homologous recombination and construct a LuxS-deleted mutant strain of S.mutans.Methods The erythromycin resistance gene(Eymr)was inserted between the two DNA fragments located in the upper and downstream of LuxS gene that had been amplified by PCR.Then the two DNA fragments along with the inserted Eymr were engineered into pUCl9 plasmid to construct the recombination plasmid pUCluxKO.Electrotransformation of S. mutans cells with pUCluxKO-mutant resulted in the isolation of erythromycin resistant S.mutans, transformants, which was then subjected to polymerase chain reaction,Vibrio harveyi BBl70 luminescence bioassay and sequencing analysis. Results Restriction endonuclease analysis showed that pUCluxKOmutant vector had been successfully recombined. The deletion of LuxS of S. mutans mutants was confirmed bv PCR with primers specific for the genes of LuxS and the erythromycin resistance.S.mutans mutant could not induce bioluminescence.indicating the mutant had been successfully recombined. The constructed Chinese S.mutans showed good stability after 20 generations of cultivation. Conclusion The S.mutans gene allelic exchange plasmid is constructed correctively and a LuxS-negative mutant of S.mutans has been constructed.which can be helpful for further study of the role of LuxS in the pathogenesis of S.mutans.
