Journal of Medical Postgraduates 2003;0(03):-
Objective: To study the effects of glycosaminoglycan from scallop skirt (SS-GAG) on the formation of foam cells from porcine artery smooth muscle cells (SMCs) and the expression of the total superoxide dismutase(SOD)and glutathione peroxidase(GSH-PX)activity, NO production and the mechanism of anti-atherosclerosis action of SS-GAG. Methods: SMCs were incubated with 15 mg/L oxidized low-density lipoprotein (Ox-LDL) for 72 h to establish a smooth muscle cell-derived foam cell model. In addition, SMC cells were divided into 6 groups:①blank group,②model(Ox-LDL) group, ③Ox-LDL+200 mg/L SS-GAG group,④Ox-LDL+400 mg/L SS-GAG group,⑤Ox-LDL+800 mg/L SS-GAG group, ⑥Ox-LDL+Heparin 100 mg/L group. After 72 h incubation, intracellular total cholesterol (TC), free cholesterol (FC), cholesteryl ester (CE) content and CE/TC ratio were measured through enzymatic method. The SOD, GSH-PX and NO concentration in the medium were also determined through Xanthine oxidase method or TBARs. Results: TC, CE, CE/TC in model group significantly increased, while FC, GSH-PX and NO concentration in the medium significantly decreased compared with blank group. After treatment with heparin (100 mg/L) and different concentrations of SS-GAG (200 mg/L, 400 mg/L,800 mg/L), TC, CE, and CE/TC significantly decreased (P

The specific anti-rat LH in the peroxidase anti-peroxidase (PAP) immunohistochemical method was used to demonstrate the pituitary gonadotropic cells in the pregnant rats, and then the morphometric analysis method was used to study the change of the number, size and nucleo-cytoplasmic ratio parameters of the gonadotropic cells in pregnant rats after being injected by a high dose of LHRH-A.It was observed that on 24h after injection the number of gonadotropic cells decreased, their size became smaller and their nucleo-cytoplasmic ratio larger: the cell number per mm~3 pituitary tissue was from (4.25?0.21)?10~4 to (2.49?0.14)?10~4; the cell size from 1295?35?m~3 to 895?31?m~3; the nucleo-cytoplasmic ratio from 0.144?0.005 to 0.229?0.010. On 48h after injection these changes were in the process of recovery. On 168h after injection all parameters were recovered. Our observations suggest that after being injected by a high dose of LHRH-A the change of gonadotropic cells in pregnant rats results from the exogenous high dose of LHRH-A which has stimulated gonadotropic cells to release a great amount of LH. It is believed, however, that the changes of gonadotropic cells caused by a high dose of LHRH-A can still return to normal.

OBJECTIVE:To discuss the drug resistance situation and influential factors of sputum culture positive tuberculosis patients in a specialized subject hospital. METHODS:The information of sputum culture positive tuberculosis inpatients were col-lected from Heze infectious disease hospital during Jan. 1st 2014 to Apr. 30th 2015;the results of drug sensitive test were analyzed retrospectively by using absolute concentration method. RESULTS:Among 259 patients,137 cases were drug-resistant(52.90%), among including 69(26.64%) mono-drug resistant cases,52(20.08%) poly-drug resistant cases,15(5.79%)multi-drug resistant cases and 1(0.39%)extensively drug resistant case;streptomycin (31.66%) and amikacin (15.44%) were drugs with the highest drug-resistant rate;there was no statistical significance in resistant rate between different genders and ages(P>0.05),the rate of drug resistance in first-treatment patients was 47.21%,and that of the re-treatment patients was 70.97%,the difference was statisti-cal significant(P<0.05). In addition to ethambutol and kanamycin, other all had mono-drug resistance, there were 27 combinations of poly-drug resistance and 9 combinations of multi-drug resistance. Logistic regression analysis showed that treatment history was a risk factor for drug-resistance,the risk of re-treatment patient was 2.734 times of first-treatment patients. CONCLUSIONS:Sputum culture positive tuberculosis inpatients patients have high drug resistance and complex spectrum in the hospital;we should pay great attention to the risk of drug resistance in senile patients and re-treatment patients, and rational use of streptomycin,amikacin,isoni-azid and capreomycin.
