1.Effects of glycosaminoglycan from scallop skirt on foam cell formation and its function

Objective: To study the effects of glycosaminoglycan from scallop skirt (SS-GAG) on the formation of foam cells from porcine artery smooth muscle cells (SMCs) and the expression of the total superoxide dismutase(SOD)and glutathione peroxidase(GSH-PX)activity, NO production and the mechanism of anti-atherosclerosis action of SS-GAG. Methods: SMCs were incubated with 15 mg/L oxidized low-density lipoprotein (Ox-LDL) for 72 h to establish a smooth muscle cell-derived foam cell model. In addition, SMC cells were divided into 6 groups:①blank group,②model(Ox-LDL) group, ③Ox-LDL+200 mg/L SS-GAG group,④Ox-LDL+400 mg/L SS-GAG group,⑤Ox-LDL+800 mg/L SS-GAG group, ⑥Ox-LDL+Heparin 100 mg/L group. After 72 h incubation, intracellular total cholesterol (TC), free cholesterol (FC), cholesteryl ester (CE) content and CE/TC ratio were measured through enzymatic method. The SOD, GSH-PX and NO concentration in the medium were also determined through Xanthine oxidase method or TBARs. Results: TC, CE, CE/TC in model group significantly increased, while FC, GSH-PX and NO concentration in the medium significantly decreased compared with blank group. 2.Retrospective analysis on 56 ischemic colitis

Objective To analyze the clinical features,endoscopic characteristics and methods for diagnosis and treatment of ischemic colitis.Methods The clinical data of 56 patients with ischemic colitis were retrospectively analyzed.Results Ischemic colitis occurred more often in elderly patients.Male and female ratio was 1 ∶1.8.The patients often had concomitant basic diseases,such as diabetes mellitus (53.6%,30/56),hypertension (39.3%,22/56),coronary heart disease (32.1%,18/56).The main manifestations of ischemic colitis were abdominal pain (100.0%,56/56) and bloody stools (89.3%,50/56).The lesions revealed by colonoscopy were mainly located in the left colon and distributed in a segmental pattern,and the pathological changes included mucosal edema,congestion,erosion and ulceration.The prognosis was good in most cases.Conclusions Ischemic colitis should be suspected in elderly patients with acute abdominal pain and bloody stools.Early colonoscopy is helpful for accurate diagnosis and appropriate treatment of ischemic colitis.
3.Resection Arthroplasty of the Sternoclavicular Joint to Treat Dislocation of the Sternoclavicular Joint

To investigate the possibility of resection arthroplasty of the sternoclavicular joint in treatment of dislocation of this joint.Methods:Five patients who had had resection arthroplasty of the sternoclavicular joint and in whom the costoclavicular ligament had to be reconstructed,follow-up averaged 1.8 years.Results:The average duration of follow-up were 1.8 years.All of the patients had an excellent result,no cases had infection、pain and abnormalities.Conclusion:We believe that resection arthroplasty of the sternoclavicular joint and preservation reconstruction of the costoclavicular joint is essential,and it is a satisfactory and safe way for the treatment of dislocation of the sternoclavicular joint.
4.A study of correlation between insulin resistance and the clinical pathology characters in chronic hepatitis B patients

Objective To investigate the relationship between insulin resistance index (HOMA-IR) and the clinical pathology fea-tures in chronic hepatitis B(CHB) patients. Methods ALT and hepatitis B virus loading level and HOMA-IR were measured in 80 CHB pa-tients. Correlation analysis was performed with pathological features of liver tissue. Result Insulin resistance(IR) was much more obvious in patients with severer degree of liver inflammation and fibrosis . HOMA-IR and the insulin level of CHB patients in G3 grade(15.24±7.41,3.51 ± 1.91) and S3 stage (16.65±6. 46,3.79 ± 1.50) were higher than those in G2 grade (10.33 ± 6.28,2.25 ± 1.39) and S2 stage(9.06 ± 4.61,2.02 ± 1.11). The HOMA-IR was positive correlated with the level of age and ALT. Conclusion The development of IR in CHB patients was correlated with age and the degree of liver inflammation and fibrosis.
5.Construction of LuxS gene deletion of Streptococcus mutans and evaluation of the acid tolerance of the mutant

Objective To construct Streptococcus mutans UA159 mutants with deletion of LuxS gene related to quorum-sensing pathway and evaluate the aciduricity of the mutants. Methods Using S. mutans UA159 as materials, the PCR fragments of the upstream and downstream regions of LuxS and erythromycin resistance(Eymr) gene of PJT10 were cloned into plasmid PUC19. The resulting constructs were integrated into the chromosome of S. mutans. LuxS gene deletion mutant was then constructed in S. mutans by means of allelic exchange and selected for resistance to erythromycin. The aciduric ability of the mutant under different pH was measured and S. mutans UA159 was used as control. Results The LuxS-deleted status of S. mutans mutants were confirmed by various PCR and DNA sequencing. The results showed that Eymr gene take the place of LuxS gene, while the mutant can not induce bioluminescenece. The LuxS mutant strain displayed a decreased growth ability with the decreasing pH values compared to those of the wild-type strain UA159. Conclusion A LuxS-negative mutants of S. mutans is constructed. The LuxS quorum sensing system is involved in the regulation of aciduricity of S. mutans UA159.

Objective:To observe the expression of PCNA and morphological changes of submandibular gland of db/db mutant diabetic mice. Methods:Fifty C57BL/Ksj-db/+m mice were randomly divided into 2 groups, db/db diabetic group and control group. The morphological changes of submandibular gland and the expression of PCNA were observed by using light microscope (LM) and immunohistochemistry. Results:Submandibular gland atrophy and the significant morphological changes in acini and ducts were observed in db/db diabetic mice. 7.Antitumor effects of CIK cells against hepatocellular carcinoma cells in vitro and in vivo

Purpose:To evaluate the antitumor effects of human cytokine-induced killer (CIK) cells from healthy donors and patients with primary hepatocellular carcinoma (HCC) in vitro and in vivo .Methods:Peripheral blood mononuclear cells (PBMC) from healthy donors and patients were induced to become CIK cells by in vitro incubation. The CIK cells were identified at different intervals by flow cytometry analysis. The cytotoxicity of CIK cells against autologous primary HCC was determined by 51Cr Release Assays. The antitumor activity of the CIK, LAK and PBMC cells were evaluated in Balb/c nude mice bearing BEL-7402liver cancer.Results:The flow cytometry analysis showed that CIK cells from patients with HCC proliferated and expanded more than 300-fold in vitro incubation by day 28;the percentage of CD3+CD56+ cells increased from 0.23% on day 0 to 17.8% on day 21. CIK cells generated from patients with HCC patients possessed a higher antitumor cytotoxic activity on autologous HCC cells in vitro than autologous PBMC. 8.DETECTION OF HIV CORECEPTOR CCR5 MUTATION IN UIGUR POPULATION FROM SINKIANG UIGUR AUTONOMOUS REGION

Whole blood samples were collected from 316 Uigur subjects inhabiting Kashi City, Sinkiang Uigur Autonomous Region, and their genomic DNA was extracted using QIAgen Blood Kit. DNA fragment spanning CCR5?32 and CCR5m303 mutation points were amplified by PCR and genotyped by directed electrophoresis and RFLP with HinCII respectively. In all 316 samples only 22 heterozygotes of CCR5?32 were found, in accordance with Hardy Weinberg equilirium. No CCR5m303 mutation was found. No difference existed between male and female individuals. The frequency of CCR5?32 in Uigur population is 3.48%, similar to Medi Asia ethnic groups (higher than that of Chinese Han people and lower than that of Caucasian people), i.e, in continuity regarding district distribution. Due to its slow down effect on clinical course of AIDS , high frequency of CCR5?32 is highly significant in prevention and treatment of AIDS in Uigur population.

Objective To identify the property and implications of dendritic cells of peripheral blood monocytes (PBMC) in patients with persistent HBV infection. Methods The PBMC from 18 patients with persistent HBV infection and 10 healthy subjects were incubated and induced into mature dendritic cells (DC) in the completed medium containing GM CSF、IL 4、FLt3 L and TNF ? cytokines. The flow cytometric analysis was employed to detect the expression of surface markers on DC. ELISA test was used to determine the level of interleukin 12 (IL 12) and IL 10 cytokines produced by DCs. The stimulatory capacity of DC were also determined in allogenic mixed leukocyte reaction (MLR). Results A typical morphology of DCs was observed when the PBMCs from healthy subjects and HBV infected patients were induced for 6 7 day in vitro incubation, but there were some significant differences of DCs as follows: (1) the proliferation ability and number of DCs significantly decreased, in particular, the expression levels of HLA DR,CD80(B7 1),CD86(B7 2) and CD1? on DC surface were simultaneously much lower in patients compared to healthy subjects ( P

It was reported that 249I 280M haplotype of HIV coreceptor CX 3 CR1 was associated with accelerated AIDS progression. In this study, genomic DNA of 119 Uigur subjects (98 normal, 21 HIV 1 infected) was purified from PBMC, and 249I and 280M allelic frequencies were identified by PCR RFLP. All data were tested by ? 2 analysis.It was found that allelic frequency of 249I and 280M was 15 8% and 13 8%, respectively, and there was no difference between normal and HIV infected groups. Both population and sex distribution were in accordance with Hardy Weinberg equilibrium. Frequency of 249I280M haplotype was 13 8%, close to that of Caucasian people.
