2.CHARACTERISTICS AND CLINICAL IMPLICATION OF HIV-1 CORECEPTOR CX_3CR1 ALLELIC POLYMORPHISM IN UIGUR ETHNIC GROUP FROM SINKIANG UIGUR AUTONOMOUS REGION

It was reported that 249I 280M haplotype of HIV coreceptor CX 3 CR1 was associated with accelerated AIDS progression. In this study, genomic DNA of 119 Uigur subjects (98 normal, 21 HIV 1 infected) was purified from PBMC, and 249I and 280M allelic frequencies were identified by PCR RFLP. All data were tested by ? 2 analysis.It was found that allelic frequency of 249I and 280M was 15 8% and 13 8%, respectively, and there was no difference between normal and HIV infected groups. Both population and sex distribution were in accordance with Hardy Weinberg equilibrium. Frequency of 249I280M haplotype was 13 8%, close to that of Caucasian people. No linkage was found between CX 3 CR1 and CCR5?32, but strong linkage existed between 249I and 280M ( P
3.Characterization of variants in the RANTES gene in Han and Uiygur Chinese population and its implication in HIV-1 infection

Objective To detect the single nucleotide polymorphism(SNP) of chemokine RANTES promoter of healthy and HIV 1 infected individuals of Han and Uiygur Chinese population, and to evaluate the correlation of their SNPs with HIV 1 infection. Methods Case control study was adopted ,Genotypies of RANTES promoter 403 and 28 from 863 samples were detected by DNA sequencing or by polymerase chain reaction restriction fragment length polymorphism. Results There were two SNPs and six genotypes in Chinese Han and Uiygur. 403 and 28 allelic frequencies were high, but there was no significant difference between the two healthy populations. Strong linkage disequilibrium was observed between the two SNPs. RANTES genotypes were AC/AC, AC/AG, AC/GC, AG/GC, GC/GC, and AG/AG. The haplotypes were GC 62 7% in both rases, AC 28 7% and 30 4%, AG 8 6% and 6 8% in Han and Uiygur, respectively. Compared with AC/AC, odd ratio (OR) of RANTES genotypes AC/AG, AC/GC, AG/GC, and GC/GC was associated with reduced susceptibility to HIV 1 infection. Conclusion Same high RANTES promoter mutation was found in Han and Uiygur populations. Compared with AC/AC, odd ratio (OR) of RANTES genotypes AC/AG, AC/GC, AG/GC 9.9%, and GC/GC might be associated with reduced susceptibility to HIV 1 infection. 4.DETECTION OF HIV CORECEPTOR CCR5 MUTATION IN UIGUR POPULATION FROM SINKIANG UIGUR AUTONOMOUS REGION

Whole blood samples were collected from 316 Uigur subjects inhabiting Kashi City, Sinkiang Uigur Autonomous Region, and their genomic DNA was extracted using QIAgen Blood Kit. DNA fragment spanning CCR5?32 and CCR5m303 mutation points were amplified by PCR and genotyped by directed electrophoresis and RFLP with HinCII respectively. In all 316 samples only 22 heterozygotes of CCR5?32 were found, in accordance with Hardy Weinberg equilirium. No CCR5m303 mutation was found. No difference existed between male and female individuals. The frequency of CCR5?32 in Uigur population is 3.48%, similar to Medi Asia ethnic groups (higher than that of Chinese Han people and lower than that of Caucasian people), i.e, in continuity regarding district distribution. Due to its slow down effect on clinical course of AIDS , high frequency of CCR5?32 is highly significant in prevention and treatment of AIDS in Uigur population.
5.Expression of bmp3,bmp4 and bmp7 during dog permanent tooth root formation

Objective:To evaluate the physiological roles of bmp3,bmp4 and bmp7 during the formation of permanent tooth roots in dog. Methods:The expression of bmp3,bmp4 and bmp7 mRNA at different stages of the development of permanent tooth roots was examined by in situ hybridization in 3 dogs aged 12-18 weeks. Results:bmp3 was found in dental sac surrounding the germs at the early stage of tooth root development, and in cementoblasts and periodontal cells at the later stage. bmp4 was found in odontoblasts, dental papilla and osteoblasts. bmp7 positive signals was found only in epithelial cells of root sheath around cervical circulus at early stage, then located in cementoblasts and odontoblasts at later stage. Conclusion:The spatiotemporal expressions of bmp3,bmp4 and bmp7 are widely diverse, indicating that they participate in the regulation of tooth root development.

Object To explore the beneficial effect of phytoecdysone (EDS) on myocardial infarction and its mechanism of action Methods Rat myocardial infarction model was prepared by ligating the left anterior descending coronary artery, and EDS was injected ip for seven consecutive days Serum creatine phosphokinase (CPK) glutamic oxalacetic transaminase (GOT), lactic dehydrogenase (LDH) activities, infarct size(IS), coronary blood flow, capillary vessel density and vascular endothelial growth factor (VEGF) expression were determined Results 0 5, 5, and 50 mg/kg of phytoecdysone were able to effect the activities of serum CPK, GOT, LDH in a dose depending manner with an optimal effect for improving cardiac zymogram at the dose of 5 mg/kg ip At this dosage EDS can markedly reduce IS, increase coronary blood flow, capillary vessel density and the expression of VEGF Conclusion ESD can alleviate myocardial infarction symptoms The mechanism of such beneficial effect may due to its ability to promote VEGF expression regeneration of capillary vessels and increase coronary blood flow

8.Effect of carbon monoxide on permeability of brain blood barrier in cerebral local ischemia rats

AIM: To evaluate the effect of carbon monoxide(CO) on the permeability of brain blood barrier(BBB) in cerebral ischemic rats. METHODS: SD rats were divided into three groups. Saline, hemin or ZnPP were injected intraperitoneally 12 h before middle cerebral artery occlusion (MCAO), respectively. The concentration of blood CO and the permeability of BBB at 24 h after MCAO were measured. RESULTS: The CO concentration in blood in hemin group was higher than that in saline group( P 0.05). CONCLUSION: CO reduced the permeability of BBB as a messenger gas molecular when its intrinsic concentration was elevated.
9.Effects of Wallerian degeneration on biological characteristics and secretory function of Schwann cells in rats with sciatic nerve injury

BACKGROUND:Studies have shown that Schwann cells form a Bunger band in the basement tube and guide the extension of regenerating axons after peripheral nerve injury, but the exact mechanism remains to be explored.
OBJECTIVE:To explore the effect of Wal erian degeneration on biological characteristics and secretory function of Schwann cells in rats with sciatic nerve injury.
METHODS:A rat model of sciatic nerve injury was established and divided into two groups:sciatic nerve transection group and surgical control group. Schwann cells were isolated and cultured from sciatic nerve segments by one enzyme digestion. The cellmorphology was observed under light microscope and S-100 protein expression was determined by immunofluorescence staining. After subculture, the first generation of Schwann cells were chosen to draw the growth curve by the counting method within 14 days. The cellactivity was detected by MTT assay. The adhesion of Schwann cells was examined by acid phosphatase analysis and the concentration of nerve growth factor was detected by ELISA method.
RESULTS AND CONCLUSION:At 14 days after primary culture, a great number of Schwann cells were observed near the edges of nerve segments in the sciatic nerve transection group, but only smal number of Schwann cells scattered around nerve segments in the control group. Schwann cells in both groups showed S-100 positive expression. At 3 days after subculture, Schwann cells reached the logarithm proliferative phase, the cellnumber and proliferation absorbance values in both groups were increased along with time extension. Furthermore, the number of Schwann cells and absorbance value in the sciatic nerve transection group were significantly higher than those of control group (P<0.05). The adhesion ability in the sciatic nerve transection group was also significantly higher than those in the control group (P<0.05). ELISA results showed that, the concentrations of nerve growth factor in the sciatic nerve transection group were significantly higher than those in the control group at 4, 6, 8, 10, 12 and 14 days (P<0.05). After sciatic nerve injury, Wal erian degeneration can induce Schwann cells dedifferentiate into the precursors, significantly influence the biological function of Schwann cells, promote the proliferation of Schwann cells within the short term, secrete large amounts of neurotrophic factors, enhance celladhesion, and provide a suitable microenvironment for regenerated axons. In addition, it creates the necessary microenvironment for peripheral nerve regeneration.
10.Expressions of Transforming Growth Factor β2 and Smad3 in Human Gliomas with Different Pathologic Grade Proliferation and Its Clinical Significance

Objective To investigate the expressions of Transforming Growth Factorβ2 (TGF-β2) and Smad3 in human gliomas associated with pathologic grading. Methods The expressions of TGFβ2 and Smad3 protein were detected with SP immunhistochemistry in 80 human glioma specimens. The Kapan-Meier survival curves of progression-free survival time and overall survival time in different expression levels was compared with log-rank. Results The expression of TGFβ2 and Smad3 correlated with the pathological grading (r=0.545, r=0.570, P<0.01). Both progression-free survival time and overall survival time were significantly different between low expression group and high expression group (P<0.05). Conclusion Both TGF-β2 and Smad3 correlate well with the occurrence and differentiation of human gliomas,which help for the diagnosis, treatment and prognosis judgment.
