1.Simultaneous Determination of 4 Anthraquinones in Shugan Quzhi Capsules by RP-HPLC

Objective To establish a RP-HPLC method for simultaneous quantitative determination of 4 anthraquinones in Shugan Quzhi capsules. Methods A RP-HPLC method was used with a Diamonsil C18 column (5 ?m,4.6 mm?150 mm). The mobile phase was composed of methanol and 0.1% H3PO4 water solution (77∶23,v/v). The wavelength of UV detector was 254 nm and column temperature was 30 ℃. Flow rate was 1.0 mL/min and 20 ?L was injected every time. Results The linear ranges of rhein,emodin,chrysophanol and physcion were 0.083 2~2.08,0.100 8~2.52,0.291 2~7.28 and 0.088~2.20 ?g/mL,respectively. Their average recoveries were 97.3%,96.9%,96.5% and 95.9%,respectively. Conclusion This method was sensitive,repeatable and suitable to determine the contents of 4 anthraquinones in Shugan Quzhi capsules.
2.Neuron-specific enolase and high sensitivity C-reactive protein on neonatal hypoxic-ischemic encephalopathy's condition and prognosis

Objective To investigate the serum neuron-specific enolase (NSE) and high sensitivity C-reactive protein (hs-CRP) on neonatal hypoxic-ischemic encephalopathy (HIE) in children with illness and prognosis of assessed value. Method The serum NSE, hs-CRP levels of 40 patients with HIE (HIE group) of the acute stage and convalescence, and 20 healthy neonates (control group ) were measured and the clinical sub-degree relationships were analyzed. Results The serum NSE, hs-CRP levels in HIE group of the acute stage [ (32.88 ± 12.61 ) μ g/L, (6.43 ± 2.07) mg/L] were significantly higher than those in control group [(8.62 ± 3.58) μ g/L, (2.61 ± 0.95) mg/L](P < 0.01 ). The serum NSE, hs-CRP levels in HIE group of the severe acute stage were significantly higher than those of the mild and light acute stage (P<0.01 ). The more severe disease, the more higher NSE, hs-CRP. Conclusion The serum NSE, hs-CRP reflect the brain neuronal damage or necrosis of the objective indicators that could be used as early objective indicators to judge the pathogenetic condition and prognosis of HIE.
3.Expression of platelet membrane glycoprotein Ib/IX/V complex Sample analysis between type 2 diabetes mellitus patients and healthy people

BACKGROUND: It has been proved that platelet activation is involved in the development of diabetic angiopathy. Glycoprotein (GP) Ib/IX/V complex is one of the main platelet membrane glycoproteins, and the receptor of both von Willebrand Factor and thrombin. It plays a key role in the process of platelet activation.OBJECTIVE: To observe the expression changes of GP Ib/IX/V complex and its component GP Ibα in patients with type 2 diabetes mellitus. DESIGN: Case-control study. SETTING: Department of Integrated Chinese and Western Medicine, Union Hospital, Tongji Medical College, Huazhong University of Science and Technology.PARTICIPANTS: A total of 51 type 2 diabetic outpatients who visited Union Hospital were enrolled from December 2005 to January 2007. The diagnosis was based on the independent criteria from WHO in 1999. Of all the 51 patients, there were 23 females and 28 males, with a peripheral platelet count of over 50×109 L-1. All the subjects had no history of administrating drugs two weeks before the examination, which would potentially influence platelet count. According to disease controlling condition, the patients were assigned to well controlled patient (WCP) group (n = 25) and poorly controlled patient (PCP) group (n =26); and according to whether angiopathy was accompanied, diabetic patients were divided into vascular disease (VD) group (n =27) and non-vascular disease (NVD) group (n =24). Meanwhile 23 healthy subjects were enrolled as normal control group. Informed consents were obtained from subjects and their relatives. The experiments were approved by the ethical committee of Union Hospital.METHODS: Fasting venous blood was harvested from all the subjects' elbow on the early morning of visit day.①Biochemical analysis: Fasting plasma glucose (FPG), glycosylated hemoglobin (HbA1c) and fasting plasma insulin (FINS) was measured by the Clinical Chemistry Department in Union Hospital.② Measurement of platelet membrane GP Ib/IX/V complex and its component GP Ibα: First, 3 mL cubital fasting blood was drawn from each subject and was anti-coagulated with 38 g/L natrium citricum. After that, all samples were fixed with 10 g/L paraform for 45 minutes. Then 50 μL well-fixed blood was added into the polystyrene tube, meanwhile 20 μL monoclonal antibody, such as CD42a-b-c-d and PE-labeled CD42b, was respectively mixed gently with the blood sample and incubated at room temperature in dark for 30 minutes. Next, 20 μL FITC-labeled rat IgG was mixed with the sample containing CD42a-b-c-d and incubated equally. In the end all blood samples were analyzed by FACS420 flow cytometry and the results were expressed as mean fluorescence intensity (MFI). ③ Platelet maximun aggregation rate (MAR) was detected according to reference.MAIN OUTCOME MEASURES: ①Biochemical indicators;②The expressions of GP Ib/IX/V complex and GP Ibα;③Platelet MAR.RESULTS: Fifty-one patients with type 2 diabetes mellitus and twenty-three healthy subjects were all involved in the result analysis.①There were significant differences in FINS in WCP group and PCP group compared with normal controls (P < 0.01). FPG and HbA1c were significantly higher in PCP group compared with normal control group and WCP group (P < 0.01).②Expressions of GP Ib/IX/V complex and GP Ibα were significantly lower in WCP group and PCP group compared with normal control group (P < 0.01), significantly lower in PCP group than in WCP group (P < 0.05), and also significantly lower in VD group and NVD group compared with normal control group (P < 0.01). Moreover the expression of GP Ibα in VD group and NVD group was significantly lower than that of normal control group (P < 0.05), and it also significantly decreased in VD group compared with NVD group (P < 0.05). MFI of GP Ib/IX/V complex had an obvious negative correlation with FBG, HbA1c and FINS (r =-0.634, -0.573, -0.649, P < 0.05), and GP Ibα MFI was obviously negatively correlated with FBG and HbA1c (r =-0.602, -0.543, P < 0.05).③Platelet MAR of diabetic patients were remarkably higher than in healthy subjects (t =-3.852, P < 0.01). Platelet MAR in PCP and VD groups were respectively higher than those in WCP and NVD groups (P < 0.05). CONCLUSION: Platelet activation exists in the early stage of type 2 diabetes mellitus without diabetic angiopathy, and is more obvious after diabetic angiopathy. There is a positive correlation between platelet activation and blood glucose. As a receptor of thrombin and von Willebrand Factor, GP Ib/IX/V complex may be involved in the development of diabetic angiopathy.
4.Effect of quality control circle on reducing incidence of fecal incontinence dermatitis of intensive care unit patients

Objective To investigate the effect of quality control circle on reducing incidence of fecal incontinence dermatitis of intensive care unit (ICU) patients. Methods Quality control circle team was set up and theme was determined then retrospective analysis was carried out for the occurrence of defect in patients with fecal incontinence dermatitis from January to May 2014 before quality control circle activities. Occurrence reasons were explored and corrective measures were formulated and implemented.After the implementation of quality control circle,the occurrence of defect from June to October 2014 was observed. Results After the implementation of quality control circle,the occurrence of fecal incontinence dermatitis reduced from 24.5%(24/98) to 10.4% (10/96) and the difference was statistically significant,χ2=6.65, P<0.05. Conclusion Quality control circle is effective for reducing the occurrence of ICU fecal incontinence dermatitis and continual improvement of nursing quality.
5.Simultaneous Determination of Matrine and Oxymatrine in Human Plasma by HPLC

OBJECTIVE:To established a method for determining matrine and oxymatrine in human plasma.METHODS:The plasma was extracted with chloroform-n-butyl alcohol(98∶2) after basification and purified with neutral alumina solid-phase extraction.Then it was eluted with a Lichrosorb-NH2 column and CH3CN-CH3CH2OH-H3PO4(80∶10∶8),detected at ? 220nm.RESULTS:The linear range was 1.25mg~40mg/L and the limit of detection was 0.1mg/L(S/N=2).The average recoveries of matrine and oxymatrine were 106.96% and 105.04%,respectively.RSDs of within-day and between-day were lower than 13% and 7% respectively.CONCLUSION:The present study provides a simple and reliable method for determining concentrations of matrine and oxymatrine in human plasma.
6.Therapeutic effect of adenovirus-mediated apoptin gene transfer combined with ADM and CDDP on hepatocellular carcinoma in mice.

OBJECTIVETo study the therapeutic effect adenovirus-mediated apoptin gene transfer combined with ADM and CDDP on hepatocellular carcinoma in mice.
METHODSIn c57BL/ 6 mice bearing hepatocellular carcinoma, the changes of tumor volume, histomorphology, tumor inhibition rate and the side effects were observed after intratumoral injection of adenovirus containing apoptin gene and ADM and CDDP.
RESULTSSeven days after the treatment, the mean volume of the tumor in the mice receiving intratumoral apoptin-containing adenovirus injection combined with ADM and CDDP reduced significantly as compared with that in mice treated with adenovirus vehicle and control group. The tumor inhibition rate in the combined treatment group was 90.13%, significantly higher than that in the control group. No adverse effect of the treat was observed in the course of the experiment.
CONCLUSIONThe adenovirus vectors containing apoptin gene combined with ADM and CDDP may serve a safe treatment of hepatocellular carcinoma.

7.Prenatal diagnosis for a fetus with Walker-Warburg syndrome.

OBJECTIVE:
To explore the genetic etiology for a fetus with Walker-Warburg syndrome(WWS).
METHODS:
A fetus with WWS diagnosed at Gansu Provincial Maternity and Child Health Care Hospital in June 9, 2021 was selected as the study subject. Genomic DNA was extracted from amniotic fluid sample of the fetus and peripheral blood samples from its parents. Trio-Whole exome sequencing (trio-WES) was carried out. Candidate variants were verified by Sanger sequencing.
RESULTS:
The fetus was found to harbor compound heterozygous variants of the POMT2 gene, namely c.471delC (p.F158Lfs*42) and c.1975C>T (p.R659W), which were respectively inherited from its father and mother. Based on the guidelines from the American College of Medical Genetics and Genomics (ACMG), they were respectively rated as pathogenic (PVS1+PM2_Supporting+PP4) and likely pathogenic (PM2_Supporting+PM3+PP3_Moderate+PP4).
CONCLUSION
Trio-WES may be used for the prenatal diagnosis of WWS. The compound heterozygous variants of the POMT2 gene probably underlay the disorder in this fetus. Above finding has expanded the mutational spectrum of the POMT2 gene and enabled definite diagnosis and genetic counseling for the family.

The fall armyworm (FAW), Spodoptera frugiperda, is a destructive pest native to America and has recently become an invasive insect pest in China. Because of its rapid spread and great risks in China, understanding of FAW genetic background and pesticide resistance is urgent and essential to develop effective management strategies. Here, we assembled a chromosome-level genome of a male FAW (SFynMstLFR) and compared re-sequencing results of the populations from America, Africa, and China. Strain identification of 163 individuals collected from America, Africa and China showed that both C and R strains were found in the American populations, while only C strain was found in the Chinese and African populations. Moreover, population genomics analysis showed that populations from Africa and China have close relationship with significantly genetic differentiation from American populations. Taken together, FAWs invaded into China were most likely originated from Africa. Comparative genomics analysis displayed that the cytochrome p450 gene family is extremely expanded to 425 members in FAW, of which 283 genes are specific to FAW. Treatments of Chinese populations with twenty-three pesticides showed the variant patterns of transcriptome profiles, and several detoxification genes such as AOX, UGT and GST specially responded to the pesticides. These findings will be useful in developing effective strategies for management of FAW in China and other invaded areas.
