BACKGROUND: The injury of peripheral nerve may cause electroneurographic changes. However, what is the correlation between electroneurographic changes and the extent of nerve injury?OBJECTIVE: To probe into the changes of evoked electroneurography in early stage after nerve injury for clinical application and reference.DESIGN: Paired t-test in three groups of samples.SETTING: Department of Traumatic Orthopaedics People' s Hospital of Beijing University.MATERIALS:The trial was conducted in the Animal Center of the People's Hospital of Beijing University from August 2003 to December 2003. The subjects were 8 adult New Zealand rabbits of SPF grade(weighing 1.5 kg) and either gender from the Animal Center of Medical College of Beijing University.METHODS:The model of nerve injury was made in the 16 tibial nerves of the 8 rabbits, in which one third or two thirds of each nerve was ligated at the same transverse part. The samples were thus divided into three groups: non-injury (normal) group, 1/3 nerve injury group and 2/3 nerve injury group.MAIN OUTCOME MEASURES: The indices of evoked electroneurography, i. e. motor nerve condution velocity, negative M wave amplitude, negative M wave area, maximal M wave amplitude and maximal M wave area were measured in the three groups. The correlation between the extent of nerve injury and the 5 indices was analyzed.RESULTS: All the 5 indices of the 3 groups were significantly different( P< 0.05). The mean values of the 5 indices were 1.00 ±0.0,0.97 ±0.18,1.01 ±0.22;0.98 ±0.16 and 0.98 ±0.19 in normal group, 0.76 ±0.12,0.41 ± 0.24, 0.47 ± 0.30, 0.48 ± 0.27 and 0.50 ± 0.29 in 1/3 nerve injury group, and 0. 62±0. 10, 0. 11 ±0. 11, 0. 11 ±0. 10, 0. 12 ±0. 14 and 0.13± 0. 12 in 2/3 nerve injury group. The coefficient of correlation between the 5 indices and the injury were 0. 902, 0. 938, 0. 936, 0. 907, and 0. 914, respectively.CONCLUSION: In the early stage after peripheral nerve injury, the electroneurographic indices negatively correlate with the extent of nerve injury. The negative M wave amplitude and area are relatively more closely correlated with the extent of injury.
3.Correlation of 24 h microalbuminuria and cerebral microbleeds in patients with small artery occlusion: a retrospective case series study

Objective To investigate the risk factors for cerebral microbleeds (CMBs) and its correlation with the 24 h microalbuminuria (mALB) in patients with small artery occlusion (SAO).Methods The patients with SAO were enrolled.CMBs were detected with susceptibility-weighted imaging.The demographic and clinical characteristics and 24 h mALB of the patients were compared.Multivariate logistic regression analysis was used to identify the independent risk factors for CMB in patients with SAO.Spearman correlation analysis was used to investigate the correlation between the 24 h mALB and the degree of CMBs.Results A total of 90 patients with SAO were enrolled and 35 patients (38.89%) had CMBs.CMBs mainly distributed in basal ganglia/thalamus and infratentorial (62%) regions.The Age (70.8 ± 5.4 vs.67.3 ± 8.1; t =2.461,P =0.016),proportion of hypertension (80.0% vs.52.7% ;x2 =6.851,P =0.009),and 24 h mALB levels (16.257 ± 6.031 mg/24 h vs.11.910 ±5.458 mg/24 h; t =3.536,P =0.001) in the CBM group were significantly higher than those in the non-CMB group.Spearman rank correlation analysis showed that the 24 h mALB and the severity of CMB in patients with SAO showed a significant positive correlation (rs =0.795,P =0.000).The higher the 24 h mALB level was,the more severe the CMB degree would be.Multivariate logistic regression analysis showed that only 24 h mALB was the only independent risk factor for CMBs in patients with SAO (odds ratio,1.100,95% confidence interval 1.031-1.176; P =0.002).Conclusions The 24 h mALB is an independent risk factor for CMB in patients with SAO.The 24 h mALB level is positively correlated with the severity of CMB,and it may be used as a marker for small vascular injury.
4.Clinical efficacy of posterior component separation with Sublay mesh repair for complex abdominal incisional hernia

Objective To explore the clinical efficacy of posterior component separation (PCS) with Sublay mesh repair for complex abdominal incisional hernia.Methods The retrospective cross-sectional study was conducted.The clinical data of 30 patients with complex abdominal incisional hernia who were admitted to the Beijing Chao-Yang Hospital of Capital Medical University from July 2016 to March 2017 were collected.Patients intraoperatively received PCS with Sublay mesh repair.Observation indicators:(1) intra-and post-operative situations:defect area of incisional hernia,operation time,volume of intraoperative blood loss,time of postoperative drainage-tube removal,postoperative complications and duration of postoperative hospital stay;(2) follow-up situation.Follow-up using outpatient examination and telephone interview was performed to detect recurrence of hernia and mesh-related complications up to July 2017.Outpatient examination was done once at postoperative month 1,3 and 6 and telephone interview was done at 1 year postoperatively.Measurement data with normal distribution were represented as x±s and measurement data with skewed distribution were described as M (range).Results (1) Intra-and post-operative situations:30 patients received successful PCS with Sublay mesh repair for complex abdominal incisional hernia.Defect area of incisional hernia,operation time,volume of intraoperative blood loss and time of postoperative drainage tube removal were respectively (222± 124)cm2,100 minutes (range,40-235 minutes),80 mL (range,50-200 mL) and 5 days (range,2-15 days).Of 7 patients with postoperative complications,3 were complicated with shallow surgical site infection,including 1 with wound healing by vacuum sealing drainage and 2 with delayed healing by debridement and drainage;2 with postoperative seroma were improved by aspiration and local pressurization after 1 months;1 with fat liquefaction of abdominal incision was improved by symptomatic treatment;1 with postoperative active hemorrhage was confirmed with arteriolar hemorrhage of muscular layer and then received hemostasis by ligation.Time of postoperative hospital stay of 30 patients was 15 days (range,10-57 days).(2) Follow-up situation:30 patients were followed up for (7± 3) months,without occurrences of hernia recurrence,intestinal fistula and mesh-related complications.Conclusion PCS with Sublay mesh repair for complex abdominal incisional hernia is safe and feasible,with good clinical efficacies.
5.Mediating effect of psychological resilience and depression between childhood abuse and relapse tendency of drug abusers

Objective:To explore the influence of childhood abuse experience on relapse tendency of compulsory drug abusers, and the intermediary role of resilience and depression between them.Methods:A total of 261 drug abusers were investigated with childhood trauma questionnaire(CTQ), the Connor-Davidson resilience scale (CD-RISC), self-rating depression scale (SDS) and relapse tendency psychological questionnaire.All data processing and analysis were carried out by SPSS 25.0 software.Correlation analysis, Bootstrap analysis and other methods were used to test the mediating effect.Results:The scores of childhood abuse treatment, resilience, depression and relapse tendency were 44.690±18.550, 60.210±22.790, 48.370±12.450, and 15.300±9.833 respectively.Childhood abuse was negatively correlated with resilience( r=-0.396, P<0.01), and positively correlated with depression and relapse tendency( r=0.584, 0.298, P<0.01).Resilience was negatively correlated with depression and relapse tendency( r=-0.558, -0.147, P<0.05).Depression had a significant positive correlation with relapse tendency( r=0.286, P<0.01).Childhood abuse could directly predict the tendency of relapse( β=0.202, P<0.01), and it could also affect the tendency of relapse through the mediation of depression( β=0.082, 95% CI=0.007-0.165) and the chain mediation of resilience and depression( β=0.029, 95% CI=0.002-0.064). Conclusion:The experience of childhood abuse leads to the decrease of resilience of strong abstinence personnel and increases the level of depression, which finally increases the tendency of relapse.
6.Molecular Cloning and Characterization of a Glutathione S-transferase Gene Repressed by Phenylacetic Acid From Penicillium chrysogenum

Glutathione S-transferase (GST) gene, PcgstA was cloned from the penicillin producing strain Penicillium chrysogenum,which is important for understanding the industrial fermentation process. PcgstA gene has an open-reading-frame of 840 bp in length,which is interrupted by two introns. The deduced amino acid sequence shows about 50% identity to several characterized filamentous fungi GSTs. The recombinant PcGSTA in Escherichia coli were overexpressed and purified. Enzymatic assays showed that the recombinant PcGSTA had a specific activity with 1-chloro-2, 4-dinitrobenzene of (0.159±0.031) μmol/(min· mg). It was found that the expression level of PcgstA in the penicillin producing medium supplemented with phenylacetic acid, the side chain precursor of penicillin G, was significant down regulated than that in medium without phenylacetic acid. This result suggested that PcGST may be related to phenylacetic acid metabolism in the penicillin producing strain.
7.Research on stomach remaining-floating sustained-release preparation of compound demethylcantharidin

AIM:To make compound demethycantharidin(including Radix astragali and Rhizoma bletillae) into stomach remaining-floating sustained-release preparation. METHODS:Radix astragal and Rhizoma betillae pulverized to superfine powders were combined with hydrophilic gel to manufacture stomach remaining-floating sustained-release tablet(floating tablet). RESULTS:The tablet started to drift in 3 min in the artificial gastric juice and float lasted for more than 8 h. By isotopic tracering,the floating tablets given on an empty stomach were retarded for 2-3 h and on large meal for 4-5 h in combination with ordinary tablet only retarded for 1 h. A pharmacopoeia dissolution method was set up to determine the release. 50% of tablets were disintegration in 5 h,70% in 8 h. The t 1/2 was 5.1 h(2-4 h for ordinary tablets),and the MRT was 8.3 h(3.3 h for ordinary tablets),and the remaining-floating sustained-release tablets showed a relative bioavailability of 230.71%. CONCLUSION:Pretreatment of raw herbs from compound demethylcantharidin and selection of the excipients will help to produce the herb-containing floating tablet.

Objective To improve the degree of substitution of the cholesteryl by purifying cholesteryl succinyl ester (CHS) ,one of the raw materials .Methods CHS was purified by recrystallization ,the pure CHS structure was characterizate by differential scanning calorimetry (DSC) and X ray diffraction (XRD) ,the free cholesterol content of the pure and crude were determinated by high performance liquid chromatography (HPLC) ,the degree of substitution of the cholesteryl was determina-ted before and after the purified by 1 H-NMR .Result CHS was purer after recrystallization ,with property of crystal ,which could significantly improve the degree of substitution of the cholesteryl .Conclusion The method of purification was simple and feasible ,which was the basis of improving hydrophobically modified Bletilla striata polysaccharide′s degree of substitution of the cholesteryl .
9.KAT8/MOF-Mediated Anti-Cancer Mechanism of Gemcitabine in Human Bladder Cancer Cells

Histone acetylation is a well-characterized epigenetic modification controlled by histone acetyltransferases (HATs) and histone deacetylases (HDACs). Imbalanced histone acetylation has been observed in many primary cancers. Therefore, efforts have been made to find drugs or small molecules such as HDAC inhibitors that can revert acetylation levels to normal in cancer cells. We observed dose-dependent reduction in the endogenous and exogenous protein expression levels of KAT8 (also known as human MOF), a member of the MYST family of HATs, and its corresponding histone acetylation at H4K5, H4K8, and H4K16 in chemotherapy drug gemcitabine (GEM)-exposed T24 bladder cancer (BLCA) cells. Interestingly, the reduction in MOF and histone H4 acetylation was inversely proportional to GEM-induced γH2AX, an indicator of chemotherapy drug effectiveness. Furthermore, pGL4-MOF-Luc reporter activities were significantly inhibited by GEM, thereby suggesting that GEM utilizes an MOF-mediated anti-BLCA mechanism of action. In the CCK-8, wound healing assays and Transwell ® experiments, the additive effects on cell proliferation and migration were observed in the presence of exogenous MOF and GEM. In addition, the promoted cell sensitivity to GEM by exogenous MOF in BLCA cells was confirmed using an Annexin V-FITC/PI assay. Taken together, our results provide the theoretical basis for elucidating the anti-BLCA mechanism of GEM.
10.P300 for the assessment of cognitive disorders in Chinese patients with Parkinson's disease: a Meta-analysis

Objective To assess the clinical utility of P300 for the assessment of cognitive disorders in Chinese patients with Parkinson's disease (PD) by meta-analysis.Methods Studies comparing the P300 between PD patients with different cognitive disorders (dementia,mild cognitive impairment (MCI),and no cognitive impairment;the former two were “cognitive impairments”,while the latter two were “nondementia”) and healthy controls were retrieved by searching within major Chinese and English databases.Studies,whose case and control groups were matched for age and gender,were rated as high quality.Weighted mean differences (WMDs) in P300 latency and amplitude between groups were calculated by using meta-analysis.Results In total,23 studies were included and 20 of which were rated high quality.Demented PD patients had statistically significant longer P300 latency (WMD:56.14,95% CI:19.06,93.23) and lower amplitude (WMD:-1.30,95%CI:-2.08,-0.51) than non-demented PD patients.PD patients with MCI had statistically significant longer P300 latency (WMD:13.71,95%CI:5.13,22.29) and lower amplitude (WMD:-1.25,95 % CI:-2.29,-0.21) than PD patients without cognitive disorders.However,no significant differences in P300 latency (WMD:4.16,95% CI:-13.85,22.17) and amplitude (WMD:0.23,95% CI:-1.72,2.17) were found between PD patients with dementia and MCI.Conclusion Among Chinese PD patients,there are significant P300 differences between those with dementia and without dementia,and between MCI and those without cognitive impairments,however,but no significant P300 differences between those with dementia and MCI.
