In this paper, we reviewed the progress in the study of the therapeutic HBV DNA vaccine, particularly on the evidences for its therapeutic effect, its mechanism of action, and to compare it with the traditional vaccines, etc. We also presented our research results on the therapeutic HBV DNA vaccine, and evaluated its clinical effect and characteristics objectively.
2.Present situation and prospect of the rapid development of hernia and abdominal wall surgery for 20 years in China

Hernia is ancient and common.With the emerging of new concepts,techniques,materials and equipments,treatment methods of hernia have undergone ever-changing changes in the 21st century.Hernia and abdominal wall surgery in China has been developing fast in the past 20 years,and has achieved remarkable progress.However,some problems still exist,such as unbalanced regional development,concept misunderstanding,classification confusions,less standardized technology,poor follow-up method,lack of innovation,and so on.Therefore,it's crucial to recognize the shortcomings of the current stagey,discuss the existing problems,sum up experiences and lessons,and clarify the direction of improvement,in order to provide a solid foundation for the sustainable development of hernia and abdominal surgery for our country.
3.STUDY ON IMMUNE RESPONSE TO THERAPEUTIC HBV DNA VACCINE

In this study,we constructed 2 eukaryotic expressing plasmids namely pcS2?S and pFP,encoding HBV preS2+S envelope protein and human IL 2/IFN ? fusion protein, respectively. The double plasmid fusion protein was used as an adjuvant in preparation of a treatment type HBV gene vaccine. To investigate its feasibility for use as a therapeutic DNA vaccine, we've evaluated the immune response after injection into healthy mice, HBV transgenic(Tg) mice, New Zealand rabbits and Rhesus monkeys. 4.Bond strength test of two resin adhesives to fiber post and the characteristics of adhesive interface between dentine and fiber post under confocal laser scanning microscopy

BACKGROUND: Resin adhesives include total-etching, self-etching and one-step adhesive. The studies concerning their adhesive properties remain different.OBJECTIVE: To compare the bonding strengths of fiber posts cemented to root canals with etch-and-rinse adhesives and self-etch adhesives, and to observe the physical interface bonding conformation using laser scanning confocal microscope so as to understand the retention of macro-and micro-morphology relationship.DESIGN, TIME AND SETTING: In vitro comparative observation was performed at the laboratories of Prosthodontics and Oral Surgery, Ninth People's Hospital, Shanghai Jiao Tong University from March 2008 to March 2009.MATERIALS: MATCHPOST fiber posts (Bisco, USA); etch-and-rinse adhesives (One-step, Bisco, USA) and self-etch adhesives (Panavia F adhesive system, Japan), fluorescein (Sigma, USA).METHODS: A total of 10 upper central incisor teeth, which had been freshly extracted for periodontal reasons, were selected,and the crown, 1 mm from the enamel-cementum junction, was cut. They were randomly divided into 2 groups with 5 specimens in each group. The fiber posts were bonded to root canals with One-Step or Panavia F which was tagged with rhodamine B.MAIN OUTCOME MEASURES: Each specimen was sectioned into two 2 mm-thick slices. A push-out test was made in a universal testing machine (EZ20), and confocal laser scanning microscopic (CLSM) analysis was performed to evaluate the primer penetration into dentin. The thickness of adhesive layer was measured by random software.RESULTS: The bonding strength of the group cemented with One-step was significantly greater than Panavia F (P < 0.05). The bonding strength of root crown was significantly greater than root apex (P < 0.05). One-step represented in CLSM image showed numerous and longer resin tags than Panavia F (P < 0.05).CONCLUSION: Etch-and-rinse adhesives (One-step) gained higher bonding strengths than self-etch adhesives (Panavia F).Resin binder to bond strength of fiber post and neck was greater than in the root apex.
5.Model of multiple seasonal autoregressive integrated moving average model and its application in prediction of the hand-foot-mouth disease incidence in Changsha

Objective: To establish multiple seasonal autoregressive integrated moving average model (ARIMA) according to the hand-foot-mouth disease incidence in Changsha, and to explore the feasibility of the multiple seasonal ARIMA in predicting the hand-foot-mouth disease incidence. Methods: EVIEWS 6.0 was used to establish multiple seasonal ARIMA according to the hand-foot-mouth disease incidence from May 2008 to August 2013 in Changsha, and the data of the hand-foot-mouth disease incidence from September 2013 to February 2014 were served as the examinedsamples of the multiple seasonal ARIMA, then the errors were compared between the forecasted incidence and the real value. Finally, the incidence of hand-foot-mouth disease from March 2014 to August 2014 was predicted by the model. Results: Atfer the data sequence was handled by smooth sequence, model identiifcation and model diagnosis, the multiple seasonal ARIMA (1, 0, 1)×(0, 1, 1)12 was established. The R2 value of the model iftting degree was 0.81, the root mean square prediction error was 8.29 and the mean absolute error was 5.83. Conclusion: hTe multiple seasonal ARIMA is a good prediction model, and the iftting degree is good. It can provide reference for the prevention and control work in hand-foot-mouth disease.
6.Study on the relationship between complement activation and inflammatory reaction in tissues of the perihematoma in patients with intracerebral hemorrhage

7.Prospective study of the treatment of adenomyosis by combining hysteroscopic transcervical resection of endometrium with levonorgestrel-releasing intrauterine system

Objective · To explore the clinical application value of hysteroscopic transcervical resection of endometrium (TCRE) combined with levonorgestrel-releasing intrauterine system (Mirena) in the treatment of adenomyosis. Methods · A total of 112 cases of adenomyosis patients were divided randomly into the combination group and Mirena group. The combination group (56 cases) was treated by TCRE endometrium endometrial resection, assisted Mirena treatment after operation. Mirena group (56 cases) was treated by Mirena only. The follow-up lasted 36 months after treatment, including measures of the volumes of menstrual bleeding, hemoglobin levels, dysmenorrhea scores, uterine volume, serum CA125 levels and incidences of complications.Results · The median follow-up duration was 42 months, and the three-year follow-up rate was 73.21% for the combination group and 50% for the Mirena group. After surgery, the volumes of menstrual bleeding of patients in 3-36 months decreased significantly, with an increase in hemoglobin level and a decrease in serum CA215 level and dysmenorrhea scores. Compared with their situations before surgery, the difference was significant (P<0.05). A comparison of uterine volume before and after surgery showed that there is a significant decrease in the uterine volume in both groups in6-12 months after surgery (P<0.05).Twenty-four months after surgery, it shows thatthe combination group has a much more significant decrease in uterine volume [(171.3±34.8) mm3] than Mirena group [(213.7±38.6) mm3] (P<0.05). The hysterectomy rate in Mirena group was significantly higher than that in the combination group (12.50% vs 5.36%); the ring expulsion rate was 16.07% in Mirena group and 5.36% in the combination group, and the break through bleeding happeningrate was 8.93% in Mirena group and 3.57% in the combination group. After 36 months an irregular small amount of vaginal bleeding rate was 62.25% in Mirena group, while it was only 12.50% in the combination group. There was significant differencewhen comparing above indices between two groups (P<0.05). There was no obvious differences in most common side effects of both groups. Conclusion · HysteroscopicTCRE combined with Mirena reduces significantly the irregular menstrual bleeding caused by merely applying Mirena. It has a prominent clinical efficacy and can be an effective approach in treatment of adenomyosis.
8.Relationship between the levels of inflammation-related cytokines and acute coronary syndrome in elderly patients

Objective By measuring the levels of serum MCP-1,sICAM-1 and expression of monocyte CD11b and CD62p in order to investigate the effects of these inflammation mediators on acute coronary syndrome and the relationship between inflammatory mediators and degree of coronary artery stenosis. Methods By means of enzyme-linked immunosorbent assay(ELISA) technique and flow cytometry technique,the changes of concentration of MCP-1,sICAM-1 and expression of CD11b and CD62p were measured in patients with acute coronary syndrome(ACS,n=33) group,stable angina(SA,n=24) group and control group(n=20)in the study. Results (1) The concentration of MCP-1 and sICAM-1 and expression of CD11b and CD62p had significant difference in statistics among three groups.Four inflammatory mediators were the highest in ACS group and the lowest in control group.(2)There were no significant differences in the levels of MCP-1,sICAM-1,CD11b,CD62p between AMI group and UA group.(3)There were no significant differences in the degree of coronary artery stenosis amomg SA,UA and AMI groups. Conclusions The results of the present study suggest that inflammation might play a role in the pathogenesis of coronary artery disease.
9.CELLULAR IMMUNE RESPONSE INDUCED BY DNA VACCINE IN BOTH HEALTHY AND HBV TRANSGENIC MICE

The peptides from HBV cytotoxic T lymphocyte(CTL) epitope were used to either stimulate or impact the immune effector (E) or CTL target (T) cells respectively, in order to investigate the cellular immune response induced by DNA vaccination in both healthy and HBV transgenic (Tg) mice. It was found that HBV DNA based immunization could induce CTL activity in healthy BALB/c mice, with intensity correlated with the E/T ratio and IFN ? secretion level in its supernatants. The target cells hit by HBsAg CTL epitope peptide(pp20) could be lysed by the active CTL induced by HBV DNA vaccine encoding preS2 and HBsAg, while the cell lysis could not be observed in the target cells impacted by the HBcAg CTL epitope peptide (pp10).
