Chinese Journal of Laboratory Medicine 2001;0(04):-
Objective To detect EB virus in peripheral blood of patients with non-hodgkin lymphoma (NHL) by ramification amplification method (RAM).To investigate the relativity of NHL with infection of EB virus. Methods RAM was used to detect EB virus with the man-made target gene sequence of promotor of EBER-1. In 120 cases of patients with NHL, EB virus were detected by RAM, and the result of RAM was compared with that of PCR.Raji cells and NB4 cells were used as positive control and negative control respectively. Results RAM could detect 10 molecular target gene of the exponent.76 cases were positive in 120 cases of NHL patients and the positive rate was 63.3%, The result is identical with that of PCR.Conclusion Ramification amplification method is sensitive, convenient and easy to perform.NHL has a high relativity with infection caused by EB virus.

Journal of Leukemia & Lymphoma 2009;18(8):452-454
Objective To explore the effective method for in vitro culture of the dendritic cells(DCs) and the specific anti-leukemic cell effect mediated by dendritic cells pulsed with acute myelogenous leukemia antigen. Methods Bone marrow mononucleas cells (BMMNCs) isolated from AML patients were induced to undergo differentiation with 500 ng/ml A23187 and pulsed with AML antigen. After 96 h, DCs and T cells were co-cultured for 5 to 7 days. The cytotoxic activity of cytotoxic T lymphocyte(CTL) to AML were detected with MTT colorimetry. Results BMMNCs isolated from AML patients treated with 100 ng/ml rhGM-CSF in combination with 500 ng/ml A23187 for 96 h exhibited typical morphology of DCs with rapidly increased expression of CD1a and CD83 (P<0.01). Dendritic cells pulsed with acid eluted tumor antigen peptides group displayed the strongest cytotoxic activity of CTL to AML. There was a significant difference between two groups(P<0.01). Conclusion BMMNCs isolated from AML patients can be successfully induced to DCs with rhGM-CSF and A23187 and dendritic cells pulsed with acid eluted tumor antigen peptides group had the strongest cytotoxic activity of CTL to AML.

Objective To observe the effects of flunarizine on the intracerebral hemorrhage (ICH) of Wistar rats at different time points. Methods An ICH rat model was established by collagenase. The concentrations of intracellular free Ca 2+ at different time points after ICH were determined by the fluorescence detector with an indicator, Fura 2/AM. The effects of flunarizine on the concentrations at different time points were also observed. Results The concentration of intracellular free Ca 2+ began to increase at 0.5 h, increased significantly at 6 h, and peaked at 24 h (about 4 folds as high as that before hemorrhage), but began to decrease at 72 h after acute intracerebral hemorrhage. Flunarizine decreased the concentration of intracellular free Ca 2+ significantly ( P

Objective To compare the vibrating perception threshold (VPT) between normal subjects and subjects with cervical spondylosis of nerve root type, and to observe the clinical efficacy of acupoint injection at Quyuan (SI13) in releasing pain in cervical spondylosis of nerve root type.Method Sixty-three patients with cervical spondylosis of nerve root type were recruited and randomized into a treatment group of 35 cases and a control group of 28 cases. The treatment group received acupoint injection at Quyuan, and the control group received acupoint injection at Jiaji (EX-B2) points. The short-form McGill Pain Questionnaire (MPQ) was adopted. From the questionnaire, the Pain Rating Index (PRI), sensory (S), affective (A), and total (T) pain rating indexes, Visual Analogue Scale (VAS), and Present Pain Index (PPI) were used to comprehensively evaluate the pain improvement and to quantify the therapeutic efficacy, and the VPT was also considered. The Clinical Assessment Scale for Cervical Spondylosis (CASCS) by West China Rehabilitation Center was also used to analyze the result in the treatment group.Result After the intervention, the MPQ and VAS scores in the treatment group were significantly lower than those in the control group (P<0.05). The total effective rate of the treatment group was significantly higher than that of the control group (P<0.05). Meanwhile, the VPT was improved after the treatment. Conclusion Acupoint injection at Quyuan can produce a more significant efficacy than at Jiaji (EX-B2) in treating cervical spondylosis of nerve root type.

Objective To evaluate the clinical value of prenatal ultrasound in the diagnosis of congenital anal atresia. Methods A retrospective analysis was studied from January 2013 to December 2015 which analyzed abnormal sonographic features by prenatal ultrasound screening in 13 cases of anal atresia. Results 13 cases of anal atresia were confirmed after abortion, miscarriage or birth which were not probed target ring signby prenatal ultrasound screening. That included 2 simple atresia cases and 11 cases associated with other malformations and abnormalities including 8 cases with bowel dilatation and 3 cases with intestine visible echogenic meconium. Conclusion Prenatal ultrasound screening for fetal congenital anal atresia can be detected. But for lacking of specific diagnostic standard, the detection rate of anal atresia remains very low.

ObjectTo develop the reliable RP-HPLC methods for the determination of salidroside, tyrosol, rosavin, rosin, and rosarin in the plants of Rhodiola L. and to evaluate their species from different habitats. Methods Method Ⅰ: methanol-water (0.5 mmol/L SDS in 1% acetic acid aqueous solution) system for the analysis of salidroside; method Ⅱ: acetonitrile-water system for rosavin; method Ⅲ: aqueous acetonitrile-phosphoric gradient system for salidroside, tyrosol, rosavin, rosin, and rosarin. Results The contents of salidroside in different species range from 0.021% to 1.420%, and those of rosavin in all species are very limited or undetected except in Rhodiola rosea L. and R. sachalinensis. The contents of the five marker ingredients are significantly species- and habitat-dependent. Conclusion Three RP-HPLC methods are established for quantitative analysis of the above five marker ingredients in the meantime, respectively. Evaluation of the quality of varied species of Rhodiola L. shows that R. rosea growing in Xinjiang Uygur Autonomous Region and R. sachalinensis growing in Jilin province are the two better species contained with abundant above-mentioned ingredients in China.

Objective To investigate the factors related to the heating effect by transarterial catheter hyperthermic infusion with the evaluation of the feasibility in controlling the tumor temperature. Methods Infusing 55-68℃ liquid at the speed of 10-40 ml/min through 6F,5F or 3F catheter with different length respectively under the similar clinical condition. The liquid temperature at the terminal exit of the catheter was measured with a digital thermometer. The factors related to the liquid temperature at the exit of the catheter were analyzed by multiple regression analysis. Results The infusion temperature, rate and the catheter length were the main related factors to the liquid temperature at the exit of the catheter as the condition similar in clinical use. When 60-65℃ liquid was infused at the rate of 20-40 ml/min through 5F catheter with length of 80 cm,the mean and 95% confidencial interval of the liquid temperature at the chetheter exit were(47.55?0.44)℃ and 44.61-48.49℃ respectively. Conclusions The liquid temperature at the exit of infusion catheter can be regulated and controlled through adjusting the liquid perfusion temperature and speed.
