1.Rapamycin reverse lipopolysaccharide-induced acute lung injury through acti-vating autophagy flux

This study was designed to examine the effects of autophagy on the lipopolysaccharide(LPS)-induced acute lung injury (ALI)and to analyze the possible mechanism.Kunming mice of clean grade were randomly divided into 6 groups:control group(saline;ip);model group(LPS 10 mg/kg;ip);RAP group(rapamycin 6 mg/kg;ip);RAP+LPS group (RAP 6 mg/kg and LPS 10 mg/kg;ip);3-MA group (3-methyladenine 300 μg/kg;ip);and 3-MA+LPS group (3-MA 300 μg/kg and LPS 10 mg/kg;ip).6 h after LPS injection;mortality was checked.Neutrophil aggregation;and the expressions of TNF-α;LC3 and P62 in lung tissue were checked by fluo-rescence microscopy and Western blot.The results revealed a higher mortality;neutrophil infiltration and TNF-αexpression and significantly increased levels of autophagy marker LC3-II and P62 in LPS group;in RAP+LPS group;pretreatment with RAP notably reversed LPS-induced neutrophil infiltration and TNF-αexpression;LC3-II were further slightly increased;while P62 was significantly decreased;in 3-MA+LPS group;pretreatment of 3-MA slightly decreased LC3-II expression;P62;neutrophil infiltration and TNF-αremained almost the same to those in LPS group.These data suggesed that;in sepsis;autophagy flux in the lung tissue is partially blocked;and RAP help to alleviate LPS-induced lung injury;which might through promoting autophagosome maturation;smoothing autophagy flux;and eventually to mitigate pulmonary inflammation.
2.Mood disorders differ after left and right cerebral hemisphere injury

Objective To explore mood disorders after left and right cerebral hemisphere injury,to search for possible mechanisms and to provide the basis for designing rehabilitation protocols and assessing prognosis.Methods Fifty-one cases with brain injury were recruited from a rehabilitation center and divided into left and right hemisphere injury groups with 26 and 25 cases,respectively.Hamilton rating scale for depression (HRSD) scores were analyzed. Results The left hemisphere patients had significantly higher HRSD total scores and higher scores indicating 15 kinds of mood disorders (depression,feelings of guilt,suicidal thoughts etc.).Right hemisphere injury patients had significantly higher diurnal variation,depersonalization or derealization and paranoid symptoms. Conclusions The lateralization of the cerebral hemispheres may display itself in mood and emotion.After left or right hemisphere injury,the depression presentation is different,so treatment,prognosis assessment and psychological intervention should be different for left and right hemisphere injuries.
3.Application of improved chromosome short-term culture method in the chromosome karyotype analysis of leukemia patients

Objective To make the chromosome karyotype analysis of 130 patients with leukemia by using the improved chromosome short-term culture method.Methods We optimized the main factors with a single factor gradient experiment in short-term culture of bone marrow chromosome, including colchicines concentration, duration of action of colchicines,and hypotonic time.On this basis,we conducted the three-factors and three-level orthogonal experiment to achieve improved bone marrow chromosome preparation system,which was later applied in 130 patients with leukemia in our hospital.Results The orthogonal experiment results showed that the optimum conditions were colchicines concentration of 0.07 μg/mL,colchicines action time of 80 min,and hypotonic time of 35 min during the preparation of the bone marrow chromosome.Using this method,the chromosome preparation success rate reached 97.69% and the detection rate of abnormal karyotype reached 82.3% in the chromosome karyotype analysis.Conclusion Bone marrow chromosome preparation system with colchicines concentration of 0.07 μg/mL and colchicines action time of 80 min,and hypotonic time of 35 min is worthy of clinical promotion.
4.Occupational health literacy and its influencing factors among key occupational populations in Fuling District, Chongqing

Background As a key to economic and social development, occupational groups with low levels of occupational health literacy (OHL) are considered vulnerable groups with high risks of work-related diseases and injuries. Therefore, improving the OHL of these groups is of great significance in reducing the burden on public health. Objective To master the four dimensional OHL levels and influencing factors of the key groups in Fuling District, Chongqing, and provide a basis for formulating health education programs for occupational groups in the future. Methods The Occupational Health Literacy Questionnaire of Key Populations was distributed online to investigate the key occupational groups in Fuling District through stratified cluster sampling. The survey collected data on basic demographic information, legal knowledge of occupational health, basic knowledge of occupational health protection, basic skills of occupational health protection, and healthy working style and behavior. To explore the role of autophagy in hair follicle cycle and whether 3-methyladenine(3-MA)could promote hair regeneration in C57BL/6 mice through inhibiting autophagy flux, hair regeneration model of C57BL/6 mice was induced on the dorsal skin by depilation, and 3-MA was intraperitoneally injected to investigate hair regrowth, meanwhile vehicle and rapamycin(RAPA)were used as the controls. Results showed that 3-MA could obviously promote hair regrowth in depilated C57BL/6 mice. Furtherly, haematoxylin-eosin staining, immunohistochemical staining, immunofluorescence and Western blot tests were used to investigate the autophagy signals and the cell proliferation. Results showed that the expression of Beclin1 and LC3B II/I ratio were significantly decreased. Expression of P62 and Ki67 were increased, as well as the CD34 and CD49f double-labeled hair follicle stem cells were obviously increased inside bulge areas in 3-MA group, while contrarily in RAPA group. These results affirmed 3-MA, an autophagy inhibitor, could promote hair regeneration in depilated C57BL/6 mice by facilitating the transformation of hair follicle from telogen to anagen. 3-MA and other analogous autophagy inhibitors probably have a potential usage in future therapy in human telogen effluvium diseases.
6.Relationship between dental calcification stages of the third molar and ages among teenagers in Chengdu.

OBJECTIVETo investigate the relationship between the stages of calcification of the third molar and age among children in Chengdu.
METHODSThe study subjects consisted of children between 2009 and 2011 at the Department of Orthodontics, West China Hospital of Stomatology, Sichuan University. Panoramic radiographs were obtained. The stages of calcification of the third molar were analyzed by Demirjian's method. The development of the third molar between the male and female was evaluated by independent samples t-test analysis. The Spearman rank correlation was applied to analyze the stages of calcification of the third molar and chronological age.
RESULTSThe development of the third molar of the male was different from that of the female at stage A of calcification of the third molar(P=0.026). The Spearman rank correlation coefficient revealed highly significant relationships between the stages of calcification of the third molar and chronological age of the three groups (male, r=0.808, P=0.000; female, r= 0.729, P=0.000; all, r=0.760, P=0.000). Furthermore, the linear regression equations were established as follows: Yall= 4.898+2.036X: Ymale=4.987+2.046X; Yfemale=4.819+2.036X.
CONCLUSIONThe stages of calcification of the third molar may be an available indicator in judging chronological age of teenager.

Objective:
To compare the clinical and imaging characteristics of traumatic acute subdural hematoma acute subdural hematoma with rapid resolution and those without rapid resolution.
Methods:
A retrospective case-control analysis was conducted on the clinical data of 60 traumatic acute subdural hematoma patients with hematoma thickness≥5 mm admitted to Second Affiliated Hospital of Shantou University Medical College from January 2011 to May 2018. There were 37 males and 23 females, aged 18-80 years [(47.0±16.9)years]. There were 27 patients in the rapid resolution group and 33 patients in the non-rapid resolution group. Coagulation function [prothrombin time (PT) and international normalized ratio (INR)] on admission, hospital stay, Glasgow outcome scale (GOS), and brain CT results were compared between the two groups.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection can cause acute respiratory distress syndrome, hypercoagulability, hypertension, and multiorgan dysfunction. Effective antivirals with safe clinical profile are urgently needed to improve the overall prognosis. In an analysis of a randomly collected cohort of 124 patients with COVID-19, we found that hypercoagulability as indicated by elevated concentrations of D-dimers was associated with disease severity. By virtual screening of a U.S. FDA approved drug library, we identified an anticoagulation agent dipyridamole (DIP) , which suppressed SARS-CoV-2 replication . In a proof-of-concept trial involving 31 patients with COVID-19, DIP supplementation was associated with significantly decreased concentrations of D-dimers ( < 0.05), increased lymphocyte and platelet recovery in the circulation, and markedly improved clinical outcomes in comparison to the control patients. In particular, all 8 of the DIP-treated severely ill patients showed remarkable improvement: 7 patients (87.5%) achieved clinical cure and were discharged from the hospitals while the remaining 1 patient (12.5%) was in clinical remission.
