1.Effects of DKK1 on breast cancer cell migration and invasion capa-bility

Objective:To analyze the DKK1 expression in breast cancer cell line MDA-MB-231 and examine its effect on the mi-gration and invasion of the cell line. Method:DKK1 expression in the breast cancer tissues was detected immunohistochemically, and DKK1 distribution in the breast cancer cells was observed using immunofluorescence. Western blot was used to investigate DKK1 ex-pression in breast cancer tissues and their matched normal breast tissues. A eukaryotic expression vector of DKK1 was constructed and transfected into MDA231 cell line. The migration and invasion ability of the cells were observed through scratch assay and Boyden chamber, respectively. Results: Immunohistochemical staining showed that DKK1 in the transfected lymph node or non-lymph node was highly expressed in the breast cancer tissues (P<0.05). The positive DKK1 expression in lymph node metastasis was less signifi-cant than those in non-lymph node transfected cells. Immunofluorescence localization results showed a remarkable bright red granular fluorescence in the cytoplasm of the MDA231 cell line. Western blot analysis showed that the DKK1 protein expression in carcinoma tissues were obviously higher than that in matched normal breast tissues. The migration and invasion ability of DKK1-overexpressed MDA231 cell was lower than those of MDA231 cell transfected with pCMV-Tag-2b empty vector. Conclusion:Negative correlation was found between DKK1 expression and the migration and invasion ability in breast cancer cells.
2.THE INFLUENCE OF QUERCETIN ON PMN METABOLISM AND FUNCTION

Quercetin (0.05~0.20 mmol/L, final concentration) obviously inhibited the aggregation and production of PGE2 and TXB2 of human PMN which were induced by AA ( 0.66 mmol/L), LTB4 ( 0.002 mmol/L) and the fourth composition of cobra venom of Guangxi Province ( 4.8mg/L) in vitro. These effects were dose dependent, the IDg50 of PMN aggregation induced by AA, LTB4 and the fourth composition of cobra venom of Guangxi Province was 68?mol/L, 59?mol/L and 61?mol/L respectively. At the concentration of 0.50?mol/L, it can also inhibit lysozyme release of PMN induced by AA, LTB4 and the fourth composition of cobra venom of Guangxi Province.
3.Study on adscription of plasma effective constituents of Beagle dogs after administrated with the extract of Polygonum multiflorum Thunb

Objective: To investigate the plasma effective constituents of Beagle dogs after oral administration with the extract of Polygonum multiflorum thunb. Methods: Based on the established HPLC fingerprints, comparison was carried out among the blank plasma, plasma containing drug, Polygonum multiflorum thunb. Simultaneously, the retention time and UV spectrogram were utilized as index to evaluate the relation among the fingerprint peaks so as to confirm the original of peaks. Result: Fourteen components were detected after oral administration. Four of them were the original components existed in Polygonum multiflorum thunb and ten of them were metabolites. Conclusion: Studying on the constituents and metabolites in plasma is useful to research the effective substances of Chinese medicine of Polygonum multiflorum thunb in vivo.
4.Determination of SBGC and Hyperin in the Effective Fraction of Jiangzhining

Objective To establish an HPLC method for determination of 2,3,5,4’-tetrahydroxystibene-2-O-β-D-glucoside(SBGC)and hyperin in the effective fraction of Jiangzhining. Methods HPLC analysis was performed on a C18 column(4.6 mm×250 mm,5 μm),with acetonitrile-0.1% methanoic acid water solution(15.5: 84.5)serving as the mobile phase. The flow rate was 1.0 mL·min-1 ,the wavelength was 340 nm,and the injection volume was 20 μL. Results The calibration curve was linear for SBGC in the range of 24-120 μg·mL-1(r = 0.999 8)and for hyperin in the range of 2.4-12.0 μg·mL-1(r= 0.999 6),respectively.Their average recoveries were 100.07% and 100.14%,respectively.The contents of SBGC and hyperin were 2.26% and 0.23%,respectively Conclusion The method is convenient,precise and reliable for determination of the content of SBGC and hyperin in the effective fraction of Jiangzhining.
5.Application of Multislice Computed Tomography Sagittal Reconstruction for Real Lumbar Spondylolisthesis

Objective To study the application of multislice computed tomography (MSCT) sagittal reconstruction for real lumbar spondylolisthesis. Methods 93 cases with lumbar spondylolisthesis accepted 64-slice MSCT. Their routine CT axial images and sagittal reconstruction images of vertebrae disc were reviewed. Results 93 cases were found with the lumbar spondylolisthesis, 74 cases in class I and 19 cases in class II. 58 cases were found with spondylolisthesis for routine axial CT image. Conclusion MSCT sagittal reconstruction can display spondylolisthesis satisfactorily
6.A Research on the Limited Amount Index of the Pesticide Residue and Heavy Metal Content of Notoginseng Medicinal Materials and Slices Based on the Pollution-Free Quality

This study analyzed the pesticide residue and heavy metal contents in Panaxnotoginseng (Burk.) F.H.Chen to provide the basis for the quality criterion of pollution-freemedicinal materials and slices of notoginseng.We randomly collected 100 samples of notoginseng from farmer's markets,producing areas of notoginseng and internet markets.We entrusted the third-party authoritative testing institutions to detect 203 items of the pesticide residues and 4 items of heavy metals.According to relative standards of Japan,Korea,the United States and the European Union,we analyzed and summarized the data in this study.We confirmed the 25 species of pesticides with high operating frequency and detection rate and the limited amount index of 4 heavy metals.In conclusion,these results enriched the limited amount index of the pesticide residues and heavy metal contents based on the previous notoginseng standard system serving as the quality criterion of pollution-free notoginseng,which was applicative and operable.

Objective To retrospectively analyse the detection results of human leukocyte antigen(HLA) B27,and discuss its diagnosis value in disease such as ankylosing spondylitis(AS).Methods HLA-B27 detection was performed on 1 335 suspected AS patients treated in the hospital from January 2015 to June 2016 by using multiparameter flow cytometry instrument.The test results showed 201 patients were diagnosed with AS(AS group),1 134 patients were diagnosed with rheumatoid arthritis and other related diseases (non AS group).The test results were shown by using mean fluorescence intensity and positive lymphocyte expression rate.n addition,120 healthy people were enrolled as the control group in the physical examination center of the hospital.Results The positive rate of HLA-B27 screening in suspected AS patients was 15.73% (210/1 335),and the positive rate of HLA-B27 screening in the control group was 2.50% (3/120).The positive rate of HLA-B27 screening,the rate of cell expression and the average fluorescence intensity of AS group were 92.03%(185/201),(85.34±17.99)%,8.74±4.20,significantly higher than the non AS group and the control group(P<0.01).There was no significant difference between the control group and the non AS group (P>0.05).HLA-B27 screening positive patients were mainly concentrated in adolescence,and the positive rate of male was higher than that of female (P<0.05).Conclusion Flow cytometry examination of HLA-B27 can provide important basis for the diagnosis and differential diagnosis of AS.
8.Methodology Comparison Study on Two Analysis Systems of Procalcitonin

Objective To evaluate the comparability of the test results of two immunoassay systems based on the electrochemical luminescence and the fluorescence lateral flow immunoassay for serum procalcitonin (PCT).Methods Roche cobas system was used as the reference system,and fluorescence lateral flow immunoassay system of Shanghai Upper biotech company was used as evaluated system.A total of 141 clinical samples during November,2015 were detected by the two systems to obtain the correlation coefficient and the Kappa values at the two cutoff values(0.5,2.0 ng/ml).Results The two systems showed high correlation (Y=1.008X-0.032,r=0.995,P<0.001) and low deviation (t=-0.230,P=0.819>0.05) without statistic significance between two methods.Kappa values were 0.944,0.943 respectively at the two cutoff values (0.5,2.0 ng/ml).Conclusion The test results showed no significant difference between the two immunoassay systems,suggesting a consistency between them for clinical detection of PCT.All the observed indicators reached the clinical diagnostic requirements and the method of quantitative detection of PCT by fluorescence lateral flow immunoassya can be applied for the quick detection of clinical human PCT.
9.Preliminary study of three-dimensional speckle tracking echocardiography in evaluation of left ventricular systolic longitudinal strain in patients with uremia

Objective To evaluate the characteristics of left ventricular systolic longitudinal strain in uremic patients and its relationship with dialysis time using three-dimensional speckle tracking echocardiography (3D-STE).Methods Forty-six uremic patients were divided into three groups according dialysis time (ranged 0-5 years):group A (≤1 year),group B (1 year>time ≤3 years),group C (3 years >time≤5 years),group D was control group.The dynamic images of apical four chamber view were displayed and apical full-volume images were obtained and saved in all subjects using a full volume matrix probe.The original data was recorded and was analyzed by using off-line 3DT analysis software (TomTec 4D MV-Assessment).The longitudinal peak systolic strain and 16 segment systolic strains and strain mean value in 16 segment of left ventricular were acquired and compared.Results The peak strain of apical segment was lower than that of the basal segment and the middle segment and it had an increasing trend from the apical segment to the basal segment in three uremia groups.Instead,it had a decreasing trend from the apical segment to the basal segment in control group.But the differences were not statistically significant.The longitudinal peak systolic strain value,16 segment strain value and mean value of 16 segment strain all were lower in three uremic groups than those of control group.Every segment of 16 segment stain value in group A was higher than those of group B and C.There was no difference in every segment between B and C group in addition to basal and middle segment of anterior septum.Conclusions 3D-STE could evaluate the left ventricular systolic function in uremic patients and the cardiac function could be partly protected by regular dialysis treatment.
10.Selection and evaluation of materials for skull repairing

OBJECTIVE: To summarize the classification, property of skull repair material and the processing of related complications in the application of repairing and remodeling the skull.METHODS: The PubMed database (http://www.ncbi.nlm.nih.gov/PubMed) and CNKI Database (www.cnki.net/index.htm) was searched by the first author using key words of "codubix, synmesh, and bone cement" both in English and Chinese. Articles whose content is related to the types, property, biocompatibility and its application effect of skull repairing materials were selected. In the same field, the articles published by authoritative journals or different races were preferred. The repetitive or obsolete literatures were excluded. After that 26 documents were included in this paper.RESULTS: The bone cement exhibited good histocompatibility, however, it is difficult to be absorbed, thus, it only be used for repairing part of skull defects. The study found that titanium had good biocompatibility and could combine with the skull. Its application had a promising prospect, but there are many inadequacies. With the continuous deepening of bio-engineering, bone tissue engineering, and cartilage tissue engineering, it will provide a broader perspective for the study of skull repair materials.CONCLUSION: There are many kinds of materials for skull repairing, and this paper only introduces some of the widely used ones. Actually, the selection of repairing materials should consider the pathogenetic condition, economic condition, local equipment and technical levels. If possible, titanium mesh and titanium screw are preferred, which is characterized by easy operation, few complication and beautiful appearance: When using autogenous bone or bone cements, if related complications can be reduced and handled effectively, the results will be satisfactory.
