1.Analysis of the correlation between autofluorescence and central vision at advanced stage of the classical retinitis pigmentosa

AIM: To investigate autofluorescence (AF) images of classical retinitis pigmentosa (RP) at advanced stage correlated with central vision. METHODS: Thirty-seven patients (70 eyes) with classical RP at advanced stage were observed. AF imaging was performed using a Heidelberg Retina Angiograph 2 ( HRA-2) scanning laser system with the field 30°, the pixels 1 536×1 536 and pupil diameters >6mm. Some patients underwent automated static peri-metry and the flash electroretinogram(FERG) were performed according to International Society for Clinical Electrophysiology of Vision( ISCEV) standard. RESULTS: All patients were classified as four patterns according to their manifestation of AF they were intact, patchy, solitary island and central hypofluorescence patterns. Different pattern showed different central vision. The data demonstrate high correlation between the central cone function and AF changes of the fundus. There is significant correlation (n= 57, r= 0.385,P<0.01) between central vision and the AF area and between age and AF area (n= 57, r= -2.67,P<0.05).CONCLUSION: AF images is valuable to the monitoring of central cone function and in predicting retention of central vision. The changes of AF images correlated with central vision and different pattern showed different vision.
2.Ultrasound-mediated PTEN gene inhibit invasion and metastasis of human ovarian carcinoma SKOV3 cell lines

Objective To determine the transfection of PTEN gene in human ovarian carcinoma SKOV3 cells by ultrasound mediating and contrast agent′s enhancing, and to explore the mechanism of PTEN gene inhibiting the tumor cells′ invasion. Methods After addition of PTEN gene and 5 ?l lipofectamine,human ovarian carcinoma SKOV3 cells on 6-well plates were explored to ultrasound for 60 s in the presence(200 ?l) of ultrasound agent. The invasiveness of transfected cells was measured quantitatively by Matrigel invasion assays(Transwell chamber) after 48 h. Results The invasiveness of SKOV3 cells transfected by ultrasound-mediated PTEN gene was significantly declined compared with that of the controls. Conclusions The transfection of PTEN gene into SKOV3 cell can inhibit their invasion.The gene transfection can be enhanced by ultrasound and contrast agent,which may be expected to be a new means of clinical gene therapy.
3.Determination of Camphor, Borneol and Bornyl Acetate in Shenling Baizhu Powder by Gas Chromatography

Objective To establish a GC method for simultaneous determine of camphor, borneol and bornyl acetate in Shenling Baizhu Powder. Methods Capillary column was used with 100% methyl polysiloxane as stationary phase. The temperature-programmed was as follows:starting temperature of 60 ℃, 5 ℃/min, rose to 130 ℃, kept for 5 minutes, followed by 20 ℃/min up to 230 ℃, and kept for 5 minutes. Results The average recovery rate of camphor was 99.87%, RSD was 1.3%, and good linear relationship was showed in the range of 0.003 77-0.150 8 μg (r=0.999 9). The average recovery of borneol was 100.71%, RSD was 2.1%, and good linear relationship was showed in the range of 0.002 411-0.096 4 μg (r=0.999 9). The average recovery rate of bornyl acetate was 101.95%, RSD was 1.3%, and good linear relationship was showed in the range of 0.003 02-0.121 1 μg (r=0.999 9). Conclusion The method is simple, reliable, accurate, and can effectively control the quality of Amomi Fructus in Shenling Baizhu Powder.
4.Clinical effects of triamcinolone acetonide combined with laser photocoagulation on patients with retinal vein occlusion

AlM: To observe the effects of the coelomic cavity injection of triamcinolone acetonide ( TA) combined with laser photocoagulation on patients with retinal vein occlusion.
METHODS:Fifty-six patients of retinal obstruction with macular edema were accepted from January 2010 to December 2012 in our hospital. All patients received iodized lecithin and Xueshuantong. And, patients with central retinal vein occlusion ( CRVO ) , hemi- central retinal vein occlusion ( hemi-CRVO ) and branch retinal vein occlusion ( BRVO ) treated by TA combined with laser photocoagulation, respectively. Follow-up period was of at least 6mo
RESULTS: After the treatment of 1, 3 and 6mo, the central foveal thickness was reduced significantly ( P<0. 05). After followed up 6-12mo, the total effective rate of CRVO, Hemi-CRVO and BRVO patients was 83% ~95% and all the patients had no significant adverse reactions.
CONCLUSlON:Basing on the traditional treatment, TA combined with laser photocoagulation is more effective in the treatment of retinal vein occlusion and is worthy of clinical usage.
5.Effects of nitroglycerine of different concentrations on rat cardiac preservation for 8h

Objective To investigate the effects of Thomas solution in combination with nitroglycerine of different concentrations on rat cardiac preservation for 8 h. Methods Thirty Wistar rats were selected,and Langendorff heart perfusion models were established.Rats were randomly divided into Thomas solution group,Thomas solution+4?10-6 mol/L nitroglycerine group and Thomas solution+4?10-5 mol/L nitroglycerine group(n=10).After hypothermic preservation(4 ℃)for 8 h,the cardiac function of each group was determined.Tissues of left ventricular wall were obtained,and changes of myocardial ultrastructure were observed by electron microscopy. Results Compared with Thomas solution group,the cardiac function was significantly improved and the myocardial ultrastructure injury was less severe in Thomas solution+4?10-5 mol/L nitroglycerine group after hypothermic preservation(4 ℃)for 8 h,while there was no significant difference in myocardial ultrastructure injury and cardiac function between Thomas solution group and Thomas solution+4?10-6 mol/L nitroglycerine group. Conclusion Thomas solution in combination with 4?10-5 mol/L nitroglycerine can significantly improve the preservation effects for isolated hearts in rats.
6.Application of Herniamesh in the treatment of adult umbilical hernia with 12 cases analysis

Objective To evaluate the application of Herniamesh in repairing adult umbilical hernia.Methods The data of 12 aduh patients with umbilical hernia were treated with Hemiamesh from January 2007 to March 2009 was analyzed retrospectively,included characteristics,methods and effects of Hemiamesh Oval Patch 0812 on the repair of hernia.Results All cases were cured.The average stay in hospital was (5±2)d.No postoperative complication,no recurrence follow-up visited 1 to 25 months.Conclusion Repairing umbilical hernia in adult with Hemiamesh Oval Patch 0812 is a kind of simple,safe,effective operation and the ring of hernia no more than 4cm.
7.Effect and mechanism of glycosylphosphatidilinoditol- specific phospholipase D on the adhesion function of bone marrow mononuclear cells separated from myeloid leukemia patients

BACKGROUND: There still is rarely report about the effect of glycosylphosphatidilinoditol-specific phospholipase D (GPI-PLD) on the adhesion function of leukemic cells through screening Medline and CNKI databases.OBJECTIVE: To observe the effect of GPI-PLD on the adhesion function of bone marrow mononuclear cells separated from myeloid leukemia patients, and to investigate the related mechanism. DESIGN, TIME AND SETTING: This study addressing cytology in vitro was conducted at the Hematological Laboratory of Xiangya Hospital from January to June 2004.MATERIALS: Bone marrow was collected from myeloid leukemia patients at the Department of Hematology, Xiangya Hospital, China.METHODS: The GPI-PLD activity of bone marrow mononuclear cells separated from myeloid leukemia patients was measured by using GPI-anchored placental alkaline phosphatase as substrate and Triton-X114 partition. By use of 1,10-phenanthroline, the activity of GPI-PLD was inhibited, the experiment was divided into 2 groups: treatment group adding phenanthroline to achieve a final concentration of 1 mmol/L, while control group adding the same amount of phosphate buffered saline. The adhesion rate to the fibronectin and CD24 expression of these cells were measured by MTT and immunohistochemical method, respectively.MAIN OUTCOME MEASURES: GPI-PLD activity of myeloid leukemic cells, cell adhesion rate, CD24 expression were all measured. RESULTS: The GPI-PLD activity of bone marrow mononuclear cells separated from myeloid leukemia patients was inhibited significantly after these cells were treated by 1 mmol/L 1,10-phenanthroline for 5 hours compared with control groups [(5.40±2.96)%, (42.08±7.21)%, P < 0.01]. At the same time, the adhesion rate of these cells were increased after the GPI-PLD activity was inhibited [(61.19±29.14)%, (49.78±26.73)%, P < 0.01], and the CD24 expression was also up-regulated [(18.5±11.14)%, (16.02±9.68), P < 0.01].CONCLUSION: The adhesion rate of bone marrow mononuclear cells separated from myeloid leukemia patients can be promoted by inhibiting GPI-PLD activity. At the same time, the CD24 expression of GPI-anchored proteins on bone marrow mononuclear cells is improved.
8.Application of total gastrectomy in treatment of gastric malignant neoplasms

Objective To investigate the effect of total gastrectomy for gastric malignancy(GM). Methods The clinical data of total gastrectomy in 113 patients with GM were analysed retrospectively. Results Radical total gastrectomy was performed in 97 cases, palliative total gastrectomy in 16 cases, combined with resection of head or tail of pancreas, splenectomy, transverse colectomy or parital hepatectomy in 19 cases. Reconstruction of digestive tract : Roux-en-Y esophagojejunostomy in 26 cases, Braun esophagojejunostomy in 19 cases, Roux-en-Y jejunal p-type anastomosis in 68 cases, Postopterative complications occurred in15 cases,including pancreatic fistula in one cases,anastomostic leakage in 3; operative death was happened in 2 cases. 86 patients (76%)have been followed up, the postoperative 1-,3-,5-year survival rate in radical total gastrectomy, combined resection organs and palliative total gastrectomy was 77.9%,45.7%,37.2%;88.8%,55.5%,33.3%; and 80%,0%,0% respectively. Conclusions The effect of total gastrectomy for gastric malignancy can be successful, if the indications are stricted and combined with rational viscers resection.
9.Assessment of coronary flow reserve in systemic lupus erythematosus patients by transthoracic echocardiography

Objective To assess the coronary flow reserve in the patients with systemic lupus erythematosus(SLE)by transthroracic echocardiography(TTE).Methods Coronary sinus blood flow of 24 SLE patients and 18 matched healthy controls were assessed by TTE.The coronary flow reserve was calculated as the ratio of hyperemic to baseline blood flow.Hyperemia was induced by intravenous administration of dipyridamole.Results Adequate recordings of blood flow in the coronary sinus under both conditions were obtained by TTE in all study subjects.Patients with SLE were found to have significantly decreased coronary flow reserve comparing with normal SUbiects(1.55±0.21 versus 2.51±0.33.P<0.01).Conclusions Coronary flow reserve can be measured by TTE in SLE patients,Coronary flow reserve is impaired suggesting that SLE patients may have subclinical coronary artery disease.
10.Establishement of phenobabital addiction rat model by modified drug admixed food method

Objective To establish the phenobarbital addiction rat model by the modified drug‐admixed‐food(DAF) method . Methods Rats were selected as the experimental animal .The modified DAF method was adopted to construct the model .The initial dose of phenobarbital 75 mg/kg was given mixed and the dase was gradually increased day by day ,which continued for 45 d .The es‐tablished model was verified by the precipitated withdrawal method and the natural withdrawal method for judging whether the model construction succeeding .Results The convulsion rate in the DAF precipitated withdrawal group was higher than that in the control group(P< 0 .01) ;the withdrawal symptoms scores in the DAF natural withdrawal group were higher than those in the con‐trol group(P< 0 .05) ;the relieving degree of body mass in the two withdrawal groups was greater than that in the control group (P< 0 .05) .The brain section in the mode group revealed obvious cellular apoptotic change .Conclusion DAF is simple and feasible for establishing the phenobarbital addiction rat model .
