Objective To discuss the significance of high-risk human papilloma virus ( HR-HPV) detection by hybrid capture 2 (Hc-2) in managing the women with atypical squamous cell of undetermined significance (ASCIIS) and low grade squamous intraepithelial lesion (LSIL) of thin prep liquid-based cytology (TCT). Methods One hundred and seventy-eight women with ASCUS and 108 women with LSIL were studied. The infection of HR-HPV in the women was analyzed. And sensitivity, specificity, positive predictive value (PPV) and negative predictive value (NPV) were used to evaluate the significance of HR-HPV detection by Hc-2 in managing the women with ASCUS and LSIL of TCT. Results Sensitivity, specificity, PPV and NPV of HR-HPV DNA detection by Hc-2 in screening CIN II or above in ASCUS were 89.83% (53/59),53.78% (64/119), 49.07% (53/108), 91.43% (64/70) respectively, and in LSIL were 97.62%(41/42),22.73%(15/66), 44.57%(41/92), 93.75%( 15/16) respectively. Conclusion HR-HPV detection by Hc-2 is an effective measure in managing the women with ASCUS and LSIL of TCT.

Objective To study the clinical diagnosis and treatment of malignant gastrointestinal stromal tumor. Methods To retrospectively analyze the clinical data of 28 cases with malignant gastrointestinal stromal tumors underweat surgical treatment . Results The malignant gastrointestinal stromal tumor in adults were more than 50 years old,71.4%(20/28) ,and common clinical symptoms were gastrointestinal bleeding, anemia, and pain. The lesion site: 19 cases of gastric bowel, 8 cases of small intestine, 1 case of colon, radical excision in 22 cases, local excision palliative resection in 5 cases, three cases were multi-visceral resection. Conclusion Malignant gastrointestinal stromal tumor could be diagnosed by the means of endoscopic imaging and clear, and preoperative diagnosis was difficult. Surgical resection was the pathology diagnosis and treatment of primary method,if necessary,to ensure multi-visceral resection of the tumor to prevent recurrence of thoroughness, had important significance.

Objective To compare the clinical efficacy and safety of domestic orlistat and imported orlistat in Chinese overweight and obese patients. Methods In a randomized, double-blinded and positive-controlled study, 228 adults (BMI 24-< 40 kg/m~2) evaluated at seven research centers were randomized to receive domestic orlistat or imported orlistat 120 mg 3 times a day with an energy-controlled diet for 24 weeks. Results After 24 weeks, domestic orlistat treated patients got significant weight-loss (5.0±3.7) kg, which was comparable with that of imported orlistat treated patients (4.5±3.5) kg (P=0.3922).Compared with the findings before treatment, there was significant decrease of systolic blood pressure (4.4±11.5)mm Hg (1 mm Hg=0.133 kPa) and serum levels of TC (0.54±0.79) mmol/L and LDL-C (0.32±0.64) mmol/L in the domestic orlistat treated group(compared with levels of baseline, P< 0. 0001). There was no significant difference between the two groups in the changes of blood pressure and lipid levels. Both groups had similar adverse event profiles, most of which were mild and transient gastrointestinal events. There were no serious adverse events in beth groups. Conclusions Domestic orlistat combined with a light low-energy diet promoted significant weight loss, which was comparable with that of imported orlistat after 24 weeks of treatment. There was also improvement in blood pressure and serum levels of TC and LDL-C. Domestic orlistat was as effective and safe as imported orlistat in the treatment of obesity.

The hypoxanthine-guanine phosphoribosyl transferase (HGPRT) gene mutation is responsible for gouty arthritis, kidney stone, and Lesch-Nyhan Syndrome (LNS). It has been reported that the expression of HGPRT is decreased or even absent in these diseases. Rabbits are an ideal model for studying the pathology of these diseases. Therefore, the development of an HGPRT-knockdown rabbit model will be highly beneficial m such studies. Stable HGPRT-knoekdown transgenie fibroblast lines were generated by transfecting rabbit fibroblasts with RNA interference (RNAi) plasmids. Polymerase chain reaction (PCR) analyses indicated that the average positive rate was 83.3%. The mRNA and protein levels of HGPRT in the transgenic fibroblast lines were significantly lower than that in the control. Transgenic rabbit blastocysts were derived after performing nuclear transfer. The results show that RNAi can be used to stably knock down expression of the HGPRT in rabbit fibroblasts and further improvements in related technologies will facilitate the use of this method for the generation of HGPRT-knockdown rabbits.
5.Ethical and Welfare Review in the Modeling Process of Disease Animal Models
Journal of Medical Biomechanics 2024;39(1):1-8
Disease animal model is an indispensable part of studying the pathogenesis and treatment of diseases.Review of the ethics and welfare is the necessary measure to ensure the quality of scientific research and promote the scientific and rational use of laboratory animals. In the review practice,it is found that most applicants are difficult to accurately understand the items listed in the application form and ethical principles related to items. Therefore,a practical and feasible set of ethical guidelines for animal experiments is necessary. This review focuses on the legal and ethical basis of developing oral disease animal models, and divides the methods of establishing oral disease animal models into physical methods, chemical methods, biological methods, and combined methods. It also elaborates on the ethical and welfare review points of different modeling methods and model evaluation methods.Hopefully, ethical censors and project applicants may get more understanding of ethical review for disease animal models, and ultimately improve the standardization and applicability of animal models.
