1.Expression and its Clinical Pathological Correlation of Heat Shocking Protein 40 in Epithelial Ovarian Cancer

Objective:The expression of Hsp40 in epithelial ovarian cancer and its correlation with clinical pathology were investigated. Methods:The expression of Hsp40 was detected by immunohistochemistry in 45 cases of epithelial ovarian cancer and 20 cases of benign ovarian tumor and 20 cases of normal ovary. Sta-tistical analysis was done to correlate the expression of Hsp40 with pathological characteristics. Results:The expression of Hsp40 in epithelial ovarian cancer was significantly higher than benign ovarian tumor and nor-mal ovary, the positive rates were 62.2% ,25.0% and 20.0% respectively ( P <0.01 ). There was no signifi-cant correlation between the expression of Hsp40 and clinical stages, histological grades,with or without me-testasis, with or without residual tumor. Kaplan-Meier survival curves and Log - rank analysis showed that the survival time in Hsp40 expression negative patients was significantly longer than those positive expression patients( P <0.01 ). Cox analysis showed residual tumor size was the only one covariant factor, the expres-sion of Hsp40 was not significantly relevant to prognosis. Conclusions. Hsp40 may play an important role in the occurrence and development of epithelial ovarian cancer.
2.Feasibility ofEucommia ulmoides gum as a root canal filling material

BACKGROUND:Eucommia ulmoides gum has good thermoplastics, liquidity, bondability and antimicrobial properties. However, the application of Eucommia ulmoides gum as a root canal filing material is rarely reported. OBJECTIVE: To compare the different contents of gas phase nano-silica on mechanical performance of composite material taking the gutta-percha as the matrix, and observe the effect of the root canal filing. METHODS:Four kind of root canal filing materials were prepared through adding barium sulfate (20%), nano-hydroxyapatite (10%) and different contents of gutta-percha and gas phase nano-silica (40% and 15%, 45% and 20%, 50% and 25%, 55% and 30%). PureEucommia ulmoidesgum served as the blank control. The mechanical properties (hardness, tensile strength and rupture elongation rate) of those five materials were detected. Furthermore, four kinds of composite materials, pureEucommia ulmoidesgum and Gutta-Percha Pelets were filed into the fresh extracted premolar root canal by using Obtura II technology. The effect of root canal filing was observed under X-ray and scanning electron microscopy.RESULTS AND CONCLUSION: Along with the increase of the content of gas phase nano-silica, the hardness of Eucommia ulmoides gum composite material increased, tensile strength increased firstly and then decreased, and rupture elongation rate decreased. PureEucommia ulmoides gum was transmitted to the X-ray and no image of filing material was obtained. Four kinds of composite materials were resistant to X-ray, and images showed that al composite materials were equaly filed into the root canal, without cacuole, and were in contact with the root canal inner wal, showing good filing effect. Scanning electron microscopy showed that, fingerlike projections were the longest at the surface of pureEucommia ulmoides gum; as the content of gas phase nano-silica increased, fingerlike projection length reduced at the surface of composite materials.Eucommia ulmoides gum composite materials could be developed as a canal filing material under heat flow condition.
3.RKIP regulates NF-κB signaling pathway in the pathogenesis of diabetic nephropathy and drug intervention

Objective To detect the expression of Raf kinase inhibitor protein (RKIP) and NF-κB in renal tissue of diabetic kidney disease (DKD) rats model,and to investigate the effect of rituximab (RTX) on the expression of RKIP in the renal tissue of DKD rats. Methods SD rats were randomly divided into normal group (N),DKD model group (M) and RTX treatment group (D).Blood glucose and 24-hour urine protein of rats were determined in three groups.RKIP protein and NF-κB protein were determined by immunohistochemistry staining. RKIP protein expression was detected by Western blotting. Results Compared with N group,blood glucose,24-hour urine protein and NF-κB expression in M group increased significantly (all P< 0.01),the expression of RKIP in M group decreased significantly (P<0.05).Compared with M group,the expression of RKIP increased significantly in D group (P<0.05),and 24-hour urine protein and NF-κB expression decreased in D group (all P<0.05).NF-κB protein expression was negatively correlated with RKIP expression in M group. Conclusions The NF-κB pathway regulated by RKIP plays an important role in the development and pathogenesis of diabetic nephropathy.Rituximab may have a role in treatment of DKD.
4.Effects of miR-1246 on proliferation,invasion and migration of cervical squamous cell carcinoma cell line SiHa and its target gene

ABSTRACT:Objective To explore the effects of miRNA-1246 (miR-1246)on cell proliferation,invasion and migration in human cervical squamous cell carcinoma (CSCC)cell line SiHa.Methods SiHa cells were assigned into 3 groups:miR-1246 analog group,miR-1246 antagonist group and control group.Transfection efficiency was determined.The MTT assay,transwell assay and wound healing assay were performed respectively to evaluate the proliferation,invasion and migration abilities of SiHa cells.Western blot was carried out to detect the expression of thrombospondin-2 (THBS2)before and after transfection.A THBS2 3’-UTR-containing dual luciferase plasmid was synthesized and co-transfected with miR-1246 into SiHa cells to observe the luciferase enzyme activity.Results MTT assay,transwell assay and wound healing assay revealed that the abilities of proliferation,migration and invasion were significantly enhanced (P<0.01)in SiHa cells transfected with miR-1246 analog,but suppressed in SiHa cells transfected with miR-1246 antagonist.Western blot showed that SiHa cells transfected with miR-1246 analog had significantly decreased THBS2 expression (gray value = 6 .2 8 ± 1 0 .2 2 , P=0 .0 1 3 ) while those transfected with miR-1246 antagonist had significantly increased THBS2 expression (gray value = 12.90±19.81, P=0.037).After co-transfected with miR-1246 and THBS2 3’-UTR-containing plasmid,SiHa cells exhibited a decreased level of luciferase enzyme expression.Conclusion miR-1246 promoted the proliferation,invasion and migration of CSCC SiHa cell, and it might promote CSCC tumorigenesis and progression by suppressing the expression of its target gene THBS2 .
5.Diagnosis and treatment value of colposcopy and loop electrosurgical excision procedure in microinvasive cervical cancer:analysis of 135 cases

Objective To explore the sensitivity and specificity of colposcopy directed biopsy (CDB) and the value of loop electrosurgical excision procedure (LEEP) for the diagnosis and treatment of microinvasive cervical cancer (MCC). Methods One hundred and thirty five patients with MCC were diagnosed with LEEP in Obstetrics and Gynecology Hospital, Fudan University from April 2008 to November 2010, and were retrospectively analyzed on CDB diagnoses and following treatment after LEEP. According to patient′s desire for preservation of fertility and cone margin status, following strategies after LEEP included follow-up, second LEEP, hysterectomy, modified radical hysterectomy and radical hysterectomy. Single and multiple factors related to residual lesions after LEEP were analysed with Pearson Chi-square test and logistic regression model, respectively. Results CDB diagnosed MCC with a sensitivity of 4.4%(6/135), specificity of 100.0%(4 680/4 680), and false negative rate of 95.6%(129/135). Among the 135 patients, 29 did not receive further treatment in our hospital and lost contact. One hundred and six patients had secondary treatment or follow-up in our hospital, 4 of among which were closely followed up;one hundred and two received further treatment, which included 6 cases with second LEEP (3 received extrafascial hysterectomy after repeat LEEP), 59 cases hysterectomy, 14 cases modified radical hysterectomy and 26 cases radical hysterectomy. For factors related to residual lesions after LEEP, single factor analysis showed that the ratio of residual lesion in patients aged 27-39, 40-49 and 50-65 years were respectively 19.0%(11/58), 15.4%(10/65) and 5/12 (χ2=4.505, P=0.105). Residual lesions occurred in 24.7%(23/93) of patients with positive LEEP margins, which was more than that 7.1%(3/42) of patients with negative LEEP margins (χ2=5.756, P=0.016). The ratio of residual lesions in patients with positive endocervical, ectocervical and deep stromal margins were respectively 29.6%(8/27), 17.1%(7/41)and 30.6%(11/36;χ2=2.275, P=0.321). Residual lesions in patients with or without lymph vascular space invasion (LVSI) were 2/7 and 18.8%(24/128), respectively (χ2=0.412, P=0.521). The ratio of residual lesions in patients with invasion depth of<1 mm was 17.1%(7/41), 1-<3 mm was 19.0%(16/84), and 3-5 mm was 3/10, with no significant difference among three groups (χ2=0.870, P=0.647). Logistic regression analysis showed positive cone margin (OR=5.069, P=0.014) and age (OR=1.080, P=0.024) were the independent risk factors of residual lesions after LEEP conization. Conclusions CDB alone is not adequate for the diagnosis of MCC. For young patients who desire to preserve fertility with a negative cone margin, close follow-up is acceptable. Cone margin status and age are two independent risk factors for residual lesions after LEEP.
6.Correlation of fetuin-B concentrations in serum and follicular fluid with outcomes offertilization.

OBJECTIVETo investigate the relationship between serum/follicular fluid fetuin B levels and outcome offertilization (IVF).
METHODSInfertility women (28 with low fertilization rates, 44 with normal fertilization rates) receiving IVF in Women's Hospital of Zhejiang University School of Medicine during June and December 2016 were enrolled in the study. Serum/follicular fluid fetuin B levels were measured with ELISA method. Correlations of serum and follicular fetuin B level with fertilization outcome of IVF were analyzed with Pearson correlation coefficient and receiver operating characteristic (ROC) curve.
RESULTSA positive correlation between serum fetuin B and follicular fluid fetuin B levels was observed (=0.675,<0.01). Both serum and follicular fluid fetuin B levels in women with low fertilization rates of IVF were lower than those in women with normal fertilization rates[(6.09±1.31) μg/mL vs. (7.13±1.47) μg/mL,=3.050,<0.05; (5.13±0.96)μg/mL vs. (6.22±1.33) μg/mL,=3.755,<0.01]. ROC analysis showed that the area under curve (AUC) of serum fetuin B level in predicting fertilization rate was 0.832 (95%:0.729-0.934,<0.01), and 6.08 μg/mL could be used as cut-off value.
CONCLUSIONSSerum fetuin B level is correlated with follicular fluid fetuin B level, and it may be used for predicting the fertilization outcome of IVF.
7.Preparation and performance evaluation of controlled-release chitosan-based microneedles.

In clinical application, a microneedle system that continuously delivers drugs is of great value for the delivery of some vaccines and hormone drugs. In this study, a controlled-release chitosan-based microneedle array (PVA/CS-MN) was designed, combining microneedle patches with drugs for controlled-release of drugs. Here we report the optimization of the preparation process of PVA/CS-MN. The appearance, morphology, mechanical properties, dissolution and swelling properties, and in vitro penetration properties of the MN arrays were characterized. The PVA/CS-MN prepared by the optimal process showed good morphology and mechanical properties. PVA/CS-MN can smoothly open microchannels on the skin and achieve controllable dissolution and swelling functions. Ascorbic acid (l-ascorbic acid) was used as a model drug to prepare a Vc-PVA/CS-MN. In vitro transdermal diffusion experiments showed that the Vc-PVA/CS-MN released about 57% of the drug within 1 h. About 66.7% of the drug was slowly released within 12 h, and a total of 92% of the drug was released after 7 days. The controllable sustained-release properties and excellent drug delivery efficiency of PVA/CS-MN provide a new option for sustained transdermal drug delivery.
