1.Dissolution Determination of Erythromycin Ethylsuccinate Granules

OBJECTIVE:To establish a method for dissolution determination of Erythromycin ethylsuccinate granules.METHODS:Paddle method was adopted and 0.1 mol?L-1 HCl solution was used as dissolution medium at rotation rate of 50 r?min-1.UV spectrophotometry was used to detect at wavelength of 482 nm.Dissolution rate was calculated.RESULTS:The linear range of erythromycin ethylsuccinate were 4~40 mg?L-1(r=0.999 0,n=5)with an average recovery of 99.19%(RSD=0.56%).Dissolution rate of erythromycin ethylsuccinate was more than 80% within 30 min.CONCLUSION:The method is rapid,simple and accurate for the quality control of Erythromycin ethylsuccinate granules.
2.Professor Chen Huade’s Clinical Experience on Treating Vertigo of Hyperactivity of Liver Yang Type by Acupuncture

Objective]To summarize Professor Chen Huade’s clinical experience on treatment of hyperactivity of liver yang type vertigo by acupuncture. [Method]By combining with expositions of past dynasties, collecting and analysis of the clinical cases of liver yang type vertigo, to summarize the etiology and pathogenesis of liver yang type vertigo and Professor Chen’s treatment characteristics, and take a case as evidence.[Result]In Professor Chen’s opinion, the basic pathogenesis of hyperactivity of liver yang type vertigo is hepatorenal Yin deficient liver Yang syndrone. Belong to excess in the upper and deficiency in the lower, asthenia in origin and asthenia in superficiality. Based on the theory of “treating disease from the root”,cure this disease by the body acupuncture plus eartip bloodletting therapy, each had a good curative effect.The evidence showed, after the body acupuncture plus eartip bloodletting therapy ten times, vertigo symptom relieved obviously, blood pressure dropped to normal range. In the following up of 3 months, none had a recurrence. [Conclusion]Because of its cheap, non-toxic side effects, significant efficacy, and the characteristics of both symptoms and root causes curing, it is gradually accepted by more and more patients.Professor Chen Huade’s treatment on hyperactivity of liver yang type vertigo by acupuncture has significant effect in clinic which is worth spreading.
3.Meta Analysis of Acupuncture Treatment for Vertigo with Excess Syndrome

Objective To systematically assess the clinical efficacy and safety of acupuncture treatment for vertigo with excess syndrome. Methods Chinese and English literature about acupuncture treatment of vertigo with excess syndrome published in recent years was comprehensively searched. The quality of the retrieved literature meeting the inclusion criteria of randomized controlled trial was assessed and its data was collected. A Meta analysis of the included studies was carried out.Results Finally, 10 articles with randomized controlled trials containing a total of 688 patients were included in the analysis. The included literature was assessed at lower quality using Cochrane evaluation member manual 5.1. A Meta analysis showed that the efficacy rate of acupuncture treatment for vertigo with excess syndrome was higher than that of Western drugs alone [M-H OR 4.84, 95%CI (2.39, 9.81),P<0.0001]. Combined acupuncture and Chinese herbal medicine was superior to Chinese herbal medicine alone [M-H OR 3.82, 95%CI (2.06, 7.10),P<0.0001]. Vertigo symptom and function scoring showed day 3 of treatment [MD 4.66, 95%CI (2.97, 6.35)], day 7 of treatment [MD 0.95, 95%CI (0.03, 1.86)] and day 14 of treatment [MD 0.89, 95%CI (0.71, 2.49)]. There were statistically significant differences in the vertigo symptom and function scores between the two groups of patients at day 3 and 7 of treatment. There were no statistically significant differences in the scores between the two groups of patients at day 14 of treatment. Conclusions Acupuncture or combined acupuncture and Chinese herbal medicine are effective and highly safe in treating vertigo with excess syndrome, but high-quality, multi-center and large-sample RCT studies still need to be conducted for validation and support.
4.Lipo-alkaloids in Sini Decoction stemmed from ester-exchange reactions

Objective To study the solubility of lipo-alkaloids from prepared aconite root(Radix Aconiti Lateralis Preparata) and identify the sources of lipo-alkaloids in Sini Decoction.Methods The electrospray ionization mass spectrometry was employed to analyze the alkaloids in crude aconite roots,Sini Decoction,the gruffs of concocted aconite roots and the decoction of prepared aconite root and pinellia tuber(Rhizoma Pinellae).Results Lipo-alkaloids were observed in Sini Decoction,however,no lipo-(alkaloids) were found in the decoction of aconite roots with pinellia tuber.Lipo-alkaloids were the dominant components in the gruffs of concocted aconite roots.Conclusion Lipo-alkaloids are barely soluble in boiling water.The alkaloids existed in the Sini Decoction stem from the ester-exchange reactions of diester-diterpenoid alkaloids and fatty acid in the boiling water.
5.Ester-exchange reactions for triester aconitines in decocting flowers of Aconitum kusnezoffii

Objective To study how and why the lipo-alkaloids changed in decocting the flowers of Aconitum kusnezoffii (FAK). [WT5HZ]Methods The alkaloids in ethanol extract of FAK, FAK decoction, and the residues of decocted FAK were analyzed and compared by electrospray ionization tandem mass spectrometry (ESI-MSn). Results No lipo-alkaloids were detected in FAK decoction, however, triester-lipo-alkaloids become the dominant components in the residues of FAK decoction. Conclusion Besides hydrolysis reactions, ester-exchange reactions happen for diester-aconitines and triester-aconitines in the decocting of FAK. It is the first time to report that C_8-acetyl is displaced by long chain fatty acyl for triester-aconitines in the ester-exchange reactions.

Objective In embryology, the pancreas is formed by fusion of the ventral pancreas(VP) and the dorsal pancreas(DP) during the seventh week of gestation. The aim of this study was to clarify whether there were differences in the clinicopathological characteristics of the carcinoma of pancreatic head(PhCa) arising from the VP and DP. MethodsResected specimens from 43 cases with PhCa were studied histologically and immunohistochemically. The VP and DP were distinguished by islet shape and amount of cells producing pancreatic polypeptide(PP) in normal pancreatic parenchyma adjacent to the main tumor. ResultsThere were 20 cases of PhCa from VP(VP-group) and 23 cases of DP (DP-group) in this study. 7.Effects of Ginseng Aqueous Extract on Neurochemicals in Diabetic Encephalopathy Rat Brain by Online Microdialysis-LC-MS/MS

To evaluate the effects of ginseng aqueous extract on learning and memory abilities and neurochemicals in diabetic encephalopathy rat brain. Diabetes mellitus model was established. Cognitive abilities were evaluated with Morris water maze test by the indexes of escape latency time ( ELT) , times of passing through the target area and the percent of swimming track in goal quadrant. We applied online MD-LC-MS/MS method to determine eight neurochemicals in rat hippocampus among different groups in MRM mode. Venusil C18 column was used for separation. Treatment with ginseng aqueous extract significantly improved the cognitive abilities of diabetic rats (p<0. 05). The method had excellent linearity (R2>0. 99). The accuracy and precision could meet the analysis requirement. Comparing with the model group, the concentrations of taurine and acetylcholine were significantly increased ( p<0 . 01 ) after treated with ginseng aqueous extract, but the glutamic acid, aspartic acid,γ-aminobutyric acid, serine, dopamine and serotonine decreased dramatically. Ginseng aqueous extract can efficiently regulate the levels of eight neurochemials in diabetic rat brain to normal.
8.Analysis of Coenzyme Q10 and Glucose in Dried Blood Spot from Phenylketonuria Newborn

Phenylketonuria (PKU) is a newborn inherited metabolic disorder caused by the genetic deficiency of hepatic enzyme phenylalanine hydroxylase (PAH) which thus in metabolic disorder of phenylalanine. In this study, ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method was used to analyze the accurate amount of coenzyme Q10 (CoQ10) and the relative amount of CoQ9 in newborn dried blood spot (DBS) collected from 5 PKU newborns (3 and 30 days after birth, respectively) and 20 healthy newborns. The content of CoQ10 was (122.1±24.9 ng/mL) and (59.0±12.0 ng/mL) in DBS from healthy newborns and PKU newborns, respectively. The relative contents of cholesterol and glucose in the DBS were determined by gas chromatography-mass spectrometry (GC-MS). In comparison with healthy newborn group, the levels of CoQ10, CoQ9, cholesterol and glucose were all significantly decreased in PKU newborns. The increased content of Phe and the decreased content of CoQ10 showed significant inverse correlation in the DBS from PKU. This study provides references for diet therapy of PKU newborns.
9.Analysis of Brain Pharmacokinetics of Hyperoside and 1,5-Dicaffeoylquinic Acid and Treatment Effects of Acanthopanax Senticosus Leaves on Cerebral Ischemia by On-line Microdialysis-Tandem Mass Spectrometry

The brain pharmacokinetics of hyperoside and 1 , 5-dicaffeoylquinic acid and the effects of acanthopanax senticosus leaves on neurotransmitters in cerebral ischemic rat brain were investigated. In the study, acute incompleteness cerebral ischemia model was developed by ligating the bilateral common carotid arteries of rats. The differences of brain pharmacokinetics of hyperoside and 1,5-dicaffeoylquinic acid between control and cerebral ischemic rats were determined by online microdialysis coupled with MS/MS method. At the same time, the contents of glutamic acid (Glu), aspartic acid (Asp), γ-aminobutyric acid (GABA), serotonin (5-HT) , dopamine ( DA) , and acetylcholine( Ach) in rat hippocampus among different groups were determined simultaneously in MRM mode. ACE 5 C18-AR column was used for separation. Results showed that the online analytical method had excellent linearity (R2>0. 99). The accuracy and precision could meet the analysis requirement. Besides, the hyperoside and 1,5-dicaffeoylquinic acid penetrated the blood-brain barrier successfully and metabolized quickly, but the absorption reduced and elimination became faster in cerebral ischemic rats; the contents of Glu, Asp, GABA and DA in brain issue of cerebral ischemic rats decreased significantly ( p<0 . 01 ) and the level of Ach increased remarkably ( p<0 . 05 ) after a preventive medication with the extract of acanthopanax senticosus leaves for one week.
