1.Preparation and Quality Control of Ganciclovir Vagina Suppository

OBJECTIVE:To prepare Ganciclovir vagina suppository and to establish the quality control standard of it.METHODS:Suppository was prepared with gelatin as base material and ganciclovir as main ingredients.The content of ganciclovir was determined by HPLC.RESULTS:The preparation was whitish transparent duck-bill suppository with proper hardness.The identification and test of suppository was in line with the standards stated in Chinese Pharmacopeia(2005 edition).The linear range of ganciclovir was 2.0~90.0?g.mL~(-1)(r=0.999 9)The average recovery was 99.28%(RSD=0.58%,n=9)CONCUlSION:The preparation of Ganciclovir vagina suppository is easy in technological process.The content determination method is simple and accurate.
2.Quality Standard of Tongtong Granule

OBJECTIVE: To establish the quality standard of Tongtong granule. METHODS: Panax notoginseng,Ginkgo biloba and Pueraria lobata were identified qualitatively by TLC and the content of total flavonol glyconsides in G. biloba was determined by HPLC. RESULTS: The TLC spots were distinctive and specific. The linear range of quercetin, kaempferide, isorhamnetin were 5.24~83.84 ?g?mL-1(r=0.999 2), 4.56~72.96 ?g?mL-1(r=0.999 1) and 2.48~39.68 ?g?mL-1(r=0.999 4), respectively. The average recoveries were 96.21%(RSD=2.06%)、99.14%(RSD=1.19%)、95.00%(RSD=1.31%),respectively. CONCLUSION:The method is reliable, accurate, and reproducible for the quality control of Tongtong granule.
4.Influence of repetitive ischemic preconditioning on metal elements during ischemic injury of spinal cord in rabbits

BACKGROUND: It has been reported that the ischemia preconditioning (IPC) had credible protective efficiency on ischemic injury of the spinal cord during aorta operation, but the mechanism of the protective efficiency of IPC had not been clarified.OBJECTIVE: To study the protective effects of repetitive IPC on ischemic injury of spinal cord and its mechanism in rabbits.DESIGN: A completely randomized controlled study based on the experimental animals.SETTING: Department of anesthesiology in a university hospital.MATERIALS: The experiment was completed in the Department of Anesthesiology, Renmin Hospital, Wuhan University during September and December 2002. Twenty-four Japanese rabbits were randomly and double-blindly divided into sham-operation group, ischemia-reperfusion group and IPC group with 8 rabbits in each group.INTERVENTIONS: In sham-operation group, abdominal aorta was not clamped. Spinal cord ischemia was induced by infra-renal aortic cross-clamp for 45 minutes in ischemia-reperfusion group. Before the 45 minutes ischemia, the rabbits in the IPC group underwent four cycles of ischemia preconditioning, i.e. clamping abdominal aorta for 5 minutes then reperfusion for 5 minutes.MAIN OUTCOME MEASURES: The concentrations of calcium, magnesium, copper and zincum in spinal cord were measured in the 7th day after operation. Postoperative neurological function, EMG of rear limb, and spinal cord histopathological changes were assessed in all groups after operation.RESULTS: The concentrations of calcium and copper in spinal cord in ischemia-reperfusion group were significantly higher than those in sham-operation group( P < 0. 05 or 0. 01 ), but magnesium and zincum significantly lower( P < 0. 05). Compared with IPC group, calcium in ischemia-reperfusion group was significantly higher( P < 0.01 ), but zincum significantly lower( P < 0.01 ) . The neurological function and histopatholohical changes in ischemia-reperfusion group were much lower than those in sham-operation group and IPC group ( P < 0.05 or P < 0.01) . And there was significantly worse change of EMG in ischemia-repeffusion group than that in IPC group(P < 0.01).CONCLUSION: Repetitive ischemic preconditioning can protect rabbit spinal cord from ischemia reperfusion injury quickly, and one possible reason for its protective effect is to maintain the balance of calcium, magnesium,copper and zincum in ischemic region.
5.The changes of CD27+ B cells in peripheral blood of the patients with HBV related disease and its clinical significance

Objective To observe the changes of peripheral blood CD27+B cells percentage in patients with HBV-related disease of different severity and the clinical significance.Methods 20 cases of chronic viral hepatitis B,23 cases of HBV-related liver cirrhosis,21 cases of liver cancer were selected,25 cases of healthy controls in the corresponding period who had the physical examination were set as the healthy control group.The peripheral blood in the healthy controls and the patients with HBV-related disease was collected.The cellular immune level changes and CD27+ B cells percentage was detected by flow cytometry,the humoral immunity was detected by immune transmission turbidty method:IgM,IgD.Results (1)Compared with the healthy control group,CD27+B cells percentage significantly decreased in the three groups of patients with HBV-related disease [(5.16 ± 0.36) % vs.(4.52 ± 0.22) %,(2.24 ± 0.15) %,(0.58 ± 0.02) %,F =4.32,P < 0.05],and the downward trend gradually obvious as the disease degree exacerbated.nnnnn(2)IgM,IgD in the three groups of HBV related disease patients rised obviously,and the increase range became more obvious as the disease degree exacerbation(F =3.29,5.23,P =0.02,0.03).Conclusion CD27+B cells has a close relationship with HBV-related disease,and rebuilding the body's immune defense system is great importance for evaluating prognosis and the clinical guidance in HBV-related disease.
6.Effect of mind mapping-oriented health education on patient's fluid intake during hemodialysis intervals

Objective To evaluate the effect of mind mapping-oriented health education on patient's fluid intake during hemodialysis intervals. Methods From June 2015 to April 2016, 72 patients with poor compliance with fluid intake were randomly divided into control group (n=36) and observation group (n=36). In the control group, routine health education was used and the observation group was treated with mind mapping-oriented health education. The two groups were compared before intervention and 6 months after that in terms of compliance with fluid intake and daily fluid intake, ultrafiltration volume and body weight gain/dry weight ratio and incidence of complications. Results In the observation group, the fluid intake compliance, daily fluid intake, ultrafiltration volume and body weight gain/dry weight ratio 6 months after intervention were superior to those of the control group (all P<0.05). The incidences of complications such as hypotension, hypertension, muscle spasms and cardiac arrhythmia were also significantly lower than those of the control group (P<0.05). Conclusion The health education based on mind mapping is helpful to enhance the degree of cognition of fluid intake compliance, improve self-efficacy of chronic diseases, control the growth of body weight during dialysis and reduce the incidence of complications.
7.Expression of TRPV1 and Electrophysiological Characteristics of Colon-specific Dorsal Root Ganglion Neurons in Rat Model of Visceral Hypersensitivity

Background:Visceral hypersensitivity is considered to be one of the major pathophysiological mechanisms of irritable bowel syndrome. Aims:To investigate the expression of TRPV1 and electrophysiological characteristics of colon-specific dorsal root ganglion( DRG)neurons in rat model of visceral hypersensitivity. Methods:Twenty 10-day-old rats were randomly divided into two groups. In model group,visceral hypersensitivity was induced by colorectal administration of acetic acid;while in control group the same amount of saline was administered. Colon-specific DRG neurons were labeled retrogradely by injection of DiI,a fluorochrome,into the colon wall. Expression of TRPV1 in DRG neurons was detected by immunofluorescence and the electrophysiological characteristics of DRG neurons was detected by using patch-clamp technique. Results:In model group,the expression rate of TRPV1 in colon-specific DRG neurons was significantly higher than that in controls(46. 1% vs. 36. 6% ,P <0. 01),the average rheobase was significantly decreased[(57. 80 ±1. 32)pA vs.(73. 45 ± 4. 51)pA,P < 0. 05],while the frequency of action potentials(APs)in response to doubling rheobase stimulation was significantly increased[(8. 20 ± 1. 10)Hz vs. (4. 54 ± 0. 66)Hz,P < 0. 05]. Score of abdominal withdrawal reflex(AWR)under a 60 mm Hg colorectal distention was positively correlated with the expression rate of TRPV1 and the frequency of APs in response to doubling rheobase stimulation(r = 0. 87 and r = 0. 73,P < 0. 01),but was negatively correlated with the rheobase(r = - 0. 81,P < 0. 01)in model group. Conclusions:Increased expression of TRPV1 and excitability in colon-specific DRG neurons might be a crucial step in formation of visceral hypersensitivity.
8.Clinical efficacy of biofeedback therapy on constipation-predominant irritable bowel syndrome

Objective To investigate the clinical effects of biofeedback therapy on constipation-predominant irritable bowel syndrome(IBS-C).Methods 30 patients with IBS-C were given to biofeedback therapy,5 times every treatment course.and then their clinical symptoms,mental state and quality of life before the treatment and at the end of treatment with biofeedback therapy were respectively evaluated by symptom scores,self-rating anxiety scale (SAS),self-rating depression scale (SDS),and short form 36 health survey questionnaire (SF-36).Results Post-treatment with biofeedback therapy,there were very significant decrease in total and subscales scores of bowel symptom including abdominal pain/discomfort,abdominal distension,abnormal appearance of defecation,abnormal process of defecation((12.31±2.01) vs (19.16±2.17),(2.95±0.57) vs (5.04±1.04),(2.64±0.92) vs (4.25±1.09),(3.66±1.09) vs (5.10±0.57),(3.06±1.08) vs (4.77±0.95) respectively,P<0.01).The integration of SAS and SDS were obviously decreased after treatment((39.53±6.39) vs (44.43±7.89),P<0.05;(40.70±8.38) vs (46.46±8.74),P<0.05),the SF-36 scores were also improved in five dimensions including rolephysical,social-functioning,vitality,role-emotional and mental health((74.16±21.25) vs (57.0±39.40),(86.21±13.54) vs (75.54±20.96),(75.16±13.42) vs (64.66±20.54),(78.87±28.36) vs (57.76±46.26),(81.60±16.08) vs (71.20±22.04) respectively,P<0.05).Conclusion Biofeedback therapy can improve the clinical symptoms,alleviate the moods of anxiety and depression,and improve the quality of life in patients with IBS-C.
9.Analysis of the Chest X-ray Manifestations in SARS Patients Treated with Compound Glycyrrhizin

OBJECTIVE:To summerize the characteristics and variability of chest X-ray manifestations in SARS patients treated with compound glycyrrhizin.METHODS:60cases of SARS were equally divided into2groups:groupⅠreceiving compound glycyrrhizin,groupⅡ(as control)receiving conventional treatment.The appearing time,site,scope and dynamic changes of the pulmonary lesions on chest radiograms were compared between2groups.RESULTS:The average period from peak to50%improvement of lesion in X-ray manifestations was shorter in groupⅠthan that in groupⅡ.In restoration stage,more patients had their X-ray findings absorbed in groupⅠcompared with the patients in groupⅡ.Compound glycyrrhizin had little influence on WBC,blood sugar and electrolytes.CONCLUSION:Glycyrrhizin may be a promising drug against SARS with less side effects.
