1.LOCALIZATION AND QUANTITATION OF ESTROGEN RECEPTORS IN SEVERAL TISSUES OF DIFFERENT STRAINS OF MICE BY AUTORADIOGRAPHY

The distributions of estrogen receptors in preoptic region of hypothalamus, anterior lobe of pituitary gland, mammary gland, thymus and liver of LACA and C57 BL strains of mice were studied by ~3H-estradiol and autoradiography in frozen sections.In both strains of mice, labelling index (LI) was more than 85% in preoptic region of hypothalamus and anterior lobe of pituitary gland, and average number of silver grains (AG) was more than 19. LI and AG of epithelial cells of mammary glands in LACA strain of mice were quite high, 96% and 12 respectively; but in C57BL strain of mice were only 5% and 5 respectively. A few hepatic cells and lymphocytes of thymus were labelled.According to the above results the reason of why mammary carcinoma of female LACA strain of mice and adenoma of pituitary gland of C57BL strain of mice could 15e easily induced by estrogenic hormones (estradiol valerate and hydroxyprogesterone caproate, EP), but no mammary carcinoma was induced in C57BL strain of mice by EP, and this can be partly explained by the presence of a large or few number of estrogen receptors. However, the presence of estrogen receptors was only a prerequisite but not a unique condition. Other factors should be considered, because many more estrogen receptors were present in preoptic region of hypothalamus of both strains of mice and anterior lobe of pituitary gland of LACA strain of mice, but few or no tumors could be induced by EP.
2.Effects of Vasopressin on Genes Involve in Vesicle-mediated Transport in the Rat Inner Ear

Objective To study the effects of vasopressin on genes involve in vesicle-mediated transport in the rats' inner ear and to identify the possible mechanism of hydrolabyrinth induced by vasopressin.Methods 16 Wistar rats were divided into experimental group and controled group.Wistar rats of experimental group were intraperitoneally injected with 50 ?g/kg arginine vasopressin once a day for one week.Rats of controled group were injected with the same doses of physiologic saline.Then bullae both groups were immediately dissected out for total RNA extraction.cDNA were synthesized from total RNA of the inner ear by reverse transcription and labeled with Cy3 or Cy5.The BiostarR-40s cDNA microarray was hybridized with the above cDNA.The changes of mRNA expression intensity in the rats' inner ear induced by vasopressin injection were analyzed.Results Seven differentially expressed genes related to vesicle-mediated transport were screened out in the rat inner ear induced by vasopressin injection by use of cDNA microarray.Ap2b1,Arf1,Arf6,Dnch1,Mlc3 and Rab7 were increased by 2-fold or more,and Stx7 was decreased to 0.5-fold or less.Conclusion Vasopressin may disturb fluid homeostasis in the inner ear via vesicle-mediated transport resulting in hydrolabyrinth.
3.Expression of Aquaporins in the Inner Ear of Normal Guinea Pigs and Guinea Pigs with Endolymphatic Hydrops

Objective To study the expression of aquaporins(AQPs) in the inner ear of normal guinea pig and guinea pigs with endolymphatic hydrops, and to investigate the mechanism and significance. Methods The guinea pigs were divided into two groups randomly: model group and control group. The guinea pigs in model group were given desmopressing 4 ?g?kg-1?d-1 introperitoneally for 1 week, while the guinea pigs in control group were given saline. After one week, the expression of AQPs in the inner ears of all guinea pigs was evaluated.Results Expression of AQP0, 1, 2, 3, 5, 7 and 8 were found in the ears of normal guinea pigs. Weak expression of AQP0 was found in stria vascularis and spiral ganglion. The distribution of AQP1 consisted of cellular lining the bony labyrinth, fibrocytes lining in the endolymphatic duct and sac, cells under the basilar membrane, fibrocytes of the spiral ligament and the spiral limbus, Corti's organ, inner and outer spiral sulcus, stria vascularis, saccular and utricular wall, and spiral ganglion. AQP2 were found in stria vascularis, Corti's organ, spiral ganglion and endolymphatic sac. AQP3, 7 and 8 were distributed in a similar manner as the surrounding membranous labyrinth, including Corti's organ, inner and outer spiral sulcus, stria vascularis, fibrocytes of the spiral ligament and the spiral limbus, saccular and utricular wall, endolymphatic sac and spiral ganglion. AQP5 was found at Corti's organ, inner and outer spiral sulcus, spiral ganglion and fibrocytes in spiral ligment. The AQP2 in stria vascularis and endolymphatic sac was stronger in the inner ears with ELH than that in normal ears.Conclusion There are various AQPs in inner ears of normal guinea pigs, and their distributions are overlapping and without obvious regional specificity. Desmopressing induces the expression of AQP2 in the inner ears of guinea pigs. The results indicate that ELH may correlate to upregulation of AQP2, but the mechanism remains unclear.
4.Application of acoustic elastography, conventional ultrasonography and mammography in the diagnosis of the breast cancer

ObjectiveTo explore the value of the diagnosis of breast cancer by acoustic elastography, conventional ultrasonography (including gray-scale ultrasonography, color Doppler flow imaging and Doppler spectrum) and mammography.MethodsOne hundred and four patients with breast disease conifrmed by surgical pathology were studied retrospectively in Zibo Maternal and Children Health Hospital. All the patients were examined by acoustic elastography, conventional ultrasonography and mammography before surgery.ResultsPathological results of 104 cases were: sixty-five malignanies and 39 benignities. The most common signs in the diagnosis of breast cancer by conventional ultrasonography were spicule sign (76.9%, 50/65), taller-than-wide sign (73.8%, 48/65), vascular resistance index>0.7 (70.8%, 46/65), calciifcation (53.8%, 35/65). And breast masses without these signs were regarded as benign lesions. The diagnostic criteria of breast cancer by acoustic elastography was 5-point method. We regarded the lesions got more than 3 points as malignancy. The most common signs in the diagnosis of breast cancer by mammography were dense shadow (73.8%, 48/65), spicule sign (70.8%, 46/65), calcification (69.2%, 45/65). And breast masses without these signs were regarded as benign lesions. The diagnostic accuracy of acoustic elastography, conventional ultrasonography, mammography, conventional ultrasound plus acoustic elastography, conventional ultrasound plus mammography and acoustic elastography plus mammography for breast tumor were 88.4% (92/104), 86.5% (90/104), 83.7% (87/104), 93.3% (97/104), 90.4% (94/104) and 89.4% (93/104), respectively. ConclusionsThe combination of any two imaging modalities can improve the diagnostic accuracy of breast cancer. And conventional ultrasound combined with acoustic elastography could be an effective method in the diagnosis of breast cancer.

BACKGROUND:In isokinetic testing researches,the effect of sport training on the development of teenager muscle groups is less reported. OBJECTIVE:To assess the muscle strength of shoulder,hip and knee joints in different teenager populations with isokinetic testing system. DESIGN,TIME AND SETTING:The contrast observation was performed at the laboratory of Institute of Physical Education,Hebei Normal University between October and December 2006. PARTICIPANTS:Control group included 30 volunteers without sport training randomly selected from four middle schools in Shijiazhuang,15 male and 15 female;training group included 30 volunteers with sprint training randomly selected from Shijiazhuang Sports School,15 male and 15 female. METHODS:We used Kinitech isokinetic testing system(Kylingk,Austrilia) to test the shoulder,hip and knee joint of the 60 participants according to 3 different angular velocities:60( ?) /s,120(?) /s and 240( ?) /s. MAIN OUTCOME MEASURES:Peak torque(PT) values of extensor and flexor groups in shoulder,hip and knee joints. RESULTS:The 60 volunteers were all included in the result analysis. 6.Expression of negative costimulatroy molecule B7-H4 in non-small cell lung cancer tissues and its clinical significance

Objective: To study the expression of negative costimulatroy molecule B7-H4 in non-small cell lung cancer (NSCLC) tissues and its relationship with the clinical features of NSCLC. Methods: Fifty-two NSCLC specimens from pa-tients who were pathologically diagnosed in our hospital during January 2008 to April 2009 were included in the present study. B7-H4 expression and infiltration of CD3~+ T cells in NSCLC tissues were detected by immunohistochemistry. The correlation between B7-H4 expression, CD3~+ T infiltration, and the clinical features of NSCLC was studied. Results: The positive rate of B7-H4 in 52 NSCLC tissues was 48.08% (25/52), and B7-H4 expression in normal lung tissues was neg-ative or low (P <0.05). B7-H4 expression was positively correlated with the clinical tumor stages and lymph node metas-tasis of NSCLC (P < 0.05), and negatively correlated with tumor infiltration of CD3~+ T cells (P < 0.05), but had no re-lationship with clinicopathologie parameters of NSCLC (P > 0.05). Conclusion: Negative costimulatroy molecule B7-H4 may play important roles in the development of NSCLC. Positive expression of BT-H4 is correlated with the clinical tumor stages and lymph node metastasis of NSCLC, which provides a foundation for diagnosis and therapy of NSCLC.
7.The clinical value of procalcitonin in patients with chemotherapy-induced febrile neutropenia

Background and purpose:Previous researches have shown that procalcitonin differentiates infec-tious from non-infectious fever and assesses the severity of infectious diseases. This study aimed to investigate the clin-ical value of procalcitonin in patients with chemotherapy-induced febrile neutropenia.Methods:A total of 147 patients with chemotherapy-induced febrile neutropenia admitted to intensive care unit from Jan. 2012 to Dec. 2014 were di-vided into infectious group and fever of unknown origin group according to clinical symptoms, signs and etiology. The infectious group was divided into sepsis, severe sepsis, and septic shock groups according to the severity of infection. The procalcitonin levels were compared between different groups.Results:A procalcitonin cut-off value>0.935 ng/mL provided a sensitivity of 90.0%, speciifcity of 90.0% and AUC=0.905. The procalcitonin level of the infectious group was signiifcantly higher than that of the fever of unknown origin group [1.805 (1.268-2.523) ng/mLvs 0.555 (0.398-0.818) ng/mL,P<0.001]. There is a signiifcant difference between the severe sepsis group and the sepsis group [13.885 (7.600-17.961) ng/mLvs 1.805 (1.268-2.563) ng/mL,P<0.001]. Compared with the severe sepsis group, the value of procalcitonin in the septic shock group was signiifcantly higher [23.800 (20.050-30.478) ng/mLvs 13.885 (4.955-19.133) ng/mL,P<0.001].Conclusion:Plasma procalcitonin is a useful marker for diagnosing neutropenia in patients with infection. Meanwhile, procalcitonin can be used to assess the severity of infection in patients with neutropenia.
8.Microarray expression analysis identified several key protein candidates as the potential mediators of total flavones of choerospondias axillaries fructus on myocardial protection

Aim To study the molecular basis of protection on myocardial ischemic injuries of total flavones of choerospondias axiaris fructus(TFC). Methods The microarray analysis for the protein expression in myocardium provide a strong tool to explore the key protein candidates involved in the pathogenesis of ischemic injury.Surface enhanced laser desorption/ionization(SELDI) mass spectrometry with protein chip IMAC3,SAX2 and NP20 was used to compare the differentially expressed protein in TFC-treated and untreated ischemic myocardium in rats and the results were analysized with Proteinchip Software 3.0.2.Results Seven differentially expressed proteins were indentified in TFC treated myocardium.These differential effects correlated with the expression of five downregulated proteins and two upregulated proteins,and four of them were discovered on the IMAC3 chip and one of them was discovered on the SAX2 chip.Conclusions The myocardial protection of TFC may be mediated by the differential expression of these proteins which could be the key protein candidates for further investigation.
9.Influence of expiratory trigger sensitivity setting on inspiratory cycling-off synchronization during noninvasive pressure support ventilation with different respiratory mechanics

Objective To compare the inspiratory cycling-off synchronization with expiratory trigger sensitivity (ETS) setting during noninvasive pressure support ventilation (PSV) under the different conditions of respiratory mechanics. Methods Bi-level pressure ventilator was connected to a lung model (Hans Rudolph Series1101 simulator). And its mechanics was set to normal, obstructive and restrictive ventilation dysfunction. Tests were performed with pressure support levels of 10 (in normal condition, 1 cmH2O = 0.098 kPa) and 15 cmH2O, 5 cmH2O positive end-expiratory pressure (PEEP). The data including cycling delay time (Td), tidal volume (TV) and airflow were collected in the presence of air leaks (24-28 L/min). Results Significant performance on airflow was observed in various respiratory mechanics conditions. The peak inspiratory flow (PIF) and peak expiratory flow (PEF) were different, and the tidal volume, PEF and inspiratory time were increased after decreasing the ETS level. Premature cycling occurred frequently in normal and restrictive model. Delay cycling was found only when the ETS setting was at the lowest level (about 5% of PIF), at which Td were (15.20 ± 0.43) ms and (105.00 ± 15.82) ms. In obstructive model, PIF and PEF were significantly decreased, and delay cycling was always present. Td were increased after the ETS setting was changed from the most sensitive level to moderate level:(51.30 ± 12.63) ms vs. (162.40 ± 15.59) ms, as cycling-off criteria were reduced from (34.52 ± 3.36)% to (16.04 ± 2.58)% of PIF, and there were statistical differences (P<0.05). Severe delay cycling was found when the ETS level was at the lowest level. Conclusions There are significant differences in performances and cycling-off synchrony in various respiratory mechanics models during PSV. For Curative Flexo ST30 bi-level pressure ventilator, its flow cycling-off criteria range is about 5%-35%of PIF. The higher ETS level might be beneficial to improve patient-ventilator synchrony in patients with obstructive airway diseases, which could result in premature cycling in patients with restrictive disease.
10.Effect of puerarin on proliferation, differentiation andmineralization of MC3T3-E1 cells and expression of TRPM3 mRNA

Aim To investigate the effect of puerarin on proliferation, differentiation, and mineralization of MC3T3-E1 cells and the expression of TRPM3 mRNA.Methods Proliferation, differentiation, and mineralization of puerarin in MC3T3-E1 cells were determined using CCK-8 assay, alkaline phosphatase(ALP) activity assay, and Alizarin Red Staining, respectively.Effect of puerarin on cell cycle and intracellular calcium concentration of MC3T3-E1 cells was detected by flow cytometry.RT-PCR was used to detect the effect of puerarin on TRPM3 mRNA expression.Resuls 0.1, 1, 10 μmol·L-1 puerarin significantly promoted the proliferation of MC3T3-E1 cells, reduced the proportion of cells in G1 phase, increased the proportion of G2 and S phase, of which 0.1 μmol·L-1 concentration effect was the most significant.Compared with control group, the ALP activity and mineralized nodule area of 0.1 μmol·L-1 puerarin group were significantly increased.The expression of TRPM3 mRNA and the intracellular calcium concentration were significantly decreased in 0.1 μmol·L-1 puerarin group.Conclusion Puerarin can promote the proliferation, differentiation, and mineralization of MC3T3-E1 cells, and reduce the expression level of TRPM3 mRNA and intracellular calcium concentration.
