1.Effect of autologous cytokine induced killer cell therapy in patients with lung cancer

Objective To evaluate the clinical effect of cytokine induced killer cellular immunotherapies ( CIK) on the patients with advanced lung cancer.Methods Peripheral blood mononuclear cell from 32 cases of patients with advanced lung cancer were isolated.Cells were cultured in DMEM supplemented with IFN-γ,IL-2,anti-CD3 monoclonal antibody and other cytokines,and transfused back into the patients in several times after 15 days of culture.Subsequently,we observed the size of tumor,the change of immunologic function by CIK treat-ment,the status of clinical symptoms,and the toxic and side effect.The clinical curative effect was also evaluated. Results The symptoms in 32 patients who received CIK therapy were reduced,and the side effects were light. Immunophenotype analysis showed that CD3 /CD4 coexpression were found in 34.4%patients,CD3 /CD8 co-expression in 68.2%,and CD3 /CD56 co-expression in 13.7% patients.CT images about 15 days after CIK cell transfusion showed that pleural effusion were decreased.Clinical evaluation effective rate was 56.0%,and disease control rate( CR +PR +SD) was 84.3%.Conclusion CIK cell immunotherapies of cancer patients can improve immunologic function of cancer patients and inhibit tumor growth and relieve the clinical symptom of patients,and improve the quality of life.
2.Detenial sigmoid neobladder report of 50 cases

Objective To investigate the clinlic therapeutic result of detenial sigmoid neobladder after radical cystectomy. Methods 50 patients with bladder cancer (41 males and 9 females;age range from 35 to 82 years) were admitted and underwent radical cystectomy and detenial sigmoid neobladder. Results All the 50 patients were followed up for a mean of 18.6 months(range from 6 to 29 months).The blood Cr and BUN levels were both in the normal range without acidosis in all the cases.Only 2 had unilateral ureteral urine reflux during cystography.Of the 50 patients 45 cases(90%) achieved continence during daytime;10 cases(20%) were incontinent at night,but 8 of the 10 cases could control urination by being woken up at night.The capacity of the neobladder was 220 to 550 ml with a mean of 350 ml and the maximal pressure of the neobladder during filling was 15 to 55 cmH 2O(1 cmH 2O=0.098 kPa) with a mean of 35 cmH 2O. Conclusions As a simple operation the detenial sigmoid neobladder can be performed easily with less complications and more reliable results.This operation may be generally applied in clinical practice.
3.Study of Human Cytomegalovirus mRNA Transcription Profile at Different Infection Stages in Infants

Objective Human cytomegalovirus (HCMV) can induce infection in multiple organs, including genitouri-nary system, central nervous system and liver. It can even lead to serious defects and death. China, as a high-incidence area of HCMV, need employ early treatment. However, the sensitivity of present HCMV diagnosis is not satisfied. Methods In the present study, we focused on transcription level of HCMV IE and pp67 mRNA using real-time quantitative PCR. Results IE mRNA was positive even in a portion of patients who were regarded as negative by traditional DNA diagnosis method. On the other hand, pp67 mRNA that were normally expressed at the late stage of HCMV infection was 100%posi-tive in the first-visit patients who were regarded as positive using traditional DNA diagnosis method. Conclusion The pres-ent study indicated that mRNA detection based on PCR method was more sensitive compared with DNA detection for assess-ing HCMV virus infection. Finally, different treatment should be performed upon mRNA transcription profile at different in-fection stages.
4.Evaluation on angiogenesis of breast carcinoma and precancerous lesion with power Doppler imaging and contrast enhanced microvascular imaging

Objective To investigate the angiogenesis characteristics of normal breast,simple hyperplasia,atypical hyperplasia,intraductal carcinoma in situ and invasive ductal carcinoma.Methods Twenty cases in normal controls,20 cases with simple hyperplasia,20 cases with atypical hyperplasia,20 cases with intraductal carcinoma in situ and 30 cases with invasive ductal carcinoma were inspected with power Doppler imaging(PDI)and contrast enhanced microvascular imaging(MVI)on the quantity,configuration and distribution of angiogenesis.The microvessel density(MVD)were detected by immunohistochemical technique.ResultsWith the progression of breast cancer,MVD and grades of PDI kept increasing.Meanwhile,the blood vessels became twisted and increased in quantity with expansion into the lesion,the configuration of blood vessels became disordered.However,the changes mentioned above were not obvious in the very early stages.they became significant from the atypical hyperplasia(AH)stage.Conclusions Contrast MVI and PDI may be useful to screen high risk population with high risk of breast cancer due to their detection of changes in angiogenesis of breast carcinoma and precancerous lesions.
5.Role of RB in the cell cycle and its relation with colorectal cancer

Since its discovery, the retinoblastoma (RB) tumor-suppressor protein has become a focal point of caner and cell cycle research, and subsequently discovered members(pRB,p107,p103) of the RBfamily have been proved to play a important role in the cell cycle and the development of cancer. RB regulates the cell growth, development and differentiation via the interactions with E2f and some other cytokines,and its alternation even make the tumor formation. In comparison with some other tumors, colorectal cancer(CRC) has the different expression of RB.We may assume that the different causes of CRC and some unique functions of RB in CRC lead to the alternation.

BACKGROUND: The inheritance tendency and immunity imbalance of atherosclerotic brain infarction (ABI) have attracted the attention of many scholars at home and abroad, which suggests that immune mechanism plays a key role in ABI development. It has been found that ABI is related to polymorphism of human immune inheritance gene (HLA), particularly HLA-DR gene.OBJECTIVE: To investigate the correlation between HLA and ABI immune heredity.DESIGN: Single sample and single factor analysis.SETTING: Sino-Japanese Friendship Hospital of Jilin University; Neurological Department of the First Hospital of Harbin City.PARTICIPANTS: Totally 31 patients in experimental group and 30 healthy people in control group, whose three generations were of the Han Nationality in north China and who had no blood relation, were selected from Department of Neurology in First Hospital of Harbin between January 1998 and December 2000.METHODS: The experiment was conducted in the Blood Research Center of Department of Neurology in First Hospital of Harbin between June and December 2003.5 mL peripheral blood in both groups was taken to extract genome DNA with the previous method. Human HLA gene map had been described at gene bank in the Internet, in which mono-nucleic acid obtained from HLA of the sixth chromosome were retrieved. Each DR and DQ allele, sense primer and antisense primer were composed selectively. Amplification of DNA fragment was checked respectively and specifically. The primer was provided by Shanghai Gene Research Institute. PCR instrument (4 800) and its related reagents produced by PE Company (the US) were used. Fourfold table exact probability method was used to calculate the relative risk (RR) and P value.which was obviously higher than that of other sites (P < 0.05). However,associated with the ABI susceptible gene of the Han Nationality in north China.

Apoptosis of neutrophils controls the duration and the intensity of an inflammatory response and therefore the extent of neutrophil- mediated tissue damage, disturbance of neutrophil apoptosis has been associated with many diseases, underlying mechanism is not elucidated. C5a is a complement fragment that has multifunctional properties, which induces neutrophil chemoattraction, an oxidative burst, enhancement of phagocytosis, release of granule enzymes, and suppress neutrophil apoptosis. Several studies have reported calpain is involved in both neutrophil functions and apoptosis and it might play a more specific role in the regulation of neutrophil apoptosis. Diffenrent isoform of calpains is activted by diffenrent stimuli through different transduction pathway. It was reported previously that calpain is required for neutrophil migration and chemotaxis induced by C5a. In addition, autophagy is a ubiquitous physiological process that occurs in all eukaryotic cells and is considered to be a survival mechanism. Atg5 promotes autophagy and is indispensable to autophagosome formation. Upon calpain activation, Atg5 is cleaved and the resulting 24 ku Atg5 mediates apoptosis while losting the property of autophagy. Therefore, Atg5 represents a molecular switch between autophagy and apoptosis. The interaction among the C5a, calpain and Atg5 was introduced and new direction for further research was provided.
9.The association of HLA-DRB1 alleles with macrovascular complication of type 2 diabetics in Han race of Chinese north area

Objective:To study the relationship between HLA-DRB1 gene polymorphisms and macrovascular complication of type 2 diabetics. Methods:HLA-DRB1 alleles in 88 cases of type 2 diabetics, including 52 ones without complication and 36 ones with macrovascular complication, were examined by PCR-SSP. Results:There were at least 11 HLA-DRB1 alleles in type 2 diabetics. The HLA-DRB1 *03 and HLA-DRB1 *09012 gene frequencies of type 2 diabetics with macrovascular complication were significant higher than those without complication.Conclusion:HLA-DRB1 *03 and HLA-DRB1 *09012 alleles or their linkage disequilibrium genes may enhance the risk of the development of type 2 diabetes macrovascular complication.

OBJECTIVE:
Through the application of eustachian tuboplasty assisted with hypothermy plasma ablation, we evaluate its efficacy in the threatment of adult refractory otitis media with effusion.
METHOD:
We retrospectively reviewed the charts of 48 cases (61 ears) suffering from adult refractory otitis media with effusion from January 2012 to December 2013. According to the admission date, the patients were divided into the control group (17 cases, 22 ears) and the treatment group (31 cases, 39 ears). In the control group, the patients were treated with drugs and physical therapy. In the treatment group, the hypothermy plasma ablation technology was used to ablates the hypertrophic tissues around the eustachian orifice besides the pharmaedogical interventions. The recurrence rate of the two groups were analyzed and compared in 1 year after treatment. Pure tone audiometry and acoustic immittance measurement were taken for the two groups in differernt periods (one week prior to operation, one months, three months, six months and one year after operation respectively) to evaluate their hearing change and the recurrence rate (within six months after treatment and one year respectively) objectively.
RESULT:
In the control group, 2 cases (2 ears) were lost to follow-up, and the recurrence rate was 65% (13/20) in 1 year. In the treatment group, four case were lost to follow-up,and the recurrence rate was 14.3% (5/35) in 1 year. The difference was statistically significant (P<0. 01). There were similar hearing improvement in the two groups after treatment in 1 month, but the hearing improvement in the treatment group increased with time. There were no complications occuring during the operation and post-operation. Within a month after operation, the majority of ears(28/35) obtained significant hearing improvement with the decreasing air-bone-gap comparision with the pre-operation (P < 0.01), and the preoperative tympanogram of 16 ears with type B or C turned to type A (P < 0.01). There were no both statistical significance in the variation of air-bone-gap and tympanogram of comparison with results between three months and six months, six months and one year postoperatively (P > 0.05).
CONCLUSION
This study confirmed the efficacy of eustachian tuboplasty assisted with hypothermy plasma ablation in the threatment of adult refractory otitis media with effusion caused by eustachian tube opening disorder was significant. It can reduce the recurrence rate significantly in one year and allow sustained hearing improvement within three months postoperatively.
