1.Inhibitive effect of peritoneal decoy-oligoligomucleotidesnucleotides targeting nuclear factor-?B on expression of inflammatory mediators in macrophages ? cells

Objective To design and synthesize targeting nuclear factor-?B (NF-?B) decoy-oligodeoxynucleotides (ODNs) and measure their anti-inflammatory actions. Methods Peritoneal macrophages ? (pM?) cells extracted from rats were randomly divided into normal control group, lipopolysaccharides (LPS)-stimulated group (Group A), decoy-ODNs treated group (Group B), non-decoy-ODNstreated group (Group C) and cationic liposome treated group (Group D). Supernatants and cells in the control group and the Group A were collected 1,2,6,12,18 and 24 hours after LPS stimulation. By using cationic liposomes in a ratio of 4:1 in quantity, pM? cells were transfected by decoy-ODNs at concentrations of 2 mg/L,4 mg/L and 8 mg/L respectively and stimulated with LPS 6 hours later. Then, supernatants and cells were collected 8, 12 and 18 hours after transfection. The expression changes and the distribution of p65 were analyzed by immunocytohistochemical method, the protein expressions of tumor necrosis factor ? (TNF?), interleukin-6 (IL-6) and interleukin-10 (IL-10) in the supernatants by enzyme-linked immunosorbent assay (ELISA) and mRNA transcriptions of TNF?,IL-6 and IL-10 by reverse transcriptase polymerase chain reaction (RT-PCR). Results Decoy-ODNs at concentration of 4 mg/L and 8 mg/L could markedly inhibit the expression and transcription of TNF? and IL-6 of pM? cells in a dose-dependent fashion but had a weak inhibitive effect on the expression and transcription of IL-10. Conclusions The targeting NF-?B Decoy-ODNs can suppress the expressions of inflammatory mediators in pM? cells.
2.Rat model of exercise-induced myocardial ischemia injury

BACKGROUND:No uniform standard for constructing the animal model of exercise-induced myocardial ischemia injury results in the incomparability among research results and impedes the development of sport medicine especial y in the cardiovascular field;thereby, it is imperative to reach an agreement in constructing criteria.
OBJECTIVE:To explore the method of establishing the rat model of myocardial ischemia induced by running.
METHODS:Total y 96 female Sprague-Dawley rats were randomly divided into rest control group, isoprenaline group and 10 exercise groups (1-and 3-time moderate-intensity exercise groups, 1-, 2-and 3-week moderate-intensity exercise groups, 1-and 3-time high-intensity exercise groups, 1-, 2-and 3-week high-intensity exercise groups). After exhaustive exercise, myocardium was col ected for morphological observation by hematoxylin-eosin staining, serum levels of myocardial enzymes and cardiac troponin I were detected, and the expressions of Bcl-2 and Bax were detected by real-time PCR.
RESULTS AND CONCLUSION:(1) Hematoxylin-eosin staining showed that the damage degree was more severe with the time of exercise, and the high-intensity exercise groups were more severe than those in the moderate-intensity exercise groups. (2) The activity of serum glutamic-oxaloacetic transaminase and lactic dehydrogenase was significantly increased after 1-week moderate-intensity exhaustive exercise (P<0.05 or P<0.01). From the beginning of the 3-time high-intensity exhaustive exercise, the activity of glutamic-oxaloacetic transaminase and lactic dehydrogenase was significantly increased (P<0.05 or P<0.01). (3) Cardiac troponin I content change trend was basical y the same as glutamic-oxaloacetic transaminase and lactic dehydrogenase changes, but cardiac troponin I content in the moderate-intensity exhaustive exercise groups was significantly higher than that in the rest control group until 2 weeks. The Bcl-2/Bax ratios in al exercise groups were significantly lower than that in the rest control group (P<0.05 or P<0.01);those in the 1-and 3-time high-intensity exercise groups were significantly higher than in the isoprenaline group (P<0.05 or P<0.01);and those in moderate-intensity groups were higher than in the isoprenaline group (P<0.05 or P<0.01). (4) In conclusion, 2-week high-intensity and 3-week moderate-intensity exhaustive exercise can induce myocardial ischemia injury, and pathological analysis, serum levels of myocardial enzymes and cardiac troponin I can be used as the evaluation indexes, while apoptosis regulation genes just as the reference index.
3.Interpersonal sensitivity and adverse childhood experiences among medical college students: the mediating role of negative emotions

Objective To explore the relationship between adverse childhood experiences,negative emotions and interpersonal sensitivity.Methods Adverse Childhood Experiences Questionnaire (ACEQ),Depression Anxiety Stress Scale (DASS-21) and the Interpersonal Sensitivity Subscale of Symptom Check-List (SCL-90) were applied to investigate the 492 new enrolled medical students.Results Boys got higher ACEQ scores than girls; Compared with only-child,non-only-child scored higher in depression,interpersonal sensitivity; Compared with students from non-rural areas,those from rural areas got higher scores in depression,anxiety stress,interpersonal sensitivity.There existed significant differences of scores on ACEQ,depression,anxiety,stress,interpersonal sensitivity among children of fathers of different educational level; while differences of scores on depression,anxiety,interpersonal sensitivity among children of mothers of different education level were statistically significant.There were significantly differences of scores on depression,anxiety,stress,interpersonal sensitivity among participants with 0,1,2 and 3 or more ACEQ.The results of correlative analysis provided evidence that adverse childhood experiences,all the factors of negative emotions and interpersonal sensitivity were significantly positively correlative.The SEM suggested that adverse childhood experiences had an indirect effect on interpersonal sensitivity through negative emotions.Conclusions Adverse childhood experiences has a great influence on negative emotions and interpersonal sensitivity.Negative emotions has a completely mediating role between adverse childhood experiences and interpersonal sensitivity.
4.Investigation of diabetes nutrition knowledge, attitude and practice in outpatients with type 2 diabetes

Objective To conduct an investigation on diabetes nutrition knowledge, attitude and practice in outpatients with type 2 diabetes, understand the nutritional knowledge and dietary behaviors in patients with type 2 diabetes to guide individualized nutrition education. Methods The diabetes nutrition Knowledge, Attitude and Practice(KAP) questionnairewas designed based onChinese dietary guidelines (2007)andChinese diabetes medical nutrition therapy guidelines (2013). Data of out-patients with type 2 diabetes from January to March 2015 were collected. The method of investigation was face-to-face interview. Results Totally 915 patients were enrolled into this survey(male 495 and female 420). The mean BMI was (27.23 ± 3.18)kg/m2, glycosylated hemoglobin A1c (HBA1c) was (8.03 ± 2.17)%. Among the questions on knowledge, only on 4 questions the rate of awareness was over 80%, and there was a misunderstanding on diet, only 54.10%of diabetes patients believed that diabetes diet should be balanced and reasonable, rather than just limiting staple food and meat. Patients generally could realize the importance of diet for diabetes treatment. For questions on practice, the rate of reaching the standard generally was not over 80%. The rate of correct answers was not significantly different among different age groups, gender, and durations. There was a linear correlation between the rate of correct answers and glycosylated hemoglobin (P=0.043), the correlation coefficient was-0.258. The higher the rate of correct answers, the lower the level of glycosylated hemoglobin. Conclusion Outpatients with type 2 diabetes had good awareness level about diabetic nutritional knowledge and attitude but the awareness level of balanced diet needs to improve, the awareness rate on behavior was relatively low.
5.Calpain-mediated Cleavage of Atg5 Determine Autophagy or Apoptosis of PMN, and C5a's Role

Apoptosis of neutrophils controls the duration and the intensity of an inflammatory response and therefore the extent of neutrophil- mediated tissue damage, disturbance of neutrophil apoptosis has been associated with many diseases, underlying mechanism is not elucidated. C5a is a complement fragment that has multifunctional properties, which induces neutrophil chemoattraction, an oxidative burst, enhancement of phagocytosis, release of granule enzymes, and suppress neutrophil apoptosis. Several studies have reported calpain is involved in both neutrophil functions and apoptosis and it might play a more specific role in the regulation of neutrophil apoptosis. Diffenrent isoform of calpains is activted by diffenrent stimuli through different transduction pathway. It was reported previously that calpain is required for neutrophil migration and chemotaxis induced by C5a. In addition, autophagy is a ubiquitous physiological process that occurs in all eukaryotic cells and is considered to be a survival mechanism. Atg5 promotes autophagy and is indispensable to autophagosome formation. Upon calpain activation, Atg5 is cleaved and the resulting 24 ku Atg5 mediates apoptosis while losting the property of autophagy. Therefore, Atg5 represents a molecular switch between autophagy and apoptosis. The interaction among the C5a, calpain and Atg5 was introduced and new direction for further research was provided.
6.Accuracy of remifentanil target-controlled infusion system in children

Objective To evaluate the accuracy of remifentanil target-controlled infusion (TCI) system in children.Methods Thirty ASA Ⅰ patients, aged 3-12 yr, weighing 10-40 kg, scheduled for elective ear-nosethroat or urological surgery, were randomly divided into 2 groups with 15 patients in each group:2 ng/ml remifentanil group (group Ⅰ) and 4 ng/ml remifentanil group (group Ⅱ). Anesthesia was induced with iv injection of propofol 2 mg/kg and TCI of remifentanil. Remifentanil was administered with a specific TCI system incorporating the pharmacokinetic parameters of Minto.The target plasma concentrations of remifentanil were set at 2 or 4 ng/ml. Tracheal intubation was facilitated with vecuronium 0.1 mg/kg after the children lost consciousness. The children were mechanically ventilated.Anesthesia was maintained with TCI of remifentanil, iv infusion of propofol and intermittent iv boluses of vecuronium. The target plasma concentration of remifentanil remained unchanged and bispectral index value was maintained at 45-65 or auditory evoked potentials index value < 30 by adusting the infusion rate of propofol.Arterial blood samples were taken at 5, 10, 20, 30, 40, 50 and 60 min after TCI remifentanil was stared for determination of blood remifentanil concentrations by high performance liquid chromatography. Median prediction performance error (MDPE),median absolute performance error (MDAPE) and wobble of remifentanil TCI system were calculated. Results The measured concentrations of remifentanil were significantly higher than the target plasma concentrations in both groups (P<0.05). The MDPE, MDAPE and wobble were 20.0% , 30.0% and 25.0% respectively in group Ⅰ , and 17.5%, 17.5% and 12.5% respectively in group Ⅱ . TheMDAPE and wobble were significantly decreased in group Ⅱ compared with group Ⅰ(P<0.05).Conclusion When remifentanil is administered using a specific TCI system incorporating the pharmacokinetic parameters of Minto in children of 3-12 years old, the accuracy is not high.
7.The abnormalities of noninvasive urodynamics in early diabetes cystopathy

Objective To study the abnormalities of noninvasive urodynamios in early diabetes eystopathy (DCP) and provide diagnosis evidences in its early stage. Methods According to the disease course (less or more than 1 year), 85 patients with type 2 diabetes mellitus (T2DM) were divided into new lydiagnosed diabetic group and non-newly diagnosed diabetic group. Thirty healthy cases were involved in normal control group. All of them were checked with the technology of noninvasive urodynamics to measure maximal flow rate, average flow rate, the volume leading to first bladder sensation and residual urine volume. Results As to the 32 newly diagnosed diabetic group, maximal flow rate was (18.4±6.9) ml/s, and average flow rate was (10.6 ± 5.3) ml/s, 18 cases were detected to have bladder residual urine, with the average residual urine volume of (13.2 ± 17.3) ml, and the DCP detection rate was 56.2%(18/32). As to the 53 non-newly diagnosed diabetic group, maximal flow rate was (14.7 ± 6.6) ml/s, and average flow rate was (9.5±4.7) ml/s,38 cases were detected to have bladder residual urine, with the average residual urine volume of (19.3 ± 18.4) ml, and the DCP detection rate was 71.7%(38/53). There was no residual urine detected in normal control group. Their maximal flow rate was (25.7 ± 5.9) ml/s, and average flow rate was (18.0 ± 4.9)ml/s. Compared with that in normal control group, maximal flow rate, average flow rote and residual urine volume decreased in both newly diagnosed diabetic group and non-newly diagnosed diabetic group(P< 0.01). As compared with that in newly diagnosed diabetic group, maximal flow rate in non-newly diagnosed diabetic group decreased obviously (P< 0.05). Conclusions The abnormalities of urodynamics may happen in the early stage of diabetes, and maximal flow rate may be as the most sensitive index. With the new technology of noninvasive urodynamies, we DCP can be diagnosed in early stage and evaluated the function of bladder dynamically.

Objective To study the changes in expression of high-sensitivity C-reactive protein (hsCRP),glutathione S-transferase Pi(GSTPi)and annexin A5 (AnxA5)in elderly patients with old myocardial infarction and the clinical significance.Methods Serum levels of GSTPi and AnxA5 were measured by ELISA and the level of hs-CRP was measured by immunoturbidimetry in elderly patients with old myocardial infarction (n =185)from December 2012 to November 2015.Results Along with the increasing coronary artery stenosis,GSTPi level was decreased and AnxA5/hs-CRP levels were increased in elderly patients with old myocardial infarction.In comparison between coronary artery stenosis > 95% group versus stenosis of 55%-65% group,GSTPi was(190.0±37.0)μg/L vs.(289.0 ±86.0)μg/L,AnxA5 was(33.9±4.0)μg/L vs.(8.1 ± 2.9) μg/L,and hs-CRP was (15.3 ± 1.3) mg/L vs.(5.9 ± 0.8) mg/L with statistically significant differences(all P<0.01).There were significant differences between LVEF 30% group[GSTPi(198.0±39.0) μg/L,AnxA5(38.9±5.1)μg/L and hs-CRP(17.9± 1.9)mg/L]and LVEF 40%-54% group[GSTPi(219.0± 61.0)μg/L,AnxA5 (12.9±3.9)μg/L and hs-CRP(10.1 ± 1.0) mg/L] (all P<0.01).There were significant differences between NYHA Ⅳ group [GSTPi (171.0 ± 43.0) μg/L,AnxA5 (18.1 ± 5.0) μg/L and hs-CRP (16.9±2.1)mg/L]and NYHAⅠgroup[GSTPi(295.0±91.0)μg/L,AnxA5(7.3±3.1)μg/L and hs-CRP (7.8± 1.3)mg/L](all P<0.01).Conclusions The expression of GSTPi,AnxA5 and hs-CRP in elderly patients with old myocardial infarction may become the new indicators to forecast the degrees of coronary artery stenosis and heart failure.
10.Low-dose Spiral CT Scan of Chest:Parameter's Optimization

Objective To probe into the optimization of scanning parameters of chest with low-dose spiral CT.Methods The phantom was scanned with different scanning parameters.The image quality obtained with the standard dose parameters and the low-dose technique were compared to evaluate the spatial resolution,low contrast resolution,noise level field uniformity and radiation dose,then the optimal scan parameters were defined.The scanning parameters were 120 kV,0.75 s,Pitch 1.0,FOV 360,RF3 reconstruction model(standard filter).Different thickness and tube electricity were used and the data were analyzed with the statistical method.Results There was significant difference about the data of the CTDI between the 160 mA conventional spiral CT and the low dose groups(30 mA,50 mA,70 mA,90 mA).There was no significant difference about the CTDI between 30 mA and 50 mA,and between 50 mA and 70 mA.There was obviously significant difference about the image noise on 1 mm slice and 10mA with other groups.The spatial resolution and low contrast resolution increased along with the increase of tube electricity,and 50mA was a inflexion of low contrast resolution.Conclusion 50mA and 5mm slice are the best scanning parameters in low-dose lung scanning and adapt to routine screening and early lung cancer screening.
