1.Peripheral blood CD~+_(34)-positive mononuclear cells participate in neovasculogenesis of human ovarian epithelial carcinoma

Objective To observe whether endothelial cell (EC) progenitors (CD~+_(34)-positive mononuclear cells) participated in neovasculogenesis of ovarian epithelial carcinoma through in vitro and in vivo experiments, and to explore the mechanism of tumor neovasculogenesis. Methods CD~+_(34)-positive mononuclear cells were isolated from peripheral blood of ovarian epithelial carcinoma patients by means of magnetic beads coated with antibody to CD~+_(34), plated on culture dishes coated with human fibronectin in endothelium medium, and examined by using RT-PCR, fluorescence-activated cell sorting (FACS) and nitric oxide (NO) assay kit for the expression of EC lineage-markers. EC-like cells were labeled with DiI ex vivo, and injected into immunodeficiency mice model with transplanted hypodermic SKOV3 by caudal vein. After 4-6 weeks, the tumor was resected and examined by confocal microscopy, and immunohistochemistry. Results In vitro, CD~+_(34)-positive mononuclear cells differentiated into ECs. In animal models of SKOV3, EC progenitors (CD~+_(34)-positive mononuclear cells) incorporated into sites of neovasculogenesis in tumor, 4-6 weeks later DiI-labeled cells incorporated into capillaries and small arteries. Conclusions The neovasculogenesis in human ovarian epithelial carcinoma involves angiogenesis and vasculogenesis.
2.Mediating effect of feeling of inadequacy on relationship between self consistency and anxiety in medical students

ObjectiveTo explore the relationship among self consistency and congruence,the feeling of inadequacy(FIS) and anxiety as well as the mediating effect of FIS between self consistency and congruence and anxiety.MethodsA total of 1164 students recruited from two medical colleges in Hebei province using cluster random sampling were investigated with scale for the feeling of inadequacy,self consistency and congruence scale and rating anxiety scale.ResultsThe relevance ratio of anxiety in Hebei medical colleges students was 42.7%.There were significant differences in the detection rate among genders(male:46.4%,female:40.5%),grade(freshman:38.9%,sophomore:41.0%,junior:47.5%)and family economic status(Preferably:38.5%,commonly:40.1%,badly:50.8%) in medical students(all P<0.05).There were no statistical significance for the incidence rates of students from different origins(P>0.05).Anxiety score was positively correlated with the score of self consistency and congruence(r=0.274,P<0.01),and was negatively correlated with the score of the feeling of inadequacy (r=-0.364,P<0.01).And the score of self consistency and congruence was negatively correlated with the score of feeling inadequacy(r=-0.335,P<0.01).Multiple stepwise regression analysis revealed that factors of the feeling of inadequacy and self consistency and congruence accounted for 35.6% of anxiety variance.The feeling of inadequacy played a mediating role in self consistency and congruence and anxiety.The mediating rate was 60.06%.ConclusionThe anxiety issue of medical students should be paid more attention.It is possible for the high level of feeling of inadequacy and low level of self consistency and congruence to increase the occurrence rate of anxiety.The feeling of inadequacy can strongly predict medical students'anxiety.
3.The Preparation and the Anti-Tumor Targeting Therapy of Adriamycin Long Circulating Temperature-Sensitive Liposome

Objective: To develop adriamycin liposome (AL) , adriamycin long circulating liposome (ALCL) and adriamycin long circulating temprerature-sensitive liposome ( ALTSL) and to study their anti-tumor effects on tumor-bearing mice. Methods: The antitumor activity was observed using the tumor weight as index. The life prolongation rate of mice was calculated according to the tumor-bearing mice survival time. The tissue distribution of adriamycin was determined by HPLC method. Tumor, heart, liver and kidney tissue of the tumor-bearing mice, were sliced and prepared to observe the tissue pathology differences. Results: Compared with free adriamycin, the anti-tumor effects of ALCL and ALTSL were remarkably increased. Their tumor growth inhibitory rates were 57. 8% and 67. 0% respectively. The study of pharmacoki-netics indicated that the adriamycin concentrations were remarkably higher in tumor tissue and blood,lower in heart and lung tissue of ALCL and ALTSL groups when compared with the free ADM group; The pathology slices indicated that tumor cells in the ALTSL group with hyperthermia were mostly destroyed; the cardiac muscle cells in the ALTSL group were similar to the normal cardiac muscle. Conclusion: ALCL and ALTSL remarkably increased the adriamycin concentration on the tumor site, significantly enhanced the anti-tumor effects, decreased the side-effects (such as cytotoxicity) when compared with free ADM, they also significantly prolonged the survival time of the tumor-bearing mice.
4.Mediating effects of self feeling of inadequacy between resilience and alexithymia in depressive undergraduates

Objective To study the relationship among resilience,self feeling of inadequacy and alexithymia in depressive undergraduates,and to provide a theoretical basis for the prevention and intervention of depression among undergraduates.Methods 500 undergraduates of two universities in Hebei Province were collected by cluster sampling method.Beck depression scale,adolescent resilience scale,Toronto alexithymia scale and self feeling of inadequacy scale were applied to undergraduates.The survey results were analyzed statistically.Results The detection rate of depression was 28%(119/426) in undergraduates.Total score of Alexithymia Scale(69.99±9.43) was higher than the norm(65.70±7.98),and the difference was statistical significant (P<0.05).The score of self feeling of inadequacy had a significant difference between male (149.88±28.00) and female(138.58±28.79) (P<0.01).Neither gender nor grade had significant difference in depression,alexithymia and resilience(P>0.05).Self feeling of inadequacy was positively correlated with alexithymia (P<0.05).Resilience was negatively correlated with self feeling of inadequacy and alexithymia(P<0.05).For the depressive undergraduates,self feeling of inadequacy served as a full mediator between the resilience and alexithymia,whereas for the non depression undergraduates it was a partial mediator.Conclusion Depression undergraduates have serious alexithymia,serious self feeling of inadequacy and poor resilience.Self feeling of inadequacy serves as a mediator between the resilience and alexithymia in depressive and non-depressive undergraduates.
5.The significance of genetic expression of IGF-Ⅰ , ERK, and GLUT4 in adipose tissue of patients with metabolic syndrome and colorectal cancer and its significance

[Summary] To explore the mRNA expression of insulin-like growth factor-Ⅰ (IGF-Ⅰ),extracellular signal regulated kinase (ERK),and glucose transporter 4 (GLUT4) in greater omental adipose tissue of patients with metabolic syndrome and colorectal cancer.The mRNA expression of IGF-Ⅰ,ERK,and GLUT4 in greater omental adipose tissue of the subjects was measured by RT-PCR.(1) The mRNA expression level of IGF-Ⅰ and ERK in the metabolic syndrome group was significantly higher than that in the control group (P<0.01),while in colorectal cancer subgroup the expression was significantly higher than that in the non-colorectal cancer subgroup (P< 0.01).The expression of GLUT4 was obviously lowered (P<0.01).(2) The expression of ERK was positively correlated with that of IGF-Ⅰ (r =0.608,P<0.01).The fasting insulin was positively correlated with the expression of ERK and IGF-Ⅰ(r =0.538,0.439,P < 0.01),and negatively with that of GLUT4 (r =-0.457,P < 0.01).There may be relationship between ERK plus IGF-Ⅰ and metabolic syndrome complicated with colorectal cancer.The lowered GLUT4 expression may be related to insulin resistance in metabolic syndrome.
6.The value of prenatal ultrasound in diagnosis of fetal meningocele or encephalocele

Objective To investigate the clinical value of prenatal ultrasound in fetal meningocele and encephalocele.Methods Thirty nine fetuses with meningocele and encephalocele confirmed by prenatal ultrasound and abortion were acquired and their clinical data and sonographic features were retrospectively analyzed.Results Ultrasound screening in 111 620 cases showed 39 fetuses were with meningocele and encephalocele,accounting for 0.35%.Among them,16 cases were diagnosed with meningocele (including 1 case with two bulging parts) and 23 cases with encephalocele.Prenatal ultrasound could clearly detect the location and size fetal skull defect,and bulging features.According to their sonographic features,meningocele or encephalocele was determined.Conclusion Ultrasound could be a reliable prenatal screening method,provide an important basis for clinical intervention and have a significant clinical value in fetal meningocele and encephalocele.
7.Study on effects and mechanism of interleukin 16 on pathogenesis of atherosclerosis

Objective To analyze the expression of interleukin 16 (IL-16) in atherosclerosis (AS) patients, and to study the roles of IL-16in the pathogenesis of AS.Methods Thirty AS patients in Affiliated Hospital of Jining Medical College from August 2015to August 2016were randomly selected as the case group and twenty-nine healthy subjects were selected as the healthy control group.Peripheral blood of the subjects were collected.IL-16levels were determined with enzyme-linked immunosorbent assay (ELISA).Reverse transcriptase-polymerase chain reaction was applied to analyze IL-16mRNA level.IL-16expression in the atherosclerotic plaque samples was detected with immunohistochemical analysis.IL-16expression in aortic atherosclerotic plaque of AS patients and atherosclerotic ApoE-/-mice were analyzed by immunohistochemical staining.The aortic plaque changes of AS mice injected intraperitoneally with recombinant IL-16were detected.Results Both IL-16protein levels and IL-16mRNA levels were higher in case group than those of healthy control group, the difference was statistically significant (P<0.05).The IL-16mRNA was highly expressed in the atherosclerotic plaque.The aortic plaque area of the mice underwent IL-16intraperitoneal injection were decreased while the plaque stability increased.Conclusion IL-16levels elevated in both AS patients and AS mice, which suggested that IL-16might play aprotective role against AS.
