1.Study of the Relationship Between Molecular Heterogeneity of BP Antigens and Clinical Features

Bullous pemphigoid (BP) antigens are of molecular heterogeneity. To find out the relationship between the antigenic molecular heterogeneity and clinical features, treatment and prognosis in BP, 122 sera of BP patients were tested with immunoblotting technique. The results showed that 94 sera reacted with BP antigens extracted from normal human epidermis. Among them, 54 sera (57.4%) had BP 230kD antibody and 83 sera (88.3%) had 180kD antibody. Based on the results of immunoblotting assay, these 94 BP patients who had the positive antibodies were divided into three groups, including group I patients with 230kD protein antibody only, group Ⅱ patients with 180kD protein antibody and other minor protein antibodies, and group Ⅲ patients with both 230kD and 180kD antibodies. The pattern of clinical presentations (mucosal involvement, scarring and milia), disease severity, the largest dose of prednisone per day and duration of hospitalization were compared in these three groups. The results of comparison showed that there was no statistically significant difference among three groups in above mentioned items, except the difference of the largest dose of prednisone between the group Ⅰ and group Ⅱ (t=2.22, P
2.Relationship between vitiligo and thyroid diseases

Objective To investigate the relationship between vitiligo and thyroid diseases .Methods Chemiluminescence was used to detect serum levels of free triiodothyronine (FT3) ,free thyroxine(FT4) ,thyrotropic stimulating hormone(TSH) ,anti‐thy‐roglobulin antibody(TG‐Ab) and anti‐thyroperoxidase antibody(TPO‐Ab) in 289 vitiligo patients and 128 healthy subjects .All re‐sults were statistically analyzed .Results The abnormal rates of FT3 ,TG‐Ab and TPO‐Ab in vitiligo patients were higher than healthy subjects (P<0 .05) ,which might be correlated with the age and gender of vilitigo patients .Conclusion Levels of thyroid function and autoantibody might be abnormal in vilitigo patients ,which could be more obvious in male and adolescent patients .It could be advantageous to screen thyroid function and antibody levels in patients with vitiligo .

The amount of acidic acid is 0. 015mol/L The output of gas,which thd main composition is CO2 and CH4, is 20ml/L. The organic acid and gas is the important reasons of decreasing the viscosity, enhancing the flow and improving the product of oil.

Pseudomonas 23-1 Can metabolize hydrocarbon to produce the surfactant which is glycolipid outside the cell. The glycolipid, whose content is 1.25g / L, is composed of rhamnose and tencarbon fatry acid. It's critical micella concentration is 200g/L. It has the good ability to emulsify the oil. The main reason of improving the product of oil, decreasing the viscosity and enhancing the flow of oil is the effect of surfastant-glycolipid.

Acid-sensing ion channels(ASICs) are a novel class of ligand-gated cation channels activated by extracellular acidification and belong to the epithelial sodium channels(DEG/ENaC) superfamily.Their biological functions have recently been found not only in the central nervous system but also relevant to the physiology and pathology of non-neuronal tissues such as taste buds, cardiovascular system and bones.This review concerns the latest research on the expression and functions of ASICs in non-neuronal tissues so as to promote the understanding of their physiological and pathological functions.
7.Culture of human nucleus pulposus cells from degenerated intervertebral discs by explants adherent method after enzymatic pre-digestion

Objective To compare the efficiency of explants adherent after enzymatic pre-digestion and traditional typeⅡ?collagenase digestion methods for culturing human nucleus pulposus cells from degenerated intervertebral discs.Methods Human nucleus pulposus tissues collected from patients'degenerated intervertebral discs were divided into Groups A and B.In Group A,the nucleus pulposus tissues were cultured using explants adherent method directly after 0.025 % typeⅡ?collagenase digestion for 30 minutes.In Group B,the tissues were firstly digested with 0.025 % typeⅡ?collagenase for five hours,and then underwent filtration,centrifugation and inoculation successively Success rate and primary cell fusion time of the two culture methods were compared.Cell morphology and expressions of sex determining region Y-box 9(Sox-9),collagenⅡ?and aggrecan were observed by HE staining,toluidine blue staining and immunofluorescence cell staining.Results There was no significant difference in the successful culture rate between the two groups(P>0.05).However,the average fusion time of primary passage in Group A was significantly shorter than that in Group B(P<0.01).In Group A,the nucleus pulposus cells could be stained as azure by toluidine blue and immunofluorescence cell staining showed positive expressions of Sox-9,collagenⅡ?and aggrecan.Conclusions Compared with traditional typeⅡ?collagenase digestion,explants adherent method after typeⅡ?collagenase pre-digestion for the culture of human nucleus pulposus cells from degenerative intervertebral discs has a high success rate and obtains a large number of cells in a short time.Meanwhile,the cells have strong expressions of Sox-9,collagenⅡ?and aggrecan.
8.Anti-fibrosis effect and mechanism of total flavones of Bidens bipinnata L on immunological liver fibrosis in rats

Aim To observe the anti-fibros is effect and mechanism of total flavones of Bidens bipinnata L(TFB) on immunological liver fibrosis in rats.Methods The pig serum was injected into abdominal cavity(0.5 ml,twice a week) for 12 weeks so as to induce hepatic fibrosis,and then the rats were treated with TFB daily for 10 weeks and killed at the 22 nd week.The HA,LN,PCⅢ and CⅣ in serum were assessed by ELISA.Liver samples collected after experiment were stained with hematoxylin-eosin(HE) staining,Masson staining and scored.The expression of ?-smooth muscle actin(?-SMA) in liver was analyzed by immunohistochemistry.The gene expression of TGF-?1 was detected by RT-PCR.Results Compared with model group,TFB treatment significantly reduced HA,LN,PCⅢ,CⅣ content in serum.Liver histology in the TFB treated rats was also improved.Moreover TFB could decrease the expression of protein ?-SMA and TGF-?1 mRNA.Conclusions TFB significantly reduced pig serum-induced liver fibrosis in rats,probably by decreasing the expression of TGF-?1 mRNA and then inhibiting the proliferation of HSC.
9.Study on in vitro killing activity of DAAO/D-Ala system to K562e cells.

OBJECTIVETo investigate the in vitro killing efficiency of D-amino acid oxidase (DAAO)/D-alanine (D-Ala) system on K562e cells.
METHODSK(DfGC) cell line stably expressing DAAO was obtained by retrovirus transfection technique. The integration and expression of DAAO gene were identified by PCR and in situ hybridization. The killing activities of D-Ala to DAAO(+) cells alone or the mixtures of DAAO(+) and DAAO(-) cells in different ratios were observed. H(2)O(2) production by K(DfGC) cell was measured by phenol red oxidation assay.
RESULTSPCR and in situ hybridization analysis confirmed the integration of DAAO gene in positive clone and its mRNA expression. There was no significant difference in cell proliferation between the two kinds of K562 cells. Ninety percent of K(DfGC) cells was killed by 12.5 mmol/L D-Ala after 24 hour-treatment and the H(2)O(2) levels were in accord with the killing activities of D-Ala. When K(DfGC) was mixed with K562e at different ratio, no significant bystander effect could be found after treating with 15.0 mmol/L D-Ala for 24 hours.
CONCLUSIONThe leukemia cell line K562e was sensitive to DAAO/D-Ala system and there was no significant bystander effects observed within this cells.

10.Relationship between preoperative frailty and postoperative pulmonary complications in elderly patients undergoing thoracoscopic lobectomy

Objective:To evaluate the relationship between preoperative frailty and postoperative pulmonary complications (PPCs) in elderly patients undergoing thoracoscopic lobectomy.Methods:The elderly patients with non-small cell lung cancer who underwent thoracoscopic lobectomy in the Affiliated Hospital of Jiangnan University were collected.The general data, frailty status, parameters of blood and operation-related parameters were recorded.The patients were divided into PPC group and non-PPC group according to whether PPCs occurred during hospitalization, and the differences between the parameters were compared.Logistic regression analysis was used to analyze the independent risk factors for PPCs.Stratification analysis and interaction test were used to further analyze the relationship between frailty and PPCs.Results:A total of 298 elderly patients with non-small cell lung cancer undergoing thoracoscopic lobectomy were included in this study, and the incidence of PPCs was 22.8%.Compared with non-PPC group, the albumin and FEV 1/FVC were decreased, and age, blood creatinine, ratio of diabetes mellitus, ratio of chronic obstructive pulmonary disease (COPD) and rate of preoperative frailty were significantly increased in PPC group ( P<0.05). The results of logistic regression analysis showed that COPD and preoperative frailty were independent risk factors for PPCs.After adjusting all the risk variables, the frail patients had a 171% increased risk of PPCs compared with non-frail patients ( OR value=2.71, 95%CI: 1.18-4.73, P<0.05). The results of subgroup analysis showed that body mass index and operation time had effect modification on the association between frailty and PPCs (interaction P<0.05). Conclusion:COPD and preoperative frailty are independent risk factors for PPCs in elderly patients undergoing thoracoscopic lobectomy, and the frail patients with obesity or long operation time are at higher risk of PPCs.
