OBJECTIVE:To investigate the effects of MTP gene polymorphism on the lipid-regulating effect of simvastatin in the treatment of T2DM complicating with lipid metabolism disorder. METHODS:120 T2DM inpatients with hypercholesterolemia were selected from our hospital during Jun. to Dec. 2015 and given Metformin hydrochloride sustained-release tablets,Enalapril ma-leate tablets and Simvastatin tablets. PCR-RFLP was used to detect MTP G493T genotype. The levels of TG,TC,LDL-C and HDL-C were detected by AU 400 automatic biochemistry analyzer before treatment and the 4th week after treatment. The effects of different genotype on the change of blood lipid level was analyzed. RESULTS:Among 120 patients,the patients with MTP G493T GG,GT,TT genotypes accounted for 61.67%,26.67% and 11.67%,respectively,meeting Hardy-Weinberg balance(P>0.05). Before treatment,there was no statistical significance in TC,TG,LDL-C and HDL-C among different genotypes(P>0.05). After 4 weeks of treatment,the decrease of TC and LDL-C in TT genotype patients were lower than that in GG and GT genotype,with statistical significance(P>0.05). There was no significant difference in the decrease of TC and LDL-C between GT and GG geno-type,the decrease of TG and HDL-C(P>0.05). There was no significant difference in the incidence of ADR among different geno-types(P>0.05). CONCLUSIONS:After simvastatin treatment,the improvement of TC and LDL-C in patients with TT genotype is poor. MTP G493T gene polymorphism may be associated with the lipid-regulating effect of simvastatin in the treatment of T2DM patients with lipid metabolism disorder.

Objective To explore the mechanisms of hypoxia-induced expression of glucose transporter 1 (GLUT-1) in rat liver cells. Methods Wild type hypoxia response element (HRE) plasmid (construct N) containing the potential HIF-1 binding site was constructed by using construct A which contains the full length of the 5′-flanking region of the rat GLUT-1 gene as a template of PCR, and the PCR product was subcloned into the reporter plasmid pGL3-Promoter. Mutation type HRE plasmid (construct M) was made using a two-step overlapping PCR strategy. Then the liver cells were transfected with constructs N and M, respectively. At 24 h after transfection, the cells were subjected to hypoxia (1% O 2) to mimic the hypoxic environment caused by burn for 12 h. The samples were harvested and the determination of the reporter gene luciferase activity and pSV-?galactosidase activity was performed. Results Constructs N and M were successfully constructed and the liver cells were successfully transfected with construct N or construct M. Objective To construct a prokaryotic expression vector for a fusion protein, TAT protein transduction domain (PTD) and the PAS-B domain of hypoxia inducible factor 1?(HIF-1?), and then express and purifr the fusion protein. Methods The expression plasmids pTAT-PAS-B, pET-PAS-B and pTAT-EGFP were constructed respectively, and transformed into E. coli. BL21(DE3)pLysS strain to be induced by IPTG. The obtained proteins were analyzed by SDS-PAGE and Western blotting. The fusion protein were purified with Ni-NTA-His affinity chromatography. Results The three recombinant plasmids were constructed successfully. The objective fusion proteins were obtained by optimizing the conditions for expression and purification. Conclusion The successful expression and purification of the fusion protein TAT-HIF-1?PAS-B has laid the foundation for using it to modulate the activity of HIF-1? in vivo.

Objective To observe the effects of quercetin on PGE 2 release and cyclooxygenase activity in IEC 6 cells stimulated by the serum of burned rats. Methods The PGE 2 levels were measured by radioimmunoassay and the changes of cyclooxygenase activity were determined by substrate fluorescence analysis. Results Stimulated by the serum of burned rats, the PGE 2 level increased first and then obviously decreased. Quercetin at 0.1 and 1.0 ?mol/L enhanced the PGE 2 level after the stimulation of IEC 6 cells for 24 h. There was no obvious change in total cyclooxygenase activity, but the activity ratio of COX 1 to COX 2 significantly decreased. Conclusion Quercetin might influence the release of cyclooxygenase metabolite PGE 2 through the regulation of COXs activity in IEC 6 cells.

Objective:To evaluate the clinical efficacy and safety of Danhong injections in the adjuvant therapy for angina pecto-ris. Methods:According to a random number table,totally 120 cases of patients with angina pectoris were randomly divided into the observation group and the control group. The control group was treated with the conventional therapy,such as oxygen uptake,anti-in-flammation,expectoration and relieving asthma,and the observation group was treated with Danhong injections(30ml added in 250ml glucose infusions,ivd,qd)additionally. The treatment course was 2 weeks. The hemorheology and blood lipid changes before and af-ter the treatment were recorded,the ECG was analyzed and the clinical symptom improvement was evaluated in the two groups. Re-sults:After the treatment,the hemorheological indices of the two groups were significantly reduced(P<0. 01),and the improvement of the observation group was much better than that of the control group(P<0. 01). After the treatment,the TC,TG and LDL were significantly lower and the HDL was notably higher than those before the treatment in the two groups(P<0. 01),and the improvement of the observation group was significantly better than that in the control group(P<0. 01). The total efficiency of ECG and clinical symptom improvement in the observation group was significantly higher than that in the control group(P<0. 05). Conclusion:Dan-hong injections in the treatment of angina pectoris can significantly improve the hemorheology and blood lipid to enhance the clinical ef-ficacy,which are worthy of promoted application.

Objective To investigate the correlation of biofilm forming ability with biofilm associated gene of the clinically isolated Staphylococcus aureus for providing basis for the further studies of the mechanisms of biofilm formation.Methods Safranine staining was conducted for the detection of the forming ability of biofilm in 96-well plates.icaAD,icaBC,sar,agr and sigB was amplified by PCR.Results Formation of biofilm could be found macroscopically in 17 out of 27 strains,especially to X387 and X409.icaAD and icaBC were amplified in 22 isolates and sar,agr and sigB in all 27 S.aureus strains.Conclusion ica operon is the key gene for biofilm formation but cooperative action of other genes is needed during the process of biofilm formation.

Objective To study the macrolide-resistance,ability of biofilm formation and icaA-genetypes of 68 stains of clinically isolated Staphylococcus epidermidis(S.epidermidis) so as to explore the efficacy of macrolide to prevent biofilm-associated infections caused by S.epidermidis.Methods Minimum inhibitory concentration(MIC) were detected by agar-plate dilution method,and microtiter-plate assay was used to investigate the ability of biofilm formation,and icaA genetype was identified by PCR.Results High macrolide-resistant rate(88.2%) was showed for clinical isolated S.epidermidis,and biofilms for macrolide-resistant strains were significantly stronger than that of macrolide-sensitive strains,but there was no dependability found between these two for icaA positive rate.Conclusion Frequently-used macrolide such as erythromycin,azithromycin,and clarithromycin may incompetence to prevent biofilm-associated infections caused by S.epidermidis.

This paper introduces a microtiter-plate scanning instrument for medical laboratory equipment,which is controlled by the embedded PC when moving along the X direction and Y direction on the microtiter-plate plane. The designs of the driving circuit and orienting circuit are included in this paper. This instrument has been used in PKU fluorescence laboratory equipment for the newborn baby.
