ObjectiveTo investigate the role of peroxisome proliferator-activated receptor gamma (PPARγ)/Caspase-8/Caspase-3 signaling pathway in the development of hyperlipidemia in rats.Methods 60 healthy male SD rats 〔4-week old,weight (110 ± 10) g〕 were randomly divided into control group,high-fat diet group,folic acid group,vitamin B12 group,folic acid and vitamin B12 group.After one week's feeding for adaptability,the groups of foloc acid,vitamine B12 and foloc acid + vitamineB12 were dealt with intraperitoneal injection of folic acid (0.5 mg/d),vitamin B12 (0.05mg/d),and folic acid (0.5 mg/d) plus vitamin B12 (0.05 mg/d),respectively,and fed with high fat diet simultaneously.Control group was dealt with intraperitoneal injection of normal saline (0.5 ml/d)and fed with normal diet.High-fat diet group was only fed with high fat diet.Reverse transcription polymerase chain reaction (RT-PCR) was used to measure the mRNA level of PPARy,Caspase-8 and Caspase-3 in aorta abdominalis dissected at 17 weekends.Results Folic acid alone or jointed with vitamin B12 could effectively increase the level of PPARγ,while decrease the mRNA levels of Caspase8 and Caspase-3 as compared with high-fat diet group (P＜0.05),and folic acid plus vitamin B12 was more effective than folic acid alone (P＜0.05).Conclusions Folic acid alone or joined with vitamin B12 can improve the mRNA levels of PPARγ,Caspase-8 and Caspase-3 in vascular wall to protect endothelial injury from hyperlipidemia.

Objective:To study the protective effect of electroacupuncture on stem cell proliferation and differentiation in dentate gyrus (DG) in aged rats with focal cerebral ischemia reperfusion injury. Methods:We duplicated focal cerebral ischemia model with MCAO,with ischemia 3h,I/R1,3,7,14,21d points. The effect of electroacupuncture on the nervous dysfunction score,the water content of cerebral constitution and the stem cell proliferation and differentiation in dentate gyrus were observed,and the non-acupoint group was as control. Objective To investigate the myocardial protection of adiponectin (ADP) /adiponectin receptor 1 (ADPR1) related-signal pathway in rats with limb ischemic preconditioning.Methods Thirty SD male rats were randomly divided into sham-operation group,myocardial ischemia reperfusion injury (MIRI) group,limb ischemic preconditioning (LIPC) group,LY294002 (the PI3-specific inhibitor) pretreatment group and LY294002+LIPC group (n=10 each).The mRNA level of myocardial ADP and ADPR1,the protein expressions of phosphatidylinositol 3-kinase (PI3k)phosphorylated Akt (p-Akt) were determined by RT-PCR and Western blot,respectively. Results As compared with sham-operation group,the mRNA levels of ADP and ADPR1 in MIRI group were significantly decreased (0.53 ± 0.07 vs.0.74 ± 0.08 and 0.52 ± 0.02 vs.0.72 ± 0.04,P＜0.05).Compared with MIRI group,the mRNA levels of ADP (0.72±0.21) and ADPRI (0.80±0.023) in LIPC group were increased,ADP(0.49±0.07) and ADPR1 (0.52± 0.02) mRNA were decreased in LY294002 group (both P＜0.05),but there were no difference in ADP(0.70±0.16) and ADPR1(0.78±0.05) mRNA between LY294002+LIPC group and MIRI group.The protein levels of Pl3k and p-Akt were lower in MIRI group than in sham-operation group (3.85±0.23 vs.2.83±0.22and 3.77±0.32 vs.2.66±0.29,P＜0.05).In contrast to MIRI group,the yield of PI3k (2.65±0.32)and p-Akt(2.26±0.27) protein (P＜0.05) were increased in LIPC group,but there were unproductive protein of PI3k (3.75 ± 0.65) and p-Akt (4.01 ± 0.71) in LY294002 group with no differences versus the levels of PI3k (3.23 ± 0.48) and p-Akt (3.17 ± 0.54) in LY294002 + LIPC group. Conclusions Limb ischemic preconditioning may protect myocardium by promoting serum adiponectin levels,improving myocardial mRNA expressions of ADP and ADPR1,activating the ADP/PI3k/Akt signaling pathway in reperfusion injury.

Objective To study the inhibitory effects of wild-type p53 on gastric cancer cell lines BGC-7901 and to explore the importance of wild-type p53 in gene therapy for gastric cancer. Methods Adenoviral vector with exogenous wild-type p53 gene was transfected into the gastric cancer cell line BGC-7901 which harbours a mutation in codon 204 of the p53 gene.The transfer efficiency and expression effect of p53 were examined by immunohistochemistry and in situ hybridization. The growth of BGC-7901 cells in vitro was determined by cell counting and MTT assay. Flow cytometric analysis was used for observing cell cycle.Results Transfer efficiency reached 100% in BGC-7901 cells when adenovirus infection ability was over 100MOI. 3 days after transfection, the introduction of exogenous wild-type p53 into the tumor cells resulted in decreased expression of the protein of mutant p53 and increased level of mRNA of wild-type p53 gene. The growth of the BGC-7901 cells was significantly slower after transfection with wild-type p53 gene than that before transfection. The percentage of G 1/G 0 phase of the cells with exogenous wild-type p53 gene was much higher than that of the cells without exogenous wild-type p53 gene.It was 56 47% and 79 40% respectively(P

Objective To analyze the genetic characteristics of ribosomal DNA ITS 2 region for differenciating anopheline mosquitoes within Hyrcanus group. Methods The ribosomal DNA ITS 2 region of both laboratory line and filed collected An. anthropophagus and An. sinensis as well as the field collected An. yatsushiroensis and An. lester i were amplified and sequenced. The sequencing data were then analyzed for the restriction mapping using Omega Sequencing analysis program. Results and Conclusion The length of the sequences of An. sinensis, An. anthropophagus, An. yatsushiroensis and An. lesteri are 472, 452, 456 and 456 bp respectively. The restriction mapping showed that there were different restriction digesting sites among the ribosomal DNA ITS2 region sequences from An. sinensis, An. anthropophagus, An. yatsushiroensis and An. lesteri . On the basis of the sequence differences among the anopheline species within Hyrcanus group, it is possible to develop new technique for genetic identification of anopheline mosquitoes.

Purpose To investigate the histopathological features of cystic lung diseases ( CLD) , and to discuss the timing of clinical interventions. Methods HE and immunohistochemical staining were performed and reviewed in 125 cases of CLD. Results 125 ca-ses of CLD aged from birth to 11 years and 6 month, with an average age of 23. 0 months, median age 15 months, of which 60 cases were less than 1 year (48. 0%). 75 cases were male and 50 cases female, with male to female ratio of 1. 5 ∶ 1. Grossly, 50 cases showed single or multiple cysts with the size 0. 5 ~8. 0 cm in diameter, which did not communicate with bronchial cavity. 18 cases showed honeycomb cysts with the diameter of 0. 1~2. 0 cm. 26 cases were solid lesions without visible cysts. 21 cases were observed lung abscess with thick and rough wall and pus inside. 7 cases of emphysema showed microcysts with crepitation. 2 cases were identi-fied cystic and solid masses, with fish-fresh like cut surface. Histopathologically, 94 cases (75. 2%) were related to congenital bron-chopulmonary dysplasia in 125 cases of CLD, in which there were 59 patients (47. 2%) of congenial pulmonary airway malformation (CPAM), including 29 cases of type 1 (49. 2%), 18 cases of type 2 (30. 5%), and 12 cases of type 4 (20. 3%), there were 26 ca-ses (20. 8%) of pulmonary sequestration, including 15 cases of intralobar type (57. 7%) and 11 of extralobar cases (42. 3%), 5 ca-ses were complicated with CPAM type 2, 8 cases were bronchial cyst (6. 4%) and 1 case of enteric cyst (0. 8%). Acquired lesions were detected in 31 cases (24. 8%), including 21 cases of infected lung abscess, 1 case of fungal abscess. 7 cases of emphysema, and 3 cases of pleuralpulmonary blastoma (typeⅠ1 case and typeⅡ2 cases). Conclusion Pediatric CLD is characterized as com-plexed categories. The prognosis depends on correct pathological diagnosis, combined with imaging evaluation and appropriate timing of surgery.

Objective To explore the relationship between the distribution of infected snails and transmission of acute schistosomiasis in hilly regions.Methods The data concerning the distribution of the infected snails and acute schistosomiasis in Shitai County,Anhui Province from 1999 to 2008 were collected and analyzed.Results The sehistosome infection rate of human increased as the distance between the settings with infected snails and activity sites of humans shortened.Conclusions Acute infection of schistosome of human is associated with the distance between the settings with infected snails and activity sites of them.Strengthening the measures of snail control in key regions,protecting key populations and carrying out health education for schistosomiasis control are important approaches to control the transmission of acute schistosomiasis in hilly regions.

Objective To explore the use of marginal kidneys from DBD (donation after brain death) donors and the indication for adult dual kidney transplantation.Methods Two pairs of graft kidneys were procured from two marginal adult donors.Dual kidney transplants were performed in two recipients.In each recipient,two kidneys were implanted in unilateral site of right lower quadrant and placed extraperitoneally,two separate extravesical ureterneocysto-anastomoses were performed.Results Delayed graft function (DGF) combined with acute rejection occurred in two cases,and all two cases recovered after treatments.Lymphocele and hematoma occurred in one case.No graft embolism and no urinary leak happened.Conclusions Adult dual kidney transplant offers an important use of kidneys from marginal donors to increase the number of organs available for transplantation.

Objective To analyze the spatial-time distribution characteristics of schistosomiasis in Jiangling County ,Hubei Province from 2006 to 2011. Methods The surveillance data of schistosomiasis of Jiangling County from 2006 to 2011 were col-lected,and a spatial database was established. The spatial-time permutation cluster analysis was performed by SaTScan 9.1.1 to detect the spatial-time cluster areas of schistosomiasis,and a risk map was drawn by ArcGIS 10.0. Results Compared to 2006, the infection rates of human,cattle and Oncomelania hupensis snails decreased by 47.67%,93.34%and 52.41%,respectively in 2011. The space-time permutation clustering analysis of the infection rates of human,cattle and snails showed 4,3 and 4 clusters,respectively,and all the clustered areas were distributed in inner embankment areas. Conclusions From 2006 to 2011,the schistosomiasis endemic situation presents a decline trend in Jiangling County. The detected cluster areas are the im-portant areas for schistosomiasis control. The space-time permutation statistics could be used in the analysis of endemic situation of schistosomiasis.

Objective To understand the variation rules of schistosomiasis endemic situation before and after schistosomia-sis transmission controlled or interrupted,so as to provide the evidence for the consolidation of control achievements. Meth-ods In Anhui Province,3 counties reaching the criteria of schistosomiasis transmission controlled or interrupted were selected and their historical endemic data were collected and analyzed statistically from 10 years before the schistosomiasis transmission controlled to 2008. Results In Tianchang City,the Oncomelania hupensis snail area was 3.54 hm2 in a part of the lake marsh-land in the year reaching the criteria of transmission controlled(1998),and no snails were found in the year of transmission in-terrupted(2008). No stool-test-positive resident was detected except there were 58 acute schistosomiasis cases in 1993,and the sero-test-positive rate of local residents remained at low levels(0-1.55%). In Taihu County,the snail area was 0.84 hm2 account-ing for 0.06%of its historical accumulative snail areas in the year of transmission controlled(1971),and no snails were found three years before the transmission interrupted (1983). However,the schistosomiasis endemic rebounded 12 years later (1995),there was an outbreak of acute schistosome infections(73 cases),and the snail area increased again to 133.7 hm2 ac-counting for 2.91%of historical accumulative snail areas. After that,the snail area kept on rising and the infection rates of resi-dents and bovine remained higher than 1%. In Guangde County,the snail area was 32.4 hm2 accounting for 1.90%of its histori-cal accumulative snail areas in the year of transmission controlled(1995). After that,the snail area increased progressively, and the schistosomiasis endemic rebounded and there were acute schistosome infections 5 years later. The Spearman tests showed that the sero-test-positive rate(Tianchang City)and the stool-test-positive rates of residents and bovine(Taihu County)had positive correlations with the snail areas(r=0.582,0.401,0.596,all P<0.05). Conclusion The snail status is a key for the consolidation of schistosomiasis transmission controlled and interrupted. Therefore,a valid surveillance system of snail situa-tion should be established as quickly as possible.
