· AIM: To explore the dynamic expression and correlation among telomerase catalytic subunit (TERT), proliferating cell nuclear antigen ( PCNA) and antiapoptosis protein Bd-2 which relate to cell proliferation in epiretinal membrane of rat traumatic proliferative vitreoretinopathy(PVR).· METHODS: S-P technique was applied for immunohistochemical staining of epiretinal membrane of traumatic PVR with TERT, PCNA and Bcl-2 antibody. HE staining was also carried out. The staining results were analyzed with image analysis system.· RESULTS: The positive rate and average A of PCNA protein were upregulated at first and then down-regulated, with the peak value in 14d Group, which was significantly different from those in 7d Group and 28d Group.The positive rate and average A of TERT and Bcl-2 were also upregulated at first and then down-regulated, with the peak value in 14d Group and 21d Group, which were significantly different from those in 7d Group. There was significant correlation among PCNA, Bcl-2 and TERT protein expression (P≤0.01).· CONCLUSTON: TERT and Bcl-2 take part in the regulation of proliferative cells in epiretinal membrane of traumatic proliferative PVR, with high correlation with the dynamic changes of cell proliferation.
4.Investigation and analysis of nursing adverse events and discussion on management strategies

Objective To explore analytical methods for nursing adverse events,in order to provide strategies for safe clinical nursing.Methods Root analysis and SHEL accident analysis were used to analyze 91 cases of adverse events from 2009 to 2011 in a three A level hospital,and to identify their characteristics.Results Nursing adverse events with the highest frequency included medication mistake (15.38%),pressure sore (14.29%) and fall (9.89%).Nurses with different qualifications had different rates of medication mistake and pressure sore,and the differences were statistically significant (x2 =9.55,9.55,respectively;P < 0.05).Nurses of different educational levels had different rates of medication mistake,pressure sore,fall and accident events,and the differences were statistically significant (x2 =11.58,10.29,11.59,8.07,respectively; P < 0.05).The top three factors which affected nursing adverse events were poor communication (14.10%),no strict enforcement of the check system (11.54%),and insufficient evaluation (11.54%).Conclusions Both the quality of nursing staff and the overall hospital nursing management level should be strengthened to enhance patients' safety.

6.Effect of delayed preconditioning induced by Sini decoction on myocardial cell apoptosis and its mitochondrial mechanism.

OBJECTIVETo investigate the mechanism of myocardial cell apoptosis during delayed preconditioning induced by Sini decoction (SND).
METHODSD rats were divide into four groups: control, sham, I/R and SND groups. The rats in I/R group, left anterior descending coronary artery (LAD) was occluded for 1h and reperfused for 1 h. The rats in SND group were pretreated with Sini decoction (5 g x kg(-1) x d(-1)) for three days, the last treatment was pretreated 24 h before the index occlusion. Cell apoptosis was measured by flow cytometry, cytochrome C and bcl-xl were detected by Western blotting and the activity of caspase-3 was detected by assay kit.
RESULTAs compared with I/R group, apoptosis rate of myocardial cell, the release of cytochrome C from mitochondria and the activity of caspase-3 were significantly decreased, and the expression of bcl-xl protein was elevated in SND group.
CONCLUSIONThe delayed preconditioning induced by Sini decoction decreased myocardial cell apoptosis. The mechanism may be related to the inhibition of mitochondria signal pathway of apoptosis.

7.Experience of implementing high quality nursing service in rehabilitation ward

Objective To promote the high quality nursing service,explore the measures and effects of the high quality nursing service.Methods Develop the nursing service model by the change of the stuff management,and improving the health education to patients.Results The degree of satisfaction from patients increased to 99% and positive service consciousness increased,and the rate of nursing chaperonage decreased to 60%.Conclusions Carrying out the high quality nursing service in rehabilitation ward could improve the nursing quality.
8.Sensitive Detection of Telomerase Based on Hybridization Chain Reaction-assisted Multiple Signal Amplification

A new electrochemical method for telomerase activity assay was developed on the basis of hybridization chain reaction ( HCR)-assisted multiple signal amplification, aiming at improving the sensitivity and specificity of telomerase assay. The experiments utilized HeLa cells as original source of the telomerase in the electrochemical studies. The telomerase primer was firstly self-assembled on the surface of gold electrode. The telomerase catalyzed the elongation of the primer, producing the complementary sequences of hairpin probe H1. In this case, HCR was then initiated by interacting with two hairpin probes H1 and H2. Because both H1 and H2 were modified by biotin, horseradish peroxidase could be captured on the electrode surface through the high-affinity interaction between biotin and streptavidin, catalyzing the oxidation of o-phenylenediamine to produce 2,3-diaminophenazine. Therefore, the telomerase assay was realized by tracing the electrochemical signals with differential pulse voltammetry. This electrochemical method was of high efficiency and feasibility for detecting telomerase activity, and could trace the telomerase activity down to 10 cells/mL HeLa cells with a wide linear range. Besides, it could also easily distinguish the target enzyme from the control proteins with high specificity.
9.Effects of myocardial platelet rich plasma injection on rats with acute myocardial infarction: 99Tcm-MIBI gated SPECT imaging evaluation results

Objective To observe the effect of direct myocardial injection of platelet rich plasma (PRP) on cardiac function,ventricular remodeling and myocardial perfusion.Methods Myocardial infarction was induced in 30 Fisher rats by left anterior descending coronary artery (LAD) ligation.One week after LAD ligation,rats in control group (n =15) received 0.5 ml saline myocardial injection and rats in PRP group (n =11 ) received 0.5 ml PRP myocardial injection.99Tcm-methoxyisobutylisonitrile (MIBI)gated single photon-emission computed tomography (SPECT) imaging was applied at 1 week post LAD ligation and prior myocardial saline/PRP injection and repeated at 4 week post LAD ligation to assess myocardial perfusion and ejection fraction (EF),left ventricular end-diastolic volume ( LVEDV ),left ventricular end-systolic volume ( LVESV ),and the number of infarct segments.Results EF remained unchanged between 1 and 4 weeks post LAD ligation in control group and was significantly higher at 4 weeks post LAD ligation than at 1 week post LAD ligation in PRP group ( P < 0.05 ).LVESV was significantly increased in the control group ( P < 0.05 ) while remained unchanged in the PAP group ( P < 0.05 ) at 4 weeks post LAD ligation compared that at 1 week post LAD ligation.LVEDV remained unchanged in the control group (P>0.05) and significantly increased in the PRP group (P<0.05) from 1 week to 4 weeks post LAD ligation.Myocardial perfusion remained unchanged in the control group and significantly improved in the PRP group at 4 week post LAD ligations.Histological examination also confirmed that PRP treatment can decrease infarct size [from (47 ± 19)% to (36 ± 11 )% ],increase ventricular wall thickness [ from (3.1 ± 0.9 )mm to ( 4.6 ± 1.8 ) mm ] ( P < 0.05 ).Conclusion Myocardial PRP injection could improve cardiac function and reperfusion in this rat model of acute myocardial infarction.
10.Multi-component comprehensive quality evaluation of Tongjing ointment

Objective To establish a method for comprehensive quality evaluation of Tongjing ointment by multi-component quantification combined with chemometrics and grey correlation analysis(GRA).Methods With curcumin as the internal reference substance,HPLC-QAMS method was used to simultaneously determine the contents of limonin,evodiamine,rutaecarpine,bisdemethoxycurcumin,demethoxycurcumin,curcumin,6-gingerol,8-gingerol,10-gingerol,tetrahydropalmatine,dehydrocorydalin,dorydaline in Tongjing ointment.The quality of Tongjing ointment was evaluated by cluster analysis,principal component analysis,orthogonal partial least square discriminant analysis and GRA.Results The determination of 12 components manifested a good linear relationship in the range of mass concentration(r≥0.999 1).The average recovery was between 96.58%and 100.19%(RSD<2.0%,n=9).There were no significant difference between the measured value of external standard method and the calculated value of HPLC-QAMS(P>0.05).Tongjing ointment samples were classified into three eategories by chemometrics and it showed that curcumin,6-gingerol,bisdemethoxycurcumin,demethoxycurcumin,limonin and tetrahydropalmatine were the main potential markers affecting the quality of Tongjing ointment.GRA showed that the relative correlation degree was in the range of 0.317 3-0.624 8,and there were some differences in the quality of Tongjing ointment.Conclusion The established method can comprehensively evaluate the quality of Tongjing ointment.
