1.In vitro antitumor immune response induced by fusion of dendritic cells and metastatic colon cancer cells

Objective To detect the antitumor activity against SW620 and syngenic colon cancer SW480 by a fusion of human metastatic colon cancer SW620 cells and human peripheral blood-derived dendritic cells (DCs). Methods SW620 cells and human peripheral blood- derived DCs were fused with polyethylene glycol(PEG). The fusion cells were confirmed by "phenotypes determine, morphologic observation, and cytotoxic T lymphocyte response induced by the fusion were studied. Results Mature DCs with highly expressed surface markers (HLA-ABC, HLA-DR and CD80, CD86, CD83) were generated in vitro. The fusion of SW620 cells with DC resulted in a hybrid cell with morphologic as well as phenotypic characteristics of both DC and tumor cells. Flow cytometry showed that the highest fusion efficiency was 27. 12%. CTL assay demonstrated that the DC/SW620 fusion induced specific cytotoxic responses against the SW620 and SW480 cells. Conclusion The fusion of tumor cells with DCs induces tumor rejection.
2.The evaluation of left ventricular diastolic function by tissue Doppler echocardiography in essential hypertension

Objective To evaluate Left ventricular(LV) diastolic function in essential hypertension by conventional pulse-wave Doppler echocardiography (cPWD) and Doppler tissue imaging (DTI) and compare the two modalities. Methods Two hundred patients with essential hypertension were classified as NLVH subgroup (n = 160) and LVH subgroup (n =40) based on left ventricular mass index (LVMI) with one hundred and sixty health subjects as control group. The mitral valve flow pattern (MVFP) was obtained.Early diastolic (E) and late velocities (A) were measured and E/A was calculated. DTI was used to obtain the left ventricular lateral wall early diastolic mitral annulus velocity (Em) and E/Em was calculated.Results Essential hypertension patients had LV diastolic dysfunction both by cPWD (higher E and lower E/A ratio) and DTI (lower Em and higher E/Em ratio) compared with healthy subjects [E:(0. 88 ±0. 18)cm/s vs (0. 76 ±0. 19) cm/s;E/A ratio:0. 86 ±0. 28 vs 1.02 ±0. 38;Em: (9. 4 ±2. 8)cm/s vs (11. 9 ±3. 8)cm/s;E/Em ratio;7. 9 ± 2. 7 vs 6. 0 ± 1. 8: with all P value <0.01]. Em was significantly reduced and E/Em was significantly elevated in LVH subgroup than NLVH subgroup [Em; (7.7 ±2. 6) cm/s vs (9. 9 ± 2. 8) cm/s, E/Em: 9. 6 ± 3. 6 vs 7. 4 ± 2. 4, P < 0. 05]. No significant difference was found in A and E/A between these two subgroups [(0. 90 ± 0. 22) cm/s vs (0. 87 ± 0. 17) cm/s; 0. 80 ± 0. 34 vs 0.88 ±0.28, P > 0.05]. Conclusions cPWD and DTI both had implications to detect diastolic dysfunction in non-hypertrophic stage hypertension. Em、E/Em could be more sensitive and precise to reflect the impairment of diastolic function in the progress of hypertension.
3.Clinical analysis of 7 cases of Patch-skill for controlling refractory bleeding in open heart surgery

Objective To evaluate the effects of 'patch-skill'in the management of refractory bleeding during the process of open heart surgery.Methods Between May 2007 to Oetober 2008,7 patients undergoing open-heart surgery experienced active bleeding.Autologous perieardial patches were used in 6 patients, and polyester patch in the other one.The patches were sutured continuously by 5-0 Prolene suture.Results The 24 hour chest tube output was less than 500 ml in all 7 cases after operation.There were no death, secondly active bleeding,or mediastinal in-faction.Conclusion The'patch-skill' can effectively control the refractory bleeding during open heart surgery.Au-tologous perieardial patehe is the first choice because it has multiple advantages.The polyester patch ean be used al-ternatively when autologous pereardiurn cannot be used (for various reasons such as edema and infection).
4.Clinical Characteristics of 10 Cases with Polymyositis and Malignancy

Objective To summarize the clinical features of patients with polymyositis and malignancy . Methods We retrospectively reviewed the clinical records of patients with polymyositis and malignancy hospital -ized at Peking Union Medical College Hospital from October 1989 to June 2013.Results Malignancy was identi-fied in 2.4% (10/424) of patients with polymyositis in the hospital during the studied period .The median age of the 10 patients (3 males and 7 females) was 57 years.All patients had significant proximal muscle weakness .Dif-ficulty in head-lifting, bucking, and hoarseness were each observed in one case .Interstitial lung disease , respirato-ry muscle involvement , and cardiac involvement were observed in 5, 2, and 6 patients, respectively .The most common types of cancer were lymphoma and lung cancer (3 cases each).Other types included esophageal , gastric, renal, and cervical cancers (1 case each).Malignancies in 7 cases were discovered within 1 year before or after polymyositis diagnosis .The initial demonstrations of malignancies included lymphadenectasis , cough and dyspnea , dysphagia, gross hematuria , and postmenopausal vaginal bleeding .Three asymptomatic patients were identified through screening based on positive feces occult blood test or lung mass .Five out of the 9 patients receiving ade-quate dose of glucocorticoid recovered completely from polymyositis within 1 month, whereas the remaining 4 pa-tients improved but did not fully recover .Of the 4 patients, 3 patients achieved complete recovery after treatment for cancer.Conclusions Malignancy may interfere with the treatment response and prognosis of polymyositis , there-fore patients with polymyositis should have a complete screening for the underlying malignancy .Early cancer treat-ment should be prescribed to better mitigate symptoms and manage conditions in patients with polymyositis and cancer .
