1.Study on the Pharmacokinetics and Relative Bioavalability of Omeprazole Capsules in Humans

OBJECTIVE: To study the pharmacokinetics and relative bioavalability of omeprazole capsules in humans.METHODS: 18 male healthy volunteers orally took domestic omeprazole capsules and losec capsulles(used as control)40mg respectively.Blood concentrations of drugs were determined by HPLC.RESULTS: Times to reach the peak levels of omeprazole and losec were (2.10± 0.64) h and (1.88± 0.70) h, the peak plasma concentrations were (895.64± 553.07) ng/ml and (974.67± 554.93) ng/ml and the areas under the drug concentration curves were (1 971.88± 1 220.98 ) ng/(h· ml) and (2 057.60± 1 306.32) ng/(h· ml) respectively.CONCLUSION: The two capsules have the same bioequivalence.
2.Methylation of PTPRG gene and its regulation in gastric cancer.

OBJECTIVETo investigate the difference in methylation of PTPRG gene between gastric primary cancer and its lymph node metastases, and its regulation by 5-Aza-2'-deoxycytidin in a gastric cancer cell line SGC7901.
METHODSMethylation-specific polymerase chain reaction (MSP) and RT-PCR were applied to identify the difference between gastric primary cancer and lymph node metastases and assess the changes of methylation in gastric cancer cell line SGC7901 treated by 5-Aza-2'-deoxycytidin.
RESULTSThere were significant differences of PTPRG gene methylation and PTPRG mRNA expression between gastric primary cancer and lymph node metastases: a linear regression analysis revealed a significant association between the quantity of metastatic lymph nodes and their methylation rate. A statistied relationship between methylation of PTPRG gene and loss of PTPRG mRNA expression was detected. PTPRG gene methylation in the gastric cancer cell line changed into negative and PTPRG mRNA expression in the cell line was recovered after 5-Aza-2'-deoxycytidin treatment.
CONCLUSIONThere is a difference of PTPRG gene methylation in gastric primary cancer and metastatic lymph nodes. 5-Aza-2'-deoxycytidin, an inhibitor of DNA methylation, can recovery the expression of PTPRG gene.

3.Effects of electroacupuncture on the ultrastructure and the Nogo-A expressions in the cerebral cortex in rats with cerebral ischemia-reperfusion.

OBJECTIVETo observe the effects of electroacupuncture (EA) on the expressions of Nogo-A and the ultrastructure in the cerebral cortex at different time points after the cerebral ischemia-reperfusion in rats.
METHODSOne hundred and thirty male Sprague Dawley (SD) rats were randomly divided into the EA group (n = 30), the sham-EA group (n = 30), the model group (n = 30), the sham-operation group (n = 30), and the blank group (n = 10). The modified ZeaLonga method was used to prepare the left middle cerebral artery occlusion (MCAO) model in the first three groups. After the operation Baihui (DU20) and Dazhui (DU14) were daily needled in the EA group. One inch beside Baihui (DU20) and Dazhui (DU14) were daily needled in the sham-EA group. Rats in the model group were only treated with MCAO ischemia/reperfusion. Rats in the sham-operation group only received surgical wound. No treatment was given to rats in the blank group. The ultrastructures of ischemic cells and the intervention of the Nogo-A expressions were observed using the immunohistochemical staining and the transmission electron microscope 1, 7, and 28 days after EA.
RESULTS(1) In the EA group, the damage of ultrastructures of neurons, gliocytes, and blood brain barrier in the ischemic region was alleviated when compared with that of the sham-EA group and the model group. (2) On the 1st, 7th and 28th day after the cerebral ischemia-reperfusion, the expressions of Nogo-A in the ischemic cortex in the EA group was lower when compared with those in the sham-EA group and the model group at the corresponding time points, showing significant difference (P < 0.05). But there was no statistical difference between the sham-EA group and the model group at the same time point (P > 0.05).
CONCLUSIONThe mechanism of EA for protecting cerebral ischemia/reperfusion might be closely associated with alleviating the damage on the ultrastructures of brain cells, and down-regulating the expressions of Nogo-A.

4.Clinical observation of arthroscopic debridement for acute gouty arthritis of the ankle.

OBJECTIVETo evaluate the effects of arthroscopic debridement for acute gouty arthritis of the ankle.
METHODSForty-one patients with acute gouty arthritis of the ankle were treated under arthroscopy from January 2010 to June 2012. All the patients were male, age in ranging from 28 to 69 years with an average of 43 years. Eighteen patients were in the left ankles and 23 in the right ankles; 12 cases were firstly attack and 29 cases were recurrent attack. Course of disease was from 2 weeks to 30 months. The American Orthopedic Foot and Ankle Society (AOFAS) Ankle-Hindfoot Scale score was used to evaluate the clinical effects. Number of acute attacks of gouty arthritis were observed.
RESULTSAll the patients were followed up at least 12 months. The mean AOFAS Ankle-Hindfoot Scale score increased from 58.44 +/- 9.45 preoperatively to 86.15 +/- 7.36, 83.41 +/- 9.22, 84.10 +/- 8.22 postoperatively at 6, 12, months and the last follow-up respectively. Swelling of the ankle were improved significantly, pain was relieved and the mean number of acute attacks of gouty arthritis decreased significantly.
CONCLUSIONArthroscopy is helpful for the diagnosis of acute gouty arthritis of the ankle and improvement of clinical symptoms and ankle function.

5.Body mass index in girls with adolescent idiopathic scoliosis.

OBJECTIVETo investigate body weight and the body mass index (BMI) in girls with adolescent idiopathic scoliosis (AIS).
METHODSSix hundred and thirteen AIS female patients admitted from January 2005 to January 2007 and 449 healthy girls were recruited for this study with a range of age from 12 to 16 years old. The direct indexes included body height, body weight and date of birth. The indirect indexes included corrected body height, BMI and corrected BMI. The corrected body heights of AIS patients were adjusted by using the greatest Cobb angle to correct for spinal deformity according to Bjure's formula. Two-tailed Student's test was used for group comparison.
RESULTSIn the AIS patients, the maximum Cobb angle was (31 +/- 11) degrees (11 degrees to 77 degrees). The corrected body height were significantly higher in AIS patients than in the normal controls from 12 to 16 years old (P < 0.05). The body weight were significantly lower in AIS girls than in normal controls from 12 to 16 years of age (P < 0.01). From 12 to 16 years of age, the BMI were (17.6 +/- 1.9), (17.9 +/- 2.5), (17.9 +/- 2.1), (18.6 +/- 2.3) and (19.0 +/- 1.9) kg/m2 in AIS patients; while the BMI were (19.5 +/- 3.4), (19.8 +/- 3.0), (20.4 +/- 2.9), (20.4 +/- 2.8) and (20.2 +/- 2.2) kg/m2 in normal controls. The BMI were significantly lower in AIS girls than the normal controls from 12 to 16 years of age (P < 0.001).
CONCLUSIONSAfter the onset of puberty, significantly longer corrected height, lower body weight and lower BMI were found in AIS patients. Results of this large-scale study revealed the presence of abnormal growth in AIS patients during peripubertal development.

6.Determination of SARS-CoV by simplified nested fluorescent RT-PCR.

OBJECTIVETo establish a simple rapid and sensitive nested RT-PCR method for detection of SARS coronavirus RNA by designing the specific primers for SARS and optimizing the parameters for PCR.
METHODSPrimers and fluorescent probes were designed according to the sequences of SARS coronavirus genes available from GenBank. The optimization of the parameters for PCR was performed in PE 7700 thermal cycle. The 36 serum samples and 40 mouthwash of SARS patients and 80 samples of healthy people were tested.
RESULTSThe positive rate of patient serum and mouthwash was 33.6%, (12/36) and 67.5%, (27/40), respectively, while the positive rate of healthy people was zero (0/160).
CONCLUSIONThe simple nested RT-PCR method was a rapid, efficient and sensitive one for SARS early diagnosis.
