3.The relevance of proprioception function and the reservation of residual fiber in the anterior cruciate ligament reconstruction

Objective The purpose of this study is to evaluate the relevance of the recovery of proprioception function and the reservation of residual torn ligamentous fiber in the anterior cruciate ligament (ACL)reconstruction.Methods A retrospective study was carried out in 65 cases of ACL injury,who were treated with double-bundle ACL reconstruction and tibial remnant preservation and tensioning.All the patients were divided into four groups according to the degree of tibial residual fiber observed in the operation,no residual fiber was found(Group A),the length of the residual fiber was less than one third of the normal ACL(Group B),the length of the residual fiber was more than one third less than two thirds of the normal (Group C),the length of the residual fiber was more than two thirds of the normal(Group D).After one year follow-up,the passive angle reproduction test was performed with magnetic angle instrument in 3 flexion ranges(flexion of 90°-60°,60°-30°,and 30°-0°).All the results were contrasted among the four groups by statistical methods.Results In the passive angle reproduction test of the 3 flexion ranges,Group A and B showed a significant difference between the test side and control side(P<0.05),and Group C showed a significant difference between the test side and control side at the flexion of 30°-0°(P<0.05),but Group D showed no significant difference at the 3 flexion ranges(P>0.05).Conclusion During double-bundle ACL reconstruction,with the reservation of tibial residual tom ligamentous and tensioning may have a good effect on the recovery of proprioception function of the knees,which ensure a better functional outcome in the reconstructed knee.
4.Biological characteristics of tumor infiltrating regulatory T cells and the application of regulatory T cells in tumor immunotherapy

Regulatory T cells (Treg cells) have a critical role in the maintenance of immune homeostasis. However, tumor infiltrating Treg (TITR) contributes to weaken anti-tumor specific immune response through enhancing immunosuppressive effect in tumor microenvironment (TME), leading to tumor escape from immune surveillance. Recently, Treg cells-based immunotherapy has achieved good anti-tumor effects with the appearance of immune checkpoint inhibitors, chemokines and their receptors antagonists, specific Treg cells selective target genes knockout and novel drugs. This paper reviews the biological characteristics of TITR and discusses the prospect and application of Treg cells-based immunotherapy.
6.Changes of arterial blood pressure variability and baroreflex sensitivity in diabetic patients

Arterial systolic pressure variab il ity (ASPV), heart beat interval variability (HRV) and baro-reflex sensitivity ( BRS) were measured by finger arteriole blood pressue monitor system and cardiova scular spectral analysis system. The diabetic patients with cardiovascular auton omic neuropathy show significant changes in ASPV and HRV and decreased BRS. BRS changes earlier than other parameters.
9. Role of MSH2 and RUNX3 gene methylation in DNA prepared from nasopharyngeal swabs in early diagnosis and outcome prediction of nasopharyngeal carcinoma

Objective To explore the role of methylation of Human MutS homolog 2 (MSH2) and Human runt-related transcription factor 3 genes (RUNX3) in DNA prepared from nasopharyngeal swabs in early diagnosis and prognosis prediction of nasopharyngeal carcinoma. Methods The methylation-specific PCR was used to detect hypermethylation of MSH2 and RUNX3 genes in DNA prepared from nasopharyngeal swabs from 54 nasopharyngeal carcinoma patients, 18 chronic nasopharyngitis patients and 20 healthy volunteers. The specificity and sensitivity in early diagnosis of nasopharyngeal carcinoma by detecting MSH2 and/or RUNX3 gene methylation were evaluated. The relationship between methylation of MSH2 or RUNX3 gene and the biological behavior of nasopharyngeal carcinoma was analyzed. Results Hypermethylation of MSH2 and RUNX3 gene was respectively detected in 38 out of 54 (70.37%) and in 28 out of 54 (51.85%) nasopharyngeal swabs obtained from nasopharyngeal carcinoma patients, while there was no methylation in nasopharyngeal swabs from 18 chronic nasopharyngitis patients and 20 healthy volunteers. The differences were statistically significant (P<0.001). The specificity and sensitivity in early diagnosis of nasopharyngeal carcinoma were respectively 100% and 70.37% by detecting MSH2 gene methylation, 100% and 51.85% respectively by detecting RUNX3 gene methylation in nasopharyngeal swabs from nasopharyngeal carcinoma patients determined by methylation-specific PCR, while parallel combined detection of MSH2 and RUNX3 gene methylation increased the diagnostic specificity and sensitivity up to 100% and 90.74%, respectively. No close correlation was found between methylation of MSH2 or RUNX3 gene and the biological behavior of nasopharyngeal carcinoma in patients (P>0.05). Conclusions Parallel combined testing of MSH2 and RUNX3 gene methylation in DNA prepared from nasopharyngeal swabs determined by methylation-specific PCR could increase the specificity and sensitivity in early diagnosis of nasopharyngeal carcinoma, and is of important clinical significance. However it may not serve as an index in evaluating the clinical prognosis of nasopharyngeal carcinoma at present.
