Objective To observe the application effect of Clampcath trocar in immune adsorption.Methods A tatol of 40 patients underwent immune adsorption in our hospital were divided into the observation group and the control group according to the random number table method,with 20 cases in each group.In the observation group,20 cases were treated with Clampcath trocar as temporary vascular access for immune adsorption,and in the control group,20 cases were treated with 16 G indwelling needle as temporary vascular access for immune adsorption.The blood flow,tube plugging rate,discount rate,indwelling time of immune adsorption of patients in each group were compared.Results The tube blocking rate and discount rate of the observation group were significantly lower than that of the control group (P＜0.05),the difference wasstatistically significant;in the observation group,the blood flow was significantly higher than that in the control group (P＜ 0.01),the indwelling time of the observation group was longer than that of the control group (P＜0.05),the difference was statistically significant.Conclusion Clampcath trocar could be used as temporary vascular access for immune adsorption,of which the operation is simple,and is cheap and affordable.Meanwhile,it has advantages include low tube plugging rate and discount rate,long indwelling time and sufficient blood flow,thus it could alleviate the suffering of patients and reduce the waste of resources,and it's worthy of clinical promotion.

BACKGROUND: There are no studies on differentiation of bone marrow mesenchymal stem cells (BMSCs) into neurons directly induced by neural cells. OBJECTIVE: To establish the co-culture system between BMSCs and neural cells in vitro, and to study the influence of neural cells on the differentiation of BMSCs into neuron in the co-culture system. DESIGN, TIME AND SETTING: The in vitro cytology control experiment was performed at the Central Laboratory of Hospital Affiliated to Medical College of Qingdao University from December 2006 to December 2007. MATERIALS: BMSCS were harvested from bone marrow of patients with diabetes meliuts, who underwent autologous stem cell transplantation at the Hospital Affiliated to Medical College of Qingdao University. Neural cells were collected from brain tissues of infants die of asphyxia during delivery. The third passage of neural cells was used in this study. Transwell double-deck culture dish was purchased from Corning Costar, with a pore diameter of less than 3.0 ?m. Cells could not traverse, but the medium could traverse. METHODS: Neural cells were incubated in Transwell double-deck culture dish at a density of 1?106 in the co-culture group. BMSCs were incubated in the upper layer in LG-DMEM medium for 4-5 days. BMSCs were incubated in both layers in the control group. MAIN OUTCOME MEASURES: The morphological changes of BMSCs were observed and the special markers of neurons cells in BMSCs were examined by immunofluorescence. RESULTS: BMSCs in the co-culture group grew slowly, showing radial processes and connected each other. Positive rate of neuron specific enolase was (32.7?11.5)%. BMSCs in the control group were flat and wide, and negative for neuron specific enolase. CONCLUSION: Microenvironment provided by neural cells promotes the differentiation of BMSCs into neurons.

BACKGROUND: There is no ideal method about adult bone marrow mesenchymal stem cells (BMSCs) differentiate into islet-secreting cells and appreciation in vitro at present. Transgene requests strict conditions and complex program. The induction using chemicals is a present-used method. OBJECTIVE: To investigate the culture conditions for inducing adult BMSCs into islet-like cells in vitro. DESIGN, TIME AND SETTING: The cytology in vitro experiment was performed at the Central Laboratory of Affiliated Hospital of Medical College of Qingdao University from January to October 2007. MATERIALS: Bone marrow was collected from diabetic patients undergoing autologous stem cell transplantation at Department of Endocrinology, Affiliated Hospital of Medical College of Qingdao University after signing the informed consent. Epidermal growth factor and basic fibroblast growth factor were obtained from Peprotech Asia. B27 adjunct was purchased from Gibco. Nicotinamide, 2-mercaptoethanol, glutamine and dithizone were bought from Sigma, USA. METHODS: The adult BMSCs were isolated by Percoll from adult bone marrow aspirates and cultured in LG-DMEM, were suspended by Trypsin and passaged for subsequent passages. At the third to fifth passages, BMSCs were incubated at a density of 1?108 L-1 and were induced differentiation into islet-like cells through three developmental stages. In the first stage, BMSCs were incubated in HG-DMEM supplemented with 2-mercaptoethanol, glutamine for 2 days. In the second stage, BMSCs were incubated in HG-DMEM supplemented with epidermal growth factor, basic fibroblast growth factor, B27 and glutamine for 6 days. In the third stage, BMSCs were incubated in HG-DMEM supplemented with nicotinamide and 2-mercaptoethanol for 6 days. BMSCS in the control group were only incubated in the HG-DMEM. MAIN OUTCOME MEASURES: Morphological changes in BMSCs were analyzed under a phase contrast microscopy. Duodenal Homeobox 1 (PDX-1) expression was detected during the second and differentiation stage by immunofluorescence assay. Islet ?-like cell clusters from the 3rd stage were identified by positive dithizone staining. The insulin secretions under the stimulation of high or low glucose were detected by electrochemiluminescence immunoassay. RESULTS: The undifferentiated BMSCs exhibited adherent long spindle-shaped cells. After induction, cells gradually became round and formed clusters. Cells expressed the PDX-1 gene at 8 days and formed islet-like cell clusters that exhibited positive dithizone staining at 14 days. OBJECTIVE To study the Candida infection,its clinical distribution and susceptibility of antifungal agents in hospital from Jan 2002 to Dec 2006.METHODS The data of the patients′ age,samples,species distribution and antifungal sensitivity tests were retrospectively analyzed with WHONET 5.RESULTS A total of 501 cases with Candida were detected in these five years,from them 89 were older than 60,accounted for 68.1%.sputum was predominant samples,accounted for 66.1%.The main strain species were Candida albicans and C.tropicalis,C.glabrata,which were accounted for 56.1%,28.5% and 8.8%,respectively.C.albicans isolates were susceptible to four antifungal agents.CONCLUSIONS The Candida infection in hospital is increasing year by year,most of them occurred in old people.The most common infection site is lower respiratory tract.Fluconazole(FLU) is good susceptible to Candida.

Objective To investigate the degree of vascular damage in patients with hypertension in various stages with transcranial Doppler (TCD), so as to provide references for the prevention and treatment of obstructive cerebral vascular disease. Methods TCD was adopted to examine bilateral anterior cerebral arteries (ACA), terminal internal carotid arteries (TICA), vertebral arteries (VA) and basal arteries (BA) in patients with hypertension (n=176) and normotensive individuals as control group (n=60). The hemodynamic data of two groups were analyzed statistically. Results In patients with stage Ⅰ hypertension, the blood stream velocity was increased in all intracranial vessels, especially in MCM and TICA. In patients with stage Ⅱ and Ⅲ hypertension, the elevation of blood pressure was followed by the decrease of blood stream velocity, especially in MCA, TICA and BA. The rate of abnormality as detected by TCD was 90 and 96 in patients with stage Ⅱ and Ⅲof hypertension, respectively. Conclusions The abnormal findings of cerebral vessels as detected by TCD seem to coincide with the clinical characters of stroke. TCD findings may be used as an early forewarning index of obstructive cerebral disease.

[Objective] To investigate the role of exenatide on angiogenesis in endometrial cancer Ishikawa xenografts in nude mice.[Method] We used the subcutaneous human endometrial cancer cell Ishikawa xenografts in nude mice model,and divided them into control group and exenatide-treated group.The harvested tumors were preserved for immunohistochemistry staining and western blot analysis.[Results] The micro-vessel density (CD31 positive) in exenatide-treated group was (13.2 ± 1.4)/400 power field,less than that in control group [(25.9 ± 5.8)/400 power field] (P ＜ 0.01).The expression level of VEGF was significantly lower in exenatide-treated group than that in control group (P ＜ 0.05).The mean density of tumor associated macrophages (F4/80 positive) is (31.4 ± 3.4)％ in exenatide-treated group and (72.1 ± 4.2)％ in control group with significant difference (P ＜ 0.01).[Conclusion] Exenatide weaks tumor angiogenesis within endometrial cancer Ishikawa xenografts in nude mice.

To explore the audiological features in children who were sever sensorineural hearing loss infected with rubella virus. There were two cases of rubella virus infection in children who were deaf, they conducted the distortion product otoacoustic emission, ABR and auditory steady-state evoked response (ASSR) examination, then analyzed the results comprehensively. Two patients' mothers were prompted to have infected rubella virus during the early three months pregnant period by history and laboratory tests. The two patients were not detected deafness gene mutation. Audiology results implied the two patients were very severe binaural sensorineural deafness, so they were recommended to equipped with hearing aids and cochlear implant surgery. Early pregnancy women infected with rubella virus can cause very severe offspring sensorineural deafness. The crowd whose mother were suspected to infect with rubella virus in early pregnancy, that should be tracked and detected hearing in order to achieve early detection, early intervention and early treatment.
