Objective To explore the reversing mechanism of multidrug resistance of ciclosporin(CsA) on K562/A02 cell line, attenuating DNA damage repair through H2AX suppression. Methods K563 and K562/A02 respectively co-cultured with adriamycin(ADM) of different concentrations and CsA.MTT assay was employed to determine the inhibitory concentration of 50 percent(IC50),the resistance times and the reversal times. The K562/A02 cells treated with CsA,and reverse transcriptase(RT)-PCR technique was used to examine the mdr1 and H2AX mRNA level.Flow cytometry was used to measure P-glycoprotein(P-gp) and H2AX expression.Neutral comet assay was used to detect the level of DNA double strands break(DSBs) of the K562/A02 treated with ?H2AX antibody. Results 2?10-3g/L CsA could increase the sensitivity of K562/A02 to ADM,and the reversal times was 5.28.Mdr1 mRNA level and H2AX mRNA level were decreased when treated with CsA(P
4. Shengmai Injection combined with conventional treatment for acute myocardial infarction: A systematic review and Meta-analysis
Chinese Traditional and Herbal Drugs 2018;49(5):1201-1210
This Meta-analysis systematically assessed the efficacy and safety of Shengmai Injection (SMI) combined with conventional treatment for acute myocardial infarction (AMI). Seven online databases were investigated, then randomized clinical trials (RCTs) of treatment effects of AMI patients, for which SMI was used in combination with conventional therapy compared with conventional therapy alone, were included. The literatures selection, data extraction, and quality assessment were in accordance with the Cochrane standards. Data analysis was carried out using RevMan software (V.5.3). RCTs (enrolling 1 258 patients) from 18 literatures published in Chinese journals were included in the systermatic review. The additional SMI could significantly reduce the risk of mortality, arrhythmia, cardiogenic shock, heart failure and improve left ventricular ejection fraction (LVEF), stroke volume (SV), vascular patency rate. The adverse effects of mild bleeding, palpitation, shortness of breath, fatigue could occur both in the control group and the treatment group. Thus, it was difficult to evaluate the safety of SMI due to the limited data and low-guality RCTs. Therefore, high-quality and adequate sample of RCTs with low risk of bias are required to demonstrate its true effects.

Objective To investigate the phosphorylation of tau protein and the effect of atorvastatin on tau protein phosphorylation in hypercholesteremic mouse hippocampus. Methods Male Kunming mice were randomly divided into normal control group, hypercholesteremia group, and low-dose atorvastatin treatment group (8mg·kg-1·d-1), middle-dose group (15mg·kg-1·d-1),and high-dose group (20mg·kg-1·d-1) with five mice in each group. The hypercholesteremia mouse model was induced by cholesterol-enriched diets. The expression level of tau protein phosphorylation in mouse hippocampus was detected by Western blot and immunohistochemistry methods. The activities of mitogen-activated protein kinase (MAPK) and cyelin dependent kinase 5 (Cdk-5) were measured by liquid scintillation counting of the hippocampus incorporated radioactivity and immunoprecipetation activity assay respectively. Results In hypercholesteremic group, the expression level of tau protein phosphorylation in mouse hippocampus was significantly increased(F=14.761,P<0.01)as compared with control group, and so were MAPK activity and Cdk-5 activity(all P<0.01). Atorvastatin treatment group showed the decreased expression level of tau protein phosphorylation at ser396/404 site in low-dose group (F=6.349,P<0.05),middle-dose group (F=10.283,P<0.01) and high-dose group (F=10.511,P<0.01) as compared with hypercholesteremia mouse group. The activities of MAPK and Cdk-5 were decreased along with the increasing atorvastatin doses. Conclusions Hypercholesteremia induces tau protein hyperphosphorylation in mouse hippocampus. Abnormal cholesterol metabolism may play an important role in the pathology process of neurodegeneration in brain. Atorvastatin might inhibit tau protein hyperphosphorylation by inhibiting the activations of MAPK and Cdk-5 in brain, atorvastatin may have the protect effect for tau protein.
