1.Study on the Interaction between Cefprozil and Bovine Serum Albumin by Spectrometry

OBJECTIVE:To study the interaction of cefprozil (CE) with bovine serum albumine (BSA). METHODS:Under the temperatures of 289,299 and 309 K,the interaction of CE with BSA for 50 min had been studied by fluorescence quenching, UV spectrometry and synchronous fluorescence spectroscopy. Quenching constant(KSV)and speed constant(Kq)were calculated by Stern-Volmer equation. Static quenching constant(KLB)was obtained by Lineweaver-Burk equation,and UV spectrogram was used to determine the type of quenching. Double logarithmic equation was used to calculate the binding constants (Kb) and the number of binding site(n). Thermodynamic equation was used to obtain ΔH,ΔS,ΔG. Hill's coefficients(nH)was obtained by Hill equa-tion. RESULTS:At three different temperatures,with CE concentration increasing,fluorescence intensity of BSA decreased regular-ly. The value of KSV,Kq,KLB,Kb and n and nH decreased with the temperature increasing. ΔH,ΔS and ΔG were lower than 0. The numbers of binding sites were approximately equal to 1 and nH<1. CONCLUSIONS:CE statically quench the fluorescence of BSA,and the binding of them have been found to certain extent. The process of binding is spontaneous exothermic process. The main binding forces include Hydrogen bonds and Van der Waals forces,and primary binding site for CE is located at sub-domain ⅡA of BSA. There was some negative cooperative effect. CE would not affect the conformation of BSA. The binding site of CE and BSA is near by tyrosine residue.
2.On the interaction between cefonicid sodium and bovine serum albumin by spectrometry

Obiective To explore the interaction of cefonicid sodium(CS) with bovine serum albumin(BSA) by fluorescence and absorption spectroscopy. Methods The rate constant(Kq), quenching constant(Ksv), static fluo-rescence quenching association constant(KLB),binding site number(n) and binding constant(Kb) were calculated using Stern-Volmer, Lineweaver-Burk and double logarithm equations. Results CS was able to bind to BSA. The probable quenching mechanism of BSA by CS was mainly static quenching due to the formation of a CS-BSA com-plex. The results of thermodynamic parameters indicated that electrostatic force plays the main role in the binding process and the binding process was spontaneous. There was a single class of binding site for the BSA with CS. The primary binding site for CS was located at sub-domainⅡA of BSA and near by tyrosine residue. There was almost some negative cooperative effect. The results obtained from synchronous fluorescence showed that CS could change the microenvironment of Tyrand Trp residues of BSA. Conclusion The interaction between CS and BSA is dynam-ic. There is a single class of binding site for the BSA with CS. The obtained results provide references for its clini-cal application.
3.Study on Interaction betweenYan-Hu-Ning and Bovine Serum Albumin by Spectrometry

The interaction betweenYan-Hu-Ning (YHN) and bovine serum albumin (BSA) was investigated in order to provide further theoretical evidences on action mechanism study between YHN and proteins within the organism. Under optimal conditions, the interaction between YHN and BSA was studied by fluorescence quenching, ultraviolet absorption spectrometry and synchronous fluorescence spectroscopy. At the temperature of 283.15 K, 298.15 K and 313.15 K, quenching constant (KSV) and speed constant (Kq) were calculated by S-V curves. Static quenching constant (KLB) was obtained by L-B double reciprocal equation. Double logarithmic equation was used to calculate the binding constants (Kb) and the number of binding site (n). Thermodynamic equation was used to obtainΔH,ΔS,ΔG. Hill’s coefficients (nH) was obtained by Hill equation. The results showed that at three different temperatures, along with the increasing of YHN concentration, the fluorescence intensity of BSA decreased regularly. The value of KSV, Kq, KLB, Kb, n and nH decreased with the increasing of temperature;ΔG < 0,ΔH < 0,ΔS < 0; n was approximately equal to 1; nH > 1. It was concluded that YHN-BSA complex quenched the intrinsic fluorescence of BSA. The mechanism of fluorescence quenching was static quenching. The main binding forces were deduced as hydrogen bonds and Van der Waals forces from calculated values of thermodynamic parameters. YHN and BSA can form a binding site, which indicated certain binding interaction between YHN and BSA. YHN can be stored and transported by protein within the body. Free energy was produced to transformΔG into negative value. It showed that the process of binding between YHN and BSA was spontaneous. The nH was more than 1. It indicated that YHN had positive cooperative effect. The primary binding site was located at subdomainⅡA. The synchronous fluorescence spectra showed certain influence on the conformation of BSA by YHN. It led to the weakening of polarity within BSA and the binding site to be closer to the tyrosine.
4.A retrospective study on two surgical techniques in the treatment of hepatocellular carcinoma with portal vein tumor thrombus and to determine the prognostic factors of overall survival

Objective To study the impact of two surgical techniques in the treatment of hepatocellular carcinoma (HCC) with portal vein tumor thrombus (PVTT).Methods A retrospective study was conducted on 143 patients with HCC with PVTT who underwent hepatectomy from January 1995 to December 2010 at the Affiliated Tumor Hospital of Guangxi Medical University.The patients were divided into two groups:group A,115 patients who underwent resection of HCC with PVTT; group B,28 patients who underwent HCC resection but the PVTT was extracted from the cut opening of the portal vein or the transected liver parenchyma.Results The median overall survival of group A was 18.0 months and the cumulative 1-,2-,3-year survival rates were 60.6%,41.0%,25.6 %,respectively; the median overall survival of group B was 7.0 months and the cumulative 1-,2-,3-year survival rates were 35.1%,13.6%,9.1%,respectively.The differences between the 2 groups were statistically significant (P < 0.001).Univariate analysis showed tumor number,PVTT types,prophylactic transcatheter arterial chemoembolization (TACE),and surgical technique to be significant risk factor of postoperative overall survival (P < 0.05).Cox multivariate analysis indicated prophylactic TACE and surgical technique to be independent prognostic factor (P < 0.05).Conclusions When compared with group B patients,group A patients had significantly better overall survival.Postoperative prophylactic TACE further improved survival of these patients.
5.Effect of preoperative antiviral therapy on HBV reactivation and postoperative liver function in perioperative patients with HBV-DNA-negative hepatocellular carcinoma

Objective To explore the effect of preoperative antiviral therapy on hepatitis B virus ( HBV ) reactivation and postoperative liver function in perioperative patients with HBV-DNA-negative hepatocellular carcinoma(HCC).Methods 74 patients with preoperative HBV-DNA-negative scheduled which were analyzed.Patients were divided into two groups according to antiviral therapy or not:20 cases in antiviral treatment group received antiviral therapy for three days, 54 cases in non-antiviral teatment group did not receive antiviral therapy, and both groups received antiviral therapy after post-operative resuming to diets.The indicators of liver function and HBV-DNA levels were detected on pre-operative, post-operative 3rd and 7th day in two groups, and HBV-DNA-positive ( HBV-DNA>500 IU/mL) was defined as reactivation, conversely as inactivation.The indicators of liver function on pre-operative, post-operative 3rd and 7th day were compared between reactivation group and inactivation group.Results The reactivative rate was 21.6%(16/74) in all patients;27.7%(15/54) in pre-operative non-antiviral teatment group, 5.0%(1/20) in antiviral teatment group, and there was significant differences in reactivative rate between two groups ( P=0.035 ).The results of Logistic regression showed that pre-operative nonantiviral therapy was an independent risk factor of post-operative HBV reactivation (OR=13.952,95% confidence interval[CI]:1.358-143.379,P=0.027).The recovery of albumin (ALB) on post-operative 3rd, 7th days in antiviral treatment group was faster than those in nonantiviral treatment group, respectively (P=0.035,0.043).The recovery of ALB and alanine aminotransferase (ALT) on post-operative 7th day in reactivation group were slower than those in inactivation group, respectively (P=0.016, 0.048).Conclusion The pre-operative nonantiviral therapy is an independent risk factor of post-operative HBV reactivation in patients with HBV-DNA-negative HCC.The pre-operative antiviral therapy could inhibit post-operative HBV reactivation effectively and accelerate the post-operative recovery of liver function.
6.Prognostic factors of survival in patients with huge hepatocellular carcinoma after hepatectomy

Objective To analyze the prognostic factors of survival in patients with huge hepatocel lular carcinoma (HCC) who underwent hepatectomy with a view to improve treatment efficacy.Methods A retrospective study was conducted on 124 patients with huge hepatocellular carcinoma who underwent hepatectomy for HCC from January 2004 to December 2010 in our hospital.Univariate and multivariate analyses were performed using statistical software (SPSS 19.0 for Windows) to identify independent prognostic factors.Results The cumulative 1-,3-and 5-year survival rates of the 124 patients were 65.1%,35.8% and 25.1% respectively.The mean survival and the median survival were 34.7 and 26.0 months respectively.In the 65 patients who underwent curative resection,the 1-,3-and 5-year disease-free survival rates were 40.2%,19.3% and 7.1% respectively.The mean disease-free survival and the median disease-free survival were 18.6 and 9.0 months respectively.Univariate analysis showed HBsAg,tumor capsule,liver cirrhosis,vascular invasion,tumor rupture,intrahepatic metastasis,curative resection and BCLC staging significantly affected postoperative survival(P < 0.05).The Cox multivariate analysis indicated HBsAg,liver cirrhosis,curative resection and intrahepatic metastasis were independent prognostic factor (P < 0.05).Conclusions Intrahepatic metastasis,liver cirrhosis and HBsAg were prognostic factors influencing survival outcome of huge HCC in patients after hepatectomy.Improving curative resection rate of huge HCC significantly extended survival for these patients.
7.Clinical effect analysis of Xiaoke-Yuzu decoction on diabetic peripheral neuropathy

Objective To observe the clinical effect of Xiaoke-Yuzu decoction on diabetic peripheral neuropathy (DPN). Methods A total of 100 DPN inpatients were recruited and randomly divided into the treatment and control groups. The two groups were both received basic therapy, while the treatment group additionally received Xiaoke-Yuzu decoction. Toronto clinical scores and Chinese medicine symptom scores of both groups were collected to evaluate the clinical effect before and after the therapy. Results The Toronto scores of treatment group were significantly lower than control group after treatment (symptoms score 1.50 ± 0.94 vs. 2.23 ± 1.01, reflection score 3.60 ± 1.77 vs. 4.27 ± 1.72, feeling test score 1.53 ± 0.63 vs. 2.10 ± 0.84,all P<0.05). Meanwhile, the Chinese medicine symptom scores of treatment group were also significantly lower than the control group (main symptom score 1.77 ± 1.17 vs. 3.17 ± 1.82, posterior symptom score 2.23 ± 1.59 vs. 4.27 ± 1.57, the tongue and pulse score 1.83 ± 0.65 vs. 2.47 ± 0.51, all P<0.05). Conclusion Xiaoke-Yuzu decoction plus basic therpy could improve the clinical symptoms of DPN patients.
8.Preparation and application of layered osteochondral composite scaffold in the repair of articular cartilage defects

BACKGROUND:Ideal osteochondral tissue-engineered scaffolds should be able to mimic the normal structure ofhuman articular cartilage. OBJECTIVE:To prepare a layered osteochondral composite scaffold based on the anatomical and physiological functions of osteochondral articular cartilage and to observe its repair effect on osteochondral defects in rabbits. METHODS:The poly (lactide-glycolide acid)/β-tricalcium phosphate organic solution was sprayed on the surface of cartilage scaffold using rapid prototyping technology. The layered osteochondral composite scaffold was formed by the“dissolving-adhesion”process. Sixty rabbits were enrol ed, modeled into left knee articular cartilage defects, and then randomly divided into three groups. The layered osteochondral composite scaffold and cartilage scaffold were implanted into experimental and control groups, respectively. Those without any treatment served as controls. Gross and histological observations of the defect region were performed at 12 and 24 weeks after implantation. RESULTS AND CONCLUSION:Gross observation:At 12 weeks after implantation, the defects in the control group were obvious and not repaired at al;the 24-week defect area was decreased, covered by newly formed tissues, but with rough surface. In the experimental group, the defect surface was flat after 12 weeks of implantation, the texture was soft, and the boundary with the surrounding tissues was unclear;at 24 weeks, the defect was covered with transparent cartilaginous tissues and the surface was smooth. Histological observation:At 12 weeks after implantation, the irregular cal us appeared in the control group, but the trabeculae were not formed;in the experimental group, the thickness of the new cartilage was similar with that of the normal cartilage and there was irregular trabecular bone under the cartilage. After 24 weeks of implantation, there were new tissues in the control group, but the thickness was irregular and uneven and the trabecular structure was irregular;while the cartilage surface was smooth and repaired wel in the experimental group. In contrast, repair effect in the control group was poor as assessed by gross and histological observations. These results show that the layered composite scaffold holds a similar structure with human articular cartilage and can promote the repair of articular cartilage defects.
9.Clinical study on renal flow perfusion using color Doppler flow imaging combined with quantitative analysis technique

Objective To explore the clinical value of color Doppler flow imaging(CDFI) combined with quantitative analysis technique to evaluate renal flow perflusion. Methods Twenty patients with different renal diseases matched with healthy control subjects were examined with Philips iU22 CDFI system. CDFI system and Qlab quantitative technique were used to measure renal restrictive index(RI), vascularization index(VI), flow index(FI), vascularization-flow index (VFI). The patients all received renal single-photon emission computed tomography (SPECT) examination. Results The RI of 32 kidney with lower flow perfusion in SPECT were higher than in the contol group (P <0.01). VI,FI, VFI were lower than those in control(VI, VFI, P <0.01 ;FI, P <0.05). Conelusious The renal blood flow perfusion of patients with renal diseases are lower than that of healthy people. It has high clinical value using CDFI combined with quantitative analysis technique to evaluate the renal perfusion of the patients with renal diseases.
10.Quantitative color Doppler sonography assessment of the blood flow in renal cortex in patients with systemic lupus erythematosus

Objective To assess the value of quantitative color Doppler sonography in patients with systemic lupus erythematosus (SLE). Methods The right renal in 21 patients with SLE and 22 normal subjects were examined with Philips IU22 ultrasonic diagnostic device. Resistence index (RI) of interlobar artery and interlobular artery were measured with color Doppler sonography, vascularity index (VI), flow index (FI) and vascularization flow index (VFI) which reflecting the renal cortical blood flow were calculated with quantitated analysis software. All the parameters between two groups were compared and analyzed. Results The color signals of renal cortical blood flow in patients with SLE were inferior to those of the normal group, especially in the area near the edge of renal. No statistical difference of RI was found between the two groups (P>0.05), while VI, FI and VFI in SLE group were significantly lower than those of the normal group (VI, VFI, P<0.01, FI, P=0.01). Conclusion Color Doppler sonography combined with blood flow quantitative technique provide a helpful, sensitive and convenient tool to detecte the renal changes in SLE patients.
