1.Efficacy and Safety Observation of Ferrous Succinate in the Treatment of Pregnancy Complicated with Iron Deficiency Anemia

OBJECTIVE:To observe the efficacy and safety of ferrous succinate in the treatment of pregnancy complicated with iron deficiency anemia. METHODS:90 pregnant women with iron deficiency anemia were randomly divided into treatment group and control group. All patients were stopped drugs related to iron supplement 2 weeks before treatment,and the blood index-es were detected,including red blood cell count (RBC),hemoglobin concentration (Hb) and serum ferritin concentration (SF), etc. The treatment group was orally given 2 Ferrous succinate tablets after a meal,twice a day;control group was orally given 2 Ferrous sulfate tablets after a meal,twice a day. The treatment course for both groups was 1 month. The clinic data was observed, including clinical efficacy,RBC,Hb and SF level before and after treatment,and the incidence of adverse reactions. RESULTS:The total effective rate in treatment group was significantly higher than control group,the difference was statistically significant(P<0.05). After treatment,the RBC,Hb and SF level in 2 groups were significantly higher than before,and treatment group was high-er than control group,the differences were statistically significant(P<0.05). The difference was not statistically significant in the in-cidence of adverse reactions(P>0.05). CONCLUSIONS:Ferrous succinate tablets has good efficacy and safety in the treatment of pregnancy complicated with iron deficiency anemia.

3.Expression and regulatory mechanism of microRNA-155 in the villi of patients with unexplained recurrent spontaneous abortion patients

Objective To study the expression and the mechanism of miR-155in the villi of patients with unexplained recurrent spontaneous abortion (URSA).Methods The expression of miR-155 in the villi of 36 cases with URSA (URSA group) and 25 women with normal early pregnancy (control group) were detected by stem-loop real-time reverse transcription (RT) qPCR.Expression of hypoxia inducible factor-1 (HIF-1α),vascular endothelial cell growth factor(VEGF) and micro lymphatic vessel density (MVD) in the villi of were measured by immnohistochemical staining among two groups.Results (1) miR-155 expression:the mean miR-155 expression were 1.456 (0.489,2.459) in URSA group and 2.833 (1.740,3.794) in control group,which reached statistical difference (P <0.05).The mean expression of miR-155 of 1.683 (0.902,2.459) in URSA group with abortion times (≤ 3) was significantly higher than 1.229 (0.489,1.719) in URSA group with more than 4 times abortion (P < 0.05).(2) Indexes:the expression of HIF-1α,VEGF and MVD value were 121 ± 12,134 ± 12,36 ± 6 in URSA group and 99 ± 10,109 ± 10,28 ±4 in control group,which reached statistical difference(P < 0.01).The expression of HIF-1α,VEGF and MVD value of 119 ± 12,134 ± 12,35 ± 5 in URSA group with less than 3 times abortion was significantly lower than 128 ± 12,138 ± 12,43 ± 6 in URSA group with more than 4 times abortion (P < 0.01).Conclusions The expression of miR-155 and HIF-1α is topically stimulated by oxygen signal.HIF-1α adjusts the transcription and translation of VEGF,which together involved in placental trophoblast invasion and placental angiogenesis.The low expression of miR-155 could interfere with expression of HIF-1α and VEGF,which might be involved in villous vascular dysplasia in URSA.
4.Genetic analysis of neonatal diabetes mellitus and its influence on treatment outcome

Fourteen neonatal diabetes mellitus(NDM)patients were recruited. 9 patients were treated with glyburide and the other 5 with insulin. ABCC8, KCNJ11, and INS genes were sequenced in 6 of them. Gene mutations were found in 2, 1, and 1 cases in these genes, respectively. One case with 6q24 hypomethylation and another without known mutation were also found. 8 out of 9 patients treated with glyburide reached euglycemia(88.9%). The other 5 patients with insulin therapy either died or lost contact. The results suggest that glyburide therapy is effective in neonatal diabetes mellitus, while insulin therapy may contribute to poor compliance.
5.Effect of triple therapy with omeprazole in the treatment of children with peptic ulcer

Objective:To observe the clinical effect of omeprazole based triple therapy in the treatment of children with peptic ulcer.Methods:From September 2016 to September 2018, 86 children with peptic ulcer in the People's Hospital of Qingyuan County were selected in this study.By using the random number table method, they were divided into two groups, with 43 cases in each group.The control group received ranitidine based triple therapy treatment, the observation group was given omeprazole based triple therapy treatment.The treatment course of the two groups was 30 d. The eradication rate, clinical effect, gastrointestinal hormone level and adverse reactions of helicobacter pylori were compared between the two groups.Results:The eradication rate and total effective rate of helicobacter pylori in the observation group were 97.67%(42/43) and 95.35%(41/43), respectively, which were higher than those in the control group[81.40%(34/43) and 79.07%(34/43)], the differences were statistically significant (χ 2=6.081, 5.108, all P<0.05). After treatment, the levels of gastrin and motilin in the observation group were (75.29±8.31)μmol/L, (164.88±15.05)ng/L, respectively, which were lower than those in the control group [(81.07±8.96)μmol/L, (204.15±17.67)ng/L], the differences were statistically significant( t=3.102, 11.095, all P<0.05). The somatostatin level of the observation group was (22.67±3.95)ng/L, which was higher than that of the control group [(18.31±3.31)ng/L], and the difference was statistically significant( t=5.548, P<0.05). There was no statistically significant difference in the incidence of adverse reactions between the two groups(χ 2=0.938, P>0.05). Conclusion:Omeprazole based triple therapy is safe and effective in the treatment of children with peptic ulcer, which is helpful to eradicate helicobacter pylori, reduce gastrin, motilin levels and improve the level of somatostatin.
6.Effects of SP600125 on Proliferation and Invasion of Human Cervical Cancer HeLa Cells

Objective To investigate the effect of SP600125 on the proliferation, cell cycle, apoptosis and invasion of human cervical cancer HeLa cells. Methods CCK-8 method was used to detect the proliferation of HeLa cells treated with different concentrations of SP600125 at different time points. The 20 μmol/L of SP600125 was determined for subsequent experiments. Cell proliferation ability was detected using plate clone formation assay; nuclear morphology was observed by DAPI staining; cell cycle and apoptosis were measured by flow cytometry; cell migration and invasion were detected by cell scratch and Transwell methods; the mRNA and protein levels of p53, Mad2L1 and CDC20 were measured by qRT-PCR and Western blot after SP600125 treatment at different time points. Results Compared with control group (0.1%DMSO), cells proliferative activity were reduced by 10, 20, 30, 40 and 50 μmol/L SP600125 treatment for 24h. 7.Functional disability and influencing factors of lumbar disc herniation patients

Objective To investigate the functional disability of lumbar disc herniation (LDH) patients and to analyze influencing factors.Methods A total of 260 LDH patients treated in the 2nd Affiliated Hospital of Wenzhou Medical University from November 2015 to April 2016, were selected by convenience sampling method and evaluated by Oswestry low back pain-disability index-Chinese vision (ODI-C) about their functional disability, with the results analyzed by single factor analysis, Pearson correlation and hierarchical regression analysis.Results 216 valid questionnaires were regained, which showed that ODI-C scores of all the 216 LDH patients were (26.00±11.83) points. It was shown in single factor analysis that age, course of disease and surgery influenced the patients′ functional disability. It was shown in Pearson correlation analysis that interference of the pain degree or the pain, on the patients′ life was positively correlated with the patients ODI-C score, and so was their condition of depression (r=0.765, 0.725, 0.708;P<0.05). It was shown in regression analysis that, after control of demographic variables, interference of the pain degree or the pain, on the patients′ life, and the condition of depression, were independent influencing factors of functional disability of LDH patients, which accounted for 39.7% of the amount of variability.Conclusions Functional disability is common in LDH patients, which can be reduced by relieving their pain and depression.
8.Correlation of stigma, coping style and self-esteem among patients after breast modified radical mastectomy

Objective To explore stigma in patients after breast modified radical mastectomy, and analyze the correlation of stigma with coping style and self-esteem.Methods From January 2015 to February 2017, totally 186 patients after breast modified radical mastectomy were selected by convenience sampling method and investigated by self-designed general demographic questionnaire, Social Impact Scale, Medical Coping Modes Questionnaire and Self-Esteem Scale. Pearson correlation analysis was used to investigate the correlation of breast cancer patients' stigma with coping style and self-esteem, and multiple linear regression was adopted to analyze the influencing factors of stigma.Results Patients' stigma after surgery was (61.04±11.99), which was at a moderate level. The stigma was negatively correlated with confrontation and self-esteem (r= -0.421, -0.574;P<0.05), and positively correlated with resignation and avoidance (r=0.564, 0.414;P<0.05). The result of multiple linear regression showed that age and resigned coping style were the influencing factors of stigma (β'=0.291,0.263;P<0.05).Conclusions The stigma of patients after breast modified radical mastectomy is at a moderate level, and should be paid attention. Nurses should encourage patients to face difficulties and challenges, and communicate with others actively. Besides, young adult patients should be taken into consideration.
9.Neutrophil to lymphocyte ratio at admission for identification of large artery atherosclerosis and small vascular occlusion in patients with minor stroke

Objective To investigate the differential effect of neutrophil to lymphocyte ratio (NLR) at admission on large artery atherosclerosis (LAA) and small vessel occlusion (SVO) in patients with minor stroke.Methods Patients with first-ever acute ischemic stroke registered in Nanjing Stroke Registration System were enrolled retrospectively.Minor stroke was defined as the National Institutes of Health Stroke Scale (NIHSS) score ≤3.Binary logistic regression was used to evaluate the independent relationship between NLR and LAA.Results A total of 417 patients with minor stroke were included,of which 242 were in a LAA group and 175 were in a SVO group.Univariate analysis showed that there were significant differences in leukocyte count,neutrophil count,lymphocyte count,NLR,glycosylated hemoglobin,highdensity lipoprotein cholesterol,onset to treatment time,and onset to NLR detection time between the patients of the LAA group and the SVO group (all P >0.05).Binary logistic regression analysis suggested that after adjusting for sex and high-density lipoprotein cholesterol,NLR (with the highest quartile as a reference,the first quartile:odds ratio [OR] 0.207,95% confidence interval [CI]0.089-0.482;P > 0.001;the second quartile:OR 0.277,95% CI 0.122-0.625,P=0.002;the third quartile:OR 0.456,95% CI 0.197-1.057;P =0.067),onset to NLR detection time (OR 1.216,95% CI 1.038-1.424;P =0.015),and glycosylated hemoglobin (OR 1.414,95% CI 1.142-0.751;P=0.002) were independently associated with LAA.The area under the ROC curve of NLR predicting the LAA was 0.611 in patients admitted within 7 d after onset,and was 0.673 in patients admitted within 24 h after onset.Conclusions The increased NLR was independently associated with the LAA in patients with minor stroke.Early NLR value may have higher predictive value for LAA.
10.Correlation between capillary index score and outcome of endovascular treatment for acute anterior circulation ischemic stroke

Objective To investigate the capillary index score (CIS) and clinical outcome after endovascular treatment in patients with anterior circulation acute ischemic stroke (AIS). Methods From March 2014 to March 2017,patients with anterior circulation AIS received endovascular treatment in Jinling Hospital and Wuhu Yijishan Hospital were enrolled retrospectively. The data of intraoperative digital subtraction angiography were collected and CIS was calculated. They were divided into either a poor CIS group (score 0-1) or a good CIS group (scores 2-3). Univariate analysis was used to compare the baseline data, clinical data, and outcomes between the 2 groups. Multivariate logistic regression analysis was used to determine the correlation between CIS and symptomatic intracerebral hemorrhage (sICH), death, and functional outcome at 90 d after endovascular treatment(modified Rankin Scale score 0 to 2 was defined as good outcome). Results A total of 157 patients were enrolled, including 91 (58.0%) had poor CIS and 66 (42.0%) had good CIS.Age(P=0.020),baseline systolic pressure(P=0.014),baseline National Institutes of Health Stroke Scale score (P=0.011), early infarct size (P<0.001), as well as the proportions of internal carotid artery occlusion(P<0.001)and embolectomy >3 times(P=0.042)of the poor CIS group were significantly higher than those of the good CIS group.The vascular successful recanalization rate(P<0.001) and good outcome rate (P<0.001) at 90 d in the good CIS group were significantly higher than those in the poor CIS group, while the incidence of sICH (P=0.002) and mortality (P<0.001) were significantly lower than those of the poor CIS group. Multivariate logistic regression analysis showed that CIS were significantly correlated with the functional outcome at 90 d (odd ratio [OR] 0.581, 95% confidence interval[CI]0.419-0.805;P=0.001)and the risk of sICH at 72 h(OR 0.611,95% CI 0.407-0.919; P=0.018) after endovascular treatment in patients with anterior circulation AIS,but it did not have a significant correlation with the risk of death (OR 0.783, 95% CI 0.492-1.246; P=0.301). Conclusions CIS was significantly correlated with the clinical outcome in patients with anterior circulation AIS after endovascular treatment. It can be used as a tool to select patients for endovascular treatment.
