1.Effects of Simvastatin on the proliferation and osteoblastic differentiation of human periodontal ligament cells

Objective: To study the effects of Simvastatin on the proliferation and osteoblastic differentiation of human periodontal ligament (PDL) cells. Methods: The fourth passage human PDLs were cultured in conditioned mineralization medium with different concentrations of Simvastatin. Proliferation ability was detected by MTT method. Alkaline phosphatase (ALP) activity was measured with 4-nitrophenyl phosphate hexahydrate (PNPP) assay. Results: Proliferation and osteoblastic differentiation of human PDLs were increased in all treatment groups with different concentrations of Simvastatin(10-9, 10-8, 10-7, 10-6 mol/L). Compared with control group, significant statistical difference was found in 10-8 mol/L, 10-7 mol/L and 10-6 mol/L groups(P<0.05). The maximum effect was observed at the concentration of 10-7 mol/L. Conclusion: Optimal concentration of Simvastatin can improve the proliferation and osteoblastic differentiation of human PDLs.
3.Clinical efficacy of conventionally fractionated versus hypofractionated three-dimensional conformal radiotherapy for hepatocellular carcinoma with portal vein tumor thrombosis

Objective To observe the efficacy and adverse effects of conventionally fractionated (CF) versus hypofractionated (HF) three-dimensional conformal radiotherapy (3DCRT) for hepatocellular carcinoma (HCC) with portal vein tumor thrombosis.Methods A retrospective analysis was performed on the clinical data of 65 patients with HCC suitable for 3DCRT from 2008 to 2012.These patients were randomly divided into HF group (n =32) and CF group (n =32).The HF group received 3DCRT at a total dose of 45-55 Gy (3-4 Gy/fraction,3-5 fractions/week),while the CF group at a total dose of 40-56 Gy (2 Gy/fraction,5 fractions/week).Results The follow-up rate was 100%.For the HF group and CF group,the short-term response rates were 72% vs.55% (P =0.034),the 1-year local control rates were 72% vs.55% (P=0.034),the 1-year overall survival rates were 59% vs.45% (P=0.042),and the numbers of individuals with grade 1-2 acute gastrointestinal reactions,deterioration of liver function,and radiation hepatitis were 14 vs.11,8 vs.8,and 1 vs.0,respectively (P =0.237).Conclusions HF 3DCRT is superior to CF 3DCRT in the treatment of HCC.
4.Application of bispectral index during sedation in the patients in ICU

ObjectiveTo evaluate the value of bispectral index(BIS) monitoring during sedation in the ICU.Methods60 patients in ICU were randomly divided into three groups. By transfusing propofol and midazolam with injecting pump, the BIS of groups Ⅰ,Ⅱ and Ⅲ were controlled within 75~85, 65~75 and 55~65, respectively. ResultsCompared with the pre-sedation, there was no remarkable change in the patients of groups Ⅰ and Ⅱ after sedation(P>0.05)while there was remarkable change in the patients of group Ⅲ(P<0.05).The average score of Ramsay in groups Ⅰ, Ⅱ and Ⅲ were 2.2, 3.4 and 4.6 while the dose of propofol were (9.54±2.43) μg/kg·min, (12.69±3.12) μg/kg·min, (14.18±2.91) μg/kg·min and the dose of midazolam were (0.23±0.09) μg/kg·min, (0.25±0.07) μg/kg·min, (0.28±0.11) μg/kg·min, respectively.ConclusionThe application of BIS can make good judgement in the sedation, which showed different choices to different needs to obtain optimistical sedation effect.
5.Effect of Continuous Blood Purification on Multiple Organ Dysfunction Syndrome:33 Cases Report

ObjectiveTo study the effect of continuous blood purification (CBP) on the multiple organ dysfunction syndrome(MODS). MethodsContinuous blood purification were used in 33 patients with MODS. Results Among 33 patients,24 mended and 8 died. The scores of Acute Physiology And Chronic Health Evaluation (APACHE Ⅱ) and MODS obviously declined(P<0.05), as well as the BUN and blood Cr (P<0.05), but the bilirubin did not (P>0.05). The haemodynamic variables were stabilized during CBP and no obvious side-effect related to CBP was found. ConclusionThe therapy of continuous blood purification can improve the prognosis to the patients with multiple organ dysfunction syndrome. The patients were able to bear at the quality better.
6.Analysis popular characteristic and drug resistance of Pseudomonas aeruginosa in a hospital of Guiyang from 201 1 to 2013

Objective To analyze the popular characteristic and drug resistance of Pseudomonas aeruginosa in a hospital of Guiy-ang from January 201 1 to December 2013.Methods The distribution characteristic and drug resistance of Pseudomonas aeruginosa isolated from clinical samples from January 201 1 to December 2013 were analyzed retrospectively.Results A total of 642 strains of Pseudomonas aeruginosa were isolated for three years in our hospital from January 201 1 to December 2013,there was 57.0% isola-ted from sputum specimens,and 27.9% isolated from excreta of wound.The infected endemic area distribution was made up of Or-thopedic Surgery,ICU,Geriatrics Department and Respiratory Department accounting for 22.9%,20.1%,18.8% and 1 5.9% re-spectively.Pseudomonas aeruginosa was most sensitive to polymyxin bacillosporin.The drug resistance rates to Trimethoprim,Am-picillin and Cefazolin were all 100.0%.The drug resistance rates to Ampicillin/sulbactam and Ceftriaxone were 90.0%. Conclusion Pseudomonas aeruginosa is the main cause of lower respiratory tract infections in patients and wound infection and show serious multi-drug resistant,so it is necessary to use drugs reasonably according to the drug susceptibility results.
7.Production of CCHF Virus-Like Particle by a Baculovirus-Insect Cell Expression System

Crimean-Congo Haemorrhagic Fever Virus(CCHFV)is a tick-born virus of the Nairovirus genus within the Bunyaviridae family,which is widespread and causes,high fatality. The nucleocapsid of CCHFV is comprised of N proteins that are encoded by the S segment. In this research,the N protein of CCHFV was expressed in insect cells using a recombinant baculovirus. Under an electron microscope,Virus-Like Particles (VLPs)with various size and morphology were observed in cytoplasmic vesicles in the infected cells.Sucrose-gradient purification of the cell lysate indicated that the VLPs were mainly located in the upper fraction after ultracentrifugation,which was confirmed by Western blot analysis and immuno-electron microscopy(IEM).

Aim To investigate whether chronically un-predictable stress (CUS)-induced depression-like be-haviors of rats is associated with the variant expression of tryptophan hydroxylase (TPH)and tyrosine hydrox-ylase (TH).Methods 30 male SD rats were ran-domly divided into depression model group(MG)and control group (CG),the former was established using CUS plus solitary condition for 28 d,whereas the latter was fed normally as five rats per cage without CUS. The open field test(OFT)and the sucrose preference test were used to evaluate depressive behaviors.Both mRNA and protein expressions of TPH and TH in hip-pocampus and forebrain cortex were determined by re-al-time fluorescent quantitative PCR and western blot (WB),respectively.Results MG rats showed obvi-ous depressive behaviors with much lower locomotive activity and sucrose preference than CG.Meanwhile, the mRNA and protein expressions of TPH and TH also significantly decreased in MG rats,compared with CG rats.Conclusion The depression behaviors of rats in-duced by CUS may be associated with down-regulation of TPH and TH expression.
9.Study on the characteristics of immunoglobulin and complement expression in centenarians in Hainan

Objective:To investigate the characteristics of serum immunoglobulin and complement levels in centenarians in Hainan.Methods:Fasting venous blood samples from 969 centenarians in Hainan province were collected and from 364 seniors serving as the control group.Levels of serum immunoglobulin A, G, M and E, immunoglobulin light chain κ and λ, and complement C3 and C4 were measured.Serum immunoglobulin and complement levels were analyzed in subjects based on age, sex, ethnicity, diet and smoking.Results:Serum levels of immunoglobulin G and E, and immunoglobulin light chain κ were higher in the centenarian group than in the control group[15.9(13.8, 18.3)g/L vs.14.9(13.1, 16.9)g/L; 270.0(85.5, 851.0) vs.180.5 kU/L(58.0, 556.2)kU/L, 4.1(3.5, 4.9)g/L vs.4.0(3.1, 4.6)g/L, P<0.05]. Serum levels of immunoglobulin M, and complement C3 and C4 were lower in the centenarian group than in the control group[1.0(0.7, 1.4)g/L vs.1.1(0.8, 1.5)g/L, 1.0(0.9, 1.2) vs.1.1(1.0, 1.3), 0.2(0.2, 0.3) vs.0.3(0.2, 0.3), P<0.05]. Serum levels of immunoglobulin M and complement C3 were higher in female centenarians than in male centenarians[1.1(0.7, 1.4) vs.1.0(0.7, 1.3), 1.0(0.9, 1.1) vs.1.0(0.8, 1.1), P<0.05]. There were significant differences in immunoglobulin A, G and E, and immunoglobulin light chain κ and λ between centenarians of different ethnicities( P<0.01). Serum immunoglobulin M levels were higher in centenarians with a long history of milk-drinking than in those without the history[1.2(0.8, 1.5) vs.1.0(0.7, 1.4), P<0.01]. Conclusions:Serum levels of immunoglobulins and complements are different among centenarians of different ages, genders, ethnicities and diet habits.Factors such as advanced age, female gender, ethnic minority and milk drinking tend to promote the expression of immunoglobulins and complements.
10.Underreporting deaths in public health surveillance areas of Zhejiang Province

Objective:
To understand the underreporting rate of death among residents in Zhejiang Province from 2016 to 2018, so as to provide a basis for improvement of death surveillance measures.
Methods:
Multi-stage random cluster sampling was used to select residents in 30 public health surveillance areas of Zhejiang Province and all the residents were investigated the general information and death conditions that occurred during the period 2016-2018 by door to door visit. The underreporting rate of death was calculated and descriptive methods were used to analyze the causes of underreporting deaths.
Results:
A total of 358 992 residents were investigated in 30 public health surveillance areas of Zhejiang Province. There were 63 underreporting deaths among 5 896 deaths, with an underreporting rate of 1.07%. The underreporting rate in men was 1.30%, which was higher than 0.75% in women ( P<0.05 ). The underreporting rate of residents aged 20 to <40 years was 6.74%, the highest among all the age groups ( P<0.05 ). The underreporting rate was 4.46% on the way to the hospital, the highest among all the places of death ( P<0.05 ). The underreporting rate in Wuxing District of Huzhou was 5.80%, the highest among all the surveillance areas. There were 25 cases of "late report or untimely review", accounting for 39.68%; 38 cases of "not report", accounting for 60.32%, of which 14 cases were caused by doctors forgetting to report, accounting for 22.22%.
Conclusion
The underreporting rate of death in Zhejiang Province from 2016 to 2018 is generally low. In response to underreporting deaths, multi-departmental collaboration should be promoted to effectively strengthen the reporting and management of routine death surveillance.
