Objective To analyse and solve the difficulties and countermeasures in continuously proving medical gas. Methods Aiming at the difficulties of continuously proving medical gas, oxygen produced are mostly canned by oxygen compressor, and are transported to every function module through connecting oxygen jars. Results Medical gas module was taken into full play to meet reasonable demand for continuously providing oxygen. Conclusion It is suitable for other field module hospital to use for reference in the action of medical succor of disaster.
2.Risk factors for surgical site infection in patients with thoracolumbar fracture

Objective To evaluate risk factors for surgical site infection(SSI) in patients with thoracolumbar fracture,and provide basis for making prevention and control measures.Methods Patients with thoracolumbar fractures operation in the department of spinal surgery of a hospital between January 2010 and March 2015 were monitored through information monitoring system,on-site visit,and telephone follow-up,incidence of SSI was investigated,risk factors were performed univariate analysis.Results A total of 326 patients undergoing thoracolumbar fracture operation were monitored,15(4.60%) had SSI.SSI were high in patients≥60 years old,underwent operation at nonlaminar air flow operation room,didn't receive antimicrobial prophylaxis 30 minutes prior to operation,preoperative hospital stay >3 days,operation duration >3 hours,as well as with diabetes mellitus and chronic respiratory disease (all P< 0.01).Conclusion Risk factors for SSI in patients undergoing thoracolumbar fractures operation were patients with older age,receive operation at non-laminar air flow operation room,didn't receive antimicrobial prophylaxis,long length of preoperative hospital stay and operation duration,as well as combined with diabetes mellitus and chronic respiratory disease,preventive measures should be taken to reduce the incidence of infection.
3.Comparison of Lewis lung cancer cells and their derived cells

Objective By comparing the biological characteristics among Lewis lung cancer cells ( LLC) , LLC or-thotopic transplantation derivative cells ( R1-LLC) and R1-LLC orthotopic transplantation derivative cells ( R2-LLC) , we e-valuate their invasion and metastatic abilities in orthotopic transplantation models.Methods In vitro, we applied CCK8, clonogenic assay, and Transwell invasion assay to detect the proliferation ability, invasion ability and morphologic structures of LLC,R1-LLC, R2-LLC cells respectively, meanwhile observing morphologic structures by transmission electron microsco-py.In vivo, we constructed LLC, R1-LLC and R2-LLC orthotopic transplantation models.Herein, we observed their tumor growth and metastasis.The tumor formation rate and tumor-forming time were recorded for statistic analysis.Results In vitro:LLC, R1-LLC and R2-LLC cells showed no significant difference in proliferation ability ( P>0.05 ) , but significant differ-ences in invasion ability:R2-LLC>R1-LLC>LLC(P<0.05).In vivo:In the orthotopic group, the tumor formation rates of LLC, R1-LLC and R2-LLC cells were 66.67%、80%、93.33%(P <0.05).Conclusions In orthotopic implantation mouse models, R2-LLC cells present higher invasion and metastatic ability than R2-LLC and LLC cells.
4.CBCT image guided radiation therapy clinical application

Objective:Study the effect about cone beam CT(CBCT) image guidance sytem to improve the positioning accuracy and reducing the set-up uncertainty in precise radiotherapy. Methods:Use CBCT system to scan head and neck tumors patients (30 cases) and chest tumor patients (40 cases), including head and neck tumor patients were scaned 90 times, chest tumor patients were scaned 113 times. Setup deviation statistics about anterior and posterior, head and feet, left and right direction. Results:The patients with head and neck cancer, the maximum error of anterior-posterior is 6mm, Three directions errors which greater than 3mm all less than 10%. Patients with chest tumor position error are in the head and foot direction, greater than 5 mm up to 21.51%. Before and after the direction of the error did not exceed 5mm. The left and right direction greater than 5 mm up to 4.53%. Conclusion:It greatly enhances the precision of radiotherapy, improved the curative effect of radiotherapy by using CBCT online image guidance system for the patient position correction. Compared with EPID, patients scaned with CBCT absorb smaller cumulative dose, the image resolution is higher, image matching is more accurate. But compared with the ordinary helical CT,the resolution and scanning range also need to improve.
5.Comparison of automatic and manual methods for preparation of leukocyte-reduced red blood cell and plasma

Objective To compare the leukocyte-reduced red blood cells and plasmas respectively prepared by automated blood components separator and manual method so as to provide theoretical basis for automatic and standardized flow of blood components preparation.MethodsThe prepared red blood cells and plasma were measured on the hemoglobin concentration,mean corpuscular volume (MCV),red blood cell count (RBC),haematocrit (HCT),mean corpuscular hemoglobin (MCH),mean corpuscular hemoglobin concentration (MCHC),average red blood cell volume distribution width,plasma weight and protein content after the first separation,preparation time and etc.These results obtained from different methods were compared and analyzed.Results The plasma weight after the first separation which was one-off prepared were (267.57 ±16.31) g (by automatic method) and (246.06±22.48) g (by manual method) (P<0.05) respectively.The preparation time were (66.66±9.01) s (by automatic method) and (162.33±22.00) s (by manual method) (P<0.05) respectively.Other results had no significant difference (P>0.05).Conclusion All blood components products in this research meet the criterion of GB 18469-2012.However,the quality and separation efficiency of products made by automatic blood component separator are obviously higher than those by manual method.

7.Investigation of Endocrine Disrupting Chemicals in Source Water and Tap Water in a City

Objective To investigate the endocrine disrupting chemicals contamination in source water and tap water in a city and to provide the scientific data for the water quality inspection,management and water treatment.Methods Fifteen samples of source water and tap water were collected in April,2006 and February,2007.Extraction of potential EDCs from samples was performed in columns packed with XAD-4 resins by using solid-phase extraction(SPE).Each sample was dried under a gentle air stream for 1 h and eluted with a mixture of acetone(20 ml) and dichloromethane(40 ml) for 3 times.The extracts were collected into a glass flask,evaporated under a gentle air stream and suspended again in 0.5 ml dichloromethane.Gas chromatography-mass spectrometry(GC-MS) was used for quantitative or qualitative analysis of the organic pollutants and the optimum conditions were determined.Results Seventy-eight organic pollutants were identified from the water samples.Most of them were phthalates,hydrocarbon,phenol,and benzene.Phthalates were found in all samples.Among them,bis(2-ethylhexyl) phthalate(CAS# 117817),di-n-butyl phthalate(CAS# 84742),and pentachlorophenol(CAS# 87865) were regarded as suspected endocrine disrupting chemicals.The concentration of bis-(2-ethylhexyl) phthalate and di-n-butyl phthalate ranged 0.004 75-4.450 ?g/L and 0.002 25-2.39 ?g/L,respectively.The concentration of pentachlorophenol was 0.727 ?g/L.Conclusion The source water and tap water in this city has been seriously polluted by organic pollutants,the main endocrine disrupting chemicals detected in the present investigation are phthalates and pentachlorophenol and the related health effects remain to be studied.

Objective To investigate the effects of rhBMP2 on the differentiation of human dental pulp stem cells and expression of Delta protein in vitro. provide a theoretical basis for research on human dental pulp stem cells. Methods Monocell suspension was separated from the human adult dental pulp with collagenase Ⅰ and dispase digestion.Clonogenic cells were observed under light microscope and the expressions of surface markers were determined with immunofluorescence. divided into experimental group (containing 50 ?g?L-1rhBMP2 in the culture medium) and negative control group (containing culture medium only).alkaline phosphatase and the expression of Delta proterin of the cells at at passage 5 were detected.Results The human dental pulp stem cells showed colony growth,the nestin and vimentin staining were positive by immunohistochemical staining.STRO-1 was positive in cells by immunofluorescence.Compared with negative control group, the activities of alkaline phosphatase increased after treatment with rhBMP2 for 7,14 and 21d(P
9.Construction and identification of bicistronic retrovirus expression vector for Delta1 and EGFP gene coexpression

Objective To construct a bicistronic retrovirus expression vector to coexpress Delta1 and EGFP genes. Methods Delta1-IRES-EGFP bicistronic retrovirus vector was constructed by linking full-length Delta1 cDNA with IRES-EGFP retrovirus vector. Transfected with the retrovirus vector by electroporation, the ecotropic- and amphotropic-packaging cell lines GP+E86 and PA317 were selected by addition of puromycin. Positive cell clones were obtained and tested for the efficiency of the retroviral supernatants. Two positive cell clones with relatively highest titers were picked up and used for ping-pong amplification assay to improve the retroviral supernatants titer. At day 2 after transfection, GFP expression in NIH3T3 was detected by FACS, and Delta1 expression was determined by RT-PCR assay. Transfected or wild type NIH3T3 were co-cultured with myoblasts cell line C2C12 to test whether the exogenous Delta1 protein was functional or not. Results The bicistronic retrovirus expression vector coexpressing Delta1 and EGFP genes was successfully constructed and verified by PCR and DNA sequencing. A high-level expression of functional Delta protein was detected in transfected NIH3T3 cells. Conclusion Retrovirus is a safe, highly efficient gene transfer system.
10.Inhibition of hepatitis B surface antigen and hepatitis B virus DNA secretions by hepatitis B immunoglobulin poly(butylcynaoacrylate)nanoparticles in vitro

Objective To investigate the inhibitive activities of hepatitis B immunoglobulin(HBIG)poly(butylcynaoacrylate)nanoparticles(HBIG-PBCA-NP)to hepatitis B surface antigen(HBsAg)and hepatitis B virus(HBV)DNA secretions using HBV infected cell model in vitro.Methods HepG 2.2.15 cells were cultured with media containing HBIG-PBCA-NP or HBIG for several days,or cultured with HBIG-PBC-NP and HBIG for 2 days and without HBIG-PBCA-NP and HBIG from day 3.The supernatants at different time points were collected for quantitative detection of HBsAg and HBV DNA.The comparisons between groups were done by variance analysis.Resalts Secretions of HBsAg and HBV DNA in supernatants of HepG2.2.15 cultured with 0.1-10.0 IU/mL of HBIG-PBCA-NP and HBIG were inhibited significantly compared with control group.HBsAg titers and HBV DNA levels in supernatants of HBIG-PBCA-NP group and HBIG group cultured with media without HBIG-PBCA-NP and HBIG kept decreasing at day 5 and 7,then rebounded at day 9 and 11.HBsAg titera in supernatants of 0.1,1.0,5.0 IU/mL HBIG-PBCA-NP group were all significantly different from those in HBIG group at day 9[(31.31±1.98)μg/L vs(40.62±2.99)μg/L,(23.79±1.31)μg/L vs(36.51±2.12)μg/L,(19.91±1.74)μg/L vs(33.03±1.65)μg/L;F=412.24,P<0.01].Couclusion HBIG-PBCA-NP can inhibit secretions of HgsAg and HBV DNA in vitro,which is more effective than HBIG.
