5.The correlation between expression of carcinoembryonic antigen mRNA in preoperative peritoneal lavage fluid and prognosis of operation in patients with gastric cancer

Objective To investigate the correlation between expression of carcinoembryonic antigen (CEA) mRNA in preoperative peritoneal lavage fluid and prognosis of operation in patients with gastric cancer.Methods The expression levels of CEA mRNA in preoperative peritoneal lavage fluid of 68 patients with gastric cancer and 30 patients with gastric benign tumor were measured by reverse transcription polymerase chain reaction.The correlation between the expression level of CEA mRNA in preoperative peritoneal lavage fluid and clinicopathological features,prognosis of postoperative were analyzed.Results The expression level of CEA mRNA in preoperative peritoneal lavage fluid of patients with gastric cancre was 1.74 ± 0.25 and with gastric benign tumor was 0.19 ± 0.04,and there was statistical difference (P < 0.05).The expression level of CEA mRNA in preoperative peritoneal lavage fluid had a correlation with the tumor infiltration depth,tumor differentiation,lymph node metastasis,distant metastasis and TNM stage (P < 0.05).There was statistical difference in the recurrent / metastasis rate and survival rate 1 year after operation between low (34 cases) and higher (34 cases) expression level of CEA mRNA in preoperative peritoneal lavage fluid of gastric cancer patients:47.06% (16/34) vs.82.35% (28/34) and 79.41% (27/34) vs.55.88% (19/34),P < 0.05.Conclusion The expression of CEA mRNA in preoperative peritoneal lavage fluid has close correlation with the clinicopathologic features,and the high expression of CEA mRNA indicats poor prognosis,indicating that CEA mRNA in preoperative peritoneal lavage fluid could be used as a prognostic marker in patients with gastric cancer.
6. Research progress on chemical constituents and pharmacological effects of Sijunzi Decoction

Sijunzi Decoction (SJZD) is a classical prescription for curing spleen deficiency in traditional Chinese medicine, consisting of Ginseng Radix et Rhizoma, Atractylodes Macrocephalae Rhizoma, Poria, and Glycyrrhizae Radix et Rhizoma Praeparate cum Melle. Modern pharmacological experiments have proved that sponin, flavonoid, and polysaccharide are the most active ingredients in SJZD. SJZD has been used to regulate gastrointestinal function and enhance immunity ability. Effects on tumor cell apoptosis and anti-aging have also been reported. But few report on SJZD polysaccharide and its therapeutic basis research was found. This review is to give a comprehensive summary of information regarding the chemical constituents and pharmacological effects of SJZD to help us take the action for reasonable clinical utilization and further researches of SJZD.

8.Role of Abro1 in LPS-induced acute lung injury

Objective To investigate the effect of Abro1 on acute respiratory distress syndrome(ARDS)/acute lung injury(ALI)in mice.Methods Abro1 knock-out(KO)mice and wild type(WT)mice were both randomly divided into two groups for intratracheal instillation of lipopolysaccharide(LPS)or normal saline.At 6 or 24 hours after treatment, the pathological changes in lung tissue were observed by HE staining.At 6 hours after treatment,inflammatory immune cells and cytokines production(IL-6)in the bronchoalveolar lavage fluid were examined.Myeloperoxidase(MPO)and the mRNA level of IL-6 in the lung tissue were compared.Results At 24 hours after treatment, compared with WT mice treated with LPS,Abro1 KO mice showed a significantly lower lung injury score.At 6 hours after treatment,Abro1 depletion resulted in reduced levels of inflammatory immune cell infiltration and cytokines production(IL-6)in the bronchoalveolar lavage fluid(P<0.05).In addition,the MPO content and the mRNA level of IL-6 in the lung tissue were much lower than those in WT mice treated with LPS for 6 hours(P<0.05).Conclusion Abro1 deficiency can attenuate LPS-induced ARDS/ALI.

10.Expression and Identification Truncated Glycoprotein G of Bovine Respiratory Syncytial Virus in Escherichia coli

Two fragments G1 and G2 of the glycoprotein G gene of bovine respiratory syncytial virus(BRSV) were selected for expression in Escherichia coli based on the analysis of glycoprotein G by DNA Star software.Then the two fragments of glycoprotein G were amplified by PCR with synthesized G gene of BRSV as the template.The amplified fragments G1 and G2 are 570bp and 308bp in length,respectively.The PCR products were cloned into pET30a vector and expressed in soluble form in E.coli after induction of cultured E.coli with IPTG.Both of the recombinant proteins G1 and G2 were purified by immobilized Ni ion affinity chromatography under native conditions.Then the purified proteins were analysed by Western blotting.The results showed that the purified recombinant protein G1 retained good antigenicity and specificity.But the purified recombinant protein G2 didn't possess biological activity.Antibodies against BRSV were detected in suspected bovine serum samples in China by using indirect ELISA and Western blotting with the purified recombinant protein G1.The purified recombinant protein G1 might be used as antigen for establishing serological methods for diagnosis of BRSV infection.And the purified recombinant protein G1 might also be used for preparing polyclonal and monoclonal antibodies for research on biological functions of glycoprotein G of BRSV.
