Aim To investigate the effects of breviscapine(Bre)on Na+,K+-ATPase activity of proximal tubule(PT) and its renal protection in early diabetic rats. Methods The rats were divided into three groups: diabetic model (DM group), Bre treatment (Bre group) and normal control (NC group). Rats were administrated with Bre(20 mg?kg-1?d-1, ip) in Bre group, with normal saline(ip) in DM and NC groups for four weeks after diabetes induction with streptozotocin (STZ 65 mg?kg-1, ip) in Bre and DM groups. The urine and blood samples were collected from two intra-ureteral cannulas and the heart, respectively, under anesthesia four weeks after diabetes induction. After one of the renal arteries being perfused, the renal cortex was incubated and PT segments were microdissected freehand under microscope before the Na+,K+-ATPase activity in the segments were assessed by liquid scintillation counter. The blood glucose, levels of creatinine in serum and in urine were assayed. The microalbumin , ?2-microglobulin(?2-MG) of urine and endogenous digitalis-like substance (EDLS) of serum were measured respectively by radioimmunoassay. Results The PT Na+,K+-ATPase activity in NC group was(959.11?117.35) pmolPi?mm-1?h-1, and that in DM group was significant higher for (1893.53?383.90) pmolPi?mm-1?h-1 than it(P

Space Medicine & Medical Engineering 2006;0(01):-
Objective To study and design a new algorithm of QT interval measurement based on multi-scale morphological derivative transform(MMDT) according to the demands of mobile monitoring platform. Methods After the MMDT of ECG signals by inducing a triangle and a short line as a pair of structure elements, they were classified into four types by their morphological characteristics and the optimal strategy for each type was described to detect the onset of Q-wave.By introducing the "wing" function, two referenced points of T peak were picked out, which was helpful to locate the peak and offset of T wave, and improve the precision and recognition rate of MMDT method when detecting the bifid T wave(90.9%)and biphasic T wave(86.7%).Results Eighty data records from CSE database were used to evaluate the availability.By contrast with the wavelet transform method, the statistical results showed that the proposed algorithm had generally less error and smaller standard deviation especially for abnormal-phase T wave. Conclusion Compared with those algorithms based on wavelet transform and self-adaptive threshold techniques, our algorithm needs less empirical parameters and calculation. It is also suitable for mobile monitoring and HOLTER system, and has a wide prospect of application.

Objective To investigate the effects of high glucose and insulin on the expression of glucose transporter 4 (GLUT4), Cbl-associated protein (CAP) and cytoskeleton protein F-actin of glomerular mesangial cells (GMCs), in order to explore the function of GLUT4, Cbl-associated protein and F-actin in the pathogenesis and development of diabetic nephropathy (DN). Methods Cultured 1097 rat glomerular mesangial cells were divided into 8 groups: control, 10-9 mol/L insulin, 10-8 mol/L insulin, 10-6 mol/L insulin, high glucose (30 mmol/L), mannitol (25 mmol/L mannitol+5 mmol/L glucose), high glucose plus 10-6 mol/L insulin, high glucose plus 10-9 mol/L insulin. Expression of CAP mRNA and GLUT4 was measured by RT-PCR and immunohistochemistry method. F-actin was stained by rhodamine-pholloidin and the fluorescent intensity was calculated by image analysis system. Results The expression of GLUT4 mRNA and protein, CAP mRNA was found in normal giomerular mesangial cells (control), and there was no significant difference in 10-9 mol/L insulin group. The expression of GLUT4 mRNA (P<0.05) and protein (P<0.01), CAP mRNA (P<0.01) level was decreased in high glucose group compared with that of control group, but there was no significant difference in mannitol group. The expression of GLUT4 and CAP mRNA up-regulated with the increase of concentration of insulin. The expressions of GLUT4 mRNA in 10-8 mol/L insulin and 10-6 mol/L insulin groups were 2.06-fold and 2.66-fold of 10-9 mol/L insulin group, of GLUT4 protein were 1.93-fold and 2.83-fold of control, and of CAP mRNA were 1.91-fold and 2.15-fold of control, respectively. The expressions of GLUT4 mRNA, GLUT4 protein, CAP mRNA in high glucose plus insulin group were 2.15-fold, 2.08-fold, 2.14-fold of high glucose group respectively. High glucose decreased the fluorescent intensity of F-actin to 44.5% (P<0.01). 10-8 mol/L insulin and 10-6 mol/L insulin groups increased to 1.224-fold (P<0.05), 1.296-fold (P<0.01) in a concentration-dependent manner. The spearman correlation coefficient between GLUT4 and F-actin was 0.929 (P=0.001), between GLUT4 mRNA and CAP mRNA was 0.905 (P=0.002). Conclusions (1) A certain expression of GLUT4 mRNA and protein, CAP mRNA from GMC is found in normal glomerular mesangial cells. (2) High glucose can inhibit the expression of GLUT4 and CAP mRNA significantly, and facilitate the depolymerization of F-aetin. (3) Insulin can reverse down-regnlation of GLUT4 and CAP mRNA caused by high glucose. (4) GLUT4, CAP and F-actin are important factors in the development of DN.

OBJECTIVE:To discuss the effect of early usage with Wulingjun powder on the behavior of rats with post-traumat-ic stress disorder(PTSD)and the level of interleukin-1β(IL-1β)and IL-6 in their hippocampi. METHODS:50 SD rats were ran-domized into control group(normal saline),model group(normal saline),positive control group(paroxetine,10 mg/kg)and the groups of low and high-dose(0.4,0.8 g/kg)Wulingjun powder. The rats in all groups except for control group, were exposed to single prolonged stress for the establishment of PTSD models. Corresponding drugs were given ig immediately after such establish-ment,qd,for 7 consecutive days. An open field test(the times of the rats’entering the central area and the percentage of the time the rats spent in exploring the central area within 10 min were recorded)was conducted 7 days after giving drugs;and an elevated plus-maze test(the times of the rats’in opening arms and the percentage of the time the rats spent in opening arms within 5 min were recorded)was carried out 8 days thereafter,to observe the behavior of the rats. Then the rats were sacrificed to determine the level of IL-1β and IL-6 in their hippocampi by ELISA. RESULTS:Compared to the normal group, the behavioral index of the rats in model group was significantly lower, and the level of IL-1β and IL-6 in their hippocampi was significantly higher(P<0.05 or P<0.01). Compared to model group, the behavioral index of the rats in positive control group and the group of high-dose Wuling-jun powder was significantly higher, and the level of IL-1β and IL-6 in their hippocampi was significantly lower (P<0.05),but no statistically significant difference was shown in all indexes in the group of low-dose Wulingjun powder (P>0.05). CONCLU-SIONS:High-dose Wulingjun power can improve the anxiety-like behavior of the rats with PTSD,which may be related to the re-duction in the level of IL-1βand IL-6 in the hippocampus.
