1.Erythropoietin suppresses myocardial inflammatory cytokine expression in acute myocardial infarction rats

BACKGROUND:Studies have shown that inflammatory cytokines may influence the prognosis after myocardial infarction, and play an important role in the process of cardiac remodeling. The non-hematopoietic effects of
erythropoietin have been confirmed:erythropoietin can reduce the inflammatory reaction through bending with
the erythropoietin on the surface of target cel membrane, thus decreasing the reperfusion injury after myocardial ischemia.
OBJECTIVE:To observe the effect of recombinant human erythropoietin on the inflammatory factor expression during cardiac remodeling in rats with acute myocardial infarction.
METHODS: Sprague Dawley rat models of acute myocardial infarction were established through the ligation of the left anterior descending coronary artery. The rats were divided into five groups:sham operation group was
injected with normal saline;operation control group was injected with normal saline after modeling;SB203580 group was injected with highly selective p38 MAPK inhibitor SB203580 after modeling;erythropoietin group was injected with
erythropoietin after modeling;the erythropoietin+SB203580 group was injected with erythropoietin+SB203580 mixed solution after modeling. The tail vein blood samples were col ected before modeling, 1 day, 1 week, 2 weeks and 4 weeks after modeling, and then enzyme-linked immunosorbent assay was used to detect the levels of interleukin-1β, interlrukin-6 and tumor necrosis factor-α.
RESULTS AND CONCLUSION:There were no significant differences in the levels of interleukin-1β, interlrukin-6 and
tumor necrosis factor-αbetween groups before modeling (P>0.05). There were no significant differences in the levels of interleukin-1β, interlrukin-6 and tumor necrosis factor-αbetween different time points in the sham operation group (P>0.05), and the levels were highest at 1 day after modeling in the other four groups, and then decreased at 4 weeks after modeling (P<0.05). After modeling, the level of serum cytokines in the operation control group were higher than those in the other four groups, while level of serum cytokines in the sham operation group was lower than that in the other four groups (P<0.05);among the three groups intervened with drugs, level of serum cytokines was lower in the
erythropoietin+SB203580 group (P<0.05), while there was no significant difference in the level of serum cytokines between erythropoietin group and SB203580 group (P>0.05). Recombinant human erythropoietin can inhibit the expressions of inflammatory factors (interleukin-1β, interlrukin-6 and the tumor necrosis factor-α) during cardiac
remodeling after rat acute myocardial infarction, and the mechanism of recombinant human erythropoietin for inhibiting the expressions of inflammatory factors may related with the transforming growth factorβ1-TAK1-p38 MAPK signal pathway.
2.Clinical observation of Guanxin Tongmai plaster combined with medicine in the treatment of stable angina pectoris of coronary heart disease

Objective:To evaluate the clinical effect of Guanxin Tongmai plaster combined with conventional western medicine in the treatment of phlegm and blood stasis syndrome of coronary heart disease and angina pectoris.Methods:A total of 60 patients in the Department of Cardiology of the Affiliated Hospital of Liaoning University of Traditional Chinese Medicine (TCM) from February to August 2020 who met the inclusion criteria were randomly divided into two groups with 30 in each group. Both groups were treated with conventional western medicine. On this basis, Guanxin Tongmai plaster was applied at the acupoints in the treatment group and placebo plaster was applied in the control group. TCM syndrome score was performed before and after treatment, angina score was evaluated from three aspects of angina attack frequency, duration and pain degree, and blood lipid TG, TC, LDL-C and HDL-C were detected by enzyme quantitative method. The blood homocysteine (Hcy) was detected by enzyme circulation method, the ECG and the nitroglycerin reduction rate were recorded, and the safety index was detected according to the ECG changes.Results:In the treatment period, 2 patients in the treatment group fell off, 3 in the control group. A total of 28 patients in the treatment group and 27 in the control group were analyzed. The total effective rate of ECG efficacy in the treatment group was 67.9% (19/28) and the control group was 48.1% (13/27). There was significant difference between the two groups ( χ2=4.46, P=0.040). After treatment, the TCM syndrome score and angina score in the treatment group were significantly lower than those in the control group ( t values were 9.12 and 4.45, P values were 0.004 and 0.042, respectively). The reduction rate of nitroglycerin in the treatment group was 82.1% (23/28) and 55.6% (15/27) in the control group. There was significant difference between the two groups ( χ 2=4.72, P=0.030). After treatment, the plasma TG, TC, LDL-C in the treatment group were significantly lower than those in the control group ( t values were 4.17, 6.57 and 6.52, P<0.05 or P<0.01), the level of HDL-C was significantly higher than that of the control group ( t=7.07, P=0.010), and the level of plasma Hcy was significantly lower than that in the control group ( t=6.70, P=0.012). There was no significant difference in liver, kidney and coagulation function between the two groups. Conclusion:Guanxin Tongmai plaster combined with conventional western medicine can improve the clinical symptoms of patients with coronary heart disease and angina pectoris, reduce the level of blood lipid and Hcy, and improve the clinical curative effect.
3.Metagenomic analysis of bat virome in several Chinese regions.

Bats are important reservoir animals and more than 60 viruses have been identified in bats with many of them highly pathogenic to human. In order to understand the natural background, genetic diversity of bat viruses in China and discover potential viral pathogens, Solexa sequencing based viral metagenomics focusing on bats tissues was established and to analyze the virome of bats collected from Jilin, Yunnan and Hunan province. By Solexa sequencing, 116 442 324 useful reads were obtained and assembled into 4 872 contigs, of which 8.2% (4 002/4 4872) were annotated to 36 viral families, including 19 vertebrate virus families, 6 plant virus families, 4 insect virus families and 4 phages. Further contigs analyses showed that some adenovirus, bocavirus, picobirnavirus, parvovirus contigs sequences were similar with known viruses. However, part of them shared limited identities to these viruses implying the discovery of new viruses. Moreover, PCR validation of adenovirus and bocavirus confirmed the results obtained by viral metagenomics. This study aimed to understand bat virome in China by viral metagenomics and could be helpful to establish effective surveillance on wildlife-associate zoonoses.
