Associations between "lipid-related" candidate genes, blood lipid concentrations and coronary artery disease (CHD) risk are not clear. We aimed to investigate the effect of three newly identified lipids loci from genome-wide association studies on CHD and blood lipid levels in Chinese Han population. The genotypes of SNPs at three newly identified lipid loci and blood lipids concentrations were examined in 1360 CHD patients and 1360 age- and sex-frequency matched controls from an unrelated Chinese Han population. Allele T of rs16996148 occurred less frequently in CHD patients with the odds ratio (OR) being 0.64 (95% CI 0.50 to 0.81), after adjusting for conventional risk factors and was associated with a 33% decreased CHD risk (P<0.01) comparing with the major allele G. Individuals with GT genotype had the lowest CHD risk. No associations were found between the polymorphisms of other two loci with CHD risk and all three SNPs had no effect on lipid profile in this population. SNP rs16996148 on chromosome 19p13 is significantly associated with lower risk for CHD in Chinese Han population. However, it remains unresolved why these lipid-related loci had significantly less effects than the correspondingly expected effects on blood lipids levels in this population.

Aim To investigate the effect of magnesi-um isoglycyrrhizinate (MgIG)on radiation -induced pulmonary fibrosis in mice and the mechanism.Meth-ods Fifty female C57BL/6 mice were randomly divid-ed into control group, irradiation (RT)group,MgIG group,RT +MgIG group and RT +dexamethasone (DXM)group, with 1 0 mice in each group.Except for control group and MgIG group, the remaining mice were given a single 1 5Gy 60 Co γray on whole lung. The mice in each group were administered 2 h before irradiation and each day after irradiation:MgIG group and RT +MgIG group were administered with MgIG (1 00 mg·kg -1 )by intraperitoneal injection;control group and RT group were administered with normal sa-line (20 mL·kg -1 )by intraperitoneal injection;RT+DXMgroup was administered with DXM(0.5 mg· kg -1 )by intraperitoneal injection. After 1 2 weeks, the mice were sacrificed and lung tissues were taken out. The degree of alveolitis and pulmonary fibrosis were observed by HE staining and Masson staining. The ex-pressions of type Ⅰ collagen, type Ⅲ collagen and TGF-β1 protein were detected by immunohistochem-isty.Results The alveolitis, pulmonary fibrosis and expressions of type Ⅰ collagen, type Ⅲ collagen, TGF-β1 ,p-Smad2,p-Smad3 increased significantly in RT group compared with control group (P <0.05 ), and were significantly lower in RT +MgIG group and RT +DXMgroup than those in RT group(P <0.05). Conclusion MgIG can improve radiation-induced pulmonary fibrosis in mouse lung tissue, and its mech-anism may be related to the influence of MgIG on TGF-βsignaling pathway.

Background Retinal ichemic-reperfusion injury (RIRI) affects vision and life quality and has no ideal treatment outcome up to now.The mechanism of RIRI is associated with apoptosis.Researches showed that subretinal transplantation of bone mesenchymal stem cells (BMSCs) can relieve RIRI,but its mechanism is unclear. Objective This study was to observe the influence of BMSCs subretinal transplantation on the expressions of apoptosis protein Fas/FasL,caspases-3 after RIRI,and to explore the neuroprotecitve mechanism of BMSCs transplantation to treat RIRI.Methods Bone marrow was isolated and extract form femur and tibia of SD rat by the density gradient centrifugation method and then the BMSCs were collected.BMSCs suspension was prepared using DMEM containing low-glucose with the cell density 1 x 106-2× 106/ml.RIRI models were established by ligation of optical nerve (Daniel M method).Sixty-four SD rats were randomly divided into the normal control group, model control group,BMSCs transplantation group and PBS control group.Twenty-four hours after RIRI,BMSCs suspension with the cells density 5× 104 cells was subretinally injected in the rats of the BMSCs transplantation group, and equal volume of PBS was injected in the same way in the rats of the PBS control group. The expressions of Fas/FasL and caspase-3 were dynamically detected using immunohistochemistry at 6,24,48 and 72 hours after BMSCs transplantation. Results The positive response cells for Hoechst33324 were seen in subretina 72 hours after BMSCs transplantation.A few of positive cells for Fas,FasL,caspase-3 proteins were expressed in the rats of the normal control group at 6,24,48 and 72 hours after BMSCs transplantation. However, the positive cells for Fas,FasL,caspase-3 protein were significantly increased in the model control group and the BMSCs transplantation group compared with the normal control group at various time points after injection (all at P<0.01),and the number of positive cells was significantly less in the BMSCs group than those of the RIRI group and the BMSCs transplantation group (P<0.05,P<0.01).No significant differences were found in the numbers of positive cells of Fas,FasL and caspase-3 proteins at various time points between the model control group and the PBS control group (all at P>0.05).Conclusions The subretinal transplantation of BMSCs down-regulate the expression of Fas/FasL and caspases-3 proteins in RIR1 rats, and thus alleviate the apoptosis of retinal neural cells.

OBJECTIVE: To assess the maturation disparity of hand-wrist bones using the BoneXpert system and Greulich and Pyle (GP) atlas in a sample of normal children from China. MATERIALS AND METHODS: Our study included 229 boys and 168 girls aged 2-14 years. The bones in the hand and wrist were divided into five groups: distal radius and ulna, metacarpals, proximal phalanges, middle phalanges and distal phalanges. Bone age (BA) was assessed separately using the automatic BoneXpert and GP atlas by two raters. Differences in the BA between the most advanced and retarded individual bones and bone groups were analyzed. RESULTS: In 75.8% of children assessed with the BoneXpert and 59.4% of children assessed with the GP atlas, the BA difference between the most advanced and most retarded individual bones exceeded 2.0 years. The BA mean differences between the most advanced and most retarded individual bones were 2.58 and 2.25 years for the BoneXpert and GP atlas methods, respectively. Furthermore, for both methods, the middle phalanges were the most advanced group. The most retarded group was metacarpals for BoneXpert, while metacarpals and the distal radius and ulna were the most retarded groups according to the GP atlas. Overall, the BAs of the proximal and distal phalanges were closer to the chronological ages than those of the other bone groups. CONCLUSION: Obvious and regular maturation disparities are common in normal children. Overall, the BAs of the proximal and distal phalanges are more useful for BA estimation than those of the other bone groups.
