High mobility group box 1 protein,a kind of damage-associated molecular patterns,is a group of nonhistone DNA-binding protein in the nuclear.It not only involves in DNA replication,recombination,repair,regulation of gene transcription,but also induces activation of innate immunity and T cell generation and activates the adaptive immune response through Toll-like receptor.It is also more closely related to autoimmune diseases,inflammatory diseases,tumors and so on.Currently there are many researches on the role of TLR-mediated HMGB1 signaling pathway in diseases,which is involved in a variety of pathological processes and has a broad clinical outlook.

Hepatitis C virus (HCV)was easily developed into hepatitis,cirrhosis and liver cancer after infecting human.Cur-rently,the traditional method of treatment of HCV was pegylated interferon and ribavirin program,which was ineffective and had poor side effects,while the new direct antiviral drugs were expensive.Therefore,looking for cheap and non-toxic side effects of treatment was the focus of current research.More and more evidence indicate that micro-Ribose Nucleic Acid (miRNAs)play an important role in the development of liver disease,regeneration and functional regulation.This review studies the relationship between miR-122 and Hepatitis C virus and hepatocellular carcinoma.

5.A Study on Unilineage Differentiation of Umbilical Cord Blood CD_(34)~+ Cells Into Granulocytes, Erythrocytes or Platelets

Objectives To establish culture system for unilineage differentiation of umbilical cord blood CD 34 + cells into granulocytes, erythrocytes or platelets. Methods After CD 34 + cells were separated by midiMACS using micro beads conjugated with anti-CD 34 monoclonal antibody, these cells were induced to specifically differentiate along granulocyte, erythrocyte or platelet by adding appropriate hematopoietic growth factors including SCF plus G-CSF, EPO or TPO. Then morphology, immunological phenotype and function analysis were performed to identify these induced cells. Results After induced unilineage differentiation, the percentages of CD 15 +, GPA + and CD 41 + cells were 81 17%, 95 35% and 77 82%, respectively. These induced cells acquired terminal cell's morphologies. The cells from the granulocytic culture had the ability to phagocytose Chinese ink and plate-like particles could aggregate under the action of thrombin. Conclusion The culture system for unilineage differentiation of CD 34 + cells into erythrocytes, granulocytes, or platelets was established, which was a basis for the research about cell treatment, gene regulation and exogenous gene expression in hemopoiesis.
6.The Effect of Yinchenwulin Powder on the c-myc mRNA Expression of VSMC in Atherosclerosis Rats

Objective To observe the effect of Yinchenwulin(YCWL) powder on the c-myc rnRNA expression in vascular smooth muscle cell (VSMC) of atherosclerosis(AS) rats. Methods 70 Wistar rats were randomly divided into four groups: normal, model, Gypenosides treatment and YCWL powder treatment groups. AS models of rats were set up by feeding with high fat diet for one month. One month after treatment the level of serum lipid and blood rheology were measured, the phathological change of aorta was observed by HE staining, the ultrastructure change of aorta was observed by transmission electronic microscope, and c-myc mRNA expression in VSMC was detected by RT-PCR. Results Compared with Gypenosides treatment, YCWL powder could obviously decreased the levels of serum TC,TG and LDL, increase serum HDL level, reduce plasma viscosity,whole blood mid-cut,hematocrit and platelet adhesion rate, maintain aorta tissue structure, and down-regulate c-myc mRNA expression. Conclusion YCWL powder has therapeutic effect for atherosclerosis by regulating c-myc mRNA expression.
7.Expression of RIG-Ⅰgene in peripheral blood of patients with chronic hepatitis C and its ;correlations with hepatitis C virus load and typeⅠinterferon gene expression

Objective To detect the expression of retinoic acid-inducible gene Ⅰ ( RIG-Ⅰ) in peripheral blood mononuclear cells (PBMCs) of patients with chronic hepatitis C (CHC), and to explore its correlations with type Ⅰinterferon gene expression and serum level of hepatitis C virus .Methods A total of 101 na?ve patients with CHC were enrolled from Renmin Hospital of Wuhan University during July 2014 and July 2015, and 69 healthy subjects served as controls .The expressions of RIG-Ⅰ, IFNαand IFNβmRNA in PBMCs and serum level of HCV RNA were detected by real-time quantitative polymerase chain reaction.Pearson correlation analysis was performed to investigate the correlations of RIG-Ⅰ mRNA expression with IFNα/βmRNA expression and serum level of HCV RNA .Results The relative expressions of RIG-Ⅰ, IFNα, IFNβmRNA to healthy controls in CHC group were 0.082, 0.022 and 0.156, respectively (t=7.83, 3.65 and 2.13, all P<0.05).RIG-Ⅰ mRNA expression in patients with HCV loads <(1 ×106 ) IU/mL was 1.79 times higher than that in patients with HCV loads≥(1 ×106 ) IU/mL (t=2.60, P <0.05).Pearson correlation analysis showed that the expression of RIG-Ⅰ mRNA was positively correlated with that of IFNαmRNA and IFNβmRNA (r=0.247 and 0.489, P<0.05), but not correlated with HCV load (r=0.187, P>0.05).Conclusion Expression of RIG-ⅠmRNA decreases in patients with CHC , and is positively correlated with that of IFNαand IFNβmRNA.

9.Study on Expression Interferon Stimulating Gene and Related Signal Pathway Gene in Peripheral Blood Mononuclear Cells of Patients with Chronic Hepatitis C

Objective To detect mRNA expressions of STING and type Ⅰ interferons(IFN-α and IFN-β) in peripheral blood mononuclear ceils (PBMCs) of patients with chronic hepatitis C(CHC).Methods 113 patients with CHC and 94 healthy controls were collected from Renmin Hospital of Wuhan University during January 2015 and January 2016.Expressions of mRNA of STING,RIG-1,IFN-α and IFN-β were detected by real-time quantitative PCR,and their relative expression values were obtained by 2-△△Ct method and the results were statistically analyzed.Results The expression of mRNA of STING,RIG-1,IFN-α and IFN-β in CHC were 0.018,0.361,0.578 and 0.573 times than its in healthy controls respectively and the differences were statistically significant (t-8.28,4.26,2.18 and 2.07,all P < 0.05).Pearson correlation analysis showed that mRNA expressions of STING in healthy controls were positively correlated with that of IFN-α mRNA and IFN-β mRNA (r=0.487 and 0.207,all P<0.05),which had no correlation in CHC (r=0.174 and 0.091,all P>0.05).The expressions of STING mRNA was positively correlated with that of RIG-1 mRNA in healthycontrols (r=0.222,P<0.05),but there was no correlation of expression in CHC between STING mRNA and RIG-1 mRNA (r=-0.029,P>0.05).Conclusion Compared with healthy controls,the expression of STING,RIG-1,IFN-α and IFN-β mRNA were all reduced.There was positive correlation between STING and RIG-1,IFN-α and IFN-β in healthy controls,but no correlation in CHC.
10.Effects of static magnetic field on the activity of glutathione peroxidase and malondialdehyde content in mouse hepatic tissue

BACKGROUND: As the end product of lipid peroxidation, malondialdehyde (MDA) content can be used for assessment lipid peroxidation injury.Glutathione peroxidase (GSH-Px) acts as a free radical scavenger. Currently the effect of static magnetic field on the organism, whether positive or negative, has not been elucidated.OBJECTIVE: To study the effect of static magnetic field on anti-oxidation capacity of mouse hepatic tissues and its intensity dependence for producing such effects.DESIGN: A controlled comparative experiment.SETTING: Laboratories of Medical Physics and Biochemistry of Jiangsu University.MATERIALS: The experiment was conducted in the Laboratories of Medical Physics and Biochemistry of Jiangsu University from January to December 2003. Totally 30 mice of either sex weighing 18-20 g were selected and subjected to magnetic filed exposure using a self-designed ferrite magnet apparatus.METHODS: The mice were equally randomized into normal control group and 4 exposure groups exposed to magnetic field of (24.6±4.2) mT,(42.0±2.1) mT, (63.5±3.0) mT, and (85.1±2.9) mT, respectively. The mice in the 4 exposure groups were exposed to static magnetic field of the specified intensity for 2 hours twice a day, while those in the normal control group were subjected to the sham exposure apparatus without magnetic field at scheduled time points every day. After 15 days of exposure, the mice were sacrificed and the GSH-Px activity and the MDA content in the hepatic tissue were assayed.MAIN OUTCOME MEASURES: GSH-Px activity and MDA content in hepatic tissue of the mice.RESULTS: Thirty mice entered the final analysis without losses. MDA content in (24.6±4.2) mT and (42.0±2.1) mT groups were obviously lower than that in the normal control group [(12.70±0.53), (12.96±0.72), and (17.62±0.91) μmol/g, respectively, F=10.4, 9.89, P < 0.01]. The GSH-Px activity in the hepatic tissue in (24.6±4.2) mT and (42.0±2.1) mT groups were obviously higher than that in the normal control group [(143.36±8.34),(150.69±12.00), (87.51±11.34) μkat/g, respectively, F=10.0, 11.3, P < 0.01].CONCLUSION: Static magnetic field of appropriate intensity can lower MDA content and enhance the GSH-Px activity in the hepatic tissue of mice, and may also improve the activity of antioxidase and reduce the production of lipid peroxidation to diminish the consequent injuries and delay the aging process.
