Journal of International Oncology 2009;36(7):554-557
Objective Studing the relationship among matrixmetal-loproteinnse2 (MMP2), the tissue inhibitors of metalloprotein-ase2 (TIMP2) and chnicopathological characteristics such as histological grade, clinicopathological stage of the rectal cancer. Assessing the significance of MMP2 and TIMP2 expression in rec-tal cancer for prognosis and treatment. Methods To test the expression level of MMP2 and TIMP2 in rectal caneer patients with immunohistochemisty method. Results were analysised with χ2 test. The Kaplan-Meier (Log-rank test) was used for univariate survival analysis. Results The positive rates of MMP2 and T1MP2 ex-pression in rectal cance are higher in comparison with normal tissues. With MMP2 have a trendence of increas-ing the positive expression of TIMP2 decreases. Conclusion MMP2 and TIMP2 relate obviously with the clini-copathological character of the rectal cancer. They may play an important role in the proliferation, invasion and of the cancer. Testing the level of expression of MMP2 and TIMP2 may be helpful to prejudge the prognosis of the patients with rectal cancer. Some new therapic methods relating to MMP2 and TIMP2 may give patients with rectal cancer a more satisfying treatment.

Objective To construct the vector that expresses the fusion protein of HIV-Tat protein and red fluorescent protein(mCherry) in mammalian cells, and observe by fluorescence microscopy the intracellular transduction and localization of recombinant protein in cells,in order to obtain a useful tool for the study of the uptake mechanism and intracellular localization of HIV-TAT.Methods With the designed primer coding mCherry sequence, the mCherry gene was amplified by PCR with the vector pmCherry-C2 as template, and inserted into vector pET14b-His-TAT to construct the expression vector pET14b-His-TAT-mCherry.The constructed vector was then transformed into E.coli BL21(DE3),which had been identified by PCR and double digested with restriction endonuclease, followed by sequencing. After IPTG induction, the recombinant protein of His-TAT-mCherry was lyzed and analyzed with SDS-PAGE.Purified His-TAT-mCherry recombinant protein was added to Hela cells and the fluorescence was observed to evaluate the transduction efficiency. Results The results of identification by PCR,digestion with restriction endonuclease and sequencing indicated that the vector His-TAT-mCherry was correctly constructed.His-TAT-mCherry fusion protein was expressed in mammalian Hela cell line and purified successfully, and the fusion protein showed cellular transduction activity. It was found by fluorescence microscopy that the red fluorescence protein located mainly over the cytoplasm, and also the membrane to some extent. Conclusion The expression vector is successfully constructed for HIV-TAT labeled with mCherry sequence.Effective expression and purification of this fusion protein is achieved. It has been observed that the constructed vector may be expressed in mammalian Hela cell under active condition. Thus,it might be useful in the study of uptake mechanism and intracellular localization of HIV-TAT.

Objective:
To analyze the results of colorectal cancer screening among residents in Baoshan District, Shanghai Municipality from 2013 to 2021, so as to provide the basis for promoting colorectal cancer screening and prevention.
Methods:
Permanent residents aged 50 to 74 years in Baoshan District from 2013 to 2021 were selected as the screening population. The initial screening was conducted using a risk assessment form and fecal occult blood test. Positive results on either the risk assessment form or fecal occult blood test were considered positive for the initial screening. Participants with positive initial screening results were invited to undergo colonoscopy. The positive rate of the initial screening, colonoscopy compliance rate, and colonoscopy results were analyzed.
Results:
A total of 264 907 individuals underwent the initial colorectal cancer screening in Baoshan District from 2013 to 2021, with 65 333 individuals (24.66%) testing positive. Among them, the positive rate of the risk assessment form was 12.16%, and the positive rate of fecal occult blood test was 14.64%. A total of 14 473 individuals completed colonoscopy, with a compliance rate of 22.15%. A total of 1 284 precancerous lesions were detected, with a detection rate of 8.87%, and 386 cases of colorectal cancer were identified, with a detection rate of 2.67%. The positive rate of the initial screening, colonoscopy compliance rate, precancerous lesion detection rate, and colorectal cancer detection rate were higher in males than in females (25.55% vs. 24.06%, 23.12% vs. 21.45%, 11.60% vs. 6.74%, 3.62% vs. 1.93%, all P<0.05). With increasing age, the positive rate of the initial screening increased, the colonoscopy compliance rate decreased, the precancerous lesion detection rate and colorectal cancer detection rate increased (all P<0.05). From 2013 to 2021, the positive rate of the initial screening among residents showed a downward trend, while the colonoscopy compliance rate showed an upward trend (both P<0.05).
Conclusions
The detection rate of precancerous lesions in colorectal cancer was 8.87%, and the detection rate of colorectal cancer was 2.67% in Baoshan District from 2013 to 2021. Male and older individuals were the key populations for screening, and the colonoscopy compliance among residents needs to be improved.
