1.STUDY ON THE PHARMACOKINETIC OF RIFAPENTINE IN VOLUNTEERS

The pharmacokinetic study of Rifapentine by daily oral administration for 10d and followed by twice weekly for another 3 weeks in 9 volunteers and in comparism with Rifandin were reported. The results showed that the absorption and excretion of Rifapentine were markedly slower than that of Rifandin, its T1/2 was 15h. The Peak plasma level and T1/2 decreased after repeated administration of the drug. This suggested that Rifapentine and Rifandin in similar to Ri-fampin were hepatic microsomal enzyme inducers.
2.Effect of Total Flavonoids of Hippophae Rhamnoides on Contractile Mechanics and Calcium Transfer in Stretched Myocyte

Objective To study the role of total flavonoids of hippophae rhamnoides in improving contactile function of stretched cardiac myocyte.Method Flavonoids were given to stretched myocytes which were proved their contractile function decline and then myocyte contractile mechanics characteristics and calcium transfer were measured.Result Flavonoids increased myocyte contractility,as indicated by myocyte shortening,velocity of shortening,peak+dL/dt and peak-dL/dt during shortening,in a concentration-dependent manner (r>0.9,P<0.001),and with no relation to the intracellular calcium transfer in the myocytes.Conclusion Flavonoids of the traditional Chinese drug hippophae rhamnoids is effective in improving the contractile function of stretched cardiac myocyte in low dosage.
3.Inhibitory effect of water-soluble preparation of curcumin on colon carcinoma C26 cell line-induced angiogenesis in mice

Objective:To investigate the inhibitory effect of the water-soluble preparation of curcumin on tumor angiogenesis induced by alginate-encapsulated colon carcinoma C26 cell line in mice. Methods: The water-soluble preparation of curcumin was made by forming inclusion compound of curcumin with hydroxypropyl-?-cyclodextrin (C-HP-?-CD). Subcutaneous imbedding of alginate-encapsulated colon carcinoma C26 cells was used to create tumor bearing model in BALB/c mice. Then the mouse models were intraperitoneally injected with C-HP-?-CD, TNF-70 (as positive control), or HP-?-CD(as negative control). The angiogenesis in the alginate beads was observed under light microscope, floresencent microscope, and transmission electron microscope. The fluorescein isothiacyanate-dextran (FITC-Dextran) tracing technique was used to observe the inhibitory effect of the inclusion compound of curcumin on angiogenesis induced by colon carcinoma C26 cells in alginate beads.Results: The C-HP-?-CD and the mouse model of subcutaneous alginate-encapsulated colon carcinoma C26 cells were successfully created. Transmission electron microscope showed angiogenesis in alginate-encapsulated colon carcinoma C26 cells; claval endothelial cells were seen in some lumens. Optical microscope and the fluorescence microscope showed that the angiogenesis density of the positive group and curcumin groups were obviously lower than those of the negative control group; no obvious difference at vessel morphosis was found among the 3 alginate-encapsulated groups. 4.The effect of exposure to low intensity infrasound on the expression of PSA-NCAM in the rat hippocampus

Objective To observe the expression of the polysialic acid neural cell adhesion molecule ( PSA-NCAM) in the hippocampus of rats after exposure to low intensity ( 16 Hz,90 dB) infrasound for different periods.Methods One hundred and twenty male Sprague-Dawley rats were randomly divided into infrasound exposure and normal groups.The exposure group was tjem further divided into 1 day,7 days,14 days and 21 days exposure groups.After exposure,the rats' brains were removed and an immunohistochemical technique was used to observe the expression of PSA-NCAM in the hippocampus after 1,7,14 or 21 days of exposure. Methods The expression of PSA-NCAM had increased significantly after exposure for 7 days. It peaked at 14 days,then had decreased by 21 days,but always remaining higher than in the normal group.After the infrasound exposure had ended,the expression of PSA-NCAM demonstrated a tendency of decrease over time,and the least was on the 21st day.The largest decrease was observed in the 14 days exposure groups. Conclusion 16 Hz,90 dB infrasound can increase the expression of PSA-NCAM in the hippocampus,at least in rats.This suggests that low intensity infrasound might initiate recovery of an injured central nervous system.Migration of neural stem cells may aid in the repair of neural injuries resulted from infrasound exposure.
5.The influence on hemodynamics of myocardial ischemic dogs and blood pressure of animals with shenfu injection.

OBJECTIVETo investigate the influence of Shenfu injection on the hemodynamic indexes of myocardial ischemic dogs and blood pressure of dogs and rats.
METHODMyocardial ischemic model was made and hypotension in the dogs was induced with ligating left front descending limb coronary artery method, and secondary hypertension by narrowing nephridium artery of rats, Shenfu injection was administered with 5, 10 mL.kg-1 to the above dogs and rats separately to investigate the influence of it on the hemodynamic indexes of myocardial ischemic dogs and blood pressure of rats.
RESULT AND CONCLUSIONShenfu injection enhanced the capacity of myocardial work markedly; it augmented the myocardium contractility and cardiac output without raising the oxygen consume, and at the same time it returned normal the blood pressure in myocardial ischemic; but it had no effects on the normal blood pressure and secondary hypertension of rats.

6.A novel biodegradable polymer-coated sirolimus-eluting stent: 1-year results of the HELIOS registry.

BACKGROUND:
The HELIOS stent is a sirolimus-eluting stent with a biodegradable polymer and titanium oxide film as the tie-layer. The study aimed to evaluate the safety and efficacy of HELIOS stent in a real-world setting.
METHODS:
The HELIOS registry is a prospective, multicenter, cohort study conducted at 38 centers across China between November 2018 and December 2019. A total of 3060 consecutive patients were enrolled after application of minimal inclusion and exclusion criteria. The primary endpoint was target lesion failure (TLF), defined as a composite of cardiac death, non-fatal target vessel myocardial infarction (MI), and clinically indicated target lesion revascularization (TLR) at 1-year follow-up. Kaplan-Meier methods were used to estimate the cumulative incidence of clinical events and construct survival curves.
RESULTS:
A total of 2998 (98.0%) patients completed the 1-year follow-up. The 1-year incidence of TLF was 3.10% (94/2998, 95% closed interval: 2.54-3.78%). The rates of cardiac death, non-fatal target vessel MI and clinically indicated TLR were 2.33% (70/2998), 0.20% (6/2998), and 0.70% (21/2998), respectively. The rate of stent thrombosis was 0.33% (10/2998). Age ≥60 years, diabetes mellitus, family history of coronary artery disease, acute myocardial infarction at admission, and device success were independent predictors of TLF at 1 year.
CONCLUSION:
The 1-year incidence rates of TLF and stent thrombosis were 3.10% and 0.33%, respectively, in patients treated with HELIOS stents. Our results provide clinical evidence for interventional cardiologists and policymakers to evaluate HELIOS stent.
