Results Under the management, all indexes reached the standard. Conclusion Only if the importance be attached to the management and maintenance of clean operation room, and the rules be strictly observed, does the clean operation room be well maintained, the hospital infection caused by operating room be effectively controlled and the quality of medical treatment be improved.
2.Bone marrow mesenchymal stem cells are induced to differentiate into cardiomyocytes by hepatocyte growth factor in vitro

AIM: To explore a new method of hepatocyte growth factor (HGF) inducing bone marrow mesenchymal stem cells (MSC) to differentiate into cardiomyocytes. METHODS: Bone marrow MSC was cultured with DMEM media (10% fetal calf serum) 4-6 passages, and induced by HGF (10 ?g/L) for 30 d. Automatical beating of the differentiated cells was observed daily with transverse microscopy, or under condition of 0.1% isoproterenol or cal (cium-deprived) incubation. Specific cardiac myosin in the cells was indentified by immunochemistry. RESULTS: At 14-20 d of differentiation, bone marrow mesenchymal stem cells formed clones, in 10%-50% of which spontaneous beating cell-mass had come to continuously exist. Isoproterenol increased the beating rate and calcium-deprived media inhibited the beating. The cells were identified to be cardiomyocytes by expression of cardiac myosin heavy chain. CONCLUSION: HGF may induce bone marrow mesenchymal stem cells into cardiomyocytes with high efficiency, but the differentiating pathway of stem cells remains to be further studied. [

Objective To study the effect of IL-6 and IL-8 on the MMP-9 secretion of endometrial stromal cells in women with endometriosis to explore the function of IL-6 and IL-8 in the adherence and planting process of ectopic endometrium. Methods The endometrial stromal cells were cultured in DMEM/F12 medium containing different concentrations of IL-6 or IL-8 for 24 hours, then the media were collected to measure the levels of MMP-9 by ELISA. 4.A discussion on the key problems and system construction of health poverty alleviation in China

Health poverty alleviation is an important part of taking targeted measures to help people lift themselves out of poverty, and it is the key link to achieve a Healthy Chinese Strategy.At present, the health problem of the poor in China is becoming increasingly prominent, and as a result, the low health status of the poor has exacerbated the level of poverty, mainly because of catastrophic disease.With this discussion paper, it was revealed that there are many problems in the system of poverty alleviation in China: The protection levels of all kinds of medical insurance are still low;the medical assistance system is still not perfect;the medical service facility of the county medical institutions is poor;the county outside referral rate is high;public health problems in poor areas are more prominent.As suggestions derived from the present discussion, China''s health and poverty alleviation system should adhere to the government led and multi-sectoral coordination of multiple health poverty alleviation.It should also strengthen health care and public health system construction in poverty-stricken areas, and establish a multi-path health security system.
5.Clinical observation of gemcitabine in the treatment of elderly patients with advanced non-small cell lung cancer

Objective To investigate the clinical efficacy of gemcitabine in the treatment of elderly patients with advanced non-small cell lung cancer.Methods 50 patients in our hospital from January 2014 to September 2015 were selected as the research object, and were randomly divided into the control group and the experimental group, each group of 25 patients each.The patients in the control group were treated with special therapy, and the patients in the experimental group were treated with gemcitabine.The clinical indexes of the experimental group and the control group were compared and analyzed.Results After the treatment, the experimental group of 25 patients, 12 cases of patients with complete remission, 8 cases partial remission, 3 cases were stable, 2 patients progressed, the effective rate was 75%, clinical benefit rate of 92%.In the control group, 4 patients had complete remission, 2 patients had partial remission, 9 patients were stable, 10 patients progressed, the treatment efficiency was 24%, and the clinical benefit rate was 60%.The effective rate and the clinical benefit rate of the experimental group were significantly higher than that of the control group (P<0.05).1 years later, the survival rate of patients in the experimental group was 22, the survival rate was 88%, and the number of patients in the control group was 14.The survival rate of the experimental group was significantly higher than that of the control group, with statistical difference (P<0.05).Conclusion Gemcitabine monotherapy in the treatment of elderly patients with advanced non-small cell lung cancer can improve the therapeutic effect to a great extent, good safety, low incidence of adverse reactions, high survival rate, with further clinical promotion and application significance.
6.Protective effect of gynostemma on myocardium in diabetes associated with hyperlipemia and its mechanism

Objective To investigate the protective effect of gynostemma on myocardium and its mechanism in patients with diabetes associated with hyperlipemia.Methods 178 patients with type 2 diabetes mellitus with hyperlipidemia from the department of endocrinology in our hospital were selected and divided into two groups, 89 cases in the control group treated with basic treatment of diabetes+atorvastatin, 89 cases in the experiment group received gynostemma on the basis of the control group, myocardial injury index, blood glucose level, blood lipid level, serum cytokine levels and serum oxidative stress response indexes were detected and compared after the treatment.Results Compared with the control group, serum levels of aspartate aminotransferase(AST), creatine kinase(CK), CK-MB, lactate dehydrogenase(LDH)were lower in the experiment group after treatment, fasting plasma glucose(FPG), 2h postprandial blood glucose(2hPG), HbA1c level were lower after treatment, serum levels of total cholesterol(TC), triglycerides( TG ) , low-density lipoprotein cholesterol ( LDL-C ) were lower, high-density lipoprotein cholesterol ( HDL-C ) level was higher after treatment, serum level of adiponectin(ADPN) was higher, tumor necrosis factor-alpha(TNF-α), interleukin-6(IL-6), interleukin-18(IL-18) levels were lower after treatment, serum levels of malondialdehyde(MDA), reactive oxygen species(ROS) were lower, glutathione peroxidase(GSH-Px), superoxide dismutase(SOD) levels were higher after treatment, with significant difference (P<0.05).Conclusion Gynostemma can significantly reduce the cardiac muscle damage of patients with diabetes associated with hyperlipemia , plays a role in myocardial protection, the mechanism may be to reduce blood glucose and blood lipid levels, inhibit the inflammatory reaction caused by inflammatory cytokines, improve the antioxidant capacity of the body and reduce oxidative stress damage.

8.Resistance reverse effect of anemoside B4 and tetrandrine on oxaliplatin-resistant human colon cancer cells and the mechanism

Background and purpose:Oxaliplatin (L-OHP) is one of the most commonly used chemotherapy drugs in colorectal cancer and L-OHP-resistance is very common in colorectal cancer therapy. This research was to discuss the reversal effect in L-OHP-resistant human colon cancer cell line LoVo/L-OHP by anemoside B4 (AB4) and tetrandrine (Tet) and to clarify their molecular mechanism. Methods:LoVo/L-OHP cells were treated for 48 h by AB4 and Tet at different concentrations to get non-toxic dose. Drug sensitivity was measured by MTT. After the treatment, the cell cycle and apoptosis of the cells were detected. Expression of P-gp mRNA, zDHHC9 mRNA and SMAD4 mRNA were detected by RT-PCR. Expression of P-gp, zDHHC9 and SMAD4 protein were detected by Western blot. Results:The IC50 of LoVo/L-OHP for L-OHP was (112.5±23.6) μg/mL, and the IC50 decreased to (62.8±21.4) μg/mL (P<0.05) and (58.9±26.3) μg/mL (P<0.05) after the treatment with 0.71 μg/mL AB4 and 0.45 μg/mL Tet (non-toxic dose) separately. The cell cycle experiment showed that the cells in G1 stage decreased and in S stage increased but with no statistical signiifcance (P>0.05). The cell apoptosis experiment showed that the apoptotic rate increased after the treatment with AB4 and Tet with non-toxic dose but with no statistical signiifcance (P>0.05). The RT-PCR experiment showed that the expressions of P-gp mRNA and zDHHC9 mRNA were increased and SMAD4 mRNA was decreased in LoVo/L-OHP cells compared with in LoVo cells (P<0.05), while it was found that P-gp mRNA in LoVo/L-OHP cells was decreased after the treatment with AB4 at non-toxic dose (P<0.05). Western blot experiment showed the protein of P-gp in LoVo/L-OHP cells was decreased after treatment with AB4 (P<0.05), which was accordant to the PCR result. The expression of zDHHC9 protein in LoVo/L-OHP cells was decreased in LoVo/L-OHP cells after treatment with Tet (P<0.05). Conclusion:AB4 and Tet have some reversal effect on resistant to L-OHP in LoVo/L-OHP cells. The molec-ular mechanism of the resistance reverse effect was related to down-regulation of P-gp for AB4 and down-regulation of zDHHC9 for Tet.
9.The clinical manifestations and genetic analysis of lisencephaly in children

Objective To explore clinical characteristics and pathogenic gene of lisencephaly. Methods The clinical manifestation, laboratory examination and gene detection results of lisencephaly in two children were analyzed retrospectively, and relevant literature were reviewed. Results Two male children with lisencephaly are at ages of 7 months, and 3 years and 4 months respectively. Both of them were admitted to hospital due to epilepsy and loss of consciousness at the time of attack. There was no obvious abnormality in laboratory examination. Both of their EEG indicated epileptic wave. Cranial MRI showed lissencelphaly. Gene analysis showed that there was a heterozygous mutation of IVS3-1G>A in PAFAH1B1 gene in a child, which resulted in the deletion of exon 4 in mRNA level by functional analysis. No mutations were found in the parents of the child. The other one had c.274A>G mutation (p.K92E) in PAFAH1B1 gene, which has not been reported before, and his parents were normal. Conclusion Patients with lissencelphaly may combine with epilepsy, and the PAFAH1B1 gene mutation is the common cause.
