Breast cancer is one of the major causes of death in women,and its incidence has been increasing year after year.The Rho GTPases,their regulatory proteins and Rho GTPases play an important role in promoting the occurrence and distant metastasis of breast cancer.Here we summarized the current knowledge of the regulation network of Rho GTPases,their regulatory proteins and Rho GTPases on the occurrence and development of breast cancer,and targeted therapy for RHO GTP enzyme pathway in breast cancer.
2.Expression ofβ1 integrin, Rac1, and RhoA in invasive micropapillary breast carcinoma

Objective:To investigate the expression and significance ofβ1 integrin, Rac1, and RhoA in invasive micropapillary breast carcinoma (IMPC). Methods:Immunohistochemical staining was performed to detect the expression ofβ1 integrin, Rac1, and RhoA in 89 patients with IMPC and 90 patients with invasive ductal carcinoma-not otherwise specified (IDC-NOS) who were treated between January 2007 and December 2008 in Tianjin Medical University Cancer Institute and Hospital. The relationship among the three proteins and the expression ofβ1 integrin, Rac1, and RhoA with clinicopathological features were determined. Results:β1 integrin (78.7%) and Rac1 (76.4%) were highly expressed in patients with IMPC. This expression was significantly higher than that in patients with IDC-NOS (63.3%and 54.4%). Statistical difference was found between the two groups (P<0.05). Positive rate of RhoA (68.5%) in patients with IMPC was also statistically higher than that in patients with IDC-NOS (44.4%, P=0.001). In patients with IMPC, the expression ofβ1 integrin and Rac1 was positively associated with lymph node metastasis (P<0.05), while the expression ofβ1 integrin was positively correlated with Rac1 expression (P=0.006). Expression of RhoA was positively correlated with lymph node involvement and ER and PR status (P<0.05). However, no correlation was found between RhoA expression and the expression ofβ1 integrin and Rac1 (P>0.05). Conclusion:Thus,β1 integrin, Rac1, and RhoA were overexpressed and might play an important role in the high frequency of metastasis in patients with IMPC. These proteins could be considered as biomarkers for the prognosis and new targets for IMPC therapy.
3.Effect of Ulinastatin on the Expression of TNF-α in the Rabbits with Acute Lung Injury Induced by Underwater Explosion

Objective The expression of TNF-α was detected in sera of rabbits treated by ulinastatin with acute lung injury in duced by underwater explosion.Methods Rabbits were randomly divided into two groups such as the injured group and ulinastatin therapy group.Established underwater explosion device was used to cause acute lung injury in rabbits.TNF-α in sera of the rabbits were measured by ELISA at 4,12 and 24 hours after the explosion.The SPSS17.0 software was used to analyze the data and P<0.05 was considered to be significant.Results There was no significant difference between the concentrations of TNF-α in the sera of rabbits in the injure group (538.20±201.43 ng/L) and that of in the ulinastatin group (386.90± 109.22 ng/L,t=2.088,P=0.051) at 4 hours after burst.However,there was evidently decreased in the level of TNF-α in the ulinastatin group (400.60 ± 78.98 ng/L) compared with the injury group (573.80 ± 178.24 ng/L,t =2.809,P =0.012) at 12 hours after burst.Moreover and TNF-α in the ulinastatin group (356.10 ± 130.99 ng/L) was also decreased compared to the injure group (552.30± 169.64 ng/L,t=2.895,P=0.010) at 24 hours after burst.Conclusion The TNF-α expression in sera of the rabbits in ulinastatin group were dramatically decreased than thai of in injury group at 12 hours after burst,which may be benefit to rabbits with acute lung injury induced by underwater explosion.
4.Effects of long-term external use of goupi gao on renal functions and lead accumulation in rats.

OBJECTIVETo observe the effect of long-term external use of Goupi Gao on renal function and lead accumulation in rats.
METHODRats were externally administered with Goupi Gao at different doses (7, 3.5 and 1.75 g x kg(-1)) for 90 d. At 45 days and 90 days after administration, the renal indicator, levels of blood urea nitrogen (BU) and creatinine (Cr) in serum, beta2-microglobulin (beta2-MG) and N-acetyl-beta-D-glucosaminidase (NAG) in urine were determined. Lead content in kidneys was detected by atomic absorption spectrometry.
RESULTA 90-day administration with Goupi Gao significantly enhanced the renal indicator, levels of NAG in urine and lead content in renal, when compared with the normal rats. However, the levels of BUN and beta2-MG as well as renal pathology in Goupi Gao treated rats were not obviously changed.
CONCLUSIONConsecutive administration of Goupi Gao for 90 days can increase the renal indicator and levels of NAG in urine, enhance the accumulation of lead in renal, but with no effect on excretory function of kidneys and organic changes.

5.Study on blood lead of rats in long-term toxicity test with goupi gao.

OBJECTIVETo observe dynamic changes of blood lead concentration in rats with long-term toxicity test with Goupi Gao by the flame atomic absorption spectrometry, in order to provide reference for safe administration of Goupi Gao.
METHODThe rats were administered with Goupi Gao by high-dose (7 g x kg(-1)), medium-dose (3.5 g x kg(-1)), low-dose (1.75 g x kg(-1)) by external use for consecutively 90 days. Then, the blood samples were collected from the rats before the administration and at 10, 30, 45, 52, 60, 90 d after the administration respectively, as well as 16 d and 28 d after the drug withdrawal. The samples were dispelled with microwave digestion system and then were determined by graphite furnace atomic absorption spectrometry for blood lead levels.
RESULTAccording to methodological study, the standard curve regression equation in this experiment was A = 0.004 9X + 0. 017, r = 0.999 5, with the detection limit up to 0. 380 microg x L(-1). The RSD was 1.4% by precision checks. Blood lead level of mixed blood samples was 175.77 microg x L(-1), whose RSD was 6. 0%. Blood lead concentration gradually increased after low-dose and medium-dose administration to rats and became stable at the 10th day and the 30th day by high-dose. Dose is directly related to blood lead concentration. Meanwhile, the blood lead concentration decreases after the drug withdrawal.
CONCLUSIONThe method of determination in this experiment is so accurate and reliable that it can be used for the determination of blood lead. Long-term and high-dose external use of Goupi Gao can increase blood lead.

6.Nursing care of a child with Beckwith-Wiedemann Syndrome frequent hypoglycemia

Objective:To summarize the experience of blood glucose management and nursing for a newborn with repeated hypoglycemia in Beckwith-Wiedemann syndrome.Method:A multidisciplinary team was formed, and formulated an individualized care plan for a large infant with Beckwith-Wiedemann syndrome. A dynamic blood glucose monitoring system was used to closely monitor blood glucose fluctuations for this children, dynamically adjusted the amount of infusion and milk according to the blood sugar situation, detected and dealed with hypoglycemia in time, strengthened the skin care of child and implemented psychological care for the family.Results:After refined treatment and care, the child′s vital signs were stable, and his blood sugar could maintain within a normal range before being discharged from the hospital.Conclusions:The use of dynamic blood glucose monitoring system under the guidance of a multidisciplinary team can effectively monitor and control the blood glucose fluctuations of children with BWS syndrome, which can provide a basis for further treatment of children.
7.Research progress in evaluating severity and prognosis of children with community acquired pneumonia by scales and biomarkers

Community acquired pneumonia(CAP)has a high morbidity and mortality rate, and can bring a heavy social and economic burden.Its etiology is complex.How to identify high-risk children, early diagnosis, prognosis prediction are the focus of clinical research.Early identification and active intervention of high-risk children who need hospitalization or admission to pediatric intensive care unit by using score scales and biomarkers are crucial to improve the survival rate.This review summarized the assessment of severity and prognosis of CAP in children by different score scales and biomarkers.

MarR family transcription regulators are ubiquitous among bacteria and archaea. They extensively control multiple cellular processes and elaborately regulate the expression of genes involved in virulence, stress response and antibiotics at translational level. In Xanthomonas campestris pv. campestris, insertional inactivation of MarR family transcription regulator HpaR (XC2827) resulted in significantly decrease in virulence and increase in the production of the extracellular proteases. Here, we reported that the genome of Xcc 8004 encodes nine MarR family transcription regulators. The MarR family transcription regulators, HpaR (XC2827) and XC0449, were heterologous expressed and purified. In vitro MST and Pull-down assay confirmed the physical interaction between HpaR and XC0449. Phenotypical assay determined that deletion of XC0449 resulted in substantial virulence attenuation. In vitro EMSA, in vivo qRT-PCR and GUS activity assay identified that HpaR and XC0449 coordinately act as the transcriptional activator to regulate the expression of the virulence-associated gene XC0705, and eventually control the bacterial virulence and the production of extracellular proteases.

9.Serum inflammatory cytokines and Tie⁃2 correlation with clinical symptoms in patients with first⁃episode schizophrenia

Objective :
To explore the differences of serum inflammatory cytokines and tyrosine kinase with immunoglobulin and epidermal growth factor homology domains⁃2(Tie⁃2) levels between patients with first⁃episode schizophrenia and healthy people , and the correlation with clinical symptoms .
Methods :
A total of 168 participants were recruited , including 86 patients with first⁃episode schizophrenia (patient group) and 82 healthy people ( control group) . Demographic data , Positive and Negative Symptom Scale (PANSS) and Brief Psychiatric Rating Scale (PANSS) were collected at baseline . Serum inflammatory cytokines and Tie⁃2 levels were determined by MesoScale Discovery (MSD) .
Results :
Compared with the control group , the levels of serum interleukin (IL) Ⅳ1β and IL⁃4 in the patient group increased (P < 0. 05) , while the level of Tie⁃2 decreased (P < 0. 05) . IL⁃1β level in the patient group was positively correlated with the total score of BPRS , the score of BPRS hostility factor and the score of PANSS positive scale factor (P < 0. 05) . The positive score of PANSS and the total score of BPRS in the patient group had a positive effect on IL⁃1β level (P < 0. 05) . PANSS negative scale factor score , general psychopathological scale factor score and total score of PANSS in patient group also had positive effects on Tie⁃2 level (P < 0. 05) . IL⁃1β level in the patient group could effectively predict the severity of clinical symptoms in patients with first⁃episode schizophrenia at the critical level of 2 127. 076 pg/L , with specificity of 0. 675 and sensitivity of 0. 639 . The AUC of IL⁃1β and Tie⁃2 were 0. 836 1 and 0. 646 2 , respectively .
Conclusion
The levels of IL⁃1β , IL⁃4 and Tie⁃2 in patients with first⁃episode schizophrenia are different from those in healthy people . IL⁃1β levels in patients with first⁃episode schizophrenia are correlated with part of clinical symptoms . IL⁃1β and Tie⁃2 levels in the patient group may be influencing factors with high sensitivity and specificity in predicting the severity of clinical symptoms in patients with first⁃episode schizophrenia .
